Application of comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry method to identify potential biomarkers of perinatal asphyxia in a non-human primate model.

Perinatal asphyxia is a leading cause of brain injury in infants, occurring in 2-4 per 1000 live births. The clinical response to asphyxia is variable and difficult to predict with current diagnostic tests. Reliable biomarkers are needed to help predict the timing and severity of asphyxia, as well as response to treatment. Two-dimensional gas chromatography-time-of-flight-mass spectrometry (GC×GC-TOFMS) was used herein, in conjunction with chemometric data analysis approaches for metabolomic analysis in order to identify significant metabolites affected by birth asphyxia. Blood was drawn befor

Ectopic pituitary transplants stimulate synthesis and release of follicle-stimulating hormone in golden hamsters.

It was previously reported that PRL can stimulate FSH release in immature female rats and PRL-deficient male dwarf mice. We have examined the effects of PRL-producing ectopic pituitary grafts on plasma FSH and LH levels in adult male golden hamsters during exposure to both long (14 h of light, 10 h of darkness) and short (5 h of flight, 19 h of darkness) photoperiods. In both groups, the presence of one pituitary graft resulted in a significant elevation of pituitary and plasma FSH levels. Although there were no statistically significant changes in the concentration of LH in the pituitary or p

Zasp52 in Drosophila melanogaster indirect flight muscles can serve as a model system to investigate the function of clinical variants causing myopathies.

The sarcomere is the basic contractile unit of muscle fibers, bordered by Z-discs. In Drosophila, Z-disc structure and maintenance rely on actinin, and the Z-band alternatively spliced PDZ-motif protein 52 (Zasp52), a member of the Alp/Enigma family. Zasp52 has been shown to bind actin, actinin, and itself, but its full range of functions remains unclear, as a null mutant has not been previously analyzed. To address this, we generated a CRISPR-based null mutant of Zasp52, deleting most of the ∼50 kb locus. Null mutants are viable but flightless and display severe defects in indirect flight mus

Choice behavior of Drosophila facing contradictory visual cues.

We studied the underlying neural mechanism of a simple choice behavior between competing alternatives in Drosophila. In a flight simulator, individual flies were conditioned to choose one of two flight paths in response to color and shape cues; after the training, they were tested with contradictory cues. Wild-type flies made a discrete choice that switched from one alternative to the other as the relative salience of color and shape cues gradually changed, but this ability was greatly diminished in mutant (mbm1) flies with miniature mushroom bodies or with hydroxyurea ablation of mushroom bod

Physiology of a microgravity environment selected contribution: effects of spaceflight during pregnancy on labor and birth at 1 G.

The events of parturition (labor, delivery, maternal care, placentophagia, and onset of nursing) were analyzed in female Norway rats (Rattus norvegicus) flown on either 11- or 9-day-long spaceflights beginning at the approximate midpoint of their pregnancies. Each space shuttle flight landed on the 20th day of the rats' pregnancies, just 48-72 h before parturition. After spaceflight, dams were continuously monitored and recorded by time-lapse videography throughout the completion of parturition and onset of nursing (days 22 and 23). Analyses of parturition revealed that, compared with ground c

Different neural pathways coordinate Drosophila flight initiations evoked by visual and olfactory stimuli.

To determine the role played by the giant fiber interneurons (GFs) in coordinating the jumping stages of visually elicited and olfactory-induced fight initiation we have recorded extracellularly from the cervical connective nerve during flight initiation. A spike is recorded from the cervical connective upon brain stimulation that has the same threshold as does activation of the tergotrochanteral muscle (TTM) and dorsal longitudinal muscles (DLMs). A consistent time interval occurs between the spike and activation of the TTM. Thus, the spike probably results from activity in the GFs. The time

Distribution and characteristics of methicillin-resistant staphylococci from non-clinical environments in southern Vietnam.

BACKGROUND: Methicillin-resistance in pathogenic staphylococci such as Staphylococcus aureus (MRSA) and Staphylococcus epidermidis (MRSE) represents a major global health concern. Acquisition of methicillin-resistance (mec) genes enables these organisms to evade β-lactam antibiotics and has facilitated their spread beyond hospitals to the wider community and environment. Non-pathogenic Methicillin-resistant staphylococci (MRS), which inhabit various environments, are considered potential reservoirs of mec genes. However, information on MRS in non-clinical settings remains limited. METHODS: Thi

Genetic control of temperament traits across species: association of autism spectrum disorder risk genes with cattle temperament.

BACKGROUND: Temperament traits are of high importance across species. In humans, temperament or personality traits correlate with psychological traits and psychiatric disorders. In cattle, they impact animal welfare, product quality and human safety, and are therefore of direct commercial importance. We hypothesized that genetic factors that contribute to variation in temperament among individuals within a species will be shared between humans and cattle. Using imputed whole-genome sequence data from 9223 beef cattle from three cohorts, a series of genome-wide association studies was undertake

Comparison of Staphylococcus pettenkoferi Isolated from Human Clinical Cases and Cat Carriers Regarding Antibiotic Susceptibility and Biofilm Production.

Staphylococcus pettenkoferi (S. pettenkoferi) is a rare opportunistic bacterium not commonly found in healthy individuals or animals. S. pettenkoferi has increasing clinical significance in both veterinary and human medicine due to its multidrug resistance and biofilm-forming ability. This study analyzed 12 isolates of S. pettenkoferi collected from humans and cats and identified them using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and 16S rRNA and partial rpoB gene sequencing. All of the S. pettenkoferi were phenotypically resistant to penicil

Characterization of Selenium Species in the Shijimi Clam.

Selenium is an essential trace element for humans and animals. Fish and shellfish are known to be rich in selenium and suppose to be an effective selenium source. In this study, we characterized the selenium species in the Shijimi clam (Corbicula japonica), which is a typical clam eaten in Japan. The Shijimi clam contains a relatively high concentration of selenium (3.5 µg-selenium/g-dry Shijimi). Approximately 30% of the total selenium in the Shijimi clam meat was extractable with water, while selenium in the Shijimi clam was hardly extracted with ethanol, chloroform and hexane. Based on an u

Mobilization of mercury from lean tissues during simulated migratory fasting in a model songbird.

The pollutant methylmercury accumulates within lean tissues of birds and other animals. Migrating birds catabolize substantial amounts of lean tissue during flight which may mobilize methylmercury and increase circulating levels of this neurotoxin. As a model for a migrating songbird, we fasted zebra finches (Taeniopygia guttata) that had been dosed with 0.0, 0.1, and 0.6 parts per million (ppm) dietary methylmercury and measured changes in blood total mercury concentrations (THg) in relation to reductions in lean mass. Birds lost 6-16% of their lean mass during the fast, and THg increased an

Early diagnosis of Irkut virus infection using magnetic bead-based serum peptide profiling by MALDI-TOF MS in a mouse model.

Early diagnosis is important for the prompt post-exposure prophylaxis of lyssavirus infections. To diagnose Irkut virus (IRKV) infection during incubation in mice, a novel method using magnetic bead-based serum peptide profiling by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) has been established. For this test, serum peptides were concentrated by adsorption to and elution from the magnetic bead-based weak cation ion exchanger. Mass spectrograms obtained by MALDI-TOF MS were analyzed using ClinProTools bioinformatics software. Construction of the

[The mechanism of releasing and bio-behavioral significance of the hypothalamo-hypophyso-adrenal gland reaction to the environment (alarm reaction)].

Laboratory experimentation has demonstrated the large increase in the rate of circulating norepinephrin (N.E.), in a parallel with an improvement in cardiovascular dynamism of suprarenalectomized rabbits under the action of injection IV of hydrocortisone. As from this ascertainment, the author attempts to make a synthesis of known experimental facts demonstrating that next to a regulation in constancy (with negative retroaction) of the C.R.F. leads to A.C.T.H. leads to cortisol system, there exists a servomechanism (control external to the system) as from the activity of the nervous systems as

Repeated Exposures to Air Travel-Relevant Hypobaria Induce Anxiety-Like Behavior and Alter Functional Connectivity and White Matter Integrity in a Ferret Model of Traumatic Brain Injury.

Under-vehicle blast (UVB) caused by landmine detonation induces a distinct traumatic brain injury (TBI) and can be accompanied by head impact. Injured soldiers often undergo multiple flights after injury to access medical care and return to duty. Previous work has shown that low air pressure during air travel (hypobaria [HB]) exacerbates neurological injury, but the effects of one or more HB exposures on chronic brain injury are unknown. We hypothesized that multiple HB exposures after TBI would result in worse outcomes than 0-1 exposures. Sedated male ferrets underwent UVB and controlled cort

Exploring Aeromonas veronii in Migratory Mute Swans (Cygnus olor): A Debut Report and Genetic Characterization.

Aeromonas veronii (A. veronii) is a ubiquitous bacterium in terrestrial and aquatic environments. It has a significant impact on animal and human health, with it becoming an emerging crucial pathogen worldwide. However, there have been no reports of mute swan infections. In the present study, after an observation of pathological changes, one bacterial strain isolated from a dead migratory mute swan was identified as A. veronii HNZZ-1/2022 based on its morphology, matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS), and sequence analysis of the 16S rRNA a

Predictive statistical models for monitoring antimicrobial resistance spread in the environment using Apis mellifera (L. 1758) colonies.

The rise of antimicrobial resistance (AMR) is one of the most relevant problems for human and animal health. According to One Health Approach, it is important to regulate the use of antimicrobials and monitor the spread of AMR in the environment as well. Apis mellifera (L. 1758) colonies were used as bioindicators thanks to their physical and behavioural characteristics. During their foraging flights, bees can intercept small particles, including atmospheric particulate matter, etc., and also microorganisms. To date, the antimicrobial surveillance network is limited to the sanitary level but l

Immunization of pigs to prevent disease in humans: construction and protective efficacy of a Salmonella enterica serovar Typhimurium live negative-marker vaccine.

Zoonotic infections caused by Salmonella enterica serovar Typhimurium pose a constant threat to consumer health, with the pig being a particularly major source of multidrug-resistant isolates. Vaccination, as a promising approach to reduce colonization and shedding, has been scarcely used, as it interferes with current control programs relying on serology as a means of herd classification. In order to overcome this problem, we set out to develop a negative-marker vaccine allowing the differentiation of infected from vaccinated animals (DIVA). Applying an immunoproteomic approach with two-dimen
