On the role of peripheral macrophages during active experimental allergic encephalomyelitis (EAE).

Experimental allergic encephalitis (EAE) is an experimental autoimmune inflammatory condition of the central nervous system (CNS) that serves as a disease model for multiple sclerosis (MS). The primary effector mechanisms of the immune system leading to tissue destruction during EAE remain still controversial. T-cells, microglia, and macrophages infiltrating the brain parenchyma are suggested to be involved. To clarify the role of these cells during disease Lewis rats were immunised with different immunisation protocols: Immunisation with myelin basic protein (MBP) in complete Freunds adjuvant

Acute disseminated encephalomyelitis in a female with hereditary neuropathy with susceptibility to pressure palsy.

A 7-year-old female presented with fever, urinary incontinence, mental regression, gait disturbance, and lethargy after diarrhea. Magnetic resonance imaging revealed multifocal T(2)-weighted hypersignal lesions supportive of acute disseminated encephalomyelitis. Her mother had been diagnosed with hereditary neuropathy with susceptibility to pressure palsy. The girl was also determined to have hereditary neuropathy with liability to pressure palsy, with a 1.5-Mb deletion in chromosome 17p11.2 encompassing the gene for peripheral myelin protein 22 detected by fluorescent in situ hybridization. H…

Early eosinophil infiltration into the optic nerve of mice with experimental allergic encephalomyelitis.

Experimental allergic encephalomyelitis is a murine model of preclinical autoimmune disease that has pathologic similarities to multiple sclerosis (MS). Although CD4+ T cells have been shown to play a crucial role in the development of disease, we recently demonstrated a link between the development of paralysis and eosinophil infiltration into the spinal cord. As such, CD4+ cells may initiate disease, but eosinophils may be the actual effector cells responsible for causing damage to myelin and causing paralysis. Because MS patients sometimes experience early visual problems, ie, optic neuriti…

Intestinal lesions in a horse associated with eastern equine encephalomyelitis virus infection.

The primary lesions of eastern equine encephalomyelitis (EEE) virus infection in the horse are limited to the brain and spinal cord. Intestinal lesions in addition to the changes in the central nervous system were found in a 6-month-old male Tennessee Walking Horse. One week prior to death, this colt was vaccinated for EEE virus, western equine encephalomyelitis virus, influenza virus, equine rhinopneumonitis virus, and tetanus. The clinical signs consisted of ataxia and rear-end weakness, with a body temperature of 102.8 F. Gross lesions consisted of yellowish discoloration, swelling, edema, …

Parainfectious optic neuritis and encephalomyelitis. A report of two cases with thalamic involvement.

Two children developed mental status alteration and bilateral profound visual loss secondary to optic neuritis. The clinical picture was consistent with parainfectious encephalomyelitis. Magnetic resonance imaging showed bilateral involvement of the thalamus in both cases. In one case the thalamic involvement was solitary and was suspected initially to represent a primary thalamic neoplasm. This was ruled out by a stereotactic biopsy of the thalamus, which showed perivascular inflammation consistent with parainfectious encephalomyelitis. The clinical and neuroimaging findings improved signific…

[New information on the histopathology of Vilyui encephalomyelitis: possible role of copper deficiency].

The nature of a pathological process in the central nervous system in Vilyui encephalomyelitis defines it as a specific entity of demyelinative encephalomyelitis with a drastic prevalence of an alternative component. The bulk of the nerve parenchyma dies from circulatory incompetence due to blood vascular diseases, which are typical of the disease, as a peculiar angiopathy and progressive reduction in the microcirculatory bed. Homeostatic disorders in the myelinic membranes of central nervous fibers were found to be of value. The maintenance of homeostasis is implicated by adequate oligodendro…

[The duration of humoral postvaccinal and postinfection immunity to the Venezuelan equine encephalomyelitis virus].

Serological studies on the levels of virus-neutralizing antibodies (VNA) against Venezuelan equine encephalomyelitis virus (VEE) were carried out by the standard method in 280 human subjects given a single primary inoculation of experimental forms of live vaccines against VEE prepared from attenuated strains No. 15 and No. 230. The VNA were found to persist in the blood of the vaccinees for 5-25 years (the observation period) from the time of vaccination practically at the same level. The VNA titres did not depend on the form of the preparation (for subcutaneous or oral administration) used fo…

[The biological and immunochemical properties of monoclonal antibodies to the Venezuelan equine encephalomyelitis virus].

Five strains of hybrid cells producing highly active monoclonal antibodies (MCA) to Venezuelan equine encephalomyelitis (VEE) virus were generated. VEEC6 MCA had high virus-neutralizing and hemagglutinating activities attesting to their direction to the "critical" site of E2 glycoprotein. MCA VEEA5 directed to the same glycoprotein did not overlap spatially with the previous one. MCA VEEB5 and VEEA4 interacted with conformational virus determinants taking part in hemagglutination.

Immunosuppression by gramicidin S of experimental autoimmune uveoretinitis, pinealitis and autoimmune encephalomyelitis.

Using an in vitro lymphocyte proliferation assay we screened several cyclic peptide antibiotics (bacitracin, oleandomycin, capreomycin, colistin, virginiamycin, and gramicidin S) for their immunosuppressive activity. Gramicidin S (GrS) was found to inhibit [3H]-thymidine incorporation into concanavalin A-stimulated and E coli lipopolysaccharide-stimulated lymphocytes. In vivo studies, experimental autoimmune uveoretinitis (EAU) and pinealitis were induced in female Lewis rats by immunization with bovine S-antigen and experimental autoimmune encephalomyelitis (EAE) were induced by immunization …

Progressive encephalomyelitis with rigidity: a case report with magnetic resonance imaging findings.

A 52 year old woman presented with pain in the back and upper limbs and progressive weakness and sensory impairment of the upper and lower limbs. She developed frequent episodes of severe generalised muscle spasms associated with piloerection and hyperventilation. Nerve conduction studies and electromyography were normal. MRI demonstrated a lesion of the cervical spinal cord and lower brainstem. A biopsy of the cord revealed perivascular cuffing with mononuclear cells and inflammatory changes in the parenchyma, with increased numbers of microglia, reactive astrocytes and plasma cells. Followin

Agents of equine viral encephalomyelitis: correlation of serum and cerebrospinal fluid antibodies.

A survey was conducted by testing 115 paired equine serum and cerebrospinal fluid samples by hemagglutination-inhibition for antibodies to P … y into one, and intravenously into four ponies. Neurological signs associated with a nonsuppurative encephalomyelitis occurred in three ponies. Antibodies to Powassan virus were detected in sera of all animals but in cerebrospinal fluids of only two. Powassan virus was isolated from brain and spinal cord of only the intracerebrally inoculated animal.

[Isolation of glycoproteins of the Venezuelan equine encephalomyelitis virus and an evaluation of their immunogenic activity].

Isolation of purified Venezuelan equine encephalomyelitis virus glycoproteins by means of a new Soviet nonionic detergent, MESK, is described. The MESK detergent was shown to permit isolation of approximately 70% of virus particle glycoproteins. The resulting preparation had a high hemagglutinating activity, contained no admixtures of foreign proteins and was not infectious. The study of the immunogenicity of purified glycoproteins in experimental mice and rabbits showed them to be capable of inducing high levels of serum antibodies. The immunogenicity of the isolated glycoproteins was compara…

Antigen-induced suppression of mitogen responses and resistance to experimental autoimmune encephalomyelitis.

Random-bred Hartley and inbred Strain 2 and Strain 13 guinea pigs were inoculated for acute experimental autoimmune encephalomyelitis (EAE). Sixty-six percent (69/103) of the Hartleys developed signs of EAE while the remaining 34% (34/103) were resistant. No Strain 2 and all Strain 13 guinea pigs developed EAE. Histologic examination of nervous tissue revealed that susceptible Hartleys and Strain 13 and Strain 2 animals had lesions characteristic of EAE. Tissue from resistant Hartleys showed fewer and less severe changes. Lymphocyte-transformation assays with EAE-inducing and noninducing antig…

[Preparation and characteristics of temperature-sensitive mutants of the Venezuelan equine encephalomyelitis virus].

Fifteen ts mutants of Venezuelan equine encephalomyelitis (VEE) virus have been produced and studied. Two of them were isolated from a virus population grown in the presence of 5-fluorouracyl and 13 from a virus suspension treated with nitrous acid. The efficiency of plating 40 degrees/35 degrees of various mutants varied from 2 X 10(-2) to 6 X 10(-6). Three ts mutants had RNA- phenotype (RNA synthesis at a nonpermissive temperature was 1.5-3.3% of that of the wild type), 11 ts mutants had RNA+ phenotype (RNA synthesis decreased insignificantly, 32-75%), and one ts mutant was intermediate (18%…

Virions and virus-associated structures within dendrites in an experimental flavovirus encephalomyelitis.

In experimental yellow fever virus encephalomyelitis of adult albino mice, virions, and virus-associated structures were observed not only inside neuronal perikarya but also within dendrites of varied size. The finding permits the following explanations: (1) either the viral agent is synthesized in the nerve cell bodies and transported intradendritically in a proximodistal direction; or (2) virus morphogenesis takes place in neuronal perikarya and dendrites as well; or (3) both possiblities are equally valid. Some incidental findings were suggestive of virus release at postsynaptic dendrite me…

Venezuelan equine encephalomyelitis in an adult animal host. An electron microscopic study.

Adult white mice were inoculated intravenously with a virulent strain of Venzuelan equine encephalomyelitis virus. They were sacrificed sequentially, and cerebral tissue was collected for light and electron microscopic examination and virus titer determination. Viruses were first seen on the fifth day postinoculation infecting and arising from endothelial cells in the cerebrum. Subsequently, oligodendrocytes became infected, giving rise to mature virions. At this time in the infection, a particular electron-dense cell, probably representing a glial cell type, phagocytized mature virions. This …
