The current study used zinc oxide nanoparticles (ZnO-NPs) to protect the tomato plant against Fusarium wilt. Gamma rays were used to synthesize ZnO-NPs, and the designed ZnO-NPs were characterized using high-resolution transmission electron microscopy (HRTEM), scanning electron microscope (SEM), dynamic light scattering (DLS), energy-dispersive X-ray spectroscopy (EDX), and ultraviolet-visible (UV-Vis.) spectroscopy. We found that the 20 kGy dose is the most effective for ZnO-NPs synthesis, with the highest O.D. = 1.65 (diluted 3 times) at 400 nm. The scale of ZnO-NPs ranged from 10.45 to 75.2…

Fusarium head blight (FHB) is a global cereal disease caused by a complex of Fusarium species. Both Fusarium graminearum and F. asiaticum are the causal agents of FHB in China. F. asiaticum is the predominant species in the Middle-Lower Reaches of the Yangtze River (MLRYR) and southwest China. Therefore, detecting F. asiaticum in a timely manner is crucial for controlling the disease and preventing mycotoxins from entering the food chain. Here, we combined rapid genomic DNA extraction, recombinase polymerase amplification, Cas12a cleavage, and lateral flow detection techniques to develop a met…

BACKGROUND: Filamentous fungi are ubiquitous in plants, water, and soil. The predominant fungi that infect the human cornea include Fusarium and Aspergillus species. The onset of fungal endophthalmitis is indolent, and typically takes weeks to months to develop after corneal infection. We report a case of Fusarium infection complicating rheumatic keratitis that acutely progressed to endophthalmitis during intravenous tocilizumab therapy.
CASE PRESENTATION: A 65-year-old female patient was referred to our department due to pain and decreased vision in her left eye. Slit-lamp examination showed…

Although it is well known that miRNAs play crucial roles in multiple biological processes, there is currently no evidence indicating that milRNAs from Fusarium oxysporum f. sp. lycopersici (Fol) interfere with tomato resistance during infection. Here, using sRNA-seq, we demonstrate that Fol-milR1, a trans-kingdom small RNA, is exported into tomato cells after infection. The knockout strain ∆Fol-milR1 displays attenuated pathogenicity to the susceptible tomato cultivar 'Moneymaker'. On the other hand, Fol-milR1 overexpression strains exhibit enhanced virulence against the resistant cultivar 'Mo…

Fusarium dieback (FD) is a new vascular disease of hardwood trees caused by Fusarium spp. and other associated fungal species which are vectored by two recently introduced and highly invasive species of ambrosia beetle (Euwallacea spp. nr. fornicatus). One of these ambrosia beetles is known as the polyphagous shot hole borer (PSHB) and the other as the Kuroshio shot hole borer (KSHB). Together with the fungi that they vector, this pest-disease complex is known as the shot hole borer-Fusarium dieback (SHB-FD) complex. Mitigation of this pest-disease complex currently relies on tree removal; how…

The suppressive potentials of Bacillus and Paenibacillus strains isolated from the tomato phyllosphere were investigated to obtain new biocontrol candidates against Fusarium crown and root rot of tomato. The suppressive activities of 20 bacterial strains belonging to these genera were examined using seedlings and potted tomato plants, and two Paenibacillus strains (12HD2 and 42NP7) were selected as biocontrol candidates against the disease. These two strains suppressed the disease in the field experiment. Scanning electron microscopy revealed that the treated bacterial cells colonized the root…

Microorganisms produce volatile organic compounds (VOCs) which mediate interactions with other organisms and may be the basis for the development of new methods to control plant-parasitic nematodes that damage coffee plants. In the present work, 35 fungal isolates were isolated from coffee plant rhizosphere, Meloidogyne exigua eggs and egg masses. Most of the fungal isolates belonged to the genus Fusarium and presented in vitro antagonism classified as mutual exclusion and parasitism against the nematode-predator fungus Arthrobotrys conoides (isolated from coffee roots). These results and the …

Fusarium graminearum is a major pathogen of wheat and barley, causing fusarium head blight (FHB) and contaminating grain with harmful mycotoxins. Azole fungicides (demethylation inhibitors) are among the key tools available for managing F. graminearum infections. In an attempt to characterize how resistance to azole fungicides arises, we performed an experimental evolution study that imposed selection by exposing F. graminearum to increasing concentrations of prothioconazole (PTZ), tebuconazole (TBF), a combination of both fungicides (CMB), or a no-drug control. All evolved lineages exceeded t…

BrlA is a multifunctional transcription factor identified in specific clades of filamentous fungi, notably within the Hyphomycetes. It is widely involved in sporulation, cell wall stress response and pathogenicity, and plays an important role in fungal growth and development. To investigate the biological function of the FsbrlA in Fusarium solani, this study employed a split-marker homologous recombination strategy to directionally construct FsbrlA knockout and complementation strains. Through analyses of growth and development phenotypes, determination of cell wall components and trehalose co…

Deoxynivalenol (DON), fumonisin B1 (FB1), and zearalenone (ZEN) are typical fusarium mycotoxins that occur worldwide in foodstuffs, posing significant health hazards to humans and animals. Single and combined exposure of DON, FB1, and ZEN leads to intestinal toxicity but the toxicology mechanism research is still limited. In this study, we explored the cytotoxicity effects of DON, FB1, ZEN, and their combination in rat intestinal epithelial cell line 6 (IEC-6) cells. Cell viability results showed that the cytotoxicity potency ranking was DON > ZEN > FB1. Furthermore, both DON + FB1 and DON + Z…

Fusarium head blight, caused by Fusarium graminearum, is one of the most threatening fungal diseases of cereals worldwide. Current practices for control of F. graminearum are not always efficient, as epidemics still occur and there is low resistance in wheat varieties. Therefore, novel antifungal targets must be discovered by analyzing the molecular interaction between F. graminearum and its host. Fungal extracellular vesicles (EVs) are small membrane-bound compartments (30-1000 nm) that carry macromolecules and support fungal virulence, hence the disruption of EV production could lead to redu…

Currently, researchers are looking for ways to replace synthetic pesticides with substances of natural origin. Essential oils are produced by plants, among other things, to protect against pathogens, which is why there is interest in their use as fungicides. This experiment assessed the composition of essential oils from a commercial source, their impact on the development of mycelium of pathogens of the Fusarium genus, and the possibility of using them as a pre-sowing treatment. Grains of winter wheat (Triticum aestivum L.) and corn (Zea mays L.) were inoculated with a suspension of mycelium …

Fusarium graminearum, which causes Fusarium head blight (FHB) in cereals, is one of the most devastating fungal diseases by causing great yield losses and mycotoxin contamination. A major bioactive ingredient, venturicidin A (VentA), was isolated from Streptomyces pratensis S10 mycelial extract with an activity-guided approach. No report is available on antifungal activity of VentA against F. graminearum and effects on deoxynivalenol (DON) biosynthesis. Here, VentA showed a high antagonistic activity toward F. graminearum with an EC50 value of 3.69 μg/mL. As observed by scanning electron micro…
