Effect of dietary arginine on the immune response and gene expression in head kidney and spleen following infection of Jian carp with Aeromonas hydrophila.

This study was conducted to test the hypothesis that elevated dietary arginine enhances immunity of juvenile Jian carp (Cyprinus carpio var. Jian). Fish were fed graded levels of dietary arginine for 9 weeks. Subsequently, a challenge test was conducted by injection of Aeromonas hydrophila. Head kidney and spleen weights, as well as erythrocyte and leukocyte counts were significantly influenced by dietary arginine levels. A similar trend was also observed for hemagglutination titre, serum lysozyme activity, IgM concentration, C3 and C4 content. The highest survival rates following A. hydrophil

Structurally conserved C-RFa revealed prolactin releasing activity in vitro and gene expression changes in pituitary of seasonally acclimatized carp.

Here we show the cloning and characterization of a novel homolog of prepro C-RFa cDNA from Cyprinus carpio. The deduced preprohormone precursor of 115 amino acids leads to a mature bioactive peptide of 20 amino acids with identical sequence to other teleost C-RFa. Modeling of the mature C-RFa peptide highlighted significant similarity to homologous human PrRP20, specifically the conserved amphipathic system defined by the C-terminal alpha-helix. Clearly, the synthetic C-RFa peptide stimulated prolactin release from primary cultured fish pituitary cells. For the first time, significant variatio

Signal transduction mechanisms for autocrine/paracrine regulation of somatolactin-α secretion and synthesis in carp pituitary cells by somatolactin-α and -β.

Pituitary hormones can act locally via autocrine/paracrine mechanisms to modulate pituitary functions, which represents an interesting aspect of pituitary regulation other than the traditional hypothalamic input and feedback signals from the periphery. Somatolactin, a member of the growth hormone (GH)/prolactin (PL) family, is a pleiotropic hormone with diverse functions, but its pituitary actions are still unknown. Recently, two SL isoforms, SLα and SLβ, have been cloned in grass carp. Based on the sequences obtained, recombinant proteins of carp SLα and SLβ with similar bioactivity in induci

Bioaccumulation and metabolism of polybrominated diphenyl ethers in carp (Cyprinus carpio) in a water/sediment microcosm: important role of particulate matter exposure.

Microcosms were built up to simulate a pond system with polybrominated diphenyl ether (PBDE) contaminated sediment and bioorganisms. The microcosms were divided into groups A and B. In group A, both benthic invertebrates (tubificid worms) and carp (Cyprinu carpio) were added, while in group B, only fish were added. After exposure for 20 d, the fish were sampled (exposure I). A net was fixed in the microcosms, and new fish were added (exposure II). These fish were prohibited from contacting the sediment by the net, and the accumulation and depuration of PBDEs in the fish were investigated. Amon

Regulation of dopamine D2 receptor expression in grass carp pituitary cells: a possible mechanism for dopaminergic modification of luteinizing hormone synthesis.

In many fishes, dopamine (DA) strongly inhibits luteinizing hormone (LH) release by direct action at the pituitary level. In this study, the effect of DA on LH synthesis was examined by detecting its β-subunit mRNA level in immature grass carp pituitary cells. Results showed that DA inhibited LHβ mRNA expression and its inhibition was antagonized by a DA D2 receptor (DRD2) antagonist, sulpiride, suggesting that DA inhibited LH synthesis via DRD2. This notion was further supported by the finding that the grass carp DRD2 (gcDRD2) immunoreactivity was observed in the proximal pars distalis of the

Evaluation of internal control genes for qRT-PCR normalization in tissues and cell culture for antiviral studies of grass carp (Ctenopharyngodon idella).

Real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) has become one of the most commonly used techniques for RNA expression. To obtain more reliable results with biological significance, it requires data normalization using an appropriate internal control gene. Here, we cloned partial sequence of elongation factor 1α (EF1α) gene from grass carp (Ctenopharyngodon idella). The stabilities of four commonly used internal control genes encoding 18S rRNA, β-actin, EF1α, and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) were integratedly assessed using the geNorm, NormF

Consequent effects of the great cormorant (Phalacrocorax carbo sinensis) predation on parasite infection and body condition of common carp (Cyprinus carpio).

Lesions ranging from surface wounds to deep tissue wounds caused by cormorant predation were observed on several species of the farmed fish in Pohořelice, Czech Republic. Two-year-old stocked common carp Cyprinus carpio harvested in late March were examined for ectoparasites and endoparasites, injuries extent, and lysozyme concentration in skin mucus. Additionally, three body condition indices were measured. Endoparasite infection occurred only scarcely. Wounded fish were more susceptible to the ectoparasites Gyrodactylus spp. and Dactylogyrus spp. (Monogenea), and Ichthyophthirius multifiliis

The haemoflagellate Trypanoplasma borreli induces the production of nitric oxide, which is associated with modulation of carp (Cyprinus carpio L.) leucocyte functions.

In an attempt to characterise the role of nitric oxide (NO) in immune responses of carp, carp leucocytes obtained during an acute T. borreli infection were examined, for their capacity to generate NO. In a second set of experiments the impact NO on viability of the parasite and on the modulation of functional carp leucocyte responses were tested in vitro. Both in carp head-kidneys and in the peripheral blood, the fractions of lymphoblasts among separated leucocytes were increased. However, the relative proportions of granulocytes among head-kidney leucocytes (HKL) significantly decreased durin

Modulatory effect of metal ions on the immune response of fish: in vivo and in vitro influence of MnCl2 on NK activity of carp pronephros cells.

The in vivo and in vitro influence of MnCl2 on carp pronephros cells was investigated. Increased cytotoxicity against both YAC-1 and P 815 target cells was observed following an intraperitoneal injection of 40, 80, or 120 micrograms MnCl2/g body wt administrated 24 hr prior to the in vitro 51Cr release assay. Similarly, in vitro treatment of carp pronephros cells, at a final concentration of 60 micrograms/culture, resulted in an increase of NK cell activity in both YAC-1 and P 815 target cell lines. However, a significant decrease in this activity was shown with lower doses of MnCl2 (40 and 20

Combined toxic effects of polyethylene microplastics (PE-MPs) and 4-nonylphenol on the common carp (Cyprinus carpio) juvenile: multi-biomarker approach.

There is inadequate knowledge of the cumulative impact of exposure to 4-nonylphenol (4-NP) and polyethylene microplastics (PE-MPs), and in particular of whether their joint action departs from additivity. The objective of this study was to determine and formally compare the single and combined effects of 4-NP and PE-MPs on multiple biomarkers in juvenile Cyprinus carpio. Fish were assigned to a fully crossed 2 × 2 factorial design comprising a control group and three treated groups receiving 10 mg/L PE-MPs, 200 µg/L 4-NP, or their binary mixture, for a two-week exposure period followed by a tw

Cloning, expression, tertiary structure, activity characteristics, design of inhibitor, hydrolysis and inhibition mechanisms analysis of carp adipose triglyceride lipase (ATGL).

Lipid hydrolysis catalyzed by endogenous enzymes is a major factor reducing the quality of meat products. Adipose triglyceride lipase (ATGL) initiates triacylglycerol hydrolysis and is therefore a key enzyme in this process. In this study, the N-terminal domain of carp ATGL was cloned. Recombinant ATGL was successfully expressed and purified, and its activity was characterized at different temperatures and pH values. The predicted tertiary structure of ATGL was analyzed. Carp ATGL is a monomeric protein composed of nine α-helices and seven β-strands, with a radius of gyration (Rg) of 1.84 nm.

Comparative analysis of non-volatile flavor compounds in salt-sugar co-cured grass carp fillets utilizing various frying techniques.

This study investigated the effects of traditional frying (TF, 0-9 min), air frying (AF, 0-17 min), and microwave frying (MF, 0-12 min) on non-volatile flavor compounds in grass carp fillets. Prolonged frying time promoted the accumulation of total free amino acids and reducing sugars, with thermal degradation acting as the primary driver of this increase. Histidine, 5'-guanosine monophosphate, and tartaric acid were identified as key flavor contributors, while succinic acid also showed significant contribution in TF and the early AF groups. The equivalent umami concentration exhibited a conti

Computational identification of cathepsin B and L inhibitors and their role in modulating protein degradation and gelation of unwashed grass carp (Ctenopharyngodon idella) mince.

Unwashed fish mince is a sustainable alternative to conventional surimi, however, residual endogenous proteases can degrade myofibrillar proteins and compromise gel quality. In this study, unwashed grass carp (Ctenopharyngodon idella) mince was used as a model system to investigate inhibitory effects and underlying mechanisms of natural compounds on gel deterioration and quality improvement. Based on structural characteristics of cathepsins, molecular docking identified naringin, apigenin, proanthocyanidins and luteolin as potential inhibitors, with proanthocyanidins exhibiting the strongest i
