Solar radiation sustains and affects all life forms on Earth. The increase in solar UV-radiation at environmental levels, due to depletion of the stratospheric ozone layer, highlights serious issues of social concern. This becomes still more dramatic in tropical and subtropical regions where radiation-intensity is still higher. Thus, there is the need to evaluate the harmful effects of solar UV-radiation on the DNA molecule as a basis for assessing the risks involved for human health, biological productivity and ecosystems. In order to evaluate the profile of DNA damage induced by this form of

Cryptosporidium parvum is a coccidian protozoan that causes diarrhea in humans, often chronic and severe in patients with AIDS. Conventionally, diagnosis is made by concentration of stools followed by acid-fast staining (AF) or immunofluorescent staining. The threshold of detection in human stool specimens by these methods may require the presence of 50,000 (immunofluorescent staining) to 500,000 (AF) oocysts per g of stool. In this study, a nested PCR assay was developed to detect C. parvum DNA directly from stool specimens. After extraction of DNA from formalinized stool, a 400-bp fragment o

Dietary components have recently received rapidly expanding attention for their potential to halt or reverse the development of many oxidative stress-mediated diseases after exposure to environmental toxicants. 7, 12 dimethylbenz(a)anthracene (DMBA) is one of the most common environmental pollutants. The present study aimed to evaluate the chemo-preventive effects of broccoli as a nutritional component against DMBA intoxication in rats. A daily dose of aqueous (1 ml/rat) and methanolic (150 mg/kg) broccoli extracts, respectively, was given to 50-day-old female rats for 26 successive weeks afte

BACKGROUND: Genetic and environmental risk factors contribute to periodontal disease, but the underlying susceptibility pathways are not fully understood. Epigenetic mechanisms are malleable regulators of gene function that can change in response to genetic and environmental stimuli, thereby providing a potential mechanism for mediating risk effects in periodontitis. The aim of this study is to identify epigenetic changes across tissues that are associated with periodontal disease.
METHODS: Self-reported gingival bleeding and history of gum disease, or tooth mobility, were used as indicators

Cancer remains the second most frequent cause of death in human populations worldwide, which has been reflected in the emphasis placed on management of risk from environmental chemicals considered to be potential human carcinogens. The formation of DNA adducts has been considered as one of the key events of cancer, and persistence and/or failure of repair of these adducts may lead to mutation, thus initiating cancer. Some chemical carcinogens can produce DNA adducts, and DNA adducts have been used as biomarkers of exposure. However, DNA adducts of various types are also produced endogenously i

African swine fever virus (ASFV) continues to challenge the pig industry worldwide, with persistence in farm environments posing a major obstacle to repopulation. This study evaluated ASFV DNA persistence and farm-level biosecurity practices on pig farms in Malaysia following cleaning and disinfection (C&D) protocols after an ASF outbreak in the farm. A structured biosecurity survey covering 5 domains (personnel entry, farm structure, C&D, carcass disposal management and water source) was conducted across 37 farms, comprising both ASF-affected and unaffected farms, while environmental swab sam

The clarification of possible exposure sources of multiple metals to identify associations between metal doses and urothelial carcinoma (UC) risk is currently limited in the literature. We sought to identify the exposure sources of 10 metals (Vanadium, chromium, manganese, cobalt, nickel, copper, zinc, arsenic, cadmium, and lead) using principal component analysis (PCA) and then linked various principal component (PC) scores with environmental characteristics, including smoking-related indices, PM2.5, and distance to the nearest bus station. In addition, urinary 8-hydroxy-2'-deoxyguanosine (8-

Human placenta and cord blood are readily available specimens that respond to maternal environmental insult and are being used to investigate metabolism, bioactivation, and transplacental transfer of procarcinogens. Enzyme-linked immunosorbent assay was used to quantitate 120 placentas and 56 cord bloods from term, uncomplicated pregnancies at Taipei Chang Gung Memorial Hospital, Taiwan, for the presence of the imidazole ring-opened form of aflatoxin B1-DNA (AFB1-DNA) adducts. Of the 120 samples of placentas, 69 (57.5%) contained AFB1-DNA adducts in levels from 0.6 to 6.3 mumol/mol DNA. Of the

Psoriasis is a chronic, recurrent, inflammatory disease that is affected by genetic, immunological, epigenetic, and environmental factors. With the development of biotechnology, research on the pathogenesis of psoriasis has deeply focused on the field of epigenetics, and great progress has been made. Epigenetics is the study of heritable changes in gene expression or cell phenotypes without altering the DNA sequence. DNA methylation (DNAm) alterations are the most common epigenetic phenomena and are widely studied. Many studies have shown that DNAm plays a key role in the pathogenesis of psori

Carboxyl group-functionalized magnetic nanoparticles were used to develop an RNase-free method for plasmid DNA (pDNA) purification directly from RNA-containing crude Escherichia coli lysates. This method takes advantage of differing adsorption behaviors of pDNA and RNA onto magnetic nanoparticle surfaces at different temperatures. Pure pDNA can be isolated between 70 and 80°C without sacrificing DNA quality and quantity, as evidenced by comparison with that obtained using organic solvents or commercial kits. This RNase-free method is rapid, simple, cost-effective, and environmentally friendly,

This study reports the results of a collaborative study undertaken by two independent research groups to (a) confirm recent PCR-based detection of Yersinia pestis DNA in human teeth from medieval plague victims in France, and (b) to extend these observations over five different European burial sites believed to contain plague victims dating from the late 13th to 17th centuries. Several different sets of primers were used, including those previously documented to yield positive results on ancient DNA extracts. No Y. pestis DNA could be amplified from DNA extracted from 108 teeth belonging to 61

Gastric cancer (GC) is a highly intricate gastrointestinal malignancy. Early detection of gastric cancer forms the cornerstone of precision medicine. Several studies have been conducted to investigate early biomarkers of gastric cancer using genomics, transcriptomics, proteomics, and metabolomics, respectively. However, endogenous substances associated with various omics are concurrently altered during gastric cancer development. Furthermore, environmental exposures and family history can also induce modifications in endogenous substances. Therefore, in this study, we primarily investigated al

Bacteriophage lambda replication complex, containing the phage-encoded O initiator protein protected from proteases by other elements of this complex, is a stable structure that can be inherited by one of the two daughter lambda DNA copies after a replication round in Escherichia coli. In normal growth conditions in bacteria bearing a plasmid derived from bacteriophage lambda, such a complex may be stable for many cell generations. However, it was found that this stable structure is disassembled under certain conditions, namely, after heat shock. Therefore, we asked whether other environmental

Environmental monitoring of microorganisms is critical for the protection and enhancement of human and ecosystem health. Even though these molecular methods have overtaken traditional culture-based methods and become more accessible, these techniques still require expensive equipment and dedicated facilities to process samples which in the context of a global pandemic, remote sampling areas or low-income countries can be extremely challenging. Sample preparation and sample homogenisation are critical steps for molecular-based techniques, especially for the extraction of DNA and RNA. This study

BACKGROUND: Coeliac disease (CD) is a autoimmune disease characterised by mucosal inflammation in the small intestine in response to dietary gluten. Genetic factors play a key role with CD individuals carrying either the HLA-DQ2 or HLA-DQ8 haplotype, however these haplotypes are present in half the general population making them necessary but insufficient to cause CD. Epigenetic modifications, including DNA methylation that can change in response to environmental exposure could help to explain how interactions between genes and environmental factors combine to trigger disease development. It is well-established that occupational benzene exposure can cause leukemia. DNA damage repair has great potential in assessing the health risk of benzene, but the role of RAD51, an executor of DNA damage repair, remains unclear in benzene-induced hematotoxicity. Here, we demonstrate that exposure to hydroquinone (HQ), a key metabolite of benzene, upregulates RAD51 expression in human lymphoblast (TK6), consequently suppressing DNA damage. Mechanistically, we demonstrate that METTL3, an m6A methyltransferase, catalyzes m6A modification on RAD51 mRNA, and that the m6A reader protein YTHDC1 sub
