In Vivo Acute on Chronic Ethanol Effects in Liver: A Mouse Model Exhibiting Exacerbated Injury, Altered Metabolic and Epigenetic Responses.

Chronic alcoholics who also binge drink (i.e., acute on chronic) are prone to an exacerbated liver injury but its mechanism is not understood. We therefore investigated the in vivo effects of chronic and binge ethanol ingestion and compared to chronic ethanol followed by three repeat binge ethanol on the liver of male C57/BL6 mice fed ethanol in liquid diet (4%) for four weeks followed by binge ethanol (intragastric administration, 3.5 g/kg body weight, three doses, 12h apart). Chronic followed by binge ethanol exacerbated fat accumulation, necrosis, decrease in hepatic SAM and SAM:SAH ratio,

Stress and gonadal steroid influences on alcohol drinking and withdrawal, with focus on animal models in females.

Sexually dimorphic effects of alcohol, following binge drinking, chronic intoxication, and withdrawal, are documented at the level of the transcriptome and in behavioral and physiological responses. The purpose of the current review is to update and to expand upon contributions of the endocrine system to alcohol drinking and withdrawal in females, with a focus on animal models. Steroids important in the hypothalamic-pituitary-gonadal and hypothalamic-pituitary-adrenal axes, the reciprocal interactions between these axes, the effects of chronic alcohol use on steroid levels, and the genomic and…

Topiramate effects on ethanol and sucrose binge-like intake, anxiety- and compulsive-like behavior in non-dependent male and female C57BL/6J mice.

RATIONALE: Repetitive binge episodes are early-stage patterns in the addiction cycle associated with elevated anxiety and compulsive traits. Despite their high comorbidity and the growing off-label use of topiramate for binge-related disorders, its effects in non-dependent individuals remain poorly characterized and studies systematically comparing both sexes under identical experimental conditions remain scarce. OBJECTIVES: To evaluate the effects of topiramate on ethanol (EtOH) and sucrose binge-like intake, anxiety- and compulsive-like responses in non-dependent male and female C57BL/6J mi

The impact of adolescent drinking on traumatic brain injury-induced cognitive deficits and alcohol preference in adult C57BL/6J mice.

BACKGROUND: Industrial workers and active military personnel within combat roles face heightened risk for blast pressure wave traumatic brain injury (bTBI). Previous studies have shown that experiencing TBI is associated with increased alcohol (EtOH) consumption and illicit substance use. Notably, alcohol use typically begins during late adolescence or early adulthood, a period that precedes many TBI incidents; moreover, early-onset drinking is further associated with heightened risk of developing an alcohol use disorder (AUD) even in the absence of TBI. Adolescent binge drinking can induce la…

Indicated Prevention of Fetal Alcohol Spectrum Disorders in South Africa: Effectiveness of Case Management.

In the Western Cape Province of South Africa (ZA) a subculture of binge drinking produces the highest global documented prevalence of fetal alcohol spectrum disorders (FASD). FASD prevention research activities in ZA use the Comprehensive Prevention approach from the United States Institute of Medicine. Case management (CM) was delivered as a method of indicated prevention to empower heavy drinking pregnant women to achieve cessation or a reduction in drinking. CM activities incorporated life management, Motivational Interviewing (MI) techniques and the Community Reinforcement Approach (CRA). …

The social context of drinking and its association with days of heavy episodic drinking in the past month among a nationally representative sample of adolescents and young adults in the United States, 2021-23.

INTRODUCTION: Prior research links solitary drinking under the age of 21 years, the legal drinking age in the USA, with adverse health later in life. This study examines associations between the social context of drinking (i.e. drinking alone vs with others) and the frequency of heavy episodic drinking (HED) days in the past 30 days among 12-20-year-olds in the USA. MATERIALS AND METHODS: We aggregated National Survey on Drug Use and Health data from 2021 to 2023 for all individuals aged 12-20 years who drank alcohol in the past month (unweighted n = 6 261). The focal independent variable of

Understanding positive heavy drinking attitudes: daily variability and their role in intervention-related changes in alcohol use.

PURPOSE: Alcohol use and its related harms remain highly prevalent among young adults, with heavy drinking and alcohol-induced blackouts posing significant public health risks. This study had two goals: first, we examined attitudes toward heavy drinking as both stable (between-person) and dynamic (within-person) predictors of alcohol use and consequences among heavy-drinking young adults. Furthermore, we explored changes in attitudes toward heavy drinking as a mediator of intervention efficacy. METHOD: The data for the current work come from a larger randomized controlled trial where particip

Neighborhood-Level Drinking Norms and Alcohol Intervention Outcomes in HIV Patients Who Are Heavy Drinkers.

BACKGROUND: Heavy alcohol consumption can be harmful, particularly for individuals with HIV. There is substantial variability in response to interventions that aim to reduce drinking. Neighborhood drinking norms may explain some of this variability among HIV-infected patients. Therefore, we investigated whether neighborhood-level drinking norms modified response to alcohol intervention among HIV-infected heavy drinkers. METHODS: Heavily-drinking HIV comprehensive care patients (n = 230) completed 1 of 3 brief alcohol interventions (an educational intervention, a motivational interviewing [MI]

Positive Childhood Experiences are Associated With Alcohol Use in Adolescent and Emerging Adult Females by Adverse Childhood Experiences Dimension.

PURPOSE: Experiencing multiple adverse childhood experiences (ACEs) is associated with alcohol use in female adolescents and emerging adults. Protective and compensatory experiences (PACEs) have been theorized to off-set the health and behavioral consequences from the accumulation of ACEs throughout childhood. This study examines the association between protective experiences and subsequent alcohol and binge alcohol use frequency over one month among female adolescent and emerging adults reporting high and low levels of two ACE dimensions (household dysfunction and emotional abuse/neglect). Differences in Alcohol-Related Variables Between Individuals Who Engage in Food and Alcohol Disturbance (FAD) Behaviors and Those Who Only Use Alcohol: The Role of FAD-Motives.

OBJECTIVE: Food and alcohol disturbance (FAD) is the use of any compensatory behavior (e.g., skipping meals) within the context of a drinking episode. FAD has two underlying motives: to enhance the effects of alcohol (FAD-AE) and/or compensate for calories consumed from alcohol (FAD-CC). Prior work finds that FAD is positively associated with alcohol-related outcomes; however, it is unclear whether FAD confers increased risk above alcohol use alone and whether there are differences in alcohol outcomes by FAD-motive. Thus, the present study evaluated alcohol use patterns (i.e., past-month quant

Recognition and prevention of fetal alcohol syndrome.

Alcohol use among women of childbearing age is prevalent in the United States, with approximately 1 in 5 nonpregnant women reporting binge drinking (5 or more drinks on any one occasion) and 1 in 25 pregnant women reporting binge drinking. Alcohol use during pregnancy results in a spectrum of adverse outcomes known as fetal alcohol spectrum disorders. Fetal alcohol syndrome (FAS) is one of these disorders. Fetal alcohol syndrome is characterized by specific facial abnormalities and significant impairments in neurodevelopment and physical growth. Early identification of children with FAS has be…
