Diagnostic Value of Nucleated Red Blood Cells in Pediatric Acute Lymphoblastic Leukemia.

This study aimed to determine whether nucleated red blood cells (NRBCs) can be used as a biomarker to support pediatric acute lymphoblastic leukemia (ALL) diagnosis. In total, 60 children newly diagnosed with ALL and 70 controls were included in this retrospective study. Males were significantly higher than females in the ALL group (P = .010). The ALL group had significantly higher NRBC#, NRBC%, WBC, lymphocyte, monocyte, mean corpuscular volume (MCV), mean platelet volume (MPV), and C-reactive protein (CRP) levels (P < .05, for all). The ALL group had significantly lower RBC, platelet, neutro…

Beyond Viral Control: Tenofovir's Hidden Impact on Blood-Brain Barrier Endothelial Cells.

UNLABELLED: The blood-brain barrier (BBB) is a dynamic endothelial interface that protects the brain from harmful agents while regulating molecular exchange. Human immunodeficiency virus (HIV) compromises BBB integrity, promoting neurological damage. Antiretroviral (ARV) therapies suppress HIV replication, preventing immune system deterioration and progression to AIDS. Although Tenofovir-based ARV regimens are vital for HIV treatment and prevention, their impact on cerebrovascular function remains unclear. AIM: This study examined Tenofovir's effects on murine brain endothelial cells using an

Cryopreserving Small Volumes of Red Blood Cells Using the High Glycerol Method.

Cryopreservation of red blood cells (RBCs) plays a critical role in blood inventory management by enabling long-term frozen storage of rare blood types. This process requires the addition of glycerol, a cryoprotectant that mitigates cellular damage caused by ice crystal formation during freezing and thawing. A high glycerol method (HGM) is commonly used, which requires an approximate concentration of 40% glycerol and enables slow freezing rates (1-3°C/min) and extended storage in low-temperature freezers (≤-65°C). While automated cell processing systems have been developed for the glycerolizat…

Animals have expanded the evolutionary legacy of unicellular ancestors in blood cells.

Blood cells are common and unique to animals, enabling them to address critical challenges of defense and transport. Thus, their evolution represents a defining innovation in metazoan multicellular life. However, their evolutionary trajectory about how blood cells emerged and diversified throughout animal history remains unclear. Here, we present a combination of bioinformatics and functional data that demonstrate that the metazoan blood cell program most likely originated through the repurposing of an ancestral premetazoan toolkit governed by Fos. This primordial program established the macro…

Red blood cells in prediabetes: Early alterations, progressive trajectories, and clinical potential.

Red blood cells are not passive oxygen carriers. In overt type 2 diabetes, they have been proposed as both victims and amplifiers of metabolic and vascular dysfunction. Whether these alterations begin earlier in the prediabetic state is only beginning to emerge. This is the first comprehensive mechanistic review to synthesize evidence on RBC dysfunction in prediabetes. We aim to answer three questions. First, what happens to RBCs in prediabetes? We synthesize evidence on altered redox status, membrane composition, morphology, rheology, RBC-indices and function. Second, do these changes follow …

Role of Lipoprotein(a) and Blood Cells Ratios in Peripheral Artery Disease.

Peripheral artery disease (PAD) is a major global health issue. This study investigated the relationship between lipoprotein(a) [Lp(a)], high-density lipoprotein cholesterol (HDL-C) to blood cells ratios, and PAD development. The study included 361 patients categorized into groups based on the presence of stenotic atherosclerosis in lower limb arteries (LLAs) diagnosed via duplex ultrasound. Group 1 (n = 238) had atherosclerosis at the first visit. A second visit involved 281 patients: 158 from Group 1, 32 new diagnoses (Group 2), and 91 with no atherosclerosis at either visit (Group 3). Labor…

Chemically Engineered Peptide Efficiently Blocks Malaria Parasite Entry into Red Blood Cells.

Chemical peptide engineering, enabled by residue insertion, backbone cyclization, and incorporation of an additional disulfide bond, led to a unique cyclic peptide that efficiently inhibits the invasion of red blood cells by malaria parasites. The engineered peptide exhibits a 20-fold enhanced affinity toward its receptor (PfAMA1) compared to the native peptide ligand (PfRON2), as determined by surface plasmon resonance. In-vitro parasite growth inhibition assay revealed augmented potency of the engineered peptide. The structure of the PfAMA1-cyclic peptide complex, predicted by the deep learn…

Highly effective strategy for isolation of mononuclear cells from frozen cord blood.

BACKGROUND AIMS: Cord blood mononuclear cells (CBMCs) comprise a variety of single-nucleated cells found in the cord blood, mainly consisting of monocytes and lymphocytes. They also include a smaller proportion of other cell types, such as hematopoietic stem and progenitor cells (HSPCs) and mesenchymal stromal cells (MSCs). CBMCs are vital for acquiring HSPCs, MSCs, and other immune cells, like natural killer cells. These cells are essential for supporting subsequent research and clinical applications. Although automated equipment for CBMC enrichment has shown promise, the high cost of these m

PURPOSE: The hematological changes in COVID-19 patients continue to receive great attention, especially in the field of public health. To our knowledge, coronavirus disease may be identified based on the severity of illness, and the study of peripheral blood smears may offer important information to facilitate the identification. Thus, we evaluated the morphological abnormalities (atypical and immature lymphocytes, lymphocytes with micronuclei, various nuclear abnormalities among erythrocytes) and quantitative changes in peripheral blood cells among 48 individuals with COVID-19 disease. METHO…

The mitochondrial function of peripheral blood mononuclear cells in frail older patients.

BACKGROUND: Frailty increases the incidence of geriatric syndromes and even the risk of death in old adults. However, the diagnostic criteria for frailty are inconsistent because of complex pathological processes and diverse clinical manifestations. To determine the effective biomarker and recognize frail status early, we investigated the correlation of mitochondrial morphology and function of human peripheral blood mononuclear cells (PBMCs) with frailty status in older adults. METHODS: This Cross-sectional study followed 393 participants (aged 25-100 years, female 31.04 %) from the First Aff

Biological roles of THRAP3, STMN1 and GNA13 in human blood cancer cells.

The nature of association between chronic otitis media (COM)-mucosal disease and sensorineural hearing loss is controversial. Identifying the risk factors which influence the bone conduction threshold in these patients can help the clinician in counselling the patients for surgery at the earliest to achieve optimum hearing outcomes. The present study was undertaken to determine the association between COM-mucosal disease and cochlear dysfunction. The study also aimed at identifying the determinants and their influence on the bone conduction thresholds of the diseased ear. In this study, 72 pat

An Explainable Vision Transformer Model Based White Blood Cells Classification and Localization.

White blood cells (WBCs) are crucial components of the immune system that play a vital role in defending the body against infections and diseases. The identification of WBCs subtypes is useful in the detection of various diseases, such as infections, leukemia, and other hematological malignancies. The manual screening of blood films is time-consuming and subjective, leading to inconsistencies and errors. Convolutional neural networks (CNN)-based models can automate such classification processes, but are incapable of capturing long-range dependencies and global context. This paper proposes an e…

Blood-brain barrier endothelial cells in neurodegenerative diseases: Signals from the "barrier".

As blood-brain barrier (BBB) disruption emerges as a common problem in the early stages of neurodegenerative diseases, the crucial roles of barrier-type brain endothelial cells (BECs), the primary part of the BBB, have been reported in the pathophysiology of neurodegenerative diseases. The mechanisms of how early vascular dysfunction contributes to the progress of neurodegeneration are still unclear, and understanding BEC functions is a promising start. Our understanding of the BBB has gone through different stages, from a passive diffusion barrier to a mediator of central-peripheral interacti

DNA Methylation Analysis in Monozygotic Twins Discordant for ALS in Blood Cells.

ALS is a fatal motor neuron disease that displays a broad variety of phenotypes ranging from early fatal courses to slowly progressing and rather benign courses. Such divergence can also be seen in genetic ALS cases with varying phenotypes bearing specific mutations, suggesting epigenetic mechanisms like DNA methylation act as disease modifiers. However, the epigenotype dictated by, in addition to other mechanisms, DNA methylation is also strongly influenced by the individual's genotype. Hence, we performed a DNA methylation study using EPIC arrays on 7 monozygotic (MZ) twin pairs discordant f

Role of red blood cells in clinically relevant bleeding tendencies and complications.

The multiple roles of red blood cells (RBCs) are often neglected as contributors in hemostasis and thrombosis. Proactive opportunities to increase RBC numbers, either acutely or subacutely in the case of iron deficiency, are critical as RBCs are the cellular elements that initiate hemostasis together with platelets and stabilize fibrin and clot structure. RBCs also possess several functional properties to assist hemostasis: releasing platelet agonists, promoting shear force-induced von Willebrand factor unfolding, procoagulant capacity, and binding to fibrin. Additionally, blood clot contracti…
