Exploration of materials suitable for single-chain variable fragment (scFv)-conjugated affinity silk and its novel application in preventing bacterial infections in ornamental fish.

The ornamental fish industry is highly prone to disease outbreaks caused by pathogenic microorganisms and parasites. Affinity silk is a material that contains a fibroin L-chain fused to a single-chain variable fragment (scFv) derived from a mouse monoclonal antibody (mAb). It is an effective affinity material with innovative potential for efficiently preventing infectious diseases in aquarium water. In this study, we explored materials suitable for developing anti-Aeromonas salmonicida affinity silk, associated devices for trapping pathogens in water, and the early detection of pathogens direc

Silk Scaffold Crypt-Villus Geometry Combined With L-WRN Feeder Cells Sustains Spatially Organized Epithelial Proliferation and Multilineage Differentiation.

The crypt-villus axis of the small intestine provides essential structural and biochemical cues that govern epithelial renewal, barrier function, and lineage specification. However, most existing in vitro models rely on flat monolayers or villus-only hydrogels, limiting their ability to capture stem cell niches and spatial organization. Here, we report silk fibroin scaffolds engineered with physiologically scaled crypt-villus topography through a combination of 3D-printed molds, vacuum-assisted silk infiltration, and water annealing. This approach reproducibly generated scaffolds with well-def

Core-shell microneedles Co-loaded with simvastatin and doxycycline for combined anti-infection and osteogenesis in diabetic periodontitis.

Diabetic periodontitis remains a formidable clinical challenge characterized by persistent infection, oxidative stress, and impaired alveolar bone regeneration. Here, we have developed an engineered core-shell microneedle (MN) patch that enables the staged release of doxycycline and simvastatin to achieve coordinated antibacterial and osteogenic effects within periodontal lesions. The outer polyvinyl alcohol (PVA) shell rapidly dissolves upon insertion into gingival tissue, enabling burst release of doxycycline for immediate bacterial eradication. Subsequently, the polyvinylpyrrolidone (PVP) c

Multi-Component Functionalized Bifidobacterium Longum Hydrogel for Multi-Target Integrated Therapy of Colitis-Associated Anxiety and Depression.

Inflammatory bowel diseases (IBDs) are frequently accompanied by anxiety and depression, largely driven by perturbed gut-brain axis signaling. However, current oral therapies remain constrained by the spatial and functional separation between intestinal inflammation and central nervous system dysfunction. Here, we present a comprehensive gut-brain dual region integrated therapeutic strategy based on functionalized Bifidobacterium longum hydrogel (INPs@BL@Gel), in which baicalin and tyrosine are coordinated with Fe(III) to form infinite coordination polymers (ICPs), coated with inulin, assemble

Defining optimal electrospun membranes to enhance biological activities of human endometrial MSCs.

INTRODUCTION: Human endometrial mesenchymal stem cells (H-EMSCs) can inhibit endometrial fibrosis and repair damaged endometrium. However, direct cell injection into dam-aged endometrium shows limited cell survival. Cell seeding onto biomaterial-based electrospun membranes could improve H-EMSCs' survival and prolong their stay at the damaged endometrium. Polycaprolactone (PCL), silk fibroin (SF) and hyaluronic acid (HA) are synthetic or natural biomaterials used by the biomedicine field, however, their effects on the biological activities of H-EMSCs remain unclear. METHODS: In this study, CD9

Probiotic nanocomposite materials with excellent resistance, inflammatory targeting, and multiple efficacies for enhanced treatment of colitis in mice.

The occurrence of inflammatory bowel disease (IBD) is relevant to impaired intestinal mucosal barrier and disordered gut microbiota, subsequently leading to excessive production of reactive oxygen species (ROS) and elevated levels of inflammatory factors. Traditional therapies focus on inhibiting inflammation, but the vast majority involve non-targeted systemic administration, whose long-term use may result in potential side effects. Oral microbial therapy has exhibited great application prospects currently in IBD treatment; however, its progress has been slowed by issues with deficient bioava

"Attractive" Treatment for Abdominal Aortic Aneurysm Repair: Magnetic Localization of Silk-Iron Packaged Extracellular Vesicles.

Abdominal aortic aneurysm (AAA) is a dilatation of the distal aorta to a diameter of 50% or more of its normal size of about 2 cm. Risk of aortic rupture can be nearly eliminated with either open surgery or endovascular repair. Procedural risks limit the value of these interventions unless the diameter of the aneurysm has reached a critical threshold (established as 5.5 cm in men or 5.0 cm in women). Thus, patients are monitored until this threshold is reached. Approximately 80% of small AAA will grow and exceed the threshold, providing a therapeutic window for altering this natural history an

Comparing Biomechanical Properties of Bioabsorbable Suture Anchors: A Comprehensive Review.

Suture anchors (SAs) are medical devices used to connect soft tissue to bone. Traditionally these were made of metal; however, in the past few decades, bio-absorbable suture anchors have been created to overcome revision surgeries and other complications caused by metallic SAs. This systematic review aims to analyze the biomechanical properties of these SAs to gain a better understanding of their safety and utilization. A comprehensive systematic review that adhered to the PRISMA guidelines was conducted. Primary outcomes were that the pull-out strength of SAs, the rate of degradation secondar

Engineering the human tympanic membrane: lessons from mechanics, modelling, and materials.

The tympanic membrane (TM) plays a pivotal role in auditory transduction by converting acoustic signals into mechanical energy. Damage to the TM, whether from chronic perforations, infections, trauma, or pathological changes such as tympanosclerosis, can significantly impair its biomechanical function and lead to conductive hearing loss. Tympanoplasty remains the standard intervention, traditionally utilising autologous grafts such as temporalis fascia or cartilage. However, these biological materials present limitations, including donor site morbidity, variable mechanical properties, and pote

Development of dual-functional core-shell electrospun mats with controlled release of anti-inflammatory and anti-bacterial agents for the treatment of corneal alkali burn injuries.

In this study, a novel dual-drug carrier for the co-administration of an anti-inflammatory and antibiotic agent consisting of core-shell nanofibers for the treatment of cornea alkali burns was designed. The core-shell nanofibers were prepared via coaxial electrospinning of curcumin-loaded silk fibroin as the core and vancomycin-loaded chitosan/polyvinyl alcohol (PVA) as the shell. Electron microscopy (SEM and TEM) images confirmed the preparation of smooth, bead-free, and continuous fibers that formed clear core-shell structures. For further studies, nanofiber mats were cross-linked by heat tr

The proteomics of the freshwater pearl powder: Insights from biomineralization to biomedical application.

The freshwater pearl is one kind of valuable organic jewelry and traditional Chinese medicine (TCM). However, the molecular basis of matrix protein in pearl biomineralization and biomedical applications are largely unknown to date. In this study, the matrix proteins of water-soluble matrix, acid-soluble matrix and acid-insoluble matrix from the freshwater seedless pearl powder were detected using liquid chromatography-tandem mass spectrometry (LC-MS/MS) respectively, and identified against the transcriptomic database of the pearl sac. The results showed that a total of 190 proteins were identi

Extensive early mineralization of pre-osteoblasts, inhibition of osteoclastogenesis and faster peri-implant bone healing in osteoporotic rat model: principle effectiveness of bone-specific delivery of Tibolone as evaluated in vitro and in vivo.

Hydrophobic drug molecules pose a significant challenge in immobilization on super-hydrophobic metallic surfaces like conventional titanium implants. Pre-coating surface modifications may yield a better platform with improved wettability for such purposes. Such modifications, as depicted in this study, were hypothesized to provide the requisite roughness to assist deposition of polymers like silk fibroin (SF) as a drug-binding matrix in addition to significant improvement in early protein adsorption, which facilitates faster cellular adhesion and proliferation. A silk-based localized drug deli

On-demand biocatalytic dual oxygen spray reprograms the hypoxia-ferroptosis axis to accelerate chronic wound healing.

Effective clinical management of chronic hypoxic wounds remains challenging due to persistent hypoxic signaling, oxidative stress, and cellular dysfunction. Hypoxia-induced ferroptosis has recently gained attention as a key molecular driver of impaired healing in chronic wounds such as ischemic and diabetic wounds. Herein, a bioinspired self-oxygenating dual spray incorporating silk fibroin, aloe vera mucilage, and hemoglobin in the primary SAH spray and hydrogen peroxide in the secondary Hp spray is developed. Upon application, the dual oxygenation spray (SAH-Hp) triggers peroxide-responsive

Niosome-mediated melatonin delivery via SilMA/GelMA microneedles promotes healing of sleep deprivation-aggravated oral ulcers.

Sleep disturbance disrupts immune homeostasis and impairs oral mucosal healing, yet effective local immunomodulatory strategies remain limited. Here, we identify excessive neutrophil extracellular traps (NETs) as a key mediator linking sleep deprivation to delayed oral wound closure. NETs-associated proteases compromise epithelial barrier function and suppress repair cell migration. To address this, we developed a melatonin-loaded niosome-incorporated silk fibroin methacrylate (SilMA)/gelatin methacryloyl (GelMA) composite hydrogel microneedle (Nio-MT@S-G MN) for localized melatonin delivery a

Engineered GelMA Microgels for Locoregional Doxorubicin Release and Apoptosis Induction in Oral Squamous Cell Carcinoma.

Localized chemotherapeutic delivery remains a critical unmet need in head and neck squamous cell carcinoma (HNSCC), where the systemic administration of chemotherapeutics such as doxorubicin is constrained by rapid clearance, low tumor retention, and dose-limiting toxicities. These challenges necessitate more effective, less invasive drug-delivery platforms to improve therapeutic outcomes and patient well-being. In this study, we engineered photocrosslinked gelatin methacrylate (GelMA) microgels via a water-in-oil emulsion approach as an injectable, biodegradable platform for sustained locoreg

Radioactive Iodine-131 aerosols induce rat pulmonary fibrosis via miR-34c-5p/ARHGAP1 axis-mediated fibroblast activation.

Following nuclear accidents, Iodine-131 (131I) released as radioactive aerosols can be inhaled into the lungs, leading to radiation-induced pulmonary fibrosis (RIPF). Our previous studies have identified miR-34c-5p as a potential biomarker for 131I internal exposure, which is significantly downregulated upon 131I irradiation and is known to negatively regulate fibrogenic processes. However, it remains unclear whether inhaled 131I aerosols induce RIPF by downregulating miR-34c-5p in fibroblasts and consequently activating these effector cells. Here, 131I-labeled silk fibroin microparticles (SFM

A glucose-triggered cascade catalytic adhesive hydrogel enables mild photothermal-biofilm eradication for infected diabetic wounds.

Delayed healing in chronic diabetic wounds is primarily driven by the hyperglycemic microenvironment and persistent bacterial biofilm infection, making biofilm eradication under mild therapeutic conditions highly challenging. Herein, we designed an injectable glucose-responsive cascade catalytic adhesive hydrogel (STU@Au) for mild-temperature photothermal-catalytic synergistic therapy of infected diabetic wounds. This hydrogel integrates gold nanoclusters with glucose oxidase-like activity and photothermal properties into an adhesive matrix (STU) constructed from silk fibroin and tannic acid v
