Therapeutic efficacy of recombinant tumor necrosis factor alpha in an experimental model of human prostatic carcinoma.

Prostatic carcinoma represents the second leading cause of cancer mortality in men and is responsible for over 25,000 deaths each year. Currently, no curative therapy is available for metastatic carcinoma of the prostate. The present studies were undertaken to assess the efficacy of recombinant tumor necrosis factor alpha (TNF) in the therapy of experimental prostatic carcinoma. TNF was cytotoxic to the prostate cancer cell lines PC3, DU145, and LNCAP but not benign prostatic epithelial and stromal cells in vitro. The sensitivity of the prostatic carcinoma lines to TNF-mediated cytotoxicity wa

Microbiological and clinical effects of a dentifrice containing zinc citrate and Triclosan in the human experimental gingivitis model.

A partial mouth experimental gingivitis model was employed to establish the effect of a dentifrice containing 0.2% Triclosan and 0.5% zinc citrate on the development of chronic gingivitis. In addition, changes in the plaque flora associated with the developing gingivitis have been monitored. Following a period of stringent oral hygiene, volunteers were allocated to 1 of 2 treatment groups. A toothshield was constructed to fit 4 posterior mandibular teeth. During the 21-day experimental period test or placebo dentifrice was applied to the experimental teeth via the tooth shield. The toothshield

Effect of human urinary colony-stimulating factor on experimental Legionella pneumophila infection in guinea pigs.

The effect of human urinary colony-stimulating factor (CSF-HU) on in-vitro phagocytosis of Legionella pneumophila, superoxide anion production and intracellular killing of Legion, pneumophila by guinea pig alveolar macrophages was studied. Alveolar macrophages when incubated with CSF-HU demonstrated enhanced phagocytic activity and superoxide production. The bactericidal activity of treated cells was enhanced when they were afterwards incubated with a low concentration of bacteria, but these cells failed to inhibit bacterial multiplication when they were incubated with bacteria in a 1:10 ratio

Human interstitial retinoid binding protein. A potent uveitopathogenic agent for the induction of experimental autoimmune uveitis.

Human interstitial retinoid binding protein (HIRBP) is a 136,000 m.w. photoreceptor cell protein which transports retinoids between the retina and the retinal pigment epithelium of the eye. The amino acid sequence of HIRBP suggests that the molecule consists of four continuous homology domains which arose by several gene duplications some 600 to 800 million years ago. When injected into susceptible animal species, including primates, it induces an experimental autoimmune uveitis (EAU), a predominantly T cell-mediated autoimmune disease of the retina and uveal tract of the eye, and the pineal g

Hepatic arterial infusion of human recombinant tumor necrosis factor-alpha. An experimental study in dogs.

Local and systemic toxicities associated with hepatic arterial infusion of human recombinant tumor necrosis factor (rTNF) were studied in healthy adult mongrel dogs. The animals received saline containing human serum albumin with or without rTNF (0.02, 0.2, or 2.0 mg/m2). Arteriograms were made, and blood samples were collected for hematologic and biochemical analyses at regular intervals. The dogs were killed at 1, 3, and 7 days postinfusion and complete necropsies were performed. Specimens were obtained from various tissues for histopathologic evaluation. Results indicated that all but the h

Rat renal mesenchymal tumor as an experimental model for human congenital mesoblastic nephroma: I. Induction.

The effect of low-dose (2 Gy) radiation on ethylnitrosourea (ENU)-induced neoplasms was studied in Sprague-Dawley and Holtzman strains of rats. With a 60 mg/kg dose of ENU administered on day 1 in Sprague-Dawley rats, 18.4% of the neoplasms induced were found in the kidney. When the same dose of ENU was given on day 10, the incidence of kidney tumors fell to 2.8%. Prior (2 Gy) radiation on day 9 enhanced kidney tumor induction to 16.1%, a trend also observed in the case of ENU-induced neural tumors. In Holtzman rats, 40 mg/kg ENU induced more kidney tumors (12.5%) when given on day 4 than on d

Normal development of the human female reproductive tract and alterations resulting from experimental exposure to diethylstilbestrol.

An in vivo model is described for the study of human uterovaginal development in the presence and absence of the teratogenic drug diethylstilbestrol (DES). Intact reproductive tracts from fragments of 29 human embryos and fetuses 5.0 to 17.7 weeks of age obtained after dilatation and curettage were grown for four weeks in vivo in athymic (nude) mice that were either untreated (control) or implanted subcutaneously with a DES pellet. Control specimens grown in vivo continued their anticipated morphogenesis for equivalent in-utero ages; the normal processes observed included fusion of the paired

Engraftment of precursor lesions of human cutaneous neoplasms onto C.B-17 SCID mice: a useful in vivo experimental model of carcinogenesis in human skin.

Using a full-thickness skin grafting technique, lesional skin from various human neoplastic and preneoplastic skin diseases was transplanted onto SCID (severe combined immunodeficiency) mice. Of 27 grafted lesions, 21 were successfully accepted by the mice and maintained in good condition. All these accepted grafts were finally excised 10-101 days after transplantation for histological examination. In most grafts, the characteristic histological configurations of each disease were well preserved. Immunohistochemical study using monoclonal antibodies to human blood group antigens ABH revealed t

The myokine irisin ameliorates the secretory dysfunction of pancreatic β cells in experimental and human type 2 diabetes.

Irisin is a myokine that enhances insulin secretion and β cell viability in both rodent and human β cells. Here, we investigated whether irisin preserves β cell functional mass in vivo in diabetic mice and ex vivo in human pancreatic islets from subjects with type 2 diabetes (T2D). In mice made diabetic by a high-fat diet and streptozotocin, irisin administration improved glycemic homeostasis by increasing islet insulin content and glucose-stimulated insulin secretion while also markedly stimulating β cell proliferation. In islets from T2D subjects, which typically display substantial structur

A methodological framework and experimental protocol for proactive human-robot collaboration with multimodal intention prediction and adaptive control.

Industry 5.0 requires collaborative robots that can anticipate operator needs to improve fluency and safety in assembly. However, many human-robot collaboration (HRC) systems still treat perception, intention inference, and control as separate components. This study presents a theoretical perception-cognition-action framework that explicitly couples multimodal intention prediction with proactive and adaptive control. Multimodal observations such as RGB-D vision, gaze, wrist force/torque, robot joint state, and previous robot action are encoded by a hybrid Convolutional Neural Network (CNN)- Lo

Generation of functional mesothelial cells from human iPSCs that restore peritoneal integrity in experimental peritoneal injury.

BACKGROUND: Long-term peritoneal injury results in loss of mesothelial integrity and impaired ultrafiltration, limiting the durability of peritoneal dialysis. Because primary human peritoneal mesothelial cells (PMCs) are difficult to obtain and expand, regenerative strategies for restoring peritoneal function have remained limited. METHODS: We developed a protocol to differentiate human induced pluripotent stem cells (iPSCs) into functional peritoneal mesothelial-like cells (iPMCs). The molecular and functional characteristics of iPMCs were evaluated using gene expression analyses and functio

A preliminary experimental study of venoactive compounds on human lymphatic endothelial cells: cellular response and therapeutic potential.

PURPOSE: Lymphedema is a common complication following surgical procedures, particularly in breast cancer patients. However, no effective medical treatment has been clearly established. Venoactive compounds (venoactive drugs, VADs) are frequently used as an alternative option, despite limited supporting evidence. This study aimed to evaluate their potential therapeutic role in lymphedema. METHODS: Human lymphatic endothelial cells (HLECs) were used to assess the effects of diosmin, calcium dobesilate, and Vitis vinifera extract. Cell viability was evaluated using the MTT assay to determine no

B3GNT3 is an oncogenic and prognostic biomarker in human tumors via pan-cancer analysis combined with experimental validation.

BACKGROUND: Beta-1,3-N-acetylglucosaminyltransferase 3 (B3GNT3) is a member of the glycosyltransferase family, which is widely distributed in the Golgi apparatus and plasma membrane. However, the role of B3GNT3 in human pan-cancer has not yet been systematically analyzed and evaluated. This study aims to assess the expression profiles of B3GNT3 in different types of cancers and its potential clinical significance through a pan-cancer analysis. METHODS: Public data were derived from The Cancer Genome Atlas (TCGA) program and The Genotype-Tissue Expression (GTEx) website. Data analysis relied o
