Investigating the association between polyunsaturated fatty acids and osteomyelitis by Mendelian randomization.

Osteomyelitis, characterized by bone inflammation and infection, poses a significant global health burden. This Mendelian randomization (MR) study investigates the causal relationship between polyunsaturated fatty acids (PUFAs) and osteomyelitis risk. By using GWAS data from 114,999 individuals, we explore specific PUFAs and their genetic variations using Inverse variance weighted (IVW), MR-Egger and weighted median methods. The results reveal a suggestive association between genetically predicted higher docosahexaenoic acid (DHA) and omega-6 levels with increased osteomyelitis risk. Conversel

Congenital myopathies: pathophysiological mechanisms and promising therapies.

Congenital myopathies (CMs) are a kind of non-progressive or slow-progressive muscle diseases caused by genetic mutations, which are currently defined and categorized mainly according to their clinicopathological features. CMs exhibit pleiotropy and genetic heterogeneity. Currently, supportive treatment and pharmacological remission are the mainstay of treatment, with no cure available. Some adeno-associated viruses show promising prospects in the treatment of MTM1 and BIN1-associated myopathies; however, such gene-level therapeutic interventions target only specific mutation types and are not

A multiancestry genome-wide association study of unexplained chronic ALT elevation as a proxy for nonalcoholic fatty liver disease with histological and radiological validation.

Nonalcoholic fatty liver disease (NAFLD) is a growing cause of chronic liver disease. Using a proxy NAFLD definition of chronic elevation of alanine aminotransferase (cALT) levels without other liver diseases, we performed a multiancestry genome-wide association study (GWAS) in the Million Veteran Program (MVP) including 90,408 cALT cases and 128,187 controls. Seventy-seven loci exceeded genome-wide significance, including 25 without prior NAFLD or alanine aminotransferase associations, with one additional locus identified in European American-only and two in African American-only analyses (P

Sex-specific genetic influence on thyroid-stimulating hormone and free thyroxine levels, and interactions between measurements: KNHANES 2013-2015.

BACKGROUND: Although a wide range of genetic influences on thyroid-stimulating hormone (TSH) and free thyroxine (fT4) levels have been reported, sex differences in the genetic influences have not been well described. METHODS: We assessed TSH and fT4 levels in 2,250 subjects without thyroid peroxidase antibody, with data obtained from the Korea National Health and Nutrition Examination Surveys (KNHANES) conducted from 2013 to 2015. Using variance decomposition methods, the variation of TSH and fT4 levels was divided into genetic and environmental components common to both sexes, and to males a

SLIT2 and ANGPTL3 as putative mediators linking obesity to atrial fibrillation: A Mendelian randomization study.

The observational association between obesity and atrial fibrillation has been well-established. However, its causal mediators underlying this association remain unclear. This study aimed to investigate the causal relationship between body mass index (BMI), as well as potential mediating factors underlying this association, using Mendelian randomization (MR). Genetically predicted BMI was significantly associated with an increased risk of atrial fibrillation (inverse-variance weighted odds ratio [OR] = 1.350, 95% confidence interval [CI] = 1.268-1.436, P < .001). Sensitivity analyses using mul

Type 2 Diabetes Mellitus and Amyotrophic Lateral Sclerosis: Genetic Overlap, Causality, and Mediation.

CONTEXT: Understanding phenotypic connection between type II diabetes (T2D) mellitus and amyotrophic lateral sclerosis (ALS) can offer valuable sight into shared disease etiology and have important implication in drug repositioning and therapeutic intervention. OBJECTIVE: This work aims to disentangle the nature of the inverse relationship between T2D mellitus and ALS. METHODS: Depending on summary statistics of T2D (n = 898 130) and ALS (n = 80 610), we estimated the genetic correlation between them and prioritized pleiotropic genes through a multiple-tissue expression quantitative trait lo

Gut microbiota-metabolite axis in gastric cancer: Genetic causality and Ruminococcus subgroup shifts.

Observational studies have long suggested an association between gut microbiota dysbiosis and gastric cancer, yet the causal relevance and underlying biological pathways remain poorly defined. Here, we applied a comprehensive Mendelian randomization (MR) framework to gut microbiota and gastric cancer GWAS summary statistics to identify 14 microbial taxa significantly associated with gastric cancer risk. Sensitivity analyses, including tests for horizontal pleiotropy, heterogeneity, and genetic co-localization, revealed no strong evidence of shared causal variants (H4 < 2%); however, convergent

The causal relationship between polycystic ovary syndrome and structural changes in brain: a Mendelian randomization study.

The impact of sex hormones on the brain has been widely recognized. Polycystic ovary syndrome (PCOS) is frequently accompanied by abnormal hormone. Furthermore, psychiatric disorders are among the most common comorbidities associated with PCOS. However, whether PCOS induces alterations in brain structure remains unclear. Mendelian randomization (MR) was conducted to determine this causal association and pinpoint specific brain regions prone to PCOS. Genome wide association study (GWAS) data from the FinnGen database, for PCOS phenotypes from 39,004 PCOS patients and 731,830 controls were inclu

Low-Density Lipoprotein Cholesterol and Dementia Risk: Integrating Mendelian Randomization and Target Trial Emulation Within the Heart-Brain Axis.

BACKGROUND: The heart-brain axis links cardiovascular and neurodegenerative disease through shared vascular and inflammatory mechanisms. Although low-density lipoprotein cholesterol (LDL-C) is an established causal factor in atherosclerotic cardiovascular disease (ASCVD), its relationship with dementia remains uncertain, with midlife elevations associated with increased risk but late-life associations often appearing null or inverse. To address this cholesterol paradox, we integrated mendelian randomization (MR) with an active-comparator new-user target trial emulation. METHODS: We applied a

Circulating inflammatory cytokines and the risk of myasthenia gravis: a bidirectional Mendelian randomization study.

BACKGROUND: Myasthenia gravis (MG) is an autoimmune disorder of the neuromuscular junction. Increasing evidence has suggested inflammation is involved in the pathogenesis of MG, but whether it is the cause or a downstream effect remains unclear. In this study, a two-sample Mendelian randomization (TSMR) analysis was performed to explore the causal relationship between 91 circulating inflammatory cytokines and MG. METHOD: In this study, the data of 91 circulating inflammatory cytokines from 4824 Europeans and the largest GWAS database of MG (1873 patients and 36370 controls) were used to scree

Investigating the causal relationship between ankylosing spondylitis and osteoporosis in the European population: a bidirectional Mendelian randomization study.

BACKGROUND: Ankylosing Spondylitis (AS) is an inflammatory condition affecting the spine, which may lead to complications such as osteoporosis (OP). Many observational studies have demonstrated a close relationship with strong evidence between OP and AS. The combination of AS and OP is already an indisputable fact, but the exact mechanism of AS complicated with OP is unclear. To better prevent and treat OP in patients with AS, it is necessary to understand the specific mechanism of OP in these patients. In addition, there is a study showing that OP is a risk factor for AS, but the causal relat

Polygenic Health Index, General Health, and Pleiotropy: Sibling Analysis and Disease Risk Reduction.

We construct a polygenic health index as a weighted sum of polygenic risk scores for 20 major disease conditions, including, e.g., coronary artery disease, type 1 and 2 diabetes, schizophrenia, etc. Individual weights are determined by population-level estimates of impact on life expectancy. We validate this index in odds ratios and selection experiments using unrelated individuals and siblings (pairs and trios) from the UK Biobank. Individuals with higher index scores have decreased disease risk across almost all 20 diseases (no significant risk increases), and longer calculated life expectan

A meta-analysis uncovers the first sequence variant conferring risk of Bell's palsy.

Bell's palsy is the most common cause of unilateral facial paralysis and is defined as an idiopathic and acute inability to control movements of the facial muscles on the affected side. While the pathogenesis remains unknown, previous studies have implicated post-viral inflammation and resulting compression of the facial nerve. Reported heritability estimates of 4-14% suggest a genetic component in the etiology and an autosomal dominant inheritance has been proposed. Here, we report findings from a meta-analysis of genome-wide association studies uncovering the first unequivocal association wi

Causal associations between hormone replacement therapy and brain structure: Evidence from large-scale Mendelian randomization and double machine learning.

BACKGROUND: Hormone replacement therapy (HRT) is widely prescribed for the management of hormone deficiency, particularly during menopause, yet its causal effects on human brain structure remain incompletely understood. Observational studies have reported heterogeneous associations, underscoring the need for robust causal inference. METHODS: We applied an integrated causal framework combining two-sample Mendelian Randomization (MR) and Double Machine Learning (DML) to evaluate the effects of four HRT-related exposures-age at initiation, age at cessation, ever-use of HRT, and a composite medic

Assessing genetic causal relationship between milk fat content and chronic kidney disease: a two-sample Mendelian randomization study.

This study investigates the genetic causal relationship between milk fat content and chronic kidney disease (CKD) using publicly available genome-wide association study (GWAS) data. Single nucleotide polymorphisms (SNPs) significantly associated with milk fat content were identified following rigorous quality control procedures. A two-sample Mendelian randomization (MR) approach was used to analyze the relationship between three milk fat levels and CKD risk. The inverse variance weighting (IVW) method with random effects served as the primary analysis, supported by MR-Egger, weighted median, s
