Down's syndrome in middle age. Topographical distribution and immunoreactivity of brain lesions in an autopsied patient.

A 45-year-old patient with Down's syndrome was autopsied. The brain, weighing 800 g, was small in size, and serial sections revealed generalized gyral atrophy and ventricular dilatation. In the gray matter, there was diffuse neuronal degeneration characterized by numerous neurofibrillary tangles (NFTs), senile plaques and frequent amyloid angiopathy. Histochemical and electron microscopical analyses of these lesions showed no qualitative difference from those in Alzheimer's disease. A topographical study of NFTs showed that they were numerous in the limbic system and cerebral neocortex. Variou

Altered effective connectivity between the prefrontal cortex and caudate in children with overweight and obesity.

OBJECTIVE: This study aimed to examine the interaction patterns between the reward and inhibitory control systems in children with overweight and obesity. METHODS: Resting-state fMRI data were collected at baseline from 38 children with overweight and obesity (OW/OB) and 68 children with normal weight (NW). We first examined the differences in seed-based functional connectivity (FC) between the two groups, focusing on eight predefined regions of interest in the reward and inhibitory control systems. Based on the FC results, we further applied the spectral dynamic causal modeling technique to

A Caudate Pathway-level Transcriptomic Signature of Translational, Bioenergetic, and Proteostatic Programs in Obsessive-compulsive Disorder.

Obsessive-compulsive disorder (OCD) is linked to cortico-striato-thalamo-cortical circuit dysfunction, but molecular programs within striatal circuit nodes remain incompletely characterized. We tested whether translation, mitochondrial bioenergetics, and proteostasis show a coordinated transcriptomic signature in the OCD caudate. We reanalyzed postmortem caudate RNA-sequencing data from GSE262138 after retaining OCD cases and unaffected controls (OCD n = 27; control n = 85). Gene-level models adjusted for RNA integrity number and sex. Ranked gene statistics were used for pathway enrichment, mo

Caudate haemorrhage caused by pseudoaneurysm of accessory middle cerebral artery.

A 68-year-old man experienced a right caudate haemorrhage with intraventricular haemorrhage. Although a subarachnoid haemorrhage was not shown clearly, our investigation demonstrated an aneurysm-like vascular pouch located in the anomalous vessel arising from the A2 segment of the right anterior cerebral artery. Rupture of the vascular pouch was considered to be the cause of the caudate haemorrhage. Neck clipping was performed. In intraoperative observation, the anomalous vessel was diagnosed as a right accessory middle cerebral artery. Histopathology of the saccular wall showed only an advent

Bilateral caudate infarct--a case report.

Caudate strokes comprise only a small proportion of all subtypes of strokes. Bilateral caudate infarcts are even rarer and only a few cases have been reported in the literature. We report an 86-year-old woman with bilateral caudate infarcts. She had no past medical history of note. She presented with headache for several days and drowsiness on day of admission. Clinical examination revealed abulia, inability to comprehend or verbalize (acute mutism), right-sided neglect and right-sided hemiparesis. Computed tomographic (CT) scan brain revealed decreased attenuation in both heads of both caudat

Primary CNS lymphoma: lymphomatous meningitis presenting as a cauda equina lesion in an AIDS patient.

Neurologic complications of the acquired immune deficiency syndrome (AIDS) most often present as brain dysfunction and/or a polyneuropathy. We describe a unique neurological problem of a woman with AIDS who presented with a cauda equina mass due to primary CNS lymphoma. She subsequently developed a fulminant lymphomatous meningitis and died. Although previously rare, primary spinal cord lymphoma is expected to be encountered more often with the current AIDS epidemic and may be difficult to distinguish from infectious mass lesions.

Spasmodic head movements produced by destruction of unilateral ventromedial tegmentum in cats.

In cats, unilateral electrolytic destruction in the dorsomedial parts of the ventral tegmental area (VTA) (Tsai) led to the appearance of torticollic rotatory head postures associated with a marked drop of the corresponding caudate serotonin. Results obtained from fluorescence histochemistry and brain dialysis also revealed that decrease in serotonin shared the leading role. However, since a chemical lesion produced by neurotoxic agents failed to induce such abnormal postures, concomitant interruption of nonaminergic fibers at VMT might also be the essential factor. Among them the interstitial

Caudate hemorrhage.

Eight patients had computed tomographic (CT) evidence of caudate hemorrhage. In all cases, the initial clinical findings were most suggestive of subarachnoid rather than intracerebral hemorrhage. The CSF was bloody and xanthochromic in all cases. In caudate hemorrhage due to ruptured carotid artery aneurysms, the CT scan showed hyperdense lesions contiguous with the anterior frontal horn of the lateral ventricles. In caudate hemorrhage related to hypertension, the CT scan showed a small hematoma that appeared contiguous with the anterior portion of the body of the lateral ventricle, and there

Distribution of striatal afferents from the mesencephalon in the cat.

In a series of cats, various quantities of horseradish peroxidase were injected in different parts of the striatum to map out the extent and the topography of the afferent projections from the mesencephalon. Following large injections in the head of the caudate nucleus and putamen, the majority of cells were retrogradely labeled in the substantia nigra pars compacta (SNc) and in its dorsal extension the retrorubral nucleus or pars dorsalis (SNd), confirming previous reports. Numerous HRP positive neurons were also present in the ventral tegmental area (VTA) and the rostral raphe nuclei, includ…

Neural activity changes correlated with central anticholinergic blockade of cholinergically-induced drinking.

In the rat, microinjections of carbachol into the septal area elicited water ingestion and increased multiple unit activity at this site and also the noninjected lateral hypothalamus. Carbachol injection into the lateral hypothalamus also elicited water ingestion, but multiple unit activity did not increase in this structure, although it did in the noninjected septal area. If carbachol was injected into one of these sites and isotonic saline into the other (conditions comparable to those for which drinking has been previously demonstrated), increased multiple unit activity was still found. Cannabis and tobacco co-use and its association with striatal brain morphometry: Leveraging data from the ENIGMA addiction working group.

BACKGROUND: The striatum plays a central role in the pathophysiology of addiction. Studies report that cannabis use is associated with greater striatal gray matter volume (GMV), while tobacco use is associated with both greater and lower striatal GMV. However, their combined effects on striatal GMV remain unclear. METHODS: Using two MRI processing methods, we investigated associations between these substances and striatal GMV. Men and women from eight ENIGMA Addiction sites (N = 302) were parsed into 4 groups; individuals with: co-use (CT, n = 38); cannabis-only use (CO, n = 34); tobacco-only

Dopaminergic, serotonergic, and noradrenergic deficits in Parkinson disease.

OBJECTIVE: People with Parkinson disease (PD) frequently develop dementia, which is associated with neocortical deposition of alpha-synuclein (α-syn) in Lewy bodies and Lewy neurites. In addition, neuronal loss and deposition of aggregated α-syn also occur in multiple subcortical nuclei that project to neocortical, limbic, and basal ganglia regions. Therefore, we quantified regional deficits in innervation from these PD-affected subcortical nuclei, by measuring the neurotransmitters and neurotransmitter transporter proteins originating from projections of dopaminergic neurons in substantia nig

Endocrine, immune, and neurochemical changes in rats during withdrawal from chronic amphetamine intoxication.

In humans who chronically abuse amphetamine (AMPH), sudden abstinence often precipitates an organic mood disorder that mimics many symptoms … crease in natural killer cell activity. Brain dopamine levels in these animals are decreased in the caudate nucleus but are increased in the nucleus accumbens. AMPH withdrawal in rats may be a useful model for studying the physiologic and neural substrates of human AMPH withdrawal states.

Extracellular Space Barrier Dysfunction Disrupts Interstitial Fluid Drainage and Is Associated with Memory Heterogeneity in Cognitive Aging.

Abnormal cognitive aging is characterized by memory decline beyond normal age-related physiological changes. Nevertheless, the intrinsic mec … nstrated that memory-impaired aged rats exhibited region-specific neurotransmitter imbalance in the caudate nucleus and thalamus, which was closely associated with age-related ECS barrier dysfunction. Such barrier dysfunction induced ISF drainage disturbance, thereby reducing local neurotransmitter concentrations in the caudate nucleus and impairing thalamocortic …
