Histopathological evaluation of the nervous system in National Toxicology Program rodent studies: a modified approach.

This article outlines the changes and underlying rationale for modifications to the histopathological evaluation of the nervous system during toxicology and carcinogenesis studies conducted by the National Toxicology Program (NTP). In the past, routine evaluation of the nervous system was mostly limited to three sections of brain, and occasionally the spinal cord and peripheral nerves. Factors such as the increasing occurrence of human neurological diseases and associated economical cost burden, the role of unidentified environmental stressors in neurodegenerative disorders, multiple therapeut

Evaluation of Marine Brown Algae and Sponges from Brazil as Anticoagulant and Antiplatelet Products.

The ischemic disorders, in which platelet aggregation and blood coagulation are involved, represent a major cause of disability and death worldwide. The antithrombotic therapy has unsatisfactory performance and may produce side effects. So, there is a need to seek molecules with antithrombotic properties. Marine organisms produce substances with different well defined ecological functions. Moreover, some of these molecules also exhibit pharmacological properties such as antiviral, anticancer, antiophidic and anticoagulant properties. The aim of this study was to evaluate, through in vitro test

The respiratory health of swimmers.

Regular physical activity is recognized as an effective health promotion measure. Among various activities, swimming is preferred by a large portion of the population. Although swimming is generally beneficial to a person's overall health, recent data suggest that it may also sometimes have detrimental effects on the respiratory system. Chemicals resulting from the interaction between chlorine and organic matter may be irritating to the respiratory tract and induce upper and lower respiratory symptoms, particularly in children, lifeguards and high-level swimmers. The prevalence of atopy, rhini

Nucleotide excision repair and human syndromes.

DNA damage is implicated in cancer and aging, and several DNA repair mechanisms exist that safeguard the genome from these deleterious consequences. Nucleotide excision repair (NER) removes a wide diversity of lesions, the main of which include UV-induced lesions, bulky chemical adducts and some forms of oxidative damage. The NER process involves the action of at least 30 proteins in a 'cut-and-paste'-like mechanism. The consequences of a defect in one of the NER proteins are apparent from three rare recessive syndromes: xeroderma pigmentosum (XP), Cockayne syndrome (CS) and the photosensitive

Rationally Designed Galactose Dendrimer for Hepatocyte-Specific Targeting and Intracellular Drug Delivery for the Treatment of Liver Disorders.

Over two million people die of liver disorders every year globally. Hepatocytes are the key cells affected in several acute and chronic liver diseases. The current clinical outcomes of liver-targeted nanoparticles are limited, necessitating the need to develop smart hepatocyte-targeted drug delivery systems. Here, we present the rational design and development of a hepatocyte-targeting glycodendrimer (GAL-24) built from biocompatible building blocks, using expedite and facile chemical methodology. GAL-24 is designed to inherently target asialoglycoprotein receptor 1 (ASGP-R) on hepatocytes and

Coexistence of systemic sclerosis, scleroderma-like syndromes and neoplastic diseases.

Coexistence of rheumatic and neoplastic diseases may take different forms. Rheumatic paraneoplastic syndromes, including systemic sclerosis, scleroderma-like changes and Raynaud's phenomenon are induced by substances secreted by neoplastic cells and immunological disturbances connected are associated with malignancy. They may precede the clinical manifestation of neoplasm, occur simultaneously or after its diagnosis. In turn, chronic course of rheumatic diseases (Sjögren's syndrome, systemic sclerosis, rheumatoid arthritis, systemic lupus erythematosus, dermatomyositis) by immunologic stimulat

[Influence of a central cholinergic mode of action on the regulation of the intact and disturbed cerebral blood flow (author's transl)].

Cerebral chemical vasomotor reactivity and autoregulation were tested in normal baboons before and after intravertebral, intravenous and (or) intracarotid infusion of atropine and neostigmine. Furthermore, disordered cholinergic neurotransmission and dysautoregulation after acute experimental cerebral infarction have also been investigated. Intravertebral injection of atropine suppressed the increase of CBF by inhalation of 5% CO2 and enhanced the decrease of CBF induced by hyperventilation, but did not appreciably affect autoregulatory response. On the other hand, cerebral autoregulatory vaso

Identification of the estrogen receptor subtypes associated with testis-ova induction in the medaka.

Exposure to estrogenic endocrine disrupting chemicals (EDCs) poses a significant threat to aquatic wildlife. They are known to induce a range of adverse reproductive and developmental disorders that include induction of oocytes in the testis (testis-ova), however, mechanisms underlying the testis-ova induction are not fully understood. In this study, applying the use of estrogen receptor (esr) 1, esr2a and esr2b knock out (KO) in medaka exposed to 17β-estradiol (E2), we investigated which Esr subtypes mediate testis-ova formation. Loss of any esr subtype did not affect gonadal differentiation.

Bisphenol S promotes hepatic fibrosis through macrophage polarization- and autophagy-mediated activation of hepatic stellate cells.

The environmental endocrine disruptor bisphenol S (BPS) has emerged as a risk factor for non-alcoholic fatty liver disease (NAFLD), the most common chronic liver disorder worldwide. However, the mechanism by which this chemical triggers fibrosis progression in NAFLD remains unclear. Using transgenic zebrafish with labeled macrophages, RAW264.7 macrophages, and human hepatic stellate cells (HSCs, LX2), this study investigated the profibrotic effects of BPS and demonstrated that it promoted hepatic fibrosis through both indirect and direct mechanisms. Results showed that 1 and 100 μg/L BPS expos

Sida tuberculata extract reduces the nociceptive response by chemical noxious stimuli in mice: Implications for mechanism of action, relation to chemical composition and molecular docking.

Sida tuberculata R.E.Fr. (Malvaceae) is a medicinal plant widely found in Southern Brazil, and popularly used for inflammatory disorders and to pain relief. A phytochemical analysis followed by an investigation about antinociceptive potential and mechanism of action were performed with leaves and roots extracts. Methanolic extracts, designated as S. tuberculata leaves extract (STLE) and S. tuberculata roots extract, were analyzed both by UHPLC–MS. The in vivo antinociceptive potential of STLE (10–300 mg kg−1) was assessed in mice subjected to the acetic acid‐induced abdominal writhes and forma

Dopamine and serotonin contribute to Paecilomyces hepiali against chronic unpredictable mild stress induced depressive behavior in Sprague Dawley rats.

Paecilomyces hepiali contains identical chemical constituents to Cordyceps sinensis, and it presents antidepressant‑like activity via regulating noradrenergic and dopaminergic systems. Behavioral despair depression models serve important roles in scientific screening and evaluation of antidepressants. The present study aims to investigate the antidepressant‑like activity of P. hepiali extract (PHC) in chronic unpredictable mild stress (CUMS)‑induced rat model of depression. Following four weeks of treatment, similar to fluoxetine at 3 mg/kg (positive drug), PHC at doses from 0.08 to 2.0 g/kg s

Modulation of musculoskeletal hyperalgesia by brown adipose tissue activity in mice.

Cold exposure and a variety of types of mild stress increase pain in patients with painful disorders such as fibromyalgia syndrome. Acutely, stress induces thermogenesis by increasing sympathetic activation of beta-3 (β3) adrenergic receptors in brown adipose tissue. Chronic stress leads to the hypertrophy of brown adipose, a phenomenon termed adaptive thermogenesis. Based on the innervation of skeletal muscle by collaterals of nerves projecting to brown adipose, we theorized an association between brown adipose tissue activity and musculoskeletal hyperalgesia and tested this hypothesis in mic

Epigallocatechin-3-gallate (EGCG) Suppresses the Trafficking of Lymphocytes to Epidermal Melanocytes via Inhibition of JAK2: Its Implication for Vitiligo Treatment.

Vitiligo is an inflammatory skin disorder in which activated T cells play an important role in its onset and progression. Epigallocatechin-3-gallate (EGCG), the major chemical constituent of green tea, exhibits remarkable anti-oxidative and anti-inflammatory properties. EGCG administration has been confirmed to decrease the risk of vitiligo; however, the underlying mechanism is undetermined. In this study, we proved that EGCG directly inhibited the kinase activity of Janus kinase 2 (JAK2). In primary cultured human melanocytes, EGCG pre-treatment attenuated interferon (IFN)-γ-induced phosphory

Multistep renal carcinogenesis as gene expression disease in tumor suppressor TSC2 gene mutant model--genotype, phenotype and environment.

Cancer is an inheritable disorder of somatic cells. Environment and heredity both operate in the origins of human cancer. These environmental and genetic determinants of cancer can be classified into four groups designated "Oncodemes" [1]. Oncodeme 1 is the irreducible "background" level of cancer due to spontaneous mutagenesis. Oncodeme 2 is "environmentally induced" cancer, whose causative agents are chemical carcinogens, radiation and viruses. Oncodeme 3 is basically "environmentally induced" cancer, but there are genetically determined differences among persons, e.g. the activation or inac

Simple acid-base disorders.

This article reviews normal acid-base regulation, related laboratory tests, and the potential disorders if the body's ability to compensate is disrupted. Acid derived from the oxidation of proteins and through tissue metabolism must be excreted or neutralized daily by the kidneys and lungs to maintain a proper acid-base balance. Acid-base homeostasis is normally maintained by chemical buffering, changes in renal hydrogen-ion excretion, and alterations in the rate and volume of alveolar ventilation. Metabolic disorders are characterized by disturbances in bicarbonate (HCO3-) concentration, and

Intraparenchymal leiomyoma of the breast: a case report.

The original 'oestrogen hypothesis' postulated that the apparent increase in human male reproductive developmental disorders (testis cancer, cryptorchidism, hypospadias, low sperm counts) might have occurred because of increased oestrogen exposure of the human foetus/neonate; five potential routes of exposure were considered. This review revisits this hypothesis in the light of the data to have emerged since 1993. It addresses whether there is a secular increasing trend in the listed disorders and highlights the limitations of available data and how these are being addressed. It considers whet

Machine Learning-Integrated Pharmacophore, DFT Analysis, and molecular dynamics of Diosmetin as a potent ache inhibitor with neuroprotective activity in a Scopolamine-Induced alzheimer's zebrafish model.

Alzheimer's Disease (AD) is a degenerative disorder of the brain that causes a gradual loss of cognitive function. The cholinergic hypothesis suggests that acetylcholine deficiency is the main cause of AD, which explains why blocking acetylcholinesterase (AChE) is the most effective way to treat AD. Nevertheless, there are some drawbacks to the currently available AChE inhibitors; thus, new molecules with better therapeutic effects and fewer side effects are needed. In this study, the anti-Alzheimer activity of diosmetin, a natural flavonoid, was investigated via an integrated computational an

New insights into mechanisms of valproic acid-induced neurodevelopmental toxicity in autism spectrum disorder: An integrative network toxicology approach combined with in vivo validation.

Autism spectrum disorder (ASD) is a neurodevelopmental condition with growing evidence linking its etiology to chemical exposures, including valproic acid. This study employed an integrative approach combining network toxicology, molecular docking, Mendelian randomization (MR), GEO data analysis, and experimental validation to systematically elucidate the mechanisms of valproic acid (VPA)-induced neurodevelopmental toxicity in ASD. Computational toxicity predictions identified hepatotoxicity, respiratory toxicity, and pronounced blood-brain barrier penetration as key features of VPA. Network t
