The craniocervical junction (CCJ) forms the bridge between the skull and the spine, a highly mobile group of joints that allows the mobility of the head in every direction. The CCJ plays a major role in protecting the inferior brainstem (bulb) and spinal cord, therefore also requiring some stability. Children are subjected to multiple constitutive or acquired diseases involving the CCJ: primary bone diseases such as in FGFR-related craniosynostoses or acquired conditions such as congenital torticollis, cervical spine luxation, and neurological disorders. To design efficient treatment plans, it

A study of the scientific literature on the topographical and anatomical features of the mandibular canal (canalis mandibulae) has been conducted with the priority of searching for classical dissection anatomical experiments. Scientific achievements of recent years have been identified: the microstructure of bone walls throughout the canal has been studied, the intracanal functional organization of the neurovascular bundle has been studied, variants of the position of the inferior alveolar artery in relation to the inferior alveolar nerve have been studied, and the microcirculatory bed has bee

Piezo1 and Piezo2 are recently discovered mechanosensory ion channels. Piezo channels transduce mechanical stimulation into cellular signaling in a variety of tissues and organ systems. The functional roles of Piezo1 and Piezo2 have been revealed in both developmental and physiological scenarios by using mouse genetic models. Mechanotransduction by Piezo1 channels regulates osteoblast/osteocyte activity and, thus, strengthens the skeleton enabling it to adapt to a wide range of mechanical loadings. Deletion of the Piezo1 gene in the developing skeleton causes bone malformations that lead to sp

Numerous publications and research studies on isoflavones have prompted a nationwide increase in the consumption of soy-based foods and supplements in the United States. Isoflavones are natural endocrine active compounds generally considered to promote health and prevent or slow the onset of certain chronic diseases such as osteoporosis. The beneficial effects of soy isoflavones on bone may, however, be life-stage specific and dependent on the estrogen receptor number and endogenous hormone milieu. Perimenopausal and early menopausal women may therefore be more receptive to the therapeutic eff

The development of the hyoid bone is a complex process that involves the coordination of multiple signaling pathways. Previous studies have demonstrated that disruption of the hedgehog pathway in mice results in a series of structural malformations. However, the specific role and critical period of the hedgehog pathway in the early development of the hyoid bone have not been thoroughly characterized. In this study, we treated pregnant ICR mice with the hedgehog pathway inhibitor vismodegib by oral gavage in order to establish a model of hyoid bone dysplasia. Our results indicate that administr

Lower extremity bowing is a normal physiologic process that commonly occurs in walking children under the age of 2 years. After this age, any significant amount of residual bowing is abnormal and may be due to infections, traumatic physeal injuries, genetic predisposition, metabolic factors, or a combination of these. Of the pathologic conditions, infantile Blount's disease is the most common and is predominantly found in obese children with an early onset of walking. Because of its frequency in occurrence and general tendency for spontaneous resolution, physiologic genu varum must be clearly

We report a 5-year, 9-month-old boy with bilateral patellar hypoplasia. The radiographic skeletal survey revealed absent ossification of the ischium and inferior pubic ramus on both sides, characteristic of the newly recognized benign bone dysplasia known as small patella syndrome, or ischiopatellar dysplasia. No nail change, iliac horn, elbow anomaly, or renal disease is associated with this condition.

Tarsal coalition was noted in 18 cases of rigid equinovarus deformity. Sixteen cases were encountered at surgery and two at morbid dissection. There were 14 patients in the series; six had associated pathologic conditions that might have caused their clubfeet to be deemed "teratologic," whereas eight did not and were considered to have congenital clubfeet. Four patients in the series had bilateral coalitions. Preoperative radiographs demonstrated the coalition in only one case. A presurgical magnetic resonance image (MRI) clearly showed the coalition in another case. Nonoperative treatment was

The skeletal dysplasias are a unique and heterogeneous group of disorders with skeletal manifestations including deformity, disproportion, and short stature. The purpose of this article is to describe the use of the Ilizarov technique in the management of these orthopedic problems in 15 patients with skeletal dysplasias. The technique is particularly appropriate in those patients requiring correction of angular deformity and limb lengthening. The greatest number of complications were encountered in those patients undergoing extended limb lengthening. Nevertheless, all patients achieved their p

Two cases of the latent form of Asphyxiating Thoracic Dystrophy or Asphyxiating Thoracic Dystrophy without respiratory disease or respiratory problems are reported. The skeletal survey was diagnostic in both of them. In the first case it was performed after the pelvic film, ordered because of externally rotated thighs, showed triradiate acetabulum. In the second case the skeletal survey was requested because of undiagnosed mental retardation syndrome.

Genetic skeletal disorders (GSDs) are a heterogeneous group characterized by an intrinsic abnormality in growth and (re-)modeling of cartilage and bone. A large subgroup of GSDs has additional involvement of other structures/organs beside the skeleton, such as the central nervous system (CNS). CNS abnormalities have an important role in long-term prognosis of children with GSDs and should consequently not be missed. Sensitive and specific identification of CNS lesions while evaluating a child with a GSD requires a detailed knowledge of the possible associated CNS abnormalities. Here, we provid

The author reports the first association of central areolar choroidal dystrophy in three brothers with pseudoachondroplastic spondyloepiphyseal dysplasia. Both central areolar choroidal dystrophy and pseudoachondroplastic spondyloepiphyseal dysplasia represent heterogeneous disorders with autosomal dominant transmission. It is hypothesized that this association is a result of a new subtype involving both the skeletal system and the eye.

BACKGROUND: Trochlear dysplasia is the leading risk factor for patellofemoral instability, but its development in humans is poorly understood. Animal models provide an avenue for improving our understanding of this condition, yet they face several limitations in replicating human anatomy. This systematic review sought to evaluate existing animal models of trochlear dysplasia, highlighting their effectiveness, limitations, and translational relevance.
METHODS: A systematic review was conducted following PRISMA guidelines. PubMed, Embase, and Scopus were searched for studies that induced trochl

Geleophysic dysplasia (GD) is a rare genetic disorder characterized by severe cardiorespiratory dysfunction and poor prognosis. With no cure available, current treatments focus on symptomatic management. Using a cellular model of ADAMTSL2 p.A165T variant, our screening of 2,321 FDA-approved drugs identified several glucocorticoids, particularly betamethasone dipropionate (BMD), which significantly enhance secretion of two crucial proteins for GD, ADAMTSL2 and FBN1 while improving extracellular matrix organization. In a mouse model carrying the Adamtsl2 p.A165T variant, we observed a high early

The knee joint is the one of the most common locations for congenital and developmental musculoskeletal abnormalities. Initial imaging of the knee joint should always begin with conventional radiographs. However, evaluation of the bone marrow, cartilaginous, ligamentous, and other soft-tissue components of the knee joint are better characterized with magnetic resonance imaging (MRI). We present the MRI findings of prevalent congenital and developmental abnormalities in the paediatric knee with particular emphasis on the components of the growing skeleton.

Rabbits obtained from a South African rabbit breeder exhibited deformities of the distal forelimbs. The clinical, radiological and histological presentation of mid-antebrachial valgus formation (a.k.a distal foreleg curvature) in these rabbits was consistent with an autosomal recessive trait associated with a chondrodystrophic lesion of the distal ulna epiphysis 1st described in the 1960s. The impact this might have on South African farming enterprises and laboratory facilities has not been established, but the heritability and welfare implications of this condition make it a concern. Mildly a
