Repeated gentle handling or maternal deprivation during the neonatal stage increases adult male rats' baseline orofacial pain responsiveness.

OBJECTIVE: Early life experiences have been found to have a long-lasting effect on brain development in adult life. The purpose of this study was to determine whether neonatal manipulation could alter orofacial pain responsiveness in adult rats METHODS: In the first 21 days of life, male rats were exposed to gentle handling or maternal deprivation (MD) procedures to establish models of handled and MD rats, respectively. The rats were assigned to three of the following experimental groups at the age of two months: intra-dental capsaicin (100 µg), intra-lip formalin (50 µL), and repeated nitrogl

Immunohistochemical evidence for proteolipid protein and nestin expression in the late bell stage of developing rodent teeth.

In this study, the expression of proteolipid protein (PLP) and nestin is studied in the late bell stage of developing rodent teeth in neonatal rats. By using immunohistochemistry, it was shown that odontoblasts, ameloblasts and the stratum intermedium are positive for PLP in regions of active matrix deposition. Reactivity for nestin could be detected in the odontoblasts, stratum intermedium and in some of the apical processes of the ameloblasts. The fact that mutations in the PLP gene can cause disturbances in tooth form, number and eruption taken together with the presence of PLP reactivity i

Distribution of Helicobacter pylori in the Oral Cavity of Patients With Tooth Extractions at Various Locations and Ages.

BACKGROUND: Helicobacter pylori is well known to cause gastric cancer, gastric ulcers and duodenal ulcers. The oral cavity has been implicated as a reservoir of H. pylori in adulthood. However, there are currently no detailed studies focused on the tooth type colonised by H. pylori and/or the age of subjects. Therefore, we examined the distribution of H. pylori in oral specimens collected from 163 Japanese adults who underwent tooth extraction. MATERIALS AND METHODS: Bacterial DNA was extracted from 163 Japanese adults, and H. pylori was detected using PCR. Surface deposits on the occlusal su

Human dental pulp stem cell adhesion and detachment in polycaprolactone electrospun scaffolds under direct perfusion.

Cell adhesion in three-dimensional scaffolds plays a key role in tissue development. However, stem cell behavior in electrospun scaffolds under perfusion is not fully understood. Thus, an investigation was made on the effect of flow rate and shear stress, adhesion time, and seeding density under direct perfusion in polycaprolactone electrospun scaffolds on human dental pulp stem cell detachment. Polycaprolactone scaffolds were electrospun using a solvent mixture of chloroform and methanol. The viable cell number was determined at each tested condition. Cell morphology was analyzed by confocal

Quantification and distribution of mast cells in oral periapical inflammatory lesions.

BACKGROUND: Periapical granuloma and periapical cysts develop as a sequela of inflammation of dental pulp, usually a consequence of an infected root canal. Mast cells (MCs) have most frequently been implicated in the pathophysiology of periapical lesions such as periapical granulomas and cysts. AIMS: The present study was undertaken to emphasize the presence and quantification of MCs in periapical lesions (granulomas and cysts) using toluidine blue stain, with an aim to elucidate their role in the pathogenesis of these lesions. MATERIALS AND METHODS: The study comprised a total of 55 previou

The Twin-Rooted Anomaly: Endodontic Management of a Rare Case of a Maxillary Central Incisor With Two Roots.

This case report describes the endodontic management of a maxillary central incisor with two roots, representing a rare anatomical variation. A 73-year-old female patient presented with a fractured tooth #21 with a carious crown. Pulp sensibility testing using cold spray elicited a sharp and prolonged painful response, consistent with a diagnosis of symptomatic irreversible pulpitis with normal apical tissues. Conventional periapical radiography suggested an unusual root morphology, which was further evaluated using cone-beam computed tomography (CBCT). CBCT imaging revealed a bifurcated root

Assessing the influence of Katana™ Cleaner on the clinical performance of bulk-fill restorations in endodontically treated teeth: study protocol for a randomized clinical study.

BACKGROUND: Complete cleaning of the pulp chamber interior is a mandatory step before the restorative process. Ethanol is the most commonly used substance for cleaning the dentin surface; however, studies report that ethanol is ineffective at completely removing residues. Although Katana™ is recommended for prosthetic purposes, in vitro studies have shown it delivers results equal to or better than ethanol. The study aims to evaluate the influence of Katana™ Cleaner on cleaning the pulp chamber of teeth treated endodontically with an epoxy or bioceramic sealer and on the clinical performance o

Biological Performance of Modified Glass Ionomer Cement: Antibacterial Activity, Cytocompatibility, and Rabbit Pulpal Response.

BACKGROUND: Conventional glass ionomer cement (GIC) is widely used as a restorative base and liner owing to its chemical adhesion to dentine, fluoride release, and favourable biocompatibility. Nevertheless, its intrinsic antibacterial activity and regenerative capacity remain limited. To enhance these properties, a series of modified GIC formulations, referred to as BIOGIC, incorporating tricalcium phosphate (TCP), TCTP-derived peptides, and 10% chlorhexidine (CHX), were developed. This study evaluated their antibacterial efficacy, cytocompatibility, and pulpal responses in a rabbit-tooth mode

Tetracycline-regulated expression of OLIG2 gene in human dental pulp stem cells lead to mouse sciatic nerve regeneration upon transplantation.

Numerous studies have indicated dental pulp stem cells (DPSCs) potency to differentiate into several types of cell lineages. Oligodendrocyte lineage transcription factor 2 (OLIG2) plays an important role in the oligodendrogenic pathway. In this study, a tetracycline (Tet)-inducible system expressing OLIG2 gene was transfected into human DPSCs to direct their differentiation toward oligodendrocyte progenitor cells (OPCs). Following induction, the expression of stage-specific markers was studied by Reverse Transcription quantitative Polymerase Chain Reaction (RT-qPCR), immunocytochemistry and we

Investigating best practice for specimen preparation for biological testing of root canal sealers.

INTRODUCTION: Biological characterization of root canal sealers is important as it assesses the ability of the root canal sealer to exert antimicrobial properties thus avoiding treatment failures caused by microbial challenge and also assess the cytotoxic effect on the periapical tissues. Assessment of the biological testing of root canal sealers necessitates the sterilisation of the materials prior to evaluation. This study aims to analyse the influence of various sterilisation techniques conducted prior to biological testing on the microstructure and surface properties of endodontic sealers.
