BACKGROUND: Optimizing deep brain stimulation (DBS) programming in Parkinson's disease (PD) requires a complex process for evaluating clinical benefit and adverse effects. Localizing maximum beta (13-30Hz) power may guide DBS programming. Novel use of in-clinic unipolar recording may improve the efficacy of neurophysiology-guided DBS programming.
OBJECTIVE: Compare the contact selection and clinical outcomes based upon either maximum beta power from unipolar recordings or standard-of-care monopolar review at initial programming in PD.
METHODS: Ten PD patients (4F/6M, post-diagnosis = 9.2 ± 3

Ubiquitin-proteasome system (UPS) dysfunction is associated with the pathology of a wide range of human diseases, including myopathies and muscular atrophy. However, the mechanistic understanding of specific components of the regulation of protein turnover during development and disease progression in skeletal muscle is unclear. Mutations in KLHL40, an E3 ubiquitin ligase cullin3 (CUL3) substrate-specific adapter protein, result in severe congenital nemaline myopathy, but the events that initiate the pathology and the mechanism through which it becomes pervasive remain poorly understood. This review describes circadian patterns in cardiovascular disease processes and analyses the findings of recent studies. The awareness of an increased incidence of myocardial infarction and sudden cardiac death in the early morning hours, shortly after waking, has stimulated an interest in the relationship of these events and the occurrence of both silent and symptomatic myocardial ischaemia. A number of studies have been reported that examine both the physiological triggers and the underlying causes of these events. Beta-adrenergic blockers have been shown to abolish the early morning peak o

Plotting hourly mean blood pressures recorded continuously by an intra-arterial technique in fully ambulant subjects on a time scale has clearly outlined a circadian biorhythm which is a useful tool for the study of antihypertensive drugs. Studies with beta-adrenoreceptor blocking drugs have shown differences which may have fundamental significance in the clinical usage of these drugs for treating hypertension.

A motion trajectory prediction (MTP) - based brain-computer interface (BCI) aims to reconstruct the three-dimensional (3D) trajectory of upper limb movement using electroencephalography (EEG). The most common MTP BCI employs a time series of bandpass-filtered EEG potentials (referred to here as the potential time-series, PTS, model) for reconstructing the trajectory of a 3D limb movement using multiple linear regression. These studies report the best accuracy when a 0.5-2Hz bandpass filter is applied to the EEG. In the present study, we show that spatiotemporal power distribution of theta (4-8

This study reports the results that were obtained in 56 cases of arterial hypertension in pregnancy solely by beta-blocking with Atenolol or Labetalol. Any pregnant woman whose arterial blood pressure rises to or exceeds 140/90 mm mercury in two successive examinations at intervals of 8 days with rest is considered to be hypertensive. As soon as treatment is started mothers' supervision is assured regularly by clinical and biological examinations and the dose of drug is adapted to each case. Fetal monitoring is ensured by ultrasound, cardiac rhythm tracings and hormone estimations. As far as t

Evidence has shown that constant light or constant darkness significantly impairs cognitive functions and affects the mood state. The present study investigated the effect of circadian rhythm disturbances (circadian rhythm disruptions, CRDs) on locomotor activity, rearing behavior, pain threshold, novel object recognition memory, and brain-derived neurotrophic factor (BDNF) and glycogen synthase kinase-3 beta (GSK-3β) expression levels in the hippocampus of both sexes of rats. CRD was done using a combination of constant light (48 h) and constant darkness (48 h) periods for 8 (short-term), 16

Disruption of the circadian timing system has been reported in the preclinical phase of Alzheimer's disease (AD) and is a well-characterized component of mid- and late-stage AD. Given the distributed nature of the body's clock, with a central pacemaker in the suprachiasmatic nucleus (SCN) and peripheral clocks throughout the brain, understanding how AD affects this system has been challenging. To investigate how AD may disrupt circadian physiology, we focused on the amyloid-beta (Aβ) peptide, a key contributor to familial early-onset AD. Using the 5xFAD mouse model and ex vivo single-cell prof

AIM: Knowledge of how circadian rhythm influences brain health remains limited. We aimed to investigate the associations of accelerometer-measured circadian rest-activity rhythm (CRAR) with incident dementia, cognitive dysfunction, and structural brain abnormalities in the general population and underlying biological mechanisms.
METHODS: Fifty-seven thousand five hundred and two participants aged over 60 years with accelerometer data were included to investigate the association of CRAR with incidental dementia. Non-parametric CRAR parameters were utilized, including activity level during acti

BACKGROUND: Hyponatraemia is easily corrected by treatment with an oral vasopressin antagonist, but these medications are costly and their use at outpatient clinics is restricted by government-managed insurance in Japan. Acetazolamide could be an alternative diuretic to a vasopressin antagonist.
CASE SUMMARY: An 83-year-old dyspnoeic male patient was emergently admitted to the hospital due to decompensated heart failure (HF), hypotension, and hyperkalaemia-associated sinus arrest with a junctional escape rhythm. Urgent treatment with a noradrenaline drip infusion and a beta stimulant adhesive

The posterior alpha rhythm (8-14 Hz), originating in occipito-parietal areas through thalamocortical generation, displays characteristics of visual activity in anticipation of visual events. Posterior alpha power is influenced by visual spatial attention via top-down control from higher order attention areas such as the frontal eye field. It covaries with visual cortex excitability, as tested through transcranial magnetic stimulation (TMS), and predicts the perceptual fate of a forthcoming visual stimulus. Yet, it is still unknown whether the nature of the relationship between this prestimulus

OBJECTIVES: The pineal gland transduces photoperiodic informations to the neuroendocrine axis through the nocturnally melatonin secretion. This hormonal message plays a major role in the biological rhythm regulation. By autoradiography, more than 130 melatonin putative targets have been reported in the central nervous system (CNS) and in peripheral tissues. However, cross-species consensus concern only a few of them like the suprachiasmatic nuclei (SCN), the master circadian clock, and the pars tuberalis of the pituitary. Recently, MT1 melatonin receptor cDNA have been cloned in several mammal

OBJECTIVES: Despite optimal pharmacological treatment a large proportion of patients with atrial fibrillation (Afib) are not arrhythmia-free, and remain at risk for complications such as stroke and cardiac morbidity. If first-line treatment fails, most patients receive second-line pharmacological treatment. The emergence of new technologies aimed at restoring and maintaining sinus rhythm, such as catheter ablation techniques, has increased the interest in the economic aspects of second-line pharmacological treatment. The objective was therefore to calculate the 5-year direct medical costs of s

Sotalol is a beta-blocking agent endowed with class III electrophysiological properties. It has proved clinically effective in the treatment of arrhythmia, but episodes of torsades de pointe have been observed, particularly (though not exclusively) in the presence of hypokalaemia. The effect of sotalol with or without hypokalaemia was studied on a recently developed model for experimental torsades de pointe. Conscious dogs with complete atrioventricular block (ventricular cycle RR = 1530 +/- 170 ms) and provided with permanent atrial and ventricular epicardial electrodes were given sotalol int

Excessive daytime sleepiness, i.e., the inability to sustain wakefulness during the day, is a common Parkinson's disease (PD) non-motor symptom. Elevated cortico-basal ganglia beta oscillatory power (8-35 Hz) is a pathophysiological signature of PD motor signs. The relationship between beta oscillatory power and daytime sleepiness, however, remains unclear. Here, we examined the relationship between beta power and daytime vigilance in parkinsonian nonhuman primates. We administered dopaminergic medication to alter cortico-basal ganglia beta power and measured its effect on daytime vigilance. T

Precision step control during gait initiation is essential for stable locomotion. This study examined the role of the central nervous system in this process. In Experiment 1, 14 healthy young adults (21.9 ± 0.9 years) performed a control task (normal gait initiation) and a precision task (targeted foot placement with the first step). Task movements were divided into three phases: planning, preparation, and swing. Event-related desynchronization (ERD) in the alpha and beta bands (α-ERD and β-ERD) was calculated at Fz, Cz, and Pz for each phase. Step accuracy was evaluated as the outcome measure

BACKGROUND: Late-life depression (LLD) often co-occurs with mild cognitive impairment (MCI), and patients with LLD and MCI (LLD-MCI) have an increased risk of progression to Alzheimer's disease (AD). However, differences in resting-state neural oscillation and cognitive impairment in LLD patients remain unclear. In this cross-sectional study, electroencephalography (EEG) was used to analyse, local rhythm activity and large-scale network communication to differentiate LLD patients with and without MCI.
METHODS: We enrolled 113 participants: 74 with LLD (50 with LLD-MCI and 24 with LLD-non-MCI)

Postoperative sleep disorder frequently occurs in patients after surgery. Sleep disturbance aggravates pain, anxiety, and delirium, which is an important risk factor for poor recovery. Circadian rhythm disorder induced by general anesthesia plays important role in postoperative sleep disorders. A large number of clinical studies have shown that various forms and duration of general anesthesia can lead to postoperative sleep disorders. In this study, the effect of prolonged propofol anesthesia on biological rhythm was comprehensively evaluated by wireless physiological telemetry system, and the

Previous studies have explored neurofeedback training for Parkinsonian patients to suppress beta oscillations in the subthalamic nucleus (STN). However, its impacts on movements and Parkinsonian tremor are unclear. We developed a neurofeedback paradigm targeting STN beta bursts and investigated whether neurofeedback training could improve motor initiation in Parkinson's disease compared to passive observation. Our task additionally allowed us to test which endogenous changes in oscillatory STN activities are associated with trial-to-trial motor performance. Neurofeedback training reduced beta

Seasonal variations of the photoperiod have been shown to regulate biological and behavioral functions, with also effects on clinical symptom and course of several psychiatric conditions. Although melatonin is considered the principal signal used to transmit informations about the light and dark cycle, a dopamine (DA) role in regulating seasonal changes has been suggested. Few studies have addressed a seasonal pattern of dopamine, and human studies have been conducted on inter-subject differences, comparing measures obtained during fall-winter with those of spring-summer. We studied within-sub
