The extracellular calcium-sensing receptor promotes porcine egg activation via calcium/calmodulin-dependent protein kinase II.

Extracellular calcium is required for intracellular Ca2+ oscillations needed for egg activation, but the regulatory mechanism is still poorly understood. The present study was designed to demonstrate the function of calcium-sensing receptor (CASR), which could recognize extracellular calcium as first messenger, during porcine egg activation. CASR expression was markedly upregulated following egg activation. Functionally, the addition of CASR agonist NPS R-568 significantly enhanced pronuclear formation rate, while supplementation of CASR antagonist NPS2390 compromised egg activation. There was…

Calcium phosphate precipitation in experimental gaps between fluoride-containing fast-setting calcium silicate cement and dentin.

A novel fast-setting calcium silicate cement containing fluoride (novel-CSC) has been developed for applications in tooth crowns. The aim of this study was to assess the ability of the novel-CSC to close the experimental gaps at the dentin-cement interface. The novel-CSC was tested against Vitrebond and GC Fuji II LC. Experimental gaps of 50 or 300 μm width were created between the materials and dentin. Specimens with the 300-μm-wide gap were immersed in phosphate-buffered saline and the closed gap area was measured during 96 h. All specimens with 50 or 300 μm gap width were analyzed by scanni…

Inulin and fructooligosaccharide affect in vitro calcium uptake and absorption from calcium-enriched gluten-free bread.

Compromised intestinal calcium absorption affecting a deterioration of bone state is a sign of coeliac disease. Experimental calcium-fortified gluten-free bread (GFB) of improved calcium bioavailability could increase calcium content in the diets of coeliac disease patients, allowing them to obtain the amount of calcium they need for therapeutic use. Prebiotics, including inulin-type fructans (IFs) have a beneficial effect on calcium bioavailability. In the present study, the in vitro model composed of the intestinal-like Caco-2 cells and the human intestinal bacteria (Lactobacillus, Enterococ…

Calcium Overload Accelerates Phosphate-Induced Vascular Calcification Via Pit-1, but not the Calcium-Sensing Receptor.

AIM: Vascular calcification (VC) is a risk factor of cardiovascular and all-cause mortality in patients with chronic kidney disease (CKD). CKD-mineral and bone metabolism disorder is an important problem in patients with renal failure. Abnormal levels of serum phosphate and calcium affect CKD-mineral and bone metabolism disorder and contribute to bone disease, VC, and cardiovascular disease. Hypercalcemia is a contributing factor in progression of VC in patients with CKD. However, the mechanisms of how calcium promotes intracellular calcification are still unclear. This study aimed to examine …

Antagonists of the TMEM16A calcium-activated chloride channel modulate airway smooth muscle tone and intracellular calcium.

BACKGROUND: Perioperative bronchospasm refractory to β agonists continues to challenge anesthesiologists and intensivists. The TMEM16A calcium-activated chloride channel modulates airway smooth muscle (ASM) contraction. The authors hypothesized that TMEM16A antagonists would relax ASM contraction by modulating membrane potential and calcium flux. METHODS: Human ASM, guinea pig tracheal rings, or mouse peripheral airways were contracted with acetylcholine or leukotriene D4 and then treated with the TMEM16A antagonists: benzbromarone, T16Ainh-A01, N-((4-methoxy)-2-naphthyl)-5-nitroanthranilic a…

Optogenetic measurement of presynaptic calcium transients using conditional genetically encoded calcium indicator expression in dopaminergic neurons.

Calcium triggers dopamine release from presynaptic terminals of midbrain dopaminergic (mDA) neurons in the striatum. However, calcium transients within mDA axons and axon terminals are difficult to study and little is known about how they are regulated. Here we use a newly-developed method to measure presynaptic calcium transients (PreCaTs) in axons and terminals of mDA neurons with a genetically encoded calcium indicator (GECI) GCaMP3 expressed in transgenic mice. Using a photomultiplier tube-based system, we measured electrical stimulation-induced PreCaTs of mDA neurons in dorsolateral stria…

Evaluation of calcium-releasing and apatite-forming abilities of fast-setting calcium silicate-based endodontic materials.

AIM: To evaluate two fast-setting calcium silicate-based endodontic materials (Endocem mineral trioxide aggregate (MTA) and Endocem Zr) with regard to their ability to release calcium ions (Ca(2+)) and produce apatite-like precipitates after immersion in phosphate-buffered saline (PBS). METHODOLOGY: Endocem MTA, Endocem Zr and white ProRoot MTA (WMTA) were used. Chemical composition of the powder of each material was analysed with a wavelength-dispersive X-ray spectroscopy electron probe microanalyser with image observation function (SEM-EPMA). The amount of Ca(2+) released from water-immerse…

Impaired body calcium metabolism with low bone density and compensatory colonic calcium absorption in cecectomized rats.

An earlier study reported that cecal calcium absorption contributes less than 10% of total calcium absorbed by the intestine, although the cecum has the highest calcium transport rate compared with other intestinal segments. Thus, the physiological significance of the cecum pertaining to body calcium metabolism remains elusive. Herein, a 4-wk calcium balance study in cecectomized rats revealed an increase in fecal calcium loss with marked decreases in fractional calcium absorption and urinary calcium excretion only in the early days post-operation, suggesting the presence of a compensatory mec…

Mitochondrial calcium uptake regulates rapid calcium transients in skeletal muscle during excitation-contraction (E-C) coupling.

Defective coupling between sarcoplasmic reticulum and mitochondria during control of intracellular Ca(2+) signaling has been implicated in the progression of neuromuscular diseases. Our previous study showed that skeletal muscles derived from an amyotrophic lateral sclerosis (ALS) mouse model displayed segmental loss of mitochondrial function that was coupled with elevated and uncontrolled sarcoplasmic reticulum Ca(2+) release activity. The localized mitochondrial defect in the ALS muscle allows for examination of the mitochondrial contribution to Ca(2+) removal during excitation-contraction c

L-type calcium channels govern calcium signaling in migrating newborn neurons in the postnatal olfactory bulb.

Newborn inhibitory neurons migrate into existing neural circuitry in the olfactory bulb throughout the lifetime of adult mammals. While many factors contribute to the maturation of neural circuits, intracellular calcium is believed to play an important role in regulating cell migration and the development of neural systems. However, the factors underlying calcium signaling within newborn neurons in the postnatal olfactory bulb are not well understood. Here, we show that migrating, immature neurons in the olfactory bulb subependymal layer (SEL) undergo spontaneous and depolarization-evoked intr…

Amyotrophic lateral sclerosis immunoglobulins are ineffective in altering calcium influx through presynaptic voltage-sensitive calcium channels.

Recent work has suggested that one factor in the etiology of the neuromuscular disease, amyotrophic lateral sclerosis (ALS), may be an autoimmune mechanism in which presynaptic voltage-sensitive calcium channels are an antigenic target. We have developed a fluorescence technique to measure rapid Ca2+ influx through presynaptic calcium channels in isolated nerve terminals (synaptosomes) from rat cerebral cortex. Depolarization of the synaptosomes by elevated external K+ concentration caused a rapid increase in cytoplasmic Ca2+, as measured by a change in fluorescence of the Ca2+ chelating dye, …

Calcium binding fraction detected from the brush border membrane during intestinal calcium absorption in the rat.

A fraction which had calcium binding activity was detected from the brush border membrane during intestinal calcium absorption in the rat. In situ duodenal loop experiment showed that 45Ca activity in mucosal homogenate, detergent-solubilized whole particulate fraction and detergent-solubilized brush border membrane was maximal at 30 min after the radioactive calcium solution was injected in the loop. The calcium radioactivities did not co-precipitate with anti-IMCal (integral membrane calcium binding protein) antibody. Gel permeation HPLC showed three calcium peaks from both solubilized whole…

Sodium/calcium exchange and intracellular calcium buffering in ferret myocardium: an ion-sensitive micro-electrode study.

Measurements of the intracellular activity of Ca (aiCa), Na (aiNa) and H (pHi) ions have been made with resin-filled ion-sensitive micro-electrodes in ferret ventricular trabeculae. The mean values in quiescent muscle at 30 degrees C were: aiNa, 11.1 +/- 1.0 mM; aiCa, 58.4 +/- 6.4 nM, and pHi, 7.20 +/- 0.11. The relation between aiNa and extracellular Na activity (aoNa) is not linear and is sensitive to temperature: the Q10 for the change in aiNa in normal Tyrode solution is 1.3 +/- 0.5 and rises to 3.5 +/- 0.5 when aoNa is reduced to 1.1 mM. The addition of CN to the bathing fluid causes litt

Calcium signaling in chemotherapy-induced neuropathy.

Alterations in calcium (Ca2+) signaling is a major mechanism in the development of chemotherapy-induced peripheral neuropathy (CIPN), a side effect caused by multiple chemotherapy regimens. CIPN is associated with numbness and incessant tingling in hands and feet which diminishes quality of life during treatment. In up to 50% of survivors, CIPN is essentially irreversible. There are no approved, disease-modifying treatments for CIPN. The only recourse for oncologists is to modify the chemotherapy dose, a situation that can compromise optimal chemotherapy and impact patient outcomes. Here we fo…
