Hematological and plasma profiles and ticks and tick-borne pathogens in wild Formosan black bears (Ursus thibetanus formosanus).

BACKGROUND: The endangered Formosan black bear (Ursus thibetanus formosanus) is the largest native carnivorous mammal in Taiwan. Diseases, poor management, illegal hunting, and habitat destruction are serious threats to the survival of bear populations. However, studies on the impact of diseases on bear populations are limited. Therefore, this study aimed to establish a database of the hematological and plasma profiles of free-ranging Formosan black bears and investigate the occurrence of ectoparasites, blood parasites, and vector-borne pathogens. METHODS: Formosan black bears were captured i

A glimpse into the world of microRNAs and their putative roles in hard ticks.

Ticks are important blood feeding ectoparasites that transmit pathogens to wildlife, domestic animals, and humans. Hard ticks can feed for several days to weeks, nevertheless they often go undetected. This phenomenon can be explained by a tick's ability to release analgesics, immunosuppressives, anticoagulants, and vasodilators within their saliva. Several studies have identified extracellular vesicles (EVs) as carriers of some of these effector molecules. Further, EVs, and their contents, enhance pathogen transmission, modulate immune responses, and delay wound healing. EVs are double lipid-m

Babesia and red cell invasion.

PURPOSE OF REVIEW: Babesiosis is a zoonosis, a disease communicable from animals to humans and an important blood-borne human parasitic infection. Despite its public health impact, its study has largely been neglected. The objective of this review is to present up-to-date information on both parasite and red blood cell molecules that function at the host-parasite interface to facilitate successful invasion. RECENT FINDINGS: In the last few years, a number of parasite proteins have been identified from genome projects and from functional red cell-binding assays. However, their cognate receptor

Dogs as New Hosts for the Emerging Zoonotic Pathogen Anaplasma capra in China.

Anaplasma capra is an emerging zoonotic tick-borne pathogen with a broad host range, including many mammals. Dogs have close physical interactions with humans and regular contact with the external environment. Moreover, they have been previously reported to be hosts of Anaplasma phagocytophilum, A. platys, A. ovis, and A. bovis. To confirm whether dogs are also hosts of A. capra, pathogen DNA was extracted from blood samples of 521 dogs, followed by PCR amplification of the citrate synthase (gltA) gene, heat shock protein (groEL) gene, and major surface protein 4 (msp4) gene of the A. capra.

Studying Group B Streptococcus-Blood-Brain Barrier Interaction In Vitro Using an Induced Pluripotent Stem-Cell-Derived Brain-Like Endothelial Cell Model.

Group B Streptococcus (GBS) is a human pathogen and a leading cause of neonatal meningitis, pneumonia, and sepsis in newborns and adults. Group B Streptococcal meningitis is caused when blood-borne GBS interacts with and disrupts the blood-brain barrier, causing inflammation. The blood-brain barrier (BBB) is comprised of highly specialized brain endothelial cells (BECs) that serve as the interface between the circulation and the central nervous system (CNS), creating a tight endothelial cell barrier. Modeling the BBB using an induced pluripotent stem-cell (iPSC)-derived brain-like endothelial

Cecropin anisaxin-2S has in vitro immunomodulatory, but not antiproliferative and antiviral properties.

Helminthic host defense peptides (HDP) are pleiotropic, multifunctional effector molecules of helminth immunity, efficient against Gram-negative and Gram-positive bacteria. Among them, anisaxin-2S (A-2S), membranolytic cecropin-like HDPs produced by the zoonotic nematodes of the genus Anisakis, shows remarkable efficacy even against multidrug-resistant Gram-negative bacteria, yet its immunomodulatory, antiproliferative and antiviral properties have not been elucidated. Therefore, we tested A-2S immunomodulation in the common carp (Cyprinus carpio) blood cells exposed to two pathogens, the zoon

IxsS7: A novel biomarker for Ixodes scapularis tick bite exposure in humans.

Ixodes scapularis is a primary vector of several important tick-borne pathogens including Borrelia burgdorferi sensu lato, the causative bacterial genospecies complex of Lyme disease, Babesia microti, Anaplasma phagocytophilum, Borrelia miyamotoi, Ehrlichia muris eauclarensis, and Powassan virus. Salivary compounds secreted by I. scapularis during blood feeding are immunogenic and can elicit robust antibody responses in humans which can potentially be leveraged as surrogate markers of prior tick bite exposure. In this study, we investigate the potential of a tick secreted salivary serine prote

Surveillance of Ticks and Tick-Borne Pathogens in Suburban Natural Habitats of Central Maryland.

Lyme and other tick-borne diseases are increasing in the eastern United States and there is a lack of research on integrated strategies to control tick vectors. Here we present results of a study on tick-borne pathogens detected from tick vectors and rodent reservoirs from an ongoing 5-yr tick suppression study in the Lyme disease-endemic state of Maryland, where human-biting tick species, including Ixodes scapularis Say (Acari: Ixodidae) (the primary vector of Lyme disease spirochetes), are abundant. During the 2017 tick season, we collected 207 questing ticks and 602 ticks recovered from 327

Selective preservation of bone marrow mature recirculating but not marginal zone B cells in murine models of chronic inflammation.

Inflammation promotes granulopoiesis over B lymphopoiesis in the bone marrow (BM). We studied B cell homeostasis in two murine models of T cell mediated chronic inflammation, namely calreticulin-deficient fetal liver chimeras (FLC), which develop severe blepharitis and alopecia due to T cell hyper responsiveness, and inflammatory bowel disease (IBD) caused by injection of CD4(+) naïve T cells into lymphopenic mice. We show herein that despite the severe depletion of B cell progenitors during chronic, peripheral T cell-mediated inflammation, the population of BM mature recirculating B cells is

Differential transcriptome response of blood brain barrier spheroids to neuroinvasive Neisseria and Borrelia.

BACKGROUND: The blood-brain barrier (BBB), a highly regulated interface between the blood and the brain, prevents blood-borne substances and pathogens from entering the CNS. Nevertheless, pathogens like Neisseria meningitidis and Borrelia bavariensis can breach the BBB and infect the brain parenchyma. The self-assembling BBB-spheroids can simulate the cross talk occurring between the cells of the barrier and neuroinvasive pathogens. METHODS: BBB spheroids were generated by co-culturing human brain microvascular endothelial cells (hBMECs), pericytes and astrocytes. The BBB attributes of sphero

Zone-specific hepatocytes orchestrate the early onset of host immune defenses during Staphylococcus aureus bloodstream infection.

BACKGROUND: Bloodstream infections (BSI) are life-threatening conditions initiated by pathogens entering the circulation, with Staphylococcus aureus among the most lethal causative agents. The liver is the primary site for the initial detection and clearance of blood-borne pathogens. Emerging evidence indicates that the hepatic immune response to pathological insults is influenced by liver zonation, exhibiting both spatial and cellular specialization. Because direct experimental evidence linking liver zonation to the immune response to BSI is currently lacking, this study aimed to characterize…

Detection of Tick-Borne Pathogens in Lambs Undergoing Prophylactic Treatment Against Ticks on Two Swedish Farms.

Tick-borne pathogens (TBPs), especially Anaplasma phagocytophilum, cause disease in grazing livestock. Tick prophylaxis is, therefore, a routine practice in sheep flocks in Sweden, especially in central, southern, and coastal areas of the country where ixodid ticks (Ixodes ricinus and Haemaphysalis punctata) are present. In the present study, the status of infection by A. phagocytophilum and other TBPs in lambs treated with tick prophylaxis has been assessed serologically and with polymerase chain reaction (PCR). Blood samples (n = 78) from lambs (n = 20) subjected to regular tick prophylactic

Efficient detection of symptomatic and asymptomatic patient samples for Babesia microti and Borrelia burgdorferi infection by multiplex qPCR.

BACKGROUND: Tick-borne infections have been increasing steadily over the years, with co-infections with Borrelia burgdorferi and Babesia microti/divergens emerging as a serious health problem. B. burgdorferi is a spirochetal bacterium that causes Lyme disease while protozoan pathogens belonging to Babesia species are responsible for babesiosis. Currently used serological tests do not always detect acute Lyme disease or babesiosis, and fail to differentiate cured patients from those who get re-infected. This is a major problem for proper diagnosis particularly in regions endemic for tick-borne

Vector-borne pathogens in cats and associated fleas in southern Ethiopia.

BACKGROUND: The worldwide increment of cat populations has increased the risk of ectoparasite infestation and feline vector-borne pathogen (VBP) transmission. In low-income countries, such as Ethiopia, favorable climatic conditions and the absence of preventive measures against ectoparasites contribute to broadening VBP circulation. This study aimed to investigate the prevalence of protozoal (i.e., Hepatozoon, Babesia, and Cytauxzoon species) and bacterial (i.e., Anaplasma, Ehrlichia, Rickettsia, and Bartonella species) infections in owned cats and in their ectoparasites in southern Ethiopia.
