Autophagy-Actin Crosstalk: Implications for Cellular Homeostasis and Disease.

Actin is a key component of the cytoskeleton and plays diverse roles in cellular processes. Autophagy regulates homeostasis through various mechanisms that recycle nutrients and degrade unnecessary or harmful cellular components and aggregates. These two processes are engaged in a highly conserved crosstalk through which they regulate each other, including autophagolysosomal formation and regulation of actin dynamics. The regulation of autophagy is involved in cancer, neurodegeneration, infectious diseases, and inflammation, providing possible avenues for treatments for these diseases. In this…

Autophagy in thyroid cancer: Immune suppression and drug resistance.

Thyroid cancer exhibits a broad spectrum of clinical behaviors, ranging from indolent differentiated tumors to highly aggressive, dedifferentiated malignancies that are refractory to radioactive iodine (RAI), targeted therapy, and immunotherapy. Autophagy, a highly conserved lysosome-mediated degradation pathway, has been identified as a crucial homeostatic mechanism with either tumor-suppressive or tumor-promoting effects depending on the regulatory context. Increasing evidence suggests that while basal autophagy may help attenuate genomic instability during early tumorigenic stages, advanced…

Coronavirus NSP5 protease cleaves CCDC50 to evade antiviral autophagy.

Selective macroautophagy/autophagy is a critical component of innate antiviral defense, relying on selective autophagy receptors to recognize viral cargo and deliver it for lysosomal degradation. In our recent study, we demonstrated that porcine deltacoronavirus (PDCoV) evades this pathway through its NSP5 protease. We uncovered a previously unrecognized antiviral function of the selective autophagy receptor CCDC50, which recognizes K63-linked polyubiquitinated PDCoV envelope (E) protein at lysine 72 and mediates its autophagic degradation, thereby restricting viral replication. This antiviral…

Multilayered interplay between the cGAS-STING pathway and autophagy.

The cGAS-STING pathway detects cytosolic dsDNA to initiate innate immune responses, serving as a critical surveillance system against infection and cellular stress. Autophagy is an evolutionarily conserved catabolic process that maintains homeostasis by degrading cytoplasmic components. Although these two systems operate through distinct mechanisms, recent studies have uncovered a complex bidirectional regulatory network that intimately links them. On the one hand, STING has an evolutionarily conserved capacity to induce TBK1-independent noncanonical autophagy, and recent studies further link …

TM9SF1 Aggravates Liver Ischemia-Reperfusion Injury by Promoting Autophagy.

Liver transplantation (LT) remains the only effective treatment for end-stage liver disease. Autophagy critically regulates liver ischemia–reperfusion (I/R) injury. Transmembrane 9 superfamily member 1 (TM9SF1) is a transmembrane protein associated with the elevation of autophagy. However, the role and mechanism of this protein in liver I/R injury have not been explored. We observed upregulated TM9SF1 expression in liver I/R mice models and AML12 cells subjected to hypoxia-reoxygenation (H/R). Using TM9SF1 adeno-associated virus (AAV) to generate overexpression and knockdown (KD) mice with liv…

Autophagy-Lysosomal Axis Stimulation by Beta-Hydroxybutyrate in Astrocytes.

Beta-hydroxybutyrate (D-BHB), a ketone body with neuroprotective properties, has been previously shown to increase the autophagic flux in neurons, but its effect on autophagy induction in astrocytes remains unexplored. Astrocyte functionality is essential for neuronal support, and this includes an optimal degradation and recycling of organelles and other cellular components through autophagy. The present study shows that D-BHB exposure to cultured astrocytes elevates the conjugated form of microtubule-associated protein 1A/1B-light chain 3 (LC3-II) levels, increases the number of autophagosome…

vtRNA1-1/p62 regulates macrophages autophagy in ankylosing spondylitis.

This research aims to investigate the role and mechanisms of the vtRNA1-1/p62 molecular axis in the regulation of autophagy in AS, with the goal of identifying novel diagnostic and therapeutic targets for AS. Clinical sample analysis revealed that the transcription levels of vtRNA1-1 and p62 decreased in the peripheral blood mononuclear cells (PBMCs) of the AS group; conversely, the levels of canonical autophagy-related genes (ATG3, ATG5) and inflammatory factors (TNF-α) significantly increased. Correlation analysis revealed that vtRNA1-1 levels were positively associated with p62 but were inv…

AKAP11 links autophagy to cognitive impairment in psychiatric diseases.

Schizophrenia (SCZ) and bipolar disorder (BD) share genetic risk factors and cognitive impairments, yet the underlying mechanisms remain incompletely understood. Loss-of-function variants in AKAP11 (A-kinase anchoring protein 11) have recently emerged as major risk factors for both disorders. Our recent study demonstrates that AKAP11 deficiency in the mouse hippocampus causes cognitive deficits and synaptic dysfunction, accompanied by autophagy dysregulation. Mechanistically, AKAP11 interacts with PPP3CB (protein phosphatase 3 catalytic subunit beta) to promote TFEB (transcription factor EB) d…

Autophagy in ocular diseases: from mechanisms to therapeutic potential.

Autophagy represents a fundamental and evolutionarily preserved mechanism of degradation and metabolism in eukaryotic cells. This process is triggered by a variety of stressors, including nutrient deprivation, energy deficits, protein misfolding, low oxygen levels, and pathogen infections by pathogens. Autophagy plays a vital role in maintaining cellular equilibrium. The process of vision is notably complex, making the eye one of the most metabolically active tissues in the human body. The proper function of the eye relies on the preservation of metabolic homeostasis and the structural integri…

Autophagy-exosome crosstalk in neurodegeneration: Mechanisms and therapeutic opportunities.

Neurodegenerative diseases (NDs), including Alzheimer's, Parkinson's, Huntington's, amyotrophic lateral sclerosis, and multiple sclerosis, share a common pathogenic signature: disrupted proteostasis driven by impaired autophagy and maladaptive exosome dynamics. Under normal conditions, autophagy maintains neuronal homeostasis by clearing misfolded proteins and damaged organelles, while exosomes mediate neuroglial communication. When autophagic flux is impaired or lysosomal function is compromised, intracellular cargo handling can shift toward secretion and undegraded cargo may be redirected in…
