Investigating the effect of chitosan on the expression of P5CS, PIP, and PAL genes in rapeseed (Brassica napus L.) under salt stress.

Chitosan, a non-toxic and biodegradable compound, enhances plant growth and secondary metabolite production, presenting innovative approaches to mitigating plant stress. Salinity, a common abiotic stress, significantly impairs plant growth and development. This study investigates the effects of chitosan on the physiological, biochemical, and gene expression responses of salt-stressed Brassica napus L. exposed to NaCl concentrations of 0, 50, 100, and 150 mM. Chitosan was applied as a foliar spray at concentrations of 0, 5 and 10 mg/L. The research focuses on gene expression changes in P5CS, PI

Comprehensive validation of reference genes in Solanum nigrum under abiotic stresses: Identifying pendimethalin-responsive genes for herbicide resistance insights.

Black nightshade (Solanum nigrum L.), a globally invasive weed of the Solanaceae family, exhibits remarkable adaptability to diverse abiotic stresses. Despite its ecological and agricultural significance, validated reference genes (RGs) for reliable RT-qPCR normalization in this species remain undefined. This study systematically evaluated eight candidate RGs under six abiotic stresses, including cold, drought, salinity, heat, cadmium (Cd) exposure and pendimethalin treatment, using four computational algorithms (Delta Ct, geNorm, NormFinder and BestKeeper) integrated through RefFinder. Stress

Inhibition of NPR1 Leads to Shoot Growth Improvement under Low-Calcium Conditions in Arabidopsis.

Under low-Ca conditions, plants accumulate salicylic acid (SA) and induce SA-responsive genes. However, the relationship between SA and low-Ca tolerance remains unclear. Here, we demonstrated that the inhibition or suppression of nonexpressor of pathogenesis-related 1 (NPR1) activity, a major regulator of the SA signaling pathway in the defense response, improves shoot growth under low-Ca conditions. Furthermore, mutations in phytoalexin-deficient 4 (PAD4) or enhanced disease susceptibility 1 (EDS1), which are upstream regulators of NPR1, improved shoot growth under low-Ca conditions, suggesti

The zinc-finger transcription factor BcMF20 and its orthologs in Cruciferae which are required for pollen development.

Brassica campestris Male Fertility 20 (BcMF20) is a typical zinc-finger transcription factor that was previously isolated from flower buds of Chinese cabbage (Brassica campestris ssp. chinensis). By applying expression pattern analysis, it can be known that BcMF20 was specifically and strongly expressed in tapetum and pollen, beginning from the uninucleate stage, and was maintained during the mature-pollen stage. As BcMF20 was highly conserved in Cruciferae, it can be indicated that this zinc-finger transcription factor is important during the growth of Cruciferae. In this study, 12 C2H2-type

beta-Aminobutyric acid-induced protection of Arabidopsis against the necrotrophic fungus Botrytis cinerea.

The non-protein amino acid beta-aminobutyric acid (BABA) protects numerous plants against various pathogens. Protection of Arabidopsis plants against virulent pathogens involves the potentiation of pathogen-specific defense responses. To extend the analysis of the mode of action of BABA to necrotrophs we evaluated the effect of this chemical on Arabidopsis plants infected with the gray mold fungus Botrytis cinerea. BABA-treated Arabidopsis were found to be less sensitive to two different strains of this pathogen. BABA protected mutants defective in the jasmonate and ethylene pathways, but was

Single-nucleus RNA sequencing reveals the developmental trajectory and transcriptional regulatory networks of trichomes in Artemisia annua.

Sweet wormwood (Artemisia annua) produces and deposits artemisinin in its glandular trichomes but the molecular mechanisms of trichome development remain poorly understood. Here, we conduct single-nucleus RNA sequencing of 10-day-old A. annua seedlings, revealing the specific RNA expression of the leaf trichome cells. The reconstruction of the developmental trajectory of trichome cells not only identifies novel expressed genes, but also elucidates trichome development involved in photosynthesis, auxin biosynthesis, and cutin biosynthesis. By integrating the developmental trajectory of trichome

Integrated transcriptomic and metabolomic analyses elucidate the molecular mechanisms underlying quality formation during Rosa acicularis fruit ripening.

Fruits of Rosa acicularis are recognized for their high nutritional value owing to accumulation of diverse bioactive metabolites. However, the physiological processes and regulatory mechanisms underlying quality formation remain incompletely understood. In this study, developmental changes in fruit quality were assessed across four stages: the mature green stage (MGS), breaker stage (BS), maturity stage (MS), and ripening stage (RS), using morphological, physicochemical, and antioxidant analyses. To further elucidate the molecular regulatory basis of ripening, three representative stages (MGS,

Integrated transcriptomic and biochemical analyses reveal mechanisms of drought adaptation in strawberry (Fragaria × ananassa L.).

Drought is a major abiotic stress that limits strawberry productivity worldwide, yet the underlying physiological and molecular mechanisms of cultivar-specific tolerance remains poorly understood. This study integrated physiological, biochemical, and transcriptomic analyses to dissect drought responses of two contrasting cultivars of Fragaria × ananassa: drought-tolerant 'Kara' and drought-sensitive 'E22'. Plants were exposed to both progressive water-deficit (Def) and PEG-induced osmotic stress under controlled greenhouse conditions. Physiologically, the two cultivars diverged substantially:

Responses of grapevine cells to physiological doses of ethanol, including induced resistance to heat stress.

Grapevine is naturally exposed to stresses like heat, drought, and hypoxia. A recent study found very low oxygen levels inside grape berries, linked to ethanol content. Other studies have established a link between ethanol and tolerance to various stresses: heat, drought and high salinity. The causes of such tolerances are not well understood. In our study, 3-week-old Gamay calli, Vitis vinifera, were characterized for endogenous oxygen and ethanol concentrations. A global transcriptomic study was conducted to explore the response of grapevine cells to ethanol, which, to our knowledge, is the

Transcriptome analysis of the phosphate starvation response sheds light on strigolactone biosynthesis in rice.

Phosphorus (P) is a major element required for plant growth and development. To cope with P shortage, plants activate local and long-distance signaling pathways, such as an increase in the production and exudation of strigolactones (SLs). The role of the latter in mitigating P deficiency is, however, still largely unknown. To shed light on this, we studied the transcriptional response to P starvation and replenishment in wild-type rice and a SL mutant, dwarf10 (d10), and upon exogenous application of the synthetic SL GR24. P starvation resulted in major transcriptional alterations, such as the

Comparative transcriptome analysis reveals ecological adaption of cold tolerance in northward invasion of Alternanthera philoxeroides.

BACKGROUND: Alternanthera philoxeroides (alligator weed) is a highly invasive alien plant that has continuously and successfully expanded from the tropical to the temperate regions of China via asexual reproduction. During this process, the continuous decrease in temperature has been a key limiting environmental factor. RESULTS: In this study, we provide a comprehensive analysis of the cold tolerance of alligator weed via transcriptomics. The transcriptomic differences between the southernmost population and the northernmost population of China were compared at different time points of cold t

Nutrient depletion as a key factor for manipulating gene expression and product formation in different branches of the flavonoid pathway.

The content of flavonoids increases in response to nitrogen and phosphorus depletion in plants. Manipulation of these macronutrients may therefore be used to control the levels of desirable compounds and improve plant quality. Key enzymes in the shikimate pathway, which feeds precursors into the flavonoid pathway, are regulated post-translationally by feedback from aromatic amino acids, and possibly by redox control through photosynthesis. Use of microarrays for global transcript analysis in Arabidopsis has revealed that transcript levels are less influenced by mineral nutrients in the shikima

Characterization, expression profiling, and functional analysis of a Populus trichocarpa defensin gene and its potential as an anti-Agrobacterium rooting medium additive.

The diverse antimicrobial properties of defensins have attracted wide scientific interest in recent years. Also, antimicrobial peptides (AMPs), including cecropins, histatins, defensins, and cathelicidins, have recently become an antimicrobial research hotspot for their broad-spectrum antibacterial and antifungal activities. In addition, defensins play important roles in plant growth, development, and physiological metabolism, and demonstrate tissue specificity and regulation in response to pathogen attack or abiotic stress. In this study, we performed molecular cloning, characterization, expr

Novel epidermal-cortical pattern and root apical silencing of VcCPC promoter activity in Arabidopsis might suggest a joint contribution to hairless roots in blueberry.

BACKGROUND: Root hairs are specialized tubular extensions that play a pivotal role in nutrient absorption and plant-soil interactions. Typically, they originate from epidermal cells, which differentiate into either hair cells or non-hair cells. In blueberry (Vaccinium spp.), the roots are devoid of root hairs. To examine the mechanisms responsible for the hairless root phenotype, this study conducted anatomical and molecular biological investigations on the hairless roots of blueberry. RESULTS: The epidermal cells of blueberry roots exhibit uniformity in size and cross-sectional dimensions, w

A comparative framework for trichome and trichome-derived epidermal appendage development across model glycophytes and halophytes.

This review compares trichome and trichome-derived epidermal appendage development across model glycophytes and halophytes, revealing that conserved regulatory modules are independently rewired into lineage specific circuits to produce diverse epidermal structures. In halophytes, salt glands and epidermal bladder cells further integrate these developmental programs with ion transport and stress physiology, offering entry points for engineering crop stress resilience. Trichome-derived epidermal structures have arisen repeatedly across angiosperms, yet the mechanisms by which conserved epidermal

MdATG5a-mediated autophagy improves apple performance under low light.

Apples are sun-loving plants, and excessive canopy closure in orchards leads to poor ventilation and reduced light penetration, ultimately limiting yield and fruit quality. Autophagy is a conserved mechanism for recycling cellular materials and energy in eukaryotes and is known to participate in multiple stress responses. However, whether autophagy contributes to low-light adaptation remains largely unclear. Here, we show that MdATG5a enhances the tolerance of apple plants to low-light conditions. Under low light, MdATG5a-overexpressing (OE) plants exhibited higher photosynthetic capacity, gre

Independent recruitment of enzymes for phenylpropanoid sucrose ester biosynthesis in Polygala tenuifolia.

Plants produce a vast diversity of specialized metabolites and can independently recruit enzymes from distinct evolutionary origins to catalyze identical reactions. Sibiricose A5 and A6, two phenylpropanoid sucrose esters (PSEs) with potent antidepressant and neuroprotective activities, are commonly isolated from the roots of the medicinal plant Polygala tenuifolia. Here, we confirmed the root-specific accumulation of these PSEs and assembled a high-quality, chromosome-scale genome for P. tenuifolia (678.38 Mb), achieving a BUSCO completeness of 99.30%.
