Reduction in Gonad Development and Sperm Motility in Male Brown Planthopper Nilaparvata lugens via RNAi-Mediated Knockdown of tramtrack.

The brown planthopper Nilaparvata lugens, a major rice pest, threatens global food security through rapid reproduction. This study investigates the role of the tramtrack (ttk) gene in male reproductive development and spermatogenesis using RNA interference (RNAi). Gene expression analysis revealed higher ttk levels in testes. RNAi-mediated knockdown of ttk in fourth-instar male nymphs reduced its expression by up to 80%, leading to severely impaired gonad development. Testes, vas deferens, and accessory glands in treated males exhibited 8-89% volume reductions compared to controls, accompanied

Synthesis and Anti-insect Activity of Triazole-Containing Amide Derivatives.

The outbreak of crop diseases and insect pests, especially hemipteran piercing insects, poses a major threat to global food security. This study was designed to synthesize a series of novel flonicamid (FL) derivatives using an active substructure splicing strategy. Their insecticidal activities were evaluated against key pests, including Myzus persicae, Bemisia tabaci and Tetranychus urticae. The LC50 values of compounds 3ea, 3fa, 3ga, and 5cab against B. tabaci were 69.8 mg/L, 186 mg/L, 91.9 mg/L, and 112 mg/L, respectively. For M. persicae, the LC50 values of compounds 3aa, 5gaa, and 5fab we…

Virtual screening, molecular dynamics and binding energy-MM-PBSA studies of natural compounds to identify potential EcR inhibitors against Bemisia tabaci Gennadius.

Whitefly (Bemisia tabaci Gennadius) is a hemipteran phyto polyphagous sucking insect pest which is an important pest of cotton that causes economic losses to the crop by reducing its yield and quality. Ecdysteroids such as 20-hydroxy ecdysone (20-E), play a significant role in larval moulting, development, and reproduction in pterygota insects. Receptor of 20-E, that is Ecdysone Receptor (BtEcR) of Bemisia tabaci has been targeted to prevent fundamental developmental processes. To identify potent inhibitors of BtEcr, 98,072 natural compounds were retrieved from ZINC database. A structure-based…

Effect of the level of "Candidatus Liberibacter solanacearum" infection on the development of zebra chip disease in different potato genotypes at harvest and post storage.

Potato psyllid (Bactericera cockerelli Sulc)-transmitted "Candidatus Liberibacter solanacearum" (Lso) has been negatively impacting the potato industry in the United States as well as other potato-producing countries. Lso has been linked to a condition known as zebra chip (ZC) that affects yield and quality of potato tubers. Efforts to find sources of resistance to ZC have primarily focused on greenhouse evaluations based on a single inoculation time prior to harvest. Plant response to infection, however, could be influenced by the developmental stage of the host plant, and ZC may continue to

In vitro redox activity of haemozoin and β-haemozoin interacting with the following antimalarials: artemether, lumefantrine and quinine.

OBJECTIVE: Malaria parasites invade, grow and multiply inside erythrocytes and obtain nourishment from haemoglobin. Then, the released haem group is oxidized to haematin and inert dimeric haemozoin bio-crystals form, which provides the parasite a unique way to avoid the toxicity associated with the haem group. Therefore, antimalarial drugs are designed to inhibit dimer formation; however, recent electrochemical studies indicate that an inert dimer also promotes a toxic oxidizing environment. Therefore, this work explores drug reactivity in the presence of monomers and dimers to evaluate their

On the taxonomic structure and variability of Macropsidius involutus Dlabola, 1963 (Homoptera: Cicadellidae: Macropsinae)-morphology and calling signal pattern.

Variability of genitalia shape and male calling signal pattern in Macropsidius involutus from 10 localities in South-eastern Europe (the Crimea, the Lower Volga Region), Kazakhstan, and Kyrgyzstan was investigated. Variability of pygofer processes, penis shape, and signal pattern clearly has a geographic component, but none of these traits clearly supports subdividing this species into different subspecies unequivocally. M. involutus was found at altitudes from 0 up to ca. 2000 m a. s. l. in steppes and deserts in the plains and in steppe zones of mountains. Apparently, different environmental

Feeding of Whitefly on Tobacco Decreases Aphid Performance via Increased Salicylate Signaling.

BACKGROUND: The feeding of Bemisia tabaci nymphs trigger the SA pathway in some plant species. A previous study showed that B. tabaci nymphs induced defense against aphids (Myzus persicae) in tobacco. However, the mechanism underlying this defense response is not well understood. METHODOLOGY/PRINCIPAL FINDINGS: Here, the effect of activating the SA signaling pathway in tobacco plants through B. tabaci nymph infestation on subsequent M. persicae colonization is investigated. Performance assays showed that B. tabaci nymphs pre-infestation significantly reduced M. persicae survival and fecundity

Competition between honeydew producers in an ant-hemipteran interaction may enhance biological control of an invasive pest.

Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae), is an invasive citrus pest in southern California, which secretes honeydew and has the potential to spread a lethal bacterial disease, huanglongbing, of citrus. In urban citrus, Argentine ant, Linepithema humile (Mayr) (Hymenoptera: Formicidae), also an invasive pest, tends honeydew-producing hemipterans. We used field data to determine whether the mutualistic relationship between L. humile and six established species of honeydew producers may hinder or favor the establishment of D. citri and its biological control with Tam…

Estimating the feeding rate of the bedbug Cimex lectularius in an infested room: an inexpensive method and a case study.

The common bedbug, Cimex lectularius L. (Hemiptera: Cimicidae), is a globally re-emerging pest that is playing an increasing role in legal disputes and compensation claims as a result of its unpleasant feeding activity. However, there is little information about the feeding frequency of bedbugs outside controlled laboratory cultures. Here, we present a simple method of estimating the average time since the last bloodmeal of individual female bedbugs in a single sampling event, applicable to a single bedbug harbourage or an entire room. Using the temperature-dependent rate of decrease in the ab…

An association between phlebotomine sandflies and aphids in the Peruvian Andes.

As the composition of natural sugars in the diet of adult sandflies (Diptera: Psychodidae: Phlebotominae) may affect the development of Leishmania (Kinetoplastida: Trypanosomatidae) in sandfly guts, and so play an important role in the epidemiology of leishmaniasis, there is increasing interest in the sources of sugars for wild sandflies. Advanced chromatography techniques have provided indirect evidence that wild sandflies feed on honeydew, a substance released by aphids (Hemiptera: Aphididae) when feeding. Our objective was to determine whether sandfly density can be influenced directly by t…

MicroRNA-driven regulatory networks in aphid ecological adaptation: integrating stress tolerance, dispersal plasticity, and population expansion.

Aphids (Hemiptera: Aphididae) are important agricultural pests and exhibit strong ecological adaptability, allowing them to persist under stress, disperse to new habitats, and rapidly increase population size. Recent advances in functional genomics have identified microRNAs (miRNAs) as key post-transcriptional regulators involved in these processes, yet their roles have remained fragmented across studies. Here, we synthesize current evidence into a "three-stage framework", encompassing population maintenance under stress, dispersal to new habitats, and population expansion upon establishment. …

Nuclear receptor E75 regulates moulting and ovarian development in Nilaparvata lugens.

E75, a member of the nuclear receptor gene family, regulates diverse biological processes, including ovarian development and fat body remodelling. The brown planthopper (Nilaparvata lugens Stål) (Hemiptera:Delphacidae), a major rice pest, poses a significant threat to rice production. However, the role of Nilaparvata lugens E75 (NlE75) in development, particularly ovarian development and its underlying mechanisms, remains largely unknown. In this study, we found that downregulation of NlE75 expression significantly reduced the eclosion rate, especially in the third-, fourth- and fifth-instar n…
