Thalidomide: a novel template for anticancer drugs.

Thalidomide is an immunomodulatory agent that is approved for use in patients with cutaneous manifestations of erythema nodosum leprosum (ENL). It is currently under clinical investigation in a wide range of malignancies and immunologic-based disorders. Structural analogs of thalidomide have been synthesized in order to explore potential molecular targets of thalidomide, as well as to identify new therapeutics. Members of one class of analogs, termed immunomodulatory drugs (IMiD), are 100 to 1,000 times more potent than thalidomide in regulating cytokine production by peripheral blood mononucl

Naltrexone prevents ethanol-induced changes in rat thymus.

Ethanol is known to suppress the immune response, but the underlying mechanism accounting for the immunosuppression is not clearly elucidated yet. The purpose of this study was to investigate the effect of a single dose of ethanol on relative proportion of the four major rat thymocyte subsets and possible mechanism of its action. To this end, adult female AO rats were treated with: a) ethanol (2 or 4 g/kg, i.p.), b) naltrexone (5 mg/kg, i.p.) followed 45 min later by ethanol (2 or 4 g/kg, i.p.), c) naltrexone (5 mg/kg, i.p.), or d) only saline. Twenty hours later the rats were sacrificed and t

Coexistence of human immunodeficiency virus infection and systemic lupus erythematosus.

A variety of rheumatic disorders has been reported during the course of human immunodeficiency virus (HIV) infection; however, its association with systemic lupus erythematosus (SLE) is extremely rare. The effect of HIV infection on CD4 lymphocytes may ameliorate SLE activity and spur remission. We observed 2 patients with SLE who later developed HIV infection. Both patients' lupus went into remission, and their course was complicated by aseptic necrosis of the bone. These findings suggest that HIV infection may have an important effect on the natural course of SLE and may also have a pathogen

Immune cell signatures and preeclampsia: Unveiling causal links through genome-wide association studies.

The immune-placental interaction could potentially contribute both causally and therapeutically to preeclampsia (PE). However, previous observational studies have not demonstrated a causal association between these 2 factors. Exploring the causal relationship between immune cells and PE provides genetic evidence that immune cells play a role in PE risk. Using a large dataset of genome-wide association studies, we conducted a bidirectional Mendelian randomization (MR) analysis to determine whether 731 immune cell signatures were correlated with PE, which included 4 types of immune signatures. A

Determination of lymphocytes surface markers in patients with thermal burns and the influence of burn size on mononuclear cell subsets.

Background: Thermal burn injuries impair the host defence system. Hence, in the present study, we aimed at investigating the changes in the number and phenotype of peripheral blood lymphocyte populations (T, B, and natural killer cells) and their subpopulations in patients with thermal burns and determining the relationships with different sizes of total body surface area (TBSA). Methods: Blood samples from 67 patients, admitted to Motahary Burn Center in Tehran, with burns from 30% to more than 70% TBSA were collected on Days 3 and 7 postburn. Lymphocytes and their subpopulations were identif

Aberrant expression of p-STAT3 in peripheral blood CD4+ and CD8+ T cells related to hepatocellular carcinoma development.

Hepatocellular carcinoma (HCC) is one of the most common cancer types worldwide. The signal transducer and activator of transcription 3 (STAT3) protein is a member of the STAT transcription factor family. Oncogenesis, invasion, and metastasis of HCC are associated with activation of STAT3. However, whether aberrant expression of phosphorylated STAT3 (p-STAT3) in peripheral CD4+ and CD8+ T cells relates to HCC pathogenesis remains unclear. In this study, the expression of p-STAT3 in CD4+ and CD8+ T cells, and the levels of interferon-γ (IFN-γ), interleukin-4 (IL-4), IL-6 and IL-10 in the human

Phenotypic and functional dysregulations of CD8 + T Cells in myasthenia gravis.

Myasthenia Gravis (MG) is a heterogeneous autoimmune disorder characterized by fluctuating muscle weakness caused by autoantibodies targeting neuromuscular junction components. While the role of CD4 + T cells in MG is well established, the contribution of CD8 + T cells remains poorly understood. In this study, we analyze CD8 + T cells in 36 MG patients and 38 age- and gender-matched controls using flow cytometry to evaluate subset distribution, granzyme expression, and cytokine production. MG patients exhibit an altered CD4 + /CD8 + T cell ratio and significant changes in CD8 + T cell subsets,

Expression of vascular endothelial growth factor, apoptosis inhibitors (survivin and p16) and CCL27 in alopecia areata before and after diphencyprone treatment: an immunohistochemical study.

BACKGROUND: Alopecia areata (AA) is a relatively common inflammatory form of nonscarring hair loss of unknown pathogenesis, but possibly of autoimmune origin. Topical immunotherapy, using a potent contact allergen such as diphencyprone (DPC), is currently considered the most effective mode of treatment. However, the way in which DPC operates on hair follicles in AA still remains to be elucidated. Vascular endothelial growth factor (VEGF), essential for angiogenesis and vascular permeability, may be responsible for maintaining proper vasculature around hair follicles, and several studies provid

Modeling of ex vivo immune response reveals a central role for CD8+CD25+ cells in Adult T-cell Leukemia survival: a long-term prospective cohort study.

Adult T-cell leukemia (ATL) is a rare but aggressive CD4+CD25+ leukemia triggered by HTLV-1 infection. We quantified ex vivo levels of CD4+ and CD8+ (sub)populations and their antiproliferative, pro-apoptotic, antiviral and immunomodulatory interplay in short-term culture of primary cells from ATL patients, in a long-term prospective study (819 person-years of follow-up). We integrated clinical, cellular and molecular data into a data mining approach combining linear (consensus HIerarchical Tree clustering) and non-linear (BAYesian network) models. This HIT-BAY approach revealed an association

Circulating regulatory T cells from breast cancer patients in response to neoadjuvant chemotherapy.

BACKGROUND: Immune escape of tumor cells is a new hallmark of cancer in general, and breast cancer, in particular. Previous studies have demonstrated that the immunological profile in peripheral blood may be a prognostic and/or predictive biomarker in breast cancer. Thus, higher number of regulatory T cells (Tregs) in blood from patients with breast cancer has been reported in relation to normal donors. In the present study, we planned to evaluate the changes in different cell populations in peripheral blood: neutrophils, monocytes and lymphocytes, as well as lymphocyte subpopulations [natural
