Novel symbiotic protoplasts formed by endophytic fungi explain their hidden existence, lifestyle switching, and diversity within the plant kingdom.

Diverse fungi live all or part of their life cycle inside plants as asymptomatic endophytes. While endophytic fungi are increasingly recognized as significant components of plant fitness, it is unclear how they interact with plant cells; why they occur throughout the fungal kingdom; and why they are associated with most fungal lifestyles. Here we evaluate the diversity of endophytic fungi that are able to form novel protoplasts called mycosomes. We found that mycosomes cultured from plants and phylogenetically diverse endophytic fungi have common morphological characteristics, express similar

Mycotic keratitis caused by concurrent infections of Exserohilum mcginnisii and Candida parapsilosis.

BACKGROUND: Mycotic keratitis in human cornea has been rarely reported to be associated with a co-infection of filamentous fungi and yeast. This paper aims to report a case of mycotic keratitis concurrently infected by Exserohilum mcginnisii and Candida parapsilosis. CASE PRESENTATION: A Chinese female presented two superposed corneal infiltrates with different size and texture on her left eye. In vivo confocal microscopy showed hyper-reflective multiple linear with highly branching structures distributing in the anterior corneal stroma. Inoculations of the corneal lesion scrape concurrently

An improved method compatible with metagenomic analyses to extract genomic DNA from soils in Tuber melanosporum orchards.

AIMS: The development of high-throughput methods such as pyrosequencing and microarrays has greatly improved our understanding of the microbial diversity in complex environments such as soils. Nevertheless, albeit advancements in such techniques, the first major step is to obtain high quantity and good quality genomic DNA (gDNA). The work presented here aims to present an inherent problem with 260 : 230 nm ratio of extracted gDNA from calcareous soils of Tuber melanosporum orchards and a protocol to overcome this problem. METHODS AND RESULTS: Using two commercial gDNA extraction kits on spati

Chromoblastomycosis in Mainland China: a systematic review on clinical characteristics.

Chromoblastomycosis is one of the most frequent chronic infections caused by melanized fungi. In order to evaluate the clinical characteristics of chromoblastomycosis in Mainland China, we performed an evidence-based review of published literature. PubMed and Chinese-language database of CNKI, VIP and Wanfang data during January 1990-August 2011 were searched. Epidemiology, clinical features, laboratory findings, therapy and prognosis were analyzed. Cladophialophora carrionii was the most common causative agent in the north of the Mainland China, and Fonsecaea monophora and F. pedrosoi were th

Maxillary sinusitis caused by Lasiodiplodia theobromae.

Lasiodiplodia (monotypic) comprises a very small proportion of the fungal biota. It is a common plant pathogen in tropical and subtropical regions. Clinical reports on its association with onychomycosis, corneal ulcer and phaeohyphomycosis are available. However, Lasiodiplodia theobromae causing fungal sinusitis has not been reported. We present here a case of fungal sinusitis in a 30-year-old woman, who came to the ENT OPD (out patient department) with complaints of intermittent bleeding and nasal discharge from the left side for a week. The patient complained of headache, predominantly on th

Viability and nematophagous activity of the freeze-dried fungus Arthrobotrys robusta against Ancylostoma spp. infective larvae in dogs.

Viability and in vitro and in vivo activities of freeze-dried conidia of the predatory fungus Arthrobotrys robusta (I-31) were evaluated against infective larvae (L(3)) of Ancylostoma spp. in dogs. A. robusta conidia were lyophilized and stored at 4°C for a month. Freeze-dried conidia were diluted to 1×10(3)conidia/ml and tested in vivo. The treated group consisted of a solution containing conidia (1ml) and 1000 Ancylostoma spp. (L(3)) placed on Petri dishes plated with 2% water-agar (2% WA), at 25°C, in the dark for 10 days. The control group consisted of 1000 Ancylostoma spp. L(3), plated on

Back pain associated with endobronchial mucus impaction due to Bipolaris australiensis colonization representing atypical Allergic Bronchopulmonary Mycosis.

A 40-year-old white male had a two-year history of right sided upper back pain without dyspnea or coughing. His past history was significant for allergic rhinitis and asthma. Conservative outpatient management of the back pain including medications, rest and physical therapy were ineffective. CT scan of the chest revealed a mass in the lower lobe of the right lung. Bronchial biopsy and lavage revealed eosinophilic and neutrophilic inflammation without tissue invasion. A pure culture of Bipolaris australiensis was confirmed by DNA typing. Hematological, serological and radiological studies were

Scanning electron microscopy of Ancylostoma spp. dog infective larvae captured and destroyed by the nematophagous fungus Duddingtonia flagrans.

The interaction between the nematode-trapping fungus Duddingtonia flagrans (isolate CG768) against Ancylostoma spp. dog infective larvae (L(3)) was evaluated by means of scanning electron microscopy. Adhesive network trap formation was observed 6h after the beginning of the interaction, and the capture of Ancylostoma spp. L(3) was observed 8h after the inoculation these larvae on the cellulose membranes colonized by the fungus. Scanning electron micrographs were taken at 0, 12, 24, 36 and 48 h, where 0 is the time when Ancylostoma spp. L(3) was first captured by the fungus. Details of the capt

A skin infection mimicking chromoblastomycosis by a Capnodialean fungus.

Most black fungi that are repeatedly involved in human infection belong to the order Chaetothyriales. Capnodialean melanized fungi often thrive in extreme environments like rock surfaces and hypersaline microhabitats. They are able to grow meristematically with very thick cellular walls, resembling muriform cells of agents of chromoblastomycosis. In this report we describe a member of the order Capnodiales causing a chromoblastomycosis-like infection in human skin. However, in tissue the fungus presented with toruloid hyphae and intercalary, chlamydospore-like conidia with transversal septa, r

Ultrastructural characterization of melanosomes of the human pathogenic fungus Fonsecaea pedrosoi.

Melanin is a complex polymer widely distributed in nature and has been described as an important virulence factor in pathogenic fungi. In the majority of fungi, the mechanism of melanin formation remains unclear. In Fonsecaea pedrosoi, the major etiologic agent of chromoblastomycosis, melanin is stored in intracellular vesicles, named melanosomes. This paper details the ultrastructural aspects of melanin formation, its storage and transportation to the cell wall in the human pathogenic fungus F. pedrosoi. In this fungus, melanin synthesis within melanosomes also begins with a fibrillar matrix

Delayed-type hypersensitivity response to crude and fractionated antigens from Fonsecaea pedrosoi CMMI 1 grown in different culture media.

Chromoblastomycosis is a subcutaneous fungal disease caused by dematiaceous fungi, especially by Fonsecaea pedrosoi, regarded as its major causative agent in Brazil. In recent years there has been a decline in the use of skin testing for delayed-type hypersensitivity (DTH) in epidemiological surveys of fungal infections, mainly because of the unpredictability of positive reactions and lack of specificity of the antigens used. The aim of the present study was to assess delayed-type skin tests in guinea pigs experimentally infected with F. pedrosoi using exoantigens prepared from two culture fil

Pneumocystis.

Pneumocystis organisms can cause pneumonia in mammals that lack a strong immune defense. The genus Pneumocystis contains many different organisms that can be distinguished by DNA sequence analysis. These different organisms are different species of yeast-like fungi that are most closely related to the ascomycete, Schizosaccharomyces pombe. Each species of Pneumocystis appears to be specific for the mammal in which it is found. The species that infects humans is Pneumocystis jiroveci. P. jiroveci has not been found in any other mammal and the species of Pneumocystis found in other mammals have

A hydroxytetradecatrienoic acid from Mycosphaerella rubella.

Investigations on the acidic fractions from extracts of a culture of Mycosphaerella rubella led to the isolation of a new unsaturated dihydroxy acid, 6,13-dihydroxytetradeca-2,4,8 trienoic acid, and two of its derivatives, which are probably artifacts. Their structures were determined on the basis of H and C NMR evidence. The absolute configuration of the chiral centres of the new acid was elucidated using the modified Mosher's method.

Pseudallescheria boydii mycetoma in northern New England.

Mycetoma is a chronic subcutaneous fungal infection characterized by tumefaction, draining sinuses, and grains. It is most common in the tropics but occasionally occurs in the United States. We report a case of a mycetoma affecting the foot of a 38-year-old mentally retarded man from northern New England. The causal organism was identified as Pseudallescheria boydii, the most common cause of mycetoma in the United States. The patient showed a partial response to 8 months of ketoconazole therapy.

An ultrastructural study of pale eumycetoma grains.

The electron microscopic appearances of grains from seven patients with pale grain eumycetoma infections have been studied. The infecting fungi were Pseudallescheria boydii (2), Fusarium sp. (1), Acremonium kiliense (1), a non-sporing mould (2) and Trichophyton violaceum (1). P. boydii grown in vitro and grains from a Madurella mycetomatis infection were used for comparison. The most important ultrastructural features of the pale eumycetoma grains were the gross thickening of the fungal cell walls maximal at the periphery of the grain, the predominance of grossly disorganised hyphae and the ap
