Congenital muscular dystrophy-associated inflammatory chemokines provide axes for effective recruitment of therapeutic adult stem cell into muscles.

BACKGROUND: Congenital muscular dystrophies (CMD) are a clinically and genetically heterogeneous group of neuromuscular disorders characterized by muscle weakness. The two most prevalent forms of CMD, collagen VI-related myopathies (COL6RM) and laminin α2 deficient CMD type 1A (MDC1A), are both caused by deficiency or dysfunction of extracellular matrix proteins. Previously, we showed that an intramuscular transplantation of human adipose-derived stem cells (ADSC) into the muscle of the Col6a1-/- mice results in efficient stem cell engraftment, migration, long-term survival, and continuous pro

Costunolide mitigates inflammation and promotes extracellualr matrix integrity of thoracic aortic dissection by inhibiting NF-κB signaling.

BACKGROUND: Thoracic aortic dissection (TAD) is one of the most fatal cardiovascular diseases. One of its important pathological characteristics is the local inflammatory response. Many studies have found that Macrophage polarization plays an extremely critical role in the inflammatory progression and tissue remodeling of TAD. Costunolide (CTD) has an improving effect on oxidative stress and inflammation in the body. However, whether it can promote the integrity of extracellular matrix in Aortic dissection and its mechanism are still unclear. METHODS: The male C57BL/6J mice were used to const

Permissibility of Mongolian gerbil for Angiostrongylus cantonensis infection and utility of this animal model for anthelmintic studies.

The Mongolian gerbil (Meriones unguiculatus) has been indicated to be a useful experimental model host for studying nematode. To understand the possibility of the Mongolian gerbil as an animal model of Angiostrongylus cantonensis infection, we investigated the development, migration, and tissue distribution of A. cantonensis and pathological changes in the brain and lungs of the infected Mongolian gerbils. The first stage larvae of A. cantonensis in the stool of the infected gerbils were examined by direct smear method at 45th day postinfection (PI). In addition, a group of the infected gerbil

Granulocyte colony-stimulating factor improves cerebrovascular reserve capacity by enhancing collateral growth in the circle of Willis.

BACKGROUND AND PURPOSE: Restoration of cerebrovascular reserve capacity (CVRC) depends on the recruitment and positive outward remodeling of preexistent collaterals (arteriogenesis). With this study, we provide functional evidence that granulocyte colony-stimulating factor (G-CSF) augments therapeutic arteriogenesis in two animal models of cerebral hypoperfusion. We identified an effective dosing regimen that improved CVRC and stimulated collateral growth, thereby improving the outcome after experimentally induced stroke. METHODS: We used two established animal models of (a) cerebral hypoperf

Anti-inflammatory effects of theophylline, cromolyn and salbutamol in a murine model of pleurisy.

1. The aim of this study was to examine the effect of theophylline, cromolyn and salbutamol, three well-known anti-asthmatic drugs, on the early (4 h) and late (48 h) phases of cell migration and fluid leakage induced by carrageenin in the pleural cavity of mice. 2. In the first set of experiments, animals were pretreated (30 min) with different doses of theophylline (0.5-50 mg kg-1, i.p.), cromolyn (0.02-0.2 mg per pleural cavity) or salbutamol (0.05-50 mg kg-1, i.p.); the total and differential cell content, and also the exudate were analysed 4 h after carrageenin (1%) administration. A Comparison of in vivo Tumor-Homing Abilities of Placental-Derived and Bone Marrow-Derived Mesenchymal Stromal Cells in High-Risk Neuroblastoma.

BACKGROUND: Neuroblastoma is a highly lethal malignancy of young children. Mesenchymal stromal cells (MSCs) may represent a novel cellular delivery vehicle due to their innate tumor-homing properties. We compared in vivo homing abilities of placental-derived MSCs (PMSCs) and bone marrow-derived MSCs (BM-MSCs) in an orthotopic neuroblastoma xenograft. METHODS: 28 mice underwent direct implantation of neuroblastoma cells (cell line NB1643) into the adrenal gland followed by intraperitoneal injection of 5 × 106 MSCs (PMSC n = 13, BM-MSC n = 13, PBS controls n = 2). MSC migration was monitored wi

Lack of suppression of lymphocyte MIF production by estradiol, progesterone and human chorionic gonadotropin.

The hypothesis that hormones produced by the placenta contribute to immunosuppression during pregnancy is an attractive one. In order partially to evaluate this hypothesis, we utilized the Migration Inhibition Factor (MIF) Assay, which is an in vitro model of delayed hypersensitivity in that presensitized lymphocytes stimulated by antigen release a glycoprotein (MIF) which inhibits the normal migration of macrophages. The effect of placental hormones on this model of cellular immunity was tested using the direct and indirect MIF assays in pre-immunized guinea pigs. Estradiol (1-50 microgram/ml

Overcoming the curse of knowledge: guidelines for reporting system-specific details of host infection and migration dynamics.

Literature syntheses link knowledge across study systems and regions. However, specific details on the biological systems of the included publications may not always be explicitly outlined because of communication gaps (a.k.a. The Curse of Knowledge) and/or knowledge gaps. These gaps can lead to inappropriate comparisons across studies if researchers unfamiliar with the systems make inaccurate assumptions about aspects of study design or natural history. In the context of movement ecology, a growing number of publications explore how host infection and host migration intersect, work that is cr

Strongyloides stercoralis: eosinophil-dependent immune-mediated killing of third stage larvae in BALB/cByJ mice.

Challenge worm survival was significantly reduced when BALB/cByJ mice were vaccinated against Strongyloides stercoralis infective third stage larvae (L3) regardless of whether the challenge infections consisted of systemically migrating L3 or L3 implanted in diffusion chambers. The only cell type that increased in number in diffusion chambers in immunized mice, 1 week after booster immunizations, was the eosinophil, and maximal levels of eosinophils were coincident with parasite killing. Mice were treated with mAb to eliminate IL-5 or granulocytes to assess the role that eosinophils play in la

Decellularized extracellular matrix microparticles as a vehicle for cellular delivery in a model of anastomosis healing.

Extracellular matrix (ECM) materials from animal and human sources have become important materials for soft tissue repair. Microparticles of ECM materials have increased surface area and exposed binding sites compared to sheet materials. Decellularized porcine peritoneum was mechanically dissociated into 200 µm microparticles, seeded with fibroblasts and cultured in a low gravity rotating bioreactor. The cells avidly attached and maintained excellent viability on the microparticles. When the seeded microparticles were placed in a collagen gel, the cells quickly migrated off the microparticles

ApoER2/VLDL receptor and Dab1 in the rostral migratory stream function in postnatal neuronal migration independently of Reelin.

Postnatal migration of interneuron precursors from the subventricular zone to the olfactory bulb occurs in chains that form the substrate for the rostral migratory stream. Reelin is suggested to induce detachment of neuroblasts from the chains when they arrive at the olfactory bulb. Here we show that ApoER2 and possibly very-low-density lipoprotein receptor (VLDLR) and their intracellular adapter protein Dab1 are involved in chain formation most likely independent of Reelin. F-spondin, which is present in the stream, may act as ligand for ApoER2 and VLDLR. In mice lacking either both receptors

Identification of an essential nonneuronal function of neurotrophin 3 in mammalian cardiac development.

Neurotrophin 3 (Nt3) is one of five neurotrophin growth factors which shape the development of the nervous system by regulating neuronal survival and differentiation. Peripheral neuronal subpopulations expressing the TrkC receptor tyrosine kinase respond to Nt3 with enhanced survival, mitogenesis or cell migration and these neurons are lost in homozygous Nt3 null (-/-) mutant mice. The unexplained perinatal lethality in the Nt3-/- mice, however, suggests a wider function for this neurotrophin. Here we report that Nt3 is essential for the normal development of atria, ventricles, and cardiac out

Further studies on the effect of cyclosporin A on the course of Paragonimus infection in rats.

Paragonimus ohirai-infected rats were treated with cyclosporin A (CyA) at different times during the course of infection. CyA (5 x 80 mg/kg) affected the worm recovery, growth and maturation rates of P. ohirai with respect to control values. This tendency was most remarkable in animals treated 15 days and more after infection with CyA (groups B, +15 to +19 days; C, +25 to +29; D, +35 to +39 and E, +45 to +49). In group A (0 to +4), however, the drug did not affect markedly the growth and maturation of worms, although it significantly lowered worm recovery rates. CyA administration also affecte

Cell-mediated immunity to Dirofilaria immitis.

Cell-mediated immunity to Dirofilaria immitis (DI) in guinea pigs was confirmed by the migration inhibition test (MIT), the blast transformation test (BTT), the delayed skin reaction, and the skin reaction by passive transfer with sensitized peritoneal exudate (PE) cells. All migration inhibition (MI) positive cases were always associated with positive skin reactions and two cases showed positive skin reactions without MI. The cellular antibody confirmed by MIT first appeared on the 4th day after single sensitization, but DNA synthesis in splenic lymphocytes had already started on the 3rd day

Some considerations relating to the role of Culex pipiens fatigans Wiedemann in the transmission of human filariasis.

This paper is concerned mainly with the relationship between microfilarial periodicity and vector periodicity. The so-called "non-periodic" Pacific form of Wuchereria bancrofti in fact shows a well-marked and relatively constant periodicity. The amplitude of this periodicity is low, which may account for the difficulty of detecting it in small clinical samples. The periodicity is well adapted to the biting cycle of Aëdes polynesiensis, though less so than that of the "semi-periodic" Brugia malayi to forest Mansonioides. Microfilarial periodicity is discussed in the light of recent work on circ

The spectrin-based membrane skeleton is asymmetric and remodels during neural development in C. elegans.

Perturbation of spectrin-based membrane mechanics causes hereditary elliptocytosis and spinocerebellar ataxia, but the underlying cellular basis of pathogenesis remains unclear. Here, we introduced conserved disease-associated spectrin mutations into the Caenorhabditis elegans genome and studied the contribution of spectrin to neuronal migration and dendrite formation in developing larvae. The loss of spectrin resulted in ectopic actin polymerization outside of the existing front and secondary membrane protrusions, leading to defective neuronal positioning and dendrite morphology in adult anim

Lévy-like movement patterns of metastatic cancer cells revealed in microfabricated systems and implicated in vivo.

Metastatic cancer cells differ from their non-metastatic counterparts not only in terms of molecular composition and genetics, but also by the very strategy they employ for locomotion. Here, we analyzed large-scale statistics for cells migrating on linear microtracks to show that metastatic cancer cells follow a qualitatively different movement strategy than their non-invasive counterparts. The trajectories of metastatic cells display clusters of small steps that are interspersed with long "flights". Such movements are characterized by heavy-tailed, truncated power law distributions of persist

Juvenile-specific cathepsin proteases in Fasciola spp.: their characteristics and vaccine efficacies.

Fasciolosis, caused by Fasciola hepatica and Fasciola gigantica, is one of the most neglected tropical zoonotic diseases. One sustainable control strategy against these infections is the employment of vaccines that target proteins essential for parasites' invasion and nutrition acquiring processes. Cathepsin proteases are the most abundantly expressed proteins in Fasciola spp. that have been tested successfully as vaccines against fasciolosis in experimental as well as large animals because of their important roles in digestion of nutrients, invasion, and migration. Specifically, juvenile-spec

Studying Ancient Anthropogenic Impacts on Current Floral Biodiversity in the Southern Levant as reflected by the Philistine Migration.

Human migrations across geographic boundaries can facilitate the introduction of new husbandry practices and dispersal of plants and animals, resulting in changes in biodiversity. As previously demonstrated, the 12th century BCE Philistine migration-to the southern Levantine littoral, involved the transportation of pigs from Europe, engendering long term genetic displacement of local Near Eastern haplotypes. Building on this, and combining biogeographical methods of Floral List comparisons with archaeological data, we have elucidated the Philistine impact on Southern Levantine floral ecosystem

Development and developmental disorders of the enteric nervous system.

The enteric nervous system (ENS) arises from neural crest-derived cells that migrate into and along the gut, leading to the formation of a complex network of neurons and glial cells that regulates motility, secretion and blood flow. This Review summarizes the progress made in the past 5 years in our understanding of ENS development, including the migratory pathways of neural crest-derived cells as they colonize the gut. The importance of interactions between neural crest-derived cells, between signalling pathways and between developmental processes (such as proliferation and migration) in ensu
