Complexing of tissue plasminogen activator with PAI-1, alpha 2-macroglobulin, and C1-inhibitor: studies in patients with defibrination and a fibrinolytic state after electroshock or complicated labor.

Release of tissue plasminogen activator (t-PA) and its interaction with plasma protease inhibitors were studied in two patients with massive defibrination, one after electroshock and soft tissue injury and the other after complicated labor; both had very severe hemorrhage. Large quantities of free t-PA were present in the circulation for several hours. Complexes of t-PA with plasminogen activator inhibitor 1 (PAI-1), alpha 2-macroglobulin and C1-inhibitor were also observed. PAI-1 antigen rose dramatically in both patients, and complexes of t-PA with PAI-1 rose rapidly during the period of obs…

Synthesis and anticonvulsant activity of racemic 2-amino-N-substituted succinimide derivatives.

Several derivatives of (R,S)-2-amino-N-substituted succinimides were synthetized and evaluated in mice against seizures produced by electroshock and pentylenetetrazol. The most active compound against both electroshock- and pentylenetetrazol-induced seizures was (R,S)-N-benzyl-2-(methanesulfamido)succinimide.

Anticonvulsive activity of duloxetine: A new choice for the epileptic patients with depression.

The aim of this study was to investigate the antiepileptic effects of duloxetine in the maximal electroshock test and convulsions induced by four compounds: Pentylenetetrazole, 3-mercaptopropionic acid, thiosemicarbazide, and bicuculline. Duloxetine exhibited moderate anticonvulsive activity with an ED50 (median effective dose) of 48.21 mg/kg in the maximal electroshock test in mice. The anticonvulsive action of duloxetine was also confirmed in chemical-induced seizure tests, where this drug decreased tonic convulsions. Single administration of duloxetine (6.25-25 mg/kg) significantly increase…

Dipyrone, a novel anticonvulsant agent? Insights from three experimental epilepsy models.

Dipyrone, a non-steroidal anti-inflammatory agent, was anticonvulsant in three experimental epilepsy models. At a dose of 300 mg/kg i.p., dipyrone blocked the maximal hind limb extension in the electroshock model in Wistar rats, the tonic-clonic component of acute sound-induced seizures and the limbic component of audiogenic kindling in genetically susceptible Wistar-derived rats, all in 100% of the animals. In the electroshock model higher doses (400 and 500 mg/kg) were also effective but lower doses (100 and 200 mg/kg) were not. In this model dipyrone had no effect on the recovery of the rig…

Disruption of adenosine metabolism increases risk of seizure-induced death despite decreased seizure severity.

OBJECTIVE: Respiratory arrest plays an important role in sudden unexpected death in epilepsy (SUDEP). Adenosine is of interest in SUDEP path … . METHODS: Our approach was to examine: (1) seizure severity and seizure-induced death after 15 mA electroshock seizures and during repeated pentylenetetrazol (PTZ) administration in wild-type mice (Adk +/+) and transgenic mice with reduced adenosine metabolism (Adk +/-); (2) the postictal hypercapnic ventilatory response (HCVR) in wild-type mice (the postict …

Comparative study of retrograde amnesia in rats on active and passive avoidance tasks and spontaneous recovery of memory.

Naive and pretrained rats were trained in two active avoidance paradigms using a pole-climbing box and in a single-trial passive avoidance task using a T-maze. They were then subjected to amnestic treatments with electroshock, leptazol, pentobarbitone, or ether anesthesia. Single retention tests were given at 20-24, 44-48, or 68-96 h posttreatment. Electroshock and leptazol seizures produced retrograde amnesia in all three paradigms, provided that seizures were maximal and retention was tested before 48 h. Prior treatment with anticonvulsant drugs prevented amnesia. Ether and pentobarbitone an…

Influence of 7-nitroindazole on the anticonvulsive action of conventional antiepileptic drugs.

7-Nitroindazole (a selective neuronal nitric oxide (NO) synthase inhibitor) at 25 and 50 mg/kg, 30 min before the test, did not influence the electroconvulsive threshold. However, at 50 mg/kg, it enhanced the anticonvulsive activity of phenobarbital against maximal electroshock and did not affect that of carbamazepine, diphenylhydantoin and valproate. L-Arginine (500 mg/kg) did not modify the protective activity of phenobarbital alone or the 7-nitroindazole-induced enhancement of its anticonvulsive potency against maximal electroshock. 7-Nitroindazole did not alter the plasma levels of antiepi…

[Experimental characteristics of the neurotropic spectrum of some antiepileptic agents].

Investigation of the psychotropic spectrum of phenobarbital, diphenin, milontin, pufemide, sodium valproate (depakine), and diazeparn showed that, besides antagonism with the corazole and electroshock effects, all these drugs prevented to a considerable degree the kindling seizures in rats on a "tonsil swing" (chronic electroencephalographic partial epilepsy) model. The mechanism of this effect probably involves exciting amino acids. All anticonvulsants except for diazepam also restored memory in experimental animals with retrograde electroshock amnesia. The administration of diazepam, dipheni…

Dronedarone (a multichannel blocker) enhances the anticonvulsant potency of lamotrigine, but not that of lacosamide, pregabalin and topiramate in the tonic-clonic seizure model in mice.

Accumulating experimental evidence indicates that some recently licensed antiarrhythmic drugs, including dronedarone (a multichannel blocker … four novel antiepileptic drugs (lacosamide, lamotrigine, pregabalin and topiramate) in the maximal electroshock-induced seizure model in mice. To exclude any potential pharmacokinetic contribution of dronedarone to the observed interactions, total brain concentrations of antiepileptic drugs were measured. Dronedarone (50 mg/kg, i.p.) significantly enhanced th …

Convulsant and anticonvulsant cyclopentanones and cyclohexanones.

The convulsant and/or anticonvulsant activity of unsubstituted and mono-alkyl-substituted cyclopentanones and cyclohexanones were examined by testing the ability of these compounds to produce seizures or to inhibit seizures induced by pentylenetetrazol and maximal electroshock in CF-1 mice. In addition, these compounds were tested for their ability to bind to the picrotoxin receptor. The unsubstituted compounds, cyclopentanone and cyclohexanone, prevented both pentylnetetrazol- and maximal electroshock-induced seizures. Cyclopentanones and cyclohexanones with small (less than 3 carbon atoms) a…

An observational study on neurobehavioural effects of acute oxydemeton-methyl (insecticide and acaricide) exposure in rats.

Oxydemeton-methyl, an organophosphate insecticide and acaricide produced decrease in the exploratory behaviour and prolongation of barbitone sodium-induced hypnosis after intermittent aerosol spray inhalational exposure for 1 h in rats compared to the saline control group. Further, CD50 +/- S.E.M. value for pentylenetetrazole (PTZ) and CI50 +/- S.E.M. value for electroshock (i.e. the dose of PTZ and intensity of electroshock producing positive seizure response in 50% of rats) were significantly decreased by acute exposure to oxydemeton-methyl compared to that of saline control group. The study…

Opioid receptor mediated anticonvulsant effect of pentazocine.

Intraperitoneal (i.p.) administration of (+/-) pentazocine (10, 30 & 50 mg/kg), a Sigma opioid agonist, resulted in a dose dependent anticonvulsant action against maximal electroshock seizures in mice. This anticonvulsant effect of pentazocine was not antagonized by both the doses of naloxone (1 and 10 mg/kg) suggesting thereby that its anticonvulsant action is probably mediated by Sigma opiate binding sites. Its anticonvulsant effect was potentiated by both the anticonvulsant drugs viz. diazepam and diphenylhydantoin. Morphine, mu opioid agonist, on the other hand, failed to protect the anima…

Effects of opioid antagonists on anticonvulsant and hypnotic activity of benzodiazepines.

The influence of naloxone and naltrexone on the hypnotic and protective efficacy of diazepam, chlordiazepoxide, clonazepam and estazolam against electroshock- and pentylenetetrazole-induced seizures was studied in mice. Naloxone and naltrexone significantly decreased the anticonvulsant effects of diazepam and estazolam, but they did not changed that of chlordiazepoxide and clonazepam in electroshock-induced tonic hindlimb extension. Protective effects of benzodiazepines against pentylenetetrazole-induced seizures were slightly diminished by naloxone and naltrexone (16 mg/kg). Opioid antagonist…

Influence of arachidonyl-2'-chloroethylamide, a selective cannabinoid CB1 receptor agonist, on the anticonvulsant and acute side-effect potentials of clobazam, lacosamide, and pregabalin in the maximal electroshock-induced seizure model and chimney test in mice.

The influence of arachidonyl-2'-chloroethylamide (ACEA - a selective cannabinoid CB1 receptor agonist) on the anticonvulsant potency and acute adverse-effect potentials of clobazam, lacosamide, and pregabalin was determined in the maximal electroshock-induced seizure model and chimney test in mice. ACEA (2.5 mg/kg, i.p.) significantly enhanced the anticonvulsant potency of pregabalin in the mouse maximal electroshock-induced seizure model by decreasing the median effective dose (ED50 ) of pregabalin from 125.39 to 78.06 mg/kg (P < 0.05). In contrast, ACEA (2.5 mg/kg) had no significant impact …

Anticonvulsant activity of phenobarbital and phenytoin in combination.

The anticonvulsant properties of phenobarbital and phenytoin given individually or in various combinations were determined by the maximal electroshock seizure test in rats 2 hours after subcutaneous administration. Drug concentrations in brain and plasma obtained immediately after maximal electroshock seizure were measured by gas-liquid chromatography. Total brain anticonvulsant drug concentrations required to abolish the hindlimb extensor component of maximal electroshock seizure were similar when these drugs were employed singly or in combinations. The median effective brain concentrations (…

Evidence for a synergistic interaction between phenytoin and phenobarbital in experimental animals.

Combined anticonvulsant effects of phenytoin and phenobarbital in mice and rabbits were studied using the isobolographic method which was based on the plasma concentrations of these drugs. Anticonvulsant activity of test drugs alone or in combination was determined by the method of maximal electroshock seizure test. Response of each animal to drug action was recorded as abolition of the tonic hindlimb extensor component of maximal electroshock seizure. It was demonstrated that the minimal effective plasma concentrations of phenytoin and phenobarbital alone for protection against maximal electr…

Effectiveness of anticonvulsants in mice exposed to mixed gamma-neutron radiations.

The effectiveness of four anticonvulsants was tested in male CF1 mice exposed to 500-, 1000-, and 10,000-rad doses of mixed gamma-neutron radiations. Prevention of the hind leg extensor component of a maximal convulsion induced by electroshock was selected as the end point for effective anticonvulsant activity of diphenylhydantoin, phenobarbital, and mephenytoin. Prevention of convulsions induced by pentylenetetrazol was the end point of effective anticonvulsant activity of trimethadione. The effectiveness of the drugs was evaluated by comparing the ED50's to the ED50 value for unirradiated co…

Effects of meclofenoxate and Extr. Rhodiolae roseae L. on electroconvulsive shock-impaired learning and memory in rats.

In experiments on albino rats, the authors studied the effects of meclofenoxate and Extr. Rhodiolae roseae on the memory-impairing action of convulsant electroshock. "Step-down" passive avoidance training with negative reinforcement was used to trace the changes in memory. Meclofenoxate administered i.p. in a dose of 100 mg/kg body weight for five days prevented the retrograde amnesia observed after convulsant electroshock upon retention testing on the 3rd and 24th hr after the end of the training session. The Rhodiola extract administered orally in a dose of 0.10 ml/rat for 10 days, which in …

The effects of 5,5-diphenylbarbituric acid on experimental seizures in rats: correlation between plasma and brain concentrations and anticonvulsant activity.

Diphenylbarbituric acid was administered by gavage to male albino Sprague-Dawley rats for evaluation of anticonvulsant activity and analysis of blood plasma and brain drug concentrations. A method for gas-liquid chromatographic analysis of blood plasma and brain for diphenylbarbituric acid is described. The drug was effective in the maximum electroshock test and as an antagonist to clonic seizures and death produced by pentylenetetrazol. Brain concentrations associated with 50 to 70% protection in the maximum electroshock seizure test were of the order of 2 to 6 mug/g of brain and are thus equ…
