We assessed the impact of amyloid precursor protein (APP) gene locus duplications in early onset Alzheimer's disease in a Dutch population-based sample. Using real-time PCR and an in-house-developed multiplex amplicon quantification assay, we identified a genomic APP duplication in 1 out of 10 multigenerational families segregating early onset Alzheimer's disease. In this family, cerebral amyloid angiopathy (CAA) coincided with this disease. The duplicated genomic region included no other genes than APP and extended maximally over 0.7 Mb. In a sample of 65 familial early onset patients, we obs

BACKGROUND: Asperger syndrome is characterised by abnormalities in social interaction as well as repetitive and stereotyped behaviours and interests. The trait is thought to display complex inheritance, but in a subset of families the inheritance resembles the autosomal dominant model. Linkage to 3p14-24 has recently been reported in Asperger syndrome in Finnish families with a maximum multipoint NPL(all) of 3.32 at D3S2432.
METHODS: We have replicated linkage findings to 3p21-24 in 12 new extended Asperger syndrome families. Linkage analyses were performed separately for the 12 new families,

Progress in the past century has led to an improved understanding of the cause, pathophysiology and treatment of Marfan syndrome. The "Ghent criteria" constitute currently the most effective way of diagnosing or excluding Marfan syndrome. This system can also help to identify families with aortic aneurysms who do not have Marfan syndrome, but it should not be used to assess risk in such families. Despite the morbidity and mortality associated with Marfan syndrome, an appropriate multidisciplinary medical and surgical approach can improve and extend the lives of many patients. As knowledge of t

INTRODUCTION: New strategies have been arisen to set a rapid and effective screening for selection of microorganism with bioactive potential. This study suggests that combination of physicochemical pretreatments and taxonomic dereplication of microbial collections through MALDI-TOF MS, facilitates the detection of low abundance actinobacteria with potential as a source of antimicrobial agents.
MATERIAL AND METHODS: An unstudied microbial community from a tropical river sediment in Colombian Orinoquía is described, applying an extended cultivation strategy using physicochemical pretreatments,

The aggregation of α-synuclein (A-syn) has been implicated strongly in Parkinson's disease (PD). In vitro studies established A-syn to be a member of the intrinsically disordered protein (IDP) family. This protein undergoes structural interconversion between an extended and a compact state, and this equilibrium influences the mechanism of its aggregation. A combination of fluorescence resonance energy transfer (FRET) and fluorescence correlation spectroscopy (FCS) has been used to study the membrane induced conformational reorganization and aggregation of A-syn. Different structural and confor

Understanding how and why someone dies unexpectedly is key to bereaved family members. The coronial process in England investigates instances where the cause of death is unknown, violent or unnatural and/or occurred in state detention. Families are held to be at the centre of this process and the coroner's role has extended to concern about therapeutic jurisprudence, that is, how legal processes can minimise negative consequences for participants without jeopardising due process. Therapeutic jurisprudence involves unresolved tensions, however, and an epistemic power imbalance. Within the inque

BACKGROUND AND PURPOSE: A recurrent duplication of chromosome 16p13.1 was associated with aortic dissection as well as with cervical artery dissection. We explore the segregation of this duplication in a family with familial aortic disease.
METHODS: Whole exome sequencing (WES) analysis was performed in a patient with a family history of aortic diseases and ischemic stroke due to an aortic dissection extending into both carotid arteries.
RESULTS: The index patient, his affected father, and an affected sister of his father carried a large duplication of region 16p13.1, which was also verified

Women with a personal or family history of breast or ovarian cancer are increasingly presenting for genetic cancer risk assessment (GCRA). To explore the personal and family impact of GCRA, four focus groups were conducted of women seen for risk assessment. Participants were 22 primarily non-Latina White women with a personal or family history of breast or ovarian cancer. Analysis of the data identified new themes related to balancing time to assimilate risk information with the need to make timely healthcare decisions, physicians' lack of sufficient genetic knowledge, and concern for daughter

OBJECTIVES: The COVID-19 pandemic substantially affected population mental health. We examined predictors of increased loneliness, reduced mental wellbeing, and elevated stress in Switzerland.
METHODS: Using the 2022 Swiss Health Survey, we ran weighted binary logistic regressions (weights representing 7,182,252 residents aged ≥15 years) to test sociodemographic, psychosocial, behavioral, and pandemic-related predictors of perceived change since before the pandemic; extended models added COVID-19 health variables. Predictors were entered in blocks.
RESULTS: Increased general anxiety since th

Sesbania gum, as a member of the galactomannan family, exhibits good thickening properties. However, its slow dispersion limits its applications. To address this limitation, carboxymethylation was performed to obtain carboxymethyl sesbania gum (CMSG) with different degrees of substitution (DS). After modification, both the bulk density and zeta potential increased with DS, and all CMSG samples exhibited markedly improved dissolution rate, up to a tenfold increase in dissolution rate compared with SG. Unlike the random coil conformation of native SG in water, CMSG exhibited a more extended and

We describe a patient with unilateral lattice corneal dystrophy (LCD) associated with a previously unreported pathogenic transforming growth factor beta-induced (TGFBI) variant (c.1864A>T, p.(Asn622Tyr)), contributing to the expanding phenotypic spectrum of non-R124C TGFBI-related LCD. A 58-year-old male patient presented with complaints of recurrent epithelial erosions and progressive vision loss in his right eye for the past 20 years. On slitlamp biomicroscopic examination of the patient's right eye, thick, branched lattice lines extended from the limbus to the central cornea. The lesions ex

In oncological disease, a definition of pain as total suffering emerges. This complex phenomenon is characterized by the simultaneous involvement of several dimensions (bodily, cognitive, emotional, family, social, and cultural) held together by a bond of mutual interdependence. Cancer pain is pervasive and affects all aspects of a person's life. It alters the individual's perspective and perception of the world, creating a sense of stagnation and uncertainty characterized by anguish and precariousness. It threatens the sense of personal identity and extends its influence on the entire relatio

Homelessness impacts entire families, with potential intergenerational consequences. Addressing family homelessness provides both immediate relief and long-term societal benefits. While various programs exist to mitigate homelessness, the United States (US) Department of Veterans Affairs' (VA) Supportive Services for Veteran Families (SSVF) program offers a distinctive model for combating homelessness among veterans by supporting their families as well. We analyzed VA SSVF administrative data from 2014 to 2022, covering over 800,000 program entries from all SSVF beneficiaries in the US, to des

Hereditary nonpolyposis colorectal cancer (HNPCC) is associated with highly penetrant germline mutations in mismatch repair genes. Due to a high lifetime risk in gene carriers for synchronous and for metachronous colorectal cancer and endometrial cancer in women, prophylactic and extended surgery are considered as options for gene carriers. A 54-year-old patient with a history of metachronous rectal cancer and a family history fulfilling the Amsterdam criteria presented with carcinoma of the cecum and highly dysplastic adenomas of the splenic flexure and descending colon. Acute rheumatic fever (ARF) and rheumatic heart disease (RHD) remain significant health issues for Māori and Pacific communities in Aotearoa New Zealand (NZ). Subcutaneous injection of penicillin (SCIP) enables injections to be given 10-weekly as an alternative to the standard four-weekly intramuscular (IM) injections. As part of a clinical trial involving participants with ARF who have been on SCIP for at least one year, we aim to explore treatment adherence, pain management, and quality of life for Māori and Pacific participants and their families (whānau). A community centred approach align

PURPOSE: We extended the Breast Cancer Surveillance Consortium (BCSC) version 2 (v2) model of invasive breast cancer risk to include BMI, extended family history of breast cancer, and age at first live birth (version 3 [v3]) to better inform appropriate breast cancer prevention therapies and risk-based screening.
METHODS: We used Cox proportional hazards regression to estimate the age- and race- and ethnicity-specific relative hazards for family history of breast cancer, breast density, history of benign breast biopsy, BMI, and age at first live birth for invasive breast cancer in the BCSC co…

OBJECTIVES: We sought to identify clinical characteristics that would negatively predict shunt failure, thus potentially obviating the need for further diagnostic workup or extended periods of observation. We hypothesized that viral symptoms and a patient history of epilepsy or chronic headaches would be negative predictors of shunt failure.
METHODS: Data were retrospectively collected for children 19 years or younger with a cerebrospinal fluid diverting shunt in their medical history or problem list who underwent neuroimaging during an ED visit from March 2008 to September 2016. Patients wer

Non-syndromic syndactyly is a heterogeneous group of limb malformations involving webbing of fingers and/or toes. There are at least nine non-syndromic types described in the literature. For the clinician and the genetic counsellor not having gathered experience with this malformation, it is rather tedious to identify the correct subtype for the patient's phenotype. We therefore present a protocol for clinical use, which visualises the malformation in a graphical way and thereby simplifies typing. In addition, this protocol provides a simple documentation system for reporting clinical data for

OBJECTIVE: To explore the putative connection between inclusion body myopathy, Paget disease, frontotemporal dementia (IBMPFD) and motor neuron disease (MND).
METHODS: Clinical, genetic, and EMG characterization of 17 patients from 8 IBMPFD families.
RESULTS: Limb weakness was the most common clinical manifestation (present in 15 patients, median onset age 38 years, range 25-52), with unequivocal evidence of upper motor neuron dysfunction in 3. EMG, abnormal in all 17, was purely neurogenic in 4, purely myopathic in 6, and mixed neurogenic/myopathic in 7. Cognitive/behavioral impairment was
