Strategies to innovate emergency care of status epilepticus.

Generalized Convulsive status epilepticus (SE) is a neurological emergency because prolonged convulsions can cause respiratory compromise and neuronal injury. Compromised GABA-mediated inhibition is a defining feature of SE, and many current therapies are benzodiazepines, which are allosteric modulators of GABA-A receptors. Many patients with medically refractory epilepsy are at risk for SE. Newly available nasally delivered benzodiazepines: midazolam and diazepam given for seizure clusters may prevent SE. Although three different benzodiazepines, diazepam, lorazepam and midazolam terminate ea

Uridine decreases morphine-induced behavioral sensitization by decreasing dorsal striatal dopamine release possibly via agonistic effects at GABAA receptors.

Uridine, a potential endogenous neuromodulator, has been demonstrated to interact with the dopaminergic system and to regulate dopamine-related behaviors. The present study investigated the effects of uridine on morphine-induced hyperactivity and behavioral sensitization and on modulating dopaminergic neurotransmission in mice, which may help to understand how uridine and its metabolites act as modulators of the GABAA receptors. The results showed that either systemic (30 or 100mg/kg) or central (30, 100 or 300nM) uridine administration significantly attenuated the hyperactivity induced by acu

GABAergic modulation of diffuse noxious inhibitory controls (DNIC): a test by use of lorazepam.

Diffuse noxious inhibitory controls (DNIC) are an important supra-spinal mechanism of pain inhibition. Neurotransmitters and modulators involved in DNIC are serotonin and endogenous opioids. The influence of substances binding to the GABA(A) receptor complex, which has been suggested to play an important role in descending pain inhibition on DNIC has not yet been investigated. The aim of the present study was to find out whether the inhibitory action of DNIC might also be mediated by GABAergic mechanisms. Therefore, DNIC modulation via GABAergic mechanisms was studied in a double blind, placeb

gamma-Aminobutyric acid receptor-regulated 36Cl- flux in mouse cortical slices.

An improved neurochemical assay for gamma-aminobutyric acid (GABA) function has been developed using tracer-radioactive chloride efflux in mouse cortical slices. Careful maintenance of the brain slice viability resulted in a 3-fold stimulation of 36Cl- efflux rate by the GABA agonist muscimol (EC50 = 3 microM), comparable to electrophysiologic and other chloride flux preparations. The shape of the muscimol dose-response curve was shallow, suggestive of negative cooperativity or heterogeneous receptors, but tissue uptake of agonist, possible diffusion barriers and apparent functional desensitiz

Impact of acamprosate on plasma amyloid-β precursor protein in youth: a pilot analysis in fragile X syndrome-associated and idiopathic autism spectrum disorder suggests a pharmacodynamic protein marker.

BACKGROUND: Understanding of the pathophysiology of autism spectrum disorder (ASD) remains limited. Brain overgrowth has been hypothesized to be associated with the development of ASD. A derivative of amyloid-β precursor protein (APP), secreted APPα (sAPPα), has neuroproliferative effects and has been shown to be elevated in the plasma of persons with ASD compared to control subjects. Reduction in sAPPα holds promise as a novel molecular target of treatment in ASD. Research into the neurochemistry of ASD has repeatedly implicated excessive glutamatergic and deficient GABAergic neurotransmissio

Effects of pregnanolone and flunitrazepam on the retention of response sequences in rats.

Neuroactive steroids produce effects similar to other GABA(A) modulators (e.g., benzodiazepines and barbiturates) and have a large therapeutic potential; however, a greater understanding of the effects of these substances on learning and memory is needed. To specifically assess the effects of a neurosteroid on memory, pregnanolone (1-18 mg/kg) was administered to male Long-Evans rats responding under a repeated acquisition and delayed-performance procedure in which different 4-response sequences were acquired and then retested after varying delays. Responding was maintained under a second-orde

GABAergic mechanisms are involved in the antihyperalgesic effects of carbamazepine and oxcarbazepine in a rat model of inflammatory hyperalgesia.

BACKGROUND/AIMS: The purpose of this study was to investigate the involvement of GABAergic mechanisms in the antihyperalgesic effect of carbamazepine and oxcarbazepine by examining the effect of bicuculline (GABA(A) receptor antagonist) on these effects of antiepileptic drugs. METHODS: Rats were intraplantarly (i.pl.) injected with the proinflammatory compound concanavalin A (Con A). A paw-pressure test was used to determine: (1) the development of hyperalgesia induced by Con A; (2) the effects of carbamazepine/oxcarbazepine on Con A-induced hyperalgesia, and (3) the effects of bicuculline on

Flavonoids as GABAA receptor ligands: the whole story?

Benzodiazepines are the most widely prescribed class of psychoactive drugs in current therapeutic use, despite the important unwanted side effects that they produce, such as sedation, myorelaxation, ataxia, amnesia, and ethanol and barbiturate potentiation and tolerance. They exert their therapeutic effects via binding to the benzodiazepine binding site of gamma-aminobutyric acid (GABA) type A receptors, and allosterically modulating the chloride flux through the ion channel complex. First isolated from plants used as tranquilizers in folkloric medicine, some natural flavonoids have been shown

Combatting psoriasis with nanomedicine: gamma-amino butyric acid-chitosan nanoparticles as a targeted anti-psoriatic therapy.

The chronic inflammation and oxidative stress are the most deleterious pathogenic factors of psoriasis (PS). While gamma-aminobutyric acid (GABA) possesses well-documented immunomodulatory and antioxidant properties, its therapeutic potential is limited by bioavailability and targeting. Chitosan nanoparticles (CSNP) offer a promising drug delivery platform to overcome GABA limitations. This study aimed to augment the dermatological efficacy of GABA by encapsulation within CSNPs to overcome its skin penetration limitations, thereby creating an advanced transdermal delivery system for sustained

Elevated zero maze: a paradigm to evaluate antianxiety effects of drugs.

Elevated zero maze is a modification of the elevated plus maze model of anxiety in rodents. The novel design comprises an elevated annular platform with two opposite, enclosed quadrants and two open quadrants, removing any ambiguity in the interpretation of the time spent in the central square of the traditional design and allowing uninterrupted exploration. In the present study, we validated elevated zero maze as a tool to study antianxiety activity, using various standard anxiolytics belonging to different pharmacological groups, such as benzodiazepines, barbiturates, alcohol etc., and compa

Neurochemical and functional reorganization of the cognitive-ear link underlies cognitive impairment in presbycusis.

Recent studies suggest that the interaction between presbycusis and cognitive impairment may be partially explained by the cognitive-ear link. However, the underlying neurophysiological mechanisms remain largely unknown. In this study, we combined magnetic resonance spectroscopy (MRS) and resting-state functional magnetic resonance imaging (fMRI) to investigate auditory gamma-aminobutyric acid (GABA) and glutamate (Glu) levels, intra- and inter-network functional connectivity, and their relationships with auditory and cognitive function in 51 presbycusis patients and 51 well-matched healthy co

Accounting for individual differences in the response to tDCS with baseline levels of neurochemical excitability.

There is now considerable evidence that applying a small electrical current to the cerebral cortex can have wide ranging effects on cognition and performance, and may provide substantial benefit as a treatment for conditions such as depression. However, there is variability across subjects in the extent to which stimulation modulates behaviour, providing a challenge for the development of applications. Here, we employed an individual differences approach to test if baseline concentrations of the neurochemicals GABA and glutamate are associated with an individual's response to transcranial dire

GABAergic modulation of human social interaction in a prisoner's dilemma model by acute administration of alprazolam.

Recent work in neuroeconomics has used game theory paradigms to examine neural systems that subserve human social interaction and decision making. Attempts to modify social interaction through pharmacological manipulation have been less common. Here we show dose-dependent modification of human social behavior in a prisoner's dilemma model after acute administration of the γ-aminobutyric acid (GABA)-A modulating benzodiazepine alprazolam. Nine healthy adults received doses of placebo, 0.5, 1.0, and 2.0 mg alprazolam in a counterbalanced within-subject design, while completing multiple test bloc

The neurobiology and control of anxious states.

Fear is an adaptive component of the acute "stress" response to potentially-dangerous (external and internal) stimuli which threaten to perturb homeostasis. However, when disproportional in intensity, chronic and/or irreversible, or not associated with any genuine risk, it may be symptomatic of a debilitating anxious state: for example, social phobia, panic attacks or generalized anxiety disorder. In view of the importance of guaranteeing an appropriate emotional response to aversive events, it is not surprising that a diversity of mechanisms are involved in the induction and inhibition of anx

Research progress on the biological activity and applications of gamma-aminobutyric acid.

Gamma-aminobutyric acid (GABA) is a four‑carbon, non-proteinogenic amino acid widely found in vertebrates, plants, and tiny critters. This compound acts as the primary inhibitory neurotransmitter in the nervous system. Recent studies have shown that the physiological functions of GABA extend beyond neural regulation, as it also exhibits multiple biological activities, including antioxidant, antihypertensive, and antidiabetic effects. GABA exerts multi-targeted effects through several mechanisms, including regulating enzyme activity, modulating the expression of related signaling pathways, and

Differential effects of two major neurosteroids on cerebellar and cortical GABA(A) receptor binding and function.

Cerebellar and cerebrocortical A-type γ-aminobutyric acid (GABA(A)) receptors were examined in mice and rats. In wild-type mouse cerebellum, the agonists GABA and gaboxadol exerted heterogeneous displacement of [(3)H]ethynylbicycloorthobenzoate (EBOB) binding with nanomolar and submicromolar affinities. In mouse cerebella lacking α6 subunits (α6KO), nanomolar displacement by GABA agonists was absent, while micromolar displacement was potentiated to 12-fold by 0.3μM 5α-tetrahydrodeoxycorticosterone (5α-THDOC). In α6KO cerebellum, 60% of [(3)H]EBOB binding was neurosteroid-insensitive, while 5α-

A study comparing the actions of gabapentin and pregabalin on the electrophysiological properties of cultured DRG neurones from neonatal rats.

BACKGROUND: Gabapentin and pregabalin have wide-ranging therapeutic actions, and are structurally related to the inhibitory neurotransmitter GABA. Gabapentin, pregablin and GABA can all modulate voltage-activated Ca2+ channels. In this study we have used whole cell patch clamp recording and fura-2 Ca2+ imaging to characterise the actions of pregabalin on the electrophysiological properties of cultured dorsal root ganglion (DRG) neurones from neonatal rats. The aims of this study were to determine whether pregabalin and gabapentin had additive inhibitory effects on high voltage-activated Ca2+ c

GABA: The Peacekeeper Neurotransmitter-Gut-microbiota Derived Origins and Salivary Biomarker Detection Using Elisa.

BACKGROUND: The human body is a highly integrated biological system in which the gut flora wields regulatory control beyond simple digestion. Intestinal bacteria control neuronal and behavioral processes via the gut-brain axis; microbial gamma-aminobutyric acid (GABA) synthesis seems to be an essential controller of sleep and central nervous system activity. Increasing data show that neuroactive substances produced from bacteria influence mental health results, circadian rhythm, and sleep structure. SUMMARY: This review investigates the function of gut-derived GABA in sleep control, the part

The immediate alteration of cerebellar Glx/GABA and cerebello-thalamo-cortical connectivity in patients with schizophrenia after cerebellar TMS.

Cerebellar dysfunction is a key aspect of schizophrenia, with the cerebello-thalamo-cortical (CTC) hyperconnectivity serving as a neural signature. Abnormalities in gamma-aminobutyric acid (GABA) and glutamate + glutamine (Glx) levels also contribute to this pathology. Transcranial magnetic stimulation (TMS) applied to the cerebellum shows potential in alleviating schizophrenia symptoms, possibly by modulating functional connectivity or neurotransmitter levels. This study aims to explore the immediate effects of cerebellar TMS on CTC circuitry and neurotransmitter levels to elucidate its thera

γ-Aminobutyric Acid (GABA) enhances heat tolerance in rice via integrated antioxidant defense, metabolic homeostasis and heat shock response.

BACKGROUND: Global warming has increased the frequency and intensity of extreme heat events, posing a serious threat to crop growth and productivity. Although γ‑aminobutyric acid (GABA) is recognized as a bioactive factor functioning in plant stress adaptation, its specific role and underlying mechanisms under heat stress remain poorly understood in rice. RESULTS: In this study, we demonstrate that exogenous GABA significantly enhances heat tolerance in rice seedlings in a dose‑dependent manner. Application of 0.1 mM GABA effectively alleviated heat-induced growth inhibition and mitigated oxi
