Association between HLA class II genes and autoantibodies to cyclic citrullinated peptides (CCPs) influences the severity of rheumatoid arthritis.

OBJECTIVE: The functional role of HLA class II molecules in the pathogenesis of rheumatoid arthritis (RA) is unclear. HLA class II molecules are involved in the interaction between T and B lymphocytes required for long-lived B cell responses and generation of high-affinity IgG antibodies. We undertook this study to investigate the relationship between HLA class II gene polymorphisms and RA-specific IgG antibodies against cyclic citrullinated peptides (anti-CCP antibodies). METHODS: High-resolution HLA-DR and DQ typing and anti-CCP-2 antibody testing were performed on 268 RA patients from the

Inhibition by salicylates of urea synthesis by isolated rat hepatocytes and citrulline synthesis by isolated rat mitochondria: an effect independent of uncoupling.

1. Acetylsalicylate and salicylate inhibited urea synthesis by isolated rat hepatocytes and citrulline synthesis by isolated rat mitochondria. The effects were dose-dependent and occurred at drug concentrations seen in salicylate induced hepatoxicity. 2. Although ATP concentration was decreased in the hepatocytes the effect of the salicylates on citrulline synthesis remained after treatment with oligomycin and carbonyl cyanide m-chlorophenyl hydrazone. This suggests that the effect is independent of uncoupling of oxidative phosphorylation. 3. This in vitro inhibition of urea synthesis by salic

Long-term durability of a time-limited methotrexate intervention in patients with anti-citrullinated protein antibody-positive and anti-citrullinated protein antibody-negative arthralgia at increased risk for rheumatoid arthritis (TREAT EARLIER): 5-year data from a double-blind, randomised, placebo-controlled trial.

BACKGROUND: The TREAT EARLIER trial showed that a time-limited intervention with 12 months of methotrexate in individuals with arthralgia at risk of rheumatoid arthritis improved inflammatory burden but did not prevent rheumatoid arthritis after 2 years. Long-term follow-up is needed to assess the durability of the effects. In addition, heterogeneity in pathophysiological subtype (with or without anti-citrullinated protein antibodies [ACPA]) and disease risk was not considered in the 2-year analysis. Therefore, we investigated the long-term effect of methotrexate in reducing disease burden and

Lactiplantibacillus plantarum AS21 Attenuates Sex-Biased Glucose Intolerance in Aging Mice via Gut Mucosal Microbiota-Derived Citrulline.

Aging-related metabolic decline, particularly impaired glucose tolerance, represents a growing challenge in aging populations. Probiotics are promising modulators of host metabolism; however, how sex differences influence their efficacy in mitigating metabolic aging remains poorly understood. Here, we administered Lactiplantibacillus plantarum AS21 to naturally aged male and female mice for 6 months. We observed a pronounced sexual dimorphism in metabolic decline, with aged males exhibiting more severe glucose intolerance than females. Concurrently, circulating citrulline levels were depleted

Lacto-N-triose II alleviates metabolic dysfunction-associated Steatotic liver disease via microbiota-dependent citrulline biosynthesis.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is an increasingly recognized global metabolic disease with limited treatment options. Lacto-N-Triose II (LNT II) is an important member of the human milk oligosaccharides (HMOs) and exhibits various prebiotic properties beyond infant nutrition. In this study, we demonstrate that supplementation with probiotic Kluyveromyces lactis-derived LNT II significantly ameliorates the progression of high-fat diet (HFD)-induced MASLD in mice, as evidenced by reduced body weight gain, decreased blood lipid levels, attenuated inflammatory res

Diaphragm-specific effects of L-citrulline in mdx mice highlight its potential as adjuvant of standard therapy in Duchenne muscular dystrophy.

BACKGROUND AND PURPOSE: The absence of the protein dystrophin in Duchenne muscular dystrophy (DMD) leads to progressive muscle weakness, failing regeneration and deregulation of nitric oxide (NO) signalling. We focused on L-citrulline, a precursor of L-arginine, required for NO production in muscle, which is reduced in dystrophic mdx muscle. EXPERIMENTAL APPROACH: L-Citrulline was administered (2 mg g-1 die-1), through diet, in comparison and/or in combination with prednisolone (1 mg kg-1, 5 days per week subcutaneously) to 4- to 5-week-old mdx mice for 8 weeks. KEY RESULTS: L-Citrulline inc

Primary care anti-cyclic citrullinated protein antibody testing for rheumatoid arthritis and accompanying pathway redesign reduces unnecessary referrals.

OBJECTIVES: Rheumatoid factor (RF) and anti-cyclic citrullinated peptide (anti-CCP) antibodies are alternative diagnostic tests for RA with similar sensitivity. Anti-CCP antibodies have improved specificity compared with RF and so should result in fewer false positives. A retrospective, observational study was conducted following a change in the primary care pathway and test for inflammatory arthritis from RF to anti-CCP antibodies, assessing real-world impact on referrals. METHODS: The primary care test was changed from RF to anti-CCP antibodies as part of a service development in East and N

Impact of Chronic Nitrate and Citrulline Malate Supplementation on Performance and Recovery in Spanish Professional Female Soccer Players: A Randomized Controlled Trial.

Background: Pre-season training is critical for developing tolerance to high physical demands in professional soccer, and nitric oxide (NO) precursors such as dietary nitrate (NO3-) and citrulline malate (CM) can support performance and recovery during this demanding phase. This study aimed to examine the effects of a four-week supplementation protocol combining 500 mg of NO3- from amaranth extract and 8 g of CM (NIT + CM) on external training load and post-match recovery in professional female soccer players during pre-season. Methods: A randomized, double-blind, placebo-controlled trial was

Interaction of HLA-DRB1 shared epitope and smoking on the development of anti-citrullinated protein antibody positive rheumatoid arthritis in Greek patients.

The composition of the gastrointestinal microbiome is increasingly recognized as a crucial contributor to immune and metabolic homeostasis-deficiencies in which are characteristic of cystic fibrosis (CF) patients. The murine model (CFTR (-/-) , CF), has, in previous studies, demonstrated characteristic CF gastrointestinal (GI) manifestations including slowed transit and significant upregulation of genes associated with inflammation. To determine if characteristics of the microbiome are associated with these phenotypes we used a phylogenetic microarray to compare small intestine bacterial commu

Citrulline and kynurenine to tryptophan ratio: potential EED (environmental enteric dysfunction) biomarkers in acute watery diarrhea among children in Bangladesh.

Two emerging biomarkers of environmental enteric dysfunction (EED) include plasma citrulline (CIT), and the kynurenine (KYN): tryptophan (TRP)/ (KT) ratio. We sought to investigate the plasma concentration of CIT and KT ratio among the children having dehydrating diarrhea and examine associations between concentrations of CIT and KT ratio with concurrent factors. For this analysis, we used cross-sectional data from a total of 102, 6-36 months old male children who suffered from non-cholera acute watery diarrhea and had some dehydration admitted to an urban diarrheal hospital, in Bangladesh. L-Citrulline Supplementation Restrains Ferritinophagy-Mediated Ferroptosis to Alleviate Iron Overload-Induced Thymus Oxidative Damage and Immune Dysfunction.

L-citrulline (L-cit) is a key intermediate in the urea cycle and is known to possess antioxidant and anti-inflammation characteristics. However, the role of L-cit in ameliorating oxidative damage and immune dysfunction against iron overload in the thymus remains unclear. This study explored the underlying mechanism of the antioxidant and anti-inflammation qualities of L-cit on iron overload induced in the thymus. We reported that L-cit administration could robustly alleviate thymus histological damage and reduce iron deposition, as evidenced by the elevation of the CD8+ T lymphocyte number and

Anti-Citrullinated Peptide Antibody Expression and Its Association with Clinical Features and Outcomes in Patients with Antineutrophil Cytoplasmic Antibody-Associated Vasculitis.

Background and objectives: Anti-citrullinated peptide antibody (ACPA), a characteristic antibody detected in rheumatoid arthritis, could be linked to antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) via the formation of neutrophil extracellular traps. We investigated the rate of ACPA positivity in patients with AAV and evaluated the association of ACPAs with their clinical features and outcomes. Materials and Methods: A total of 168 AAV patients with both ACPA and ANCA results at diagnosis were identified. Clinical and laboratory variables, including the disease-specific

Variation of plasma citrulline as a predictive factor for weaning off long-term parenteral nutrition in children with neonatal short bowel syndrome.

BACKGROUND & AIMS: Long-term parenteral nutrition (PN) is the mainstay of the therapeutic strategy in intestinal failure (IF) due to neonatal short bowel syndrome (SBS). Our aim was to identify prognostic factors for PN weaning and to assess if measuring plasma citrulline concentrations over time could account for the intestinal adaptation in progress. METHODS: This retrospective study included children with neonatal SBS with surgical measurement of the residual bowel length and repeated plasma citrulline assessments during a 4-year follow-up. The degree of IF was assessed by the PN dependenc

Identification of potential autoantigens in anti-CCP-positive and anti-CCP-negative rheumatoid arthritis using citrulline-specific protein arrays.

The presence or absence of autoantibodies against citrullinated proteins (ACPAs) distinguishes two main groups of rheumatoid arthritis (RA) … binding of IgG antibodies in anti-CCP-negative plasma was also enhanced by PAD2- and PAD4-mediated citrullination of 29 and 26 proteins, respectively. For only four proteins, significantly more ACPA binding occurred after citrullination with PAD2 compared to citrullination with PAD4, while the opposite was true for one protein. We demonstrate that PAD2 and PAD4 …

Maternal supplementation with citrulline or arginine during gestation impacts fetal amino acid availability in a model of intrauterine growth restriction (IUGR).

BACKGROUND: Supplementing maternal diet with citrulline or arginine during gestation was shown to enhance fetal growth in a model of IUGR induced by maternal dietary protein restriction in the rat. OBJECTIVE: The aims of this study were to determine in the same model whether maternal supplementation with citrulline or arginine would increase 1) citrulline and arginine concentration in fetal circulation; 2) the expression of placental amino acid transporters, and 3) the fetal availability of essential amino acids. METHODS: Pregnant rats (n = 8 per group) were fed either an isocaloric control
