Genetic complementation between UV-sensitive CHO mutants and xeroderma pigmentosum fibroblasts.

The purpose of this study was to determine the feasibility of doing complementation analysis between DNA-repair mutants of CHO cells and human fibroblasts based on the recovery of hybrid cells resistant to DNA damage. Two UV-sensitive CHO mutant lines, UV20 and UV41, which belong to different genetic complementation groups, were fused with fibroblasts of xeroderma pigmentosum in various complementation groups. Selection for complementing hybrids was performed using a combination of ouabain to kill the XP cells and mitomycin C to kill the CHO mutants. Because the frequency of viable hybrid clon

High efficiency DNA-mediated transformation of primate cells.

Tissue culture cells from several mammalian species, including three primate lines, were transfected with recombinant vectors carrying Escherichia coli xanthine-guanine phosphoribosyltransferase or Tn5 aminoglycoside phosphotransferase dominant selectable markers. Human HeLa and SV40-transformed xeroderma pigmentosum cells exhibited stable transformation frequencies of at least 10(-3) (0.1 percent). CV-1, an African green monkey kidney cell line, could be stably transformed with the exceptionally high frequency of 6 X 10(-2) (6 percent).

Comparative genotoxicity studies of ethyl carbamate and related chemicals: further support for vinyl carbamate as a proximate carcinogenic metabolite.

In vivo and/or in vitro mammalian cell systems were used to evaluate sister chromatid exchange (SCE) induction and gene mutagenesis effects following exposure to ethyl carbamate (urethane), vinyl carbamate, ethyl N-hydroxycarbamate, and 2-hydroxyethyl carbamate. Although ethyl carbamate caused dose-dependent increases in SCE when injected into mice, it was ineffective for inducing SCE and gene mutation (6-thio-guanine resistance) in Chinese hamster V-79 cells cultured with or without the addition of S9 enzyme mix during treatment. Chemical-specific patterns of genotoxicity were evident for the

A screening method for isolating DNA repair-deficient mutants of CHO cells.

A simple procedure for isolating mutagen-sensitive clones of CHO cells was developed and applied in mutant hunts in which colonies were screened for hypersensitivity to killing by ultraviolet radiation (UV, ethyl methanesulfonate (EMS), or mitomycin C (MMC). Each of two UV-sensitive clones studied in detail had a D37 dose of 1.0 J/m2 compared to 7.0 J/m2 for the wild-type cells, and each was shown to have no detectable repair replication following exposure to UV doses of up to 26 J/m2. Although these mutants resemble xeroderma pigmentosum human mutants with respect to their repair defect and c

Biological and biochemical characterisation of purine analogue resistant clones of V79 Chinese hamster cells.

Purine analogue resistant clones have been selected from the closely related Chinese hamster lines V79A and V79S. Clones were of either spontaneous origin or induced by EMS or ultraviolet light. The majority of clones selected in 8-azaguanine showed stable cross resistance to 6-thioguanine. Clones derived from V79A and selected for 6-thioguanine resistance were cross resistant to 8-azaguanine: however a group of 6-thioguanine resistant mutants selected from V79S cells were 8-azaguanine sensitive. All clones except two were unable to grow in HAT medium. The two exceptions were 8-azaguanine resi

Paving the Way for a Minimal CHO ChassisExploring the Upper Limits of Megabase-Scale Deletions through DNA Repair Modulation.

Chinese hamster ovary (CHO) cells are the predominant mammalian host for therapeutic protein production, yet their large genome contains substantial non-essential DNA that may impose metabolic burden and contribute to process variability. Here, we establish a CRISPR/Cas9-based strategy for megabase-scale genomic deletions in CHO cells, targeting regions from 3 to over 100 megabases (Mb). Clones carrying deletions of up to 50 Mb were isolated, while a candidate deletion junction spanning 130 Mb was detected in the edited pool, demonstrating the potential for large-scale genome engineering in ma

Addressing the elephant in the room: A comprehensive framework to resolve selectivity, multicollinearity, and scalability challenges in Raman chemometric models for mammalian cell culture applications.

Raman spectroscopy offers significant potential for real-time bioprocess monitoring, but adoption in biologics manufacturing remains limited due to challenges in developing robust chemometric models. Key barriers include poor selectivity between structurally similar metabolites, multicollinearity among correlated analytes that confound model predictions, and insufficient training data ranges for critical quality attributes (CQAs). To overcome these barriers, a novel workflow called Spiking and Pure Analyte Characterization for Raman Chemometrics (SPARC) was developed. SPARC addresses these cha

Establishment of a reporter gene-based assay to determine ADCC activity of anti-tetanus toxin monoclonal antibody.

Tetanus remains a serious health concern, and therapeutic monoclonal antibodies targeting the tetanus toxin C-fragment (TT-C) require reliable methods to assess antibody-dependent cell-mediated cytotoxicity (ADCC) activity. In this study, we describe the development of a validated luciferase reporter ADCC assay using a stable Flp-In CHO cell line stably expressing high levels of TT-C as target cells. The generated CHO cell line exhibited stable and high-level TT-C expression, with 99.36% TT-C-positive cells. We found the assay produced a highly reproducible sigmoidal dose-response curve (R2 >

Development and non-clinical characterization of Procizumab (invobenitug): a humanized antibody neutralizing circulating DPP3.

Dipeptidyl peptidase 3 (DPP3) is an intracellular protein involved in cellular antioxidant responses. Upon acute stress and tissue damage, DPP3 is released into the circulation (circulating DPP3, cDPP3), where it induces vascular, cardiac, and immune dysregulation. Elevated cDPP3 levels have been implicated in multiple organ dysfunction during circulatory failure. Here, we describe the development and characterization of Procizumab (PCZ), a human immunoglobulin (Ig) G1 kappa monoclonal antibody targeting human cDPP3. The murine variant (mAb1967) was generated by immunizing BALB/c mice with a D

Effects of intra-tumoral cellular heterogeneity of oxygen partial pressure on biological effectiveness of hydrogen-, helium-, carbon-, oxygen-, and neon-ion beams.

Objective. The tumor microenvironment characterized by heterogeneously organized vasculatures causes intra-tumoral heterogeneity of oxygen partial pressurepat the cellular level, which cannot be measured by current imaging techniques. The intra-tumoral cellularpheterogeneity may lead to a reduction of therapeutic effects of radiation. The purpose of this study was to investigate the effects of the heterogeneity on biological effectiveness of H-, He-, C-, O-, and Ne-ion beams for different oxygenation levels, prescribed dose levels, and cell types.Approach.The intra-tumoral cellularpdistribution

Single-cycle parainfluenza virus type 5 vectors for producing recombinant proteins, including a humanized anti-V5 tag antibody.

Parainfluenza virus type 5 (PIV5) can cause either persistent or acute/lytic infections in a wide range of mammalian tissue culture cells. Here, we have generated PIV5 fusion (F)-expressing helper cell lines that support the replication of F-deleted viruses. As proof of the principle that F-deleted single-cycle infectious viruses can be used as safe and efficient expression vectors, we have cloned and expressed a humanized (Hu) version of the mouse anti-V5 tag antibody (clone SV5-Pk1). We show that multiple different cell lines can be infected and express high levels of the Hu anti-V5 antibody

Development of an attenuated glutamine synthetase (GS) selection system for the stable expression of tissue plasminogen activator in CHO-K1 cells.

Chinese hamster ovary (CHO) cells represent the most common host system for the expression of high-quality recombinant proteins. The development of stable CHO cell lines used in industrial recombinant protein production often relies on dihydrofolate reductase (DHFR) and glutamine synthetase (GS) amplification systems. Conventional approaches to develop stable cell lines lead to heterogeneous cell populations. Consequently, it is desirable to adopt innovative strategies to increase the efficiency of clone selection to reduce the time and effort invested in the cell line development process. Att

Alzheimer disease-associated tau post-translational modification mimics impact tau propagation and uptake.

As Alzheimer disease (AD) progresses, pathological tau spreads by cell-to-cell propagation of tau. This study aims to elucidate the impact of AD-associated post-translational modifications of tau-on-tau propagation. Tau propagation reporter constructs distinguishing donor cells from recipient cells were developed, and additional constructs were made with tau residues mutated from serine or threonine to aspartate to mimic the negative charge of a phosphorylation and/or from lysine to glutamine to mimic the charge-neutralizing effect of acetylation. Flow cytometry was used to quantify donor and

Application of aurintricarboxylic acid to boost CHO cell performance.

PURPOSE: Currently, a robust process using healthy Chinese hamster ovary (CHO) cells, the dominating host for biopharmaceutical protein therapeutics, is crucial for high productivity and successful scaling-up. However, issues which are originated from cell performance and detrimental to protein products such as dramatic viability drop and accumulation of toxic metabolites still exist. Aurintricarboxylic acid (ATA), reported as an antioxidant chelator to prevent apoptosis and boost cell growth, was applied as an additive in this study to address the above problems. METHODS: Two CHO cells were

Enhanced antibody production in Chinese hamster ovary cell cultures supplemented with barley shochu distillation by-product supernatant.

Antibody production in Chinese hamster ovary (CHO) cell culture was enhanced by supplementing the culture medium with barley shochu distillation by-product supernatant (BX2). To predict antibody production following BX2 addition, fed-batch culture experiments were conducted under varying BX2 conditions using a response surface methodology. BX2 supplementation was predicted to improve antibody production by 138 %, 146 %, 120 %, and 240 % in IgG-producing CHO-MK1, CHO-MK2, CHO-DG44, and Fc-fusion protein-producing CHO-DG44 cells, respectively, compared to controls without BX2. BX2 consisted prim

N-terminal domains and site-specific glycosylation regulate the secretion of avian melanocortin inverse agonists, agouti signaling protein (ASIP) and agouti-related protein (AGRP).

Agouti signaling protein (ASIP) and agouti-related protein (AGRP) are paralogous inverse agonists of melanocortin receptors with distinct physiological roles, but their structural and biochemical properties in birds remain poorly understood. Here, we characterized chicken ASIP and AGRP proteins. Analysis of available sequences revealed that a motif resembling the mammalian proprotein convertase 1/3 (PC1/3, also known as PCSK1) cleavage site is conserved across a broad range of avian orders, but Western blot analysis of transfected Chinese hamster ovary (CHO-K1) cells and chicken hypothalamus d

Optimizing Recombinant Protein Production in CHO Cells by Silencing the Caspase 8 Associated Protein 2 Gene via the CRISPR-Cas9 System.

Chinese hamster ovary (CHO) cells play a crucial role in biopharmaceutical production due to their ability to produce complex proteins. Enhancing the productivity of CHO cells is essential for meeting the growing demand for biologics. Caspase 8-Associated Protein 2 (CASP8AP2), a key regulator of apoptosis and cell survival, has been identified as a potential target to increase CHO cell productivity. To this end, CRISPR-mediated homology-independent targeted integration (HITI) was used to silence CASP8AP2. Results of the cell viability assay revealed that CASP8AP2-deficient clones (C2, C3, and

Nitric oxide regulates cytochrome P450 2D6 and 3A4 activity via concentration-dependent modulation of heme loading.

Cytochrome P450 enzymes (CYPs) play diverse roles in human health and disease, and although their activities depend on their heme contents, the cellular mechanisms governing CYP heme levels are unclear. Because CYP activities are influenced by biological nitric oxide (NO), we investigated how a range of NO exposures would impact the heme levels and activities of CYP2D6 and 3A4 expressed in Chinese hamster ovary cells and in the human liver cell line HepG2. Following expression, both CYPs were present as a 60:40 mix of heme-free and heme-bound forms. A low range of NO concentrations (approximat

Characterization of Recombinant Human Type II Collagen from CHO Cells, Functional Assessment of Chondrocytes and Alleviation of Cartilage Degeneration.

Type II collagen (Col2), a crucial structural protein in hyaline cartilage, is essential for cartilage integrity and facilitating injury repair. However, research on recombinant type II collagen still faces many challenges, such as structure and yield, which limit the application of recombinant Col2 in biomedical fields. In this study, we achieved high-yield expression of full-length human Col2 (rhCol2) in CHO cells. The physical and chemical properties of rhCol2 were very close to native Col2, including molecular weight, triple helix structure, thermal stability and self-assembly capacity. Fu
