Quantitative MRI changes in neck muscle size and composition following long-duration spaceflight.

Long-duration spaceflight poses several challenges to human health, including muscle loss and fat deposition. To mitigate these effects, exercise countermeasures are prescribed. This study aimed to measure neck muscle size and composition changes with long-duration spaceflight using quantitative magnetic resonance imaging (MRI) and examine associations with in-flight exercise modality and volume. Nine astronauts (mission duration: 7.1 ± 1.8 months) underwent pre- and post-flight 3T Dixon MRI of the cervical spine. Cross-sectional area (CSA) and muscle fat fraction (MFF) were measured bilateral…

Biological sex as a variable in immunity does not affect parabolic flight-induced alterations in immune responses.

The present era of spaceflight is accompanied by two meaningful breakthroughs. The access to in-orbit missions for civilians and the increasing enrolment of female astronauts require detailed investigations on the impact of gravitational stress on human physiology with focus on sex-specific differences. To assess the responsive capacities of innate and adaptive immunity in this context, functional characterizations were performed in women and men in a parabolic flight study. Blood and saliva were collected 1 month prior and on the day before the flight, as well as directly after flight and on …

Bone metabolism during strict head-down tilt bed rest and exposure to elevated levels of ambient CO2.

Astronauts on the International Space Station are exposed to levels of atmospheric carbon dioxide (CO2) above typical terrestrial levels. We explored the possibility that increased levels of ambient CO2 further stimulate bone resorption during bed rest. We report here data from 2 ground-based spaceflight analog studies in which 12 male and 7 female subjects were placed in a strict 6° head-down tilt (HDT) position for either 30 days at 0.5% ambient CO2 or 60 days with nominal environmental exposure to CO2. Bone mineral density (BMD) and bone mineral content (BMC) were determined using dual-ener…

Global cannabinoid receptor 1 deficiency affects disuse-induced bone loss in a site-specific and sex-dependent manner.

Bone loss during mechanical unloading increases fracture risk and is a major concern for the general population and astronauts during spaceflight. The endocannabinoid system (ECS) plays an important role in bone metabolism. One of the main ECS receptors, cannabinoid receptor 1 (CB1), has been studied in regards to basic bone metabolism; however, little is known as to how CB1 and the ECS affect bone in different mechanical environments. In this study, we analyzed the influence of global CB1 deficiency and sex on mice during disuse caused by single limb immobilization. Female mice were more sens…

Intraocular pressure during handgrip exercise: The effect of posture and hypercapnia in young males.

PURPOSE: As part of our investigations of intraocular pressure (IOP) as a potential contributing factor to the spaceflight-associated neuro- … l ocular hypertension. CONCLUSIONS: IOP in the prone 6°HDT position was similar to IOP reported in astronauts upon entering microgravity, potentially indicating that prone, rather than supine 6°HDT position might be a more suitable experimental analog for investigating the acute ocular changes that occur in microgravity.

Spinal Health during Unloading and Reloading Associated with Spaceflight.

Spinal elongation and back pain are recognized effects of exposure to microgravity, however, spinal health has received relatively little attention. This changed with the report of an increased risk of post-flight intervertebral disc (IVD) herniation and subsequent identification of spinal pathophysiology in some astronauts post-flight. Ground-based analogs, particularly bed rest, suggest that a loss of spinal curvature and IVD swelling may be factors contributing to unloading-induced spinal elongation. In flight, trunk muscle atrophy, in particular multifidus, may precipitate lumbar curvature…

Radiation carcinogenesis risk assessments for never-smokers.

Cigarette smoking, which is presently associated with more than 20% of adult deaths in the United States, is a large confounder to radiation risk estimates derived from epidemiology data. Astronauts and other exposed groups are classified as never-smokers (NS), defined as lifetime use of less than 100 cigarettes. In the past, radiation risk estimates have been made using average U.S. population rates for cancer and all causes of death, which may lead to overestimation of radiation risks for NS. In this report, age- and gender-specific radiation carcinogenesis risk calculations for NS and the a…

Exercise within lower body negative pressure partially counteracts lumbar spine deconditioning associated with 28-day bed rest.

Astronauts experience spine deconditioning during exposure to microgravity due to the lack of axial loads on the spine. Treadmill exercise in a lower body negative pressure (LBNP) chamber provides axial loads on the lumbar spine. We hypothesize that daily supine LBNP exercise helps counteract lumbar spine deconditioning during 28 days of microgravity simulated by bed rest. Twelve sets of healthy, identical twins underwent 6 degrees head-down-tilt bed rest for 28 days. One subject from each set of twins was randomly assigned to the exercise (Ex) group, whereas their sibling served as a nonexerc…

Effects of microgravity on maximal power of lower limbs during very short efforts in humans.

The maximal power of the lower limbs was determined in four astronauts (age 37-53 yr) 1) during maximal pushes of approximately 250 ms on force platforms ["maximal explosive power" (MEP)] or 2) during all-out bouts of 6-7 s on an isokinetic cycloergometer [pedal frequency 1 Hz: maximal cycling power (MCP)]. The measurements were done before and immediately after spaceflights of 31-180 days. Before flight, peak and mean values were 3.18 +/- 0.38 and 1.5 +/- 0. 13 (SD) kW for MEP and 1.17 +/- 0.12 and 0.68 +/- 0.08 kW for MCP, respectively. After reentry, MEP was reduced to 67% after 31 days and…

PhysAstro-Pose: Physics-Inspired Semi-Supervised Human Pose Estimation in Microgravity Environments.

Human pose estimation in orbit is critical for astronaut health monitoring, task assistance, and intelligent human-robot interaction aboard space stations. However, in microgravity, human poses exhibit arbitrary orientations and are often affected by severe occlusion and complex background interference, while the scarcity of annotated in-orbit data makes it difficult to directly transfer models trained on ground-based datasets. Existing semi-supervised methods also lack explicit constraints from human structural topology and pose-related physical priors, which often leads to unreasonable pseud

ESA VIVALDI Dry Immersion Microgravity Simulations Induce Increases in Immune Biomarkers Associated With Physical and Psychological Stress, and Sex-Specific Factors.

With future manned space projects involving missions of unprecedented duration, multisystem deconditioning induced by spaceflight could seriously affect the well-being and health of astronauts. Safe and easily determined in-flight biomarkers are therefore needed to monitor health status. In this study, we simulated space deconditioning with a 5-day dry immersion (DI) of 18 healthy women and 19 healthy men and evaluated the effects of this protocol on three biomarkers: the neutrophil-to-lymphocyte ratio (NLR), the granulocyte-to-lymphocyte ratio (GLR) and the platelet-to-lymphocyte ratio (PLR).…

Musculoskeletal responses to spaceflight: mechanisms, countermeasures, and key gaps.

Spaceflight and partial-gravity environments impose profound challenges to the human musculoskeletal system, driving rapid muscle atrophy, progressive bone loss, and tendon maladaptation. At the molecular level, unloading suppresses anabolic signaling, enhances proteolysis, and induces mitochondrial stress, whereas bone and tendon exhibit reduced extracellular matrix turnover and impaired mechanotransduction. Recent space-omics and cross-species studies, including rodent and Caenorhabditis elegans models, reveal that these catabolic responses are evolutionarily conserved and involve systemic p

Effects of simulated microgravity on biological features and virulence of the fungal pathogen Cryptococcus neoformans.

Cryptococcus neoformans is a fungal pathogen that causes cryptococcal meningitis, mainly in immunocompromised individuals, such as those with HIV. Its recent detection on the International Space Station raises concerns about potential health risks in space, where immune systems may be compromised. However, its behavior in space-like conditions remains unclear. In this study, we examined the effects of simulated microgravity on C. neoformans. We found that the condition enhanced the fungus's resistance to membrane and osmotic stress and increased key virulence factors, including capsule formati

Formaldehyde initiates memory and motor impairments under weightlessness condition.

During space flight, prolonged weightlessness stress exerts a range of detrimental impacts on the physiology and psychology of astronauts. These manifestations encompass depressive symptoms, anxiety, and impairments in both short-term memory and motor functions, albeit the precise underlying mechanisms remain elusive. Recent studies have revealed that hindlimb unloading (HU) animal models, which simulate space weightlessness, exhibited a disorder in memory and motor function associated with endogenous formaldehyde (FA) accumulation in the hippocampus and cerebellum, disruption of brain extrace…
