SMAD4 Palmitoylation Drives a Metabolic-Transcriptional Circuit to Promote Tumorigenesis and Confers Radiosensitivity in Pancreatic Cancer.

Pancreatic ductal adenocarcinoma (PDAC) is characterized by frequent SMAD4 inactivation and profound lipid metabolic rewiring, yet how these processes intersect especially in SMAD4+ PDAC remains elusive. Here, we identify palmitoylation as a previously unrecognized post-translational modification of SMAD4. Combining biochemical labeling, mutagenesis, and functional assays, we demonstrate that SMAD4 is palmitoylated at cysteine 363 by the acyltransferase ZDHHC22 and depalmitoylated by APT2. Mechanistically, palmitoylation enhances SMAD4 protein stability, facilitates the interaction of SMAD4 wi

Propionyl-CoA catabolism is a metabolic gatekeeper for fatty acid oxidation in pancreatic cancer.

Pancreatic ductal adenocarcinoma (PDAC) cells rely on fatty acid oxidation (FAO) for proliferation; however, the regulatory mechanisms governing this dependency and their clinical implications remain unclear. Here, we report that PDAC patients exhibit decreased plasma levels of propionyl-CoA intermediates, alongside accelerated propionyl-CoA catabolic activity within both human PDAC tumors and Pdx1-Cre/KrasG12D/+/Trp53R172H/+ (KPC) mouse tumors. Specifically, PDAC cells upregulate propionyl-CoA carboxylase (PCC) to accelerate propionyl-CoA catabolism, thereby establishing a metabolic signature

Comparison of molecular markers for determining the viability and infectivity of Cryptosporidium oocysts and validation of molecular methods against animal infectivity assay.

BACKGROUND: Globally, disinfectants are widely used to intervene in the dissemination of Cryptosporidium oocysts. However, extensive investigations of oocyst inactivation by various disinfectants are not feasible due to the limitations imposed by animal infectivity methods. Molecular techniques provide an alternative strategy; however, non-metabolic genes have been used as markers for determining viability/infectivity. METHODS: In this study we used amyloglucosidase (AG) - a metabolic protein - as a marker to determine viability/infectivity of Cryptosporidium. Oocysts were exposed to 6% hydro

n of 1 trial for an ornithine transcarbamylase deficiency carrier.

Ornithine transcarbamylase deficiency (OTCD) is an X-linked disorder of the urea cycle. It is often fatal in affected males. Treatment for affected individuals includes dietary protein restriction, activation of alternative pathways of nitrogen excretion and L-arginine supplementation. Depending on the amount of X chromosome inactivation skewing, females show variable clinical manifestations, and sometimes the need for treatment, including medications, is unclear. We conducted an n of 1 randomized controlled trial on an obligate OTC carrier. The treating physician and patient were blinded to t

Pineal metabolic reaction to retinal photostimulation in ganglionectomized rats.

The aim of the present work was to test the pineal gland metabolic reactivity to nocturnal retinal short term photic stimulation in superior cervical ganglionectomized rats. The experimental support for this work is the appearance of a transitory post synaptic hyperactivity in the pineal gland, during the anterograde degenerating process of the conarii sympathetic nerve fibers after surgical removal of the cell body. In this situation the pineal gland is deafferented from the peripheral sympathetic nervous system keeping intact, however, the direct central connections to the deep pineal/lamina

Mechanisms of first- to fourth-generation neonicotinoid resistance in Myzus persicae from Beijing peach orchards: Roles of target-site mutations and detoxification metabolism.

Myzus persicae is a globally destructive pest that has evolved severe resistance to neonicotinoids. However, the resistance status and underlying mechanisms in peach orchards of Beijing, China remain largely unknown. In this study, the resistance of six field populations collected from Beijing peach orchards against five neonicotinoids (representing first- to fourth-generation compounds) was investigated, and the molecular mechanisms were elucidated. Bioassays revealed resistance to imidacloprid (15.99-30.97-fold), acetamiprid (9.16-50.79-fold), sulfoxaflor (1.28-25.26-fold), thiamethoxam (6.1

The role of methylglyoxal detoxification in Mycobacterium tuberculosis fitness and pathogenesis.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), constitutes a major global health challenge. The pathogenesis of Mtb can be largely attributed to its sophisticated survival strategies within host macrophages. Methylglyoxal (MGO), a ubiquitous reactive dicarbonyl metabolite that spontaneously reacts with biopolymers forming advanced glycation end products (AGEs), togethers with reactive oxygen species (ROS) damage cellular events. The glyoxalase system serves as the primary metabolic pathway for MGO detoxification, mitigating carbonyl stress induced by excess MGO. The mycobacteri

Formaldehyde as a Chemical Defence Agent of Fruiting Bodies of Mycena rosea and its Role in the Generation of the Alkaloid Mycenarubin C.

Mycenarubin C, a previously unknown red pyrroloquinoline alkaloid, was isolated from fruiting bodies of the mushroom Mycena rosea and its structure was elucidated mainly by NMR spectroscopy and mass spectrometry. Unlike mycenarubin A, the major pyrroloquinoline alkaloid in fruiting bodies of M. rosea, mycenarubin C, contains an eight-membered ring with an additional C1 unit that is hitherto unprecedented for pyrroloquinoline alkaloids known in nature. Incubation of mycenarubin A with an excess of formaldehyde revealed that mycenarubin C was generated nearly quantitatively from mycenarubin A. A

Molluscicidal activity and physiological toxicity of quaternary benzo[c]phenanthridine alkaloids (QBAs) from Macleaya cordata fruits on Oncomelania hupensis.

Schistosomiasis is a serious worldwide parasitic disease. One of the best ways to control schistosomiasis is to control the population of Oncomelania hupensis snails. We sought to identify a high-efficiency biogenic molluscicide against Oncomelania with low toxicity, to avoid chemical molluscicide contamination and toxicity in aquatic organisms. We extracted quaternary benzo[c]phenanthridine alkaloids (QBAs) from Macleaya cordata fruits. Molluscicidal activity of the QBAs against Oncomelania was determined using bioassay. Our results showed that the extracted QBAs had a strong molluscicidal ef

Dietary resources shape the adaptive changes of cyanide detoxification function in giant panda (Ailuropoda melanoleuca).

The functional adaptive changes in cyanide detoxification in giant panda appear to be response to dietary transition from typical carnivore to herbivorous bear. We tested the absorption of cyanide contained in bamboo/bamboo shoots with a feeding trial in 20 adult giant pandas. We determined total cyanide content in bamboo shoots and giant panda's feces, levels of urinary thiocyanate and tissue rhodanese activity using color reactions with a spectrophotometer. Rhodanese expression in liver and kidney at transcription and translation levels were measured using real-time RT-PCR and immunohistoche

Detoxification function of the Arabidopsis sulphotransferase AtSOT12 by sulphonation of xenobiotics.

Cytosolic sulphotransferases have been implicated in inactivation of endogenous steroid hormones and detoxification of xenobiotics in human and animals. Yet, the function of plant sulphotransferases in xenobiotic sulphonation and detoxification has not been reported. In this study, we show that the Arabidopsis sulphotransferase AtSOT12 could sulphonate the bacterial-produced toxin cycloheximide. Loss-of-function mutant sot12 exhibited hypersensitive phenotype to cycloheximide, and expression of AtSOT12 protein in yeast cells conferred resistance to this toxic compound. AtSOT12 exhibited broad

Pharmacogenetics of olanzapine metabolism.

The pharmacokinetics of the atypical antipsychotic, olanzapine, display large interindividual variation leading to multiple-fold differences in drug exposure between patients at a given dose. This variation in turn gives rise to the need for individualized dosing in order to avoid concentration-dependent adverse effects or therapeutic failure. Genetically determined differences in olanzapine metabolism represent a less studied source of variability in comparison to environmental and physiological factors. In this review, we summarize available in vitro and in vivo data addressing the influence

Scopolamine detoxification technique for heroin dependence: a randomized trial.

BACKGROUND: Easing psychological symptoms associated with heroin use and heroin relapse are important goals in the treatment of heroin dependence. However, most detoxification methods are designed to decrease withdrawal-related discomfort and complications, but not to reduce the psychological effects of heroin addiction. OBJECTIVE: The objective of this study was to evaluate the efficacy of scopolamine detoxification technique (SDT) relative to standard methadone detoxification (MD) to treat heroin withdrawal and psychological symptoms associated with heroin use and relapse. METHODS: In this

Ontogeny and drug metabolism in newborns.

In pediatric age and particularly in newborn infants the drug efficacy and safety are influenced by the growth and development on drug Absorption, Distribution, Metabolism and Excretion (ADME). Thanks to the fast development of pharmacogenomics and pharmacogenetics, the drug therapy promises to be adapted to the genetic profile of the individual, reducing considerably the side effects of drugs and increasing their efficacy. Interindividual variability in drug response is well known in both adults and children. Such a variability is multifactorial considering both intrinsic and extrinsic factor

Metal toxicity, uptake and bioaccumulation in aquatic invertebrates--modelling zinc in crustaceans.

We use published data on the different patterns of the bioaccumulation of zinc by three crustaceans, the caridean decapod Palaemon elegans, the amphipod Orchestia gammarellus and the barnacle Amphibalanus amphitrite, to construct comparative biodynamic models of the bioaccumulation of zinc into metabolically available and detoxified components of accumulated zinc in each crustacean under both field and laboratory toxicity test conditions. We then link these bioaccumulation models to the onset of toxic effects on exposure of the crustaceans to high dissolved zinc bioavailabilities, using the te

Correlation between the UDP-glucuronosyltransferase (UGT1A1) TATAA box polymorphism and carcinogen detoxification phenotype: significantly decreased glucuronidating activity against benzo(a)pyrene-7,8-dihydrodiol(-) in liver microsomes from subjects with the UGT1A1*28 variant.

Of the hepatic UDP-glucuronosyltransferases (UGTs), only UGT1A1 and UGT1A9 exhibit activity against benzo(a)pyrene-trans-7R,8R-dihydrodiol [BPD(-)], precursor to the highly mutagenic anti-(+)-benzo(a)pyrene-7R,8S-dihydrodiol-9S,10R-epoxide. The UGT1A1*28 allelic variant contains an additional (TA) dinucleotide repeat in the "TATAA" box [(TA)(6)>(TA)(7)] of the UGT1A1 promoter that has been linked to decreased expression of the UGT1A1 gene and decreased bilirubin conjugation, leading to the relatively nondebilitating condition known as Gilbert's syndrome. To determine whether the UGT1A1 TATAA b

Lipid-lowering agents and myopathy.

Each of the lipid-lowering agents available today can cause myopathy. The severity of the muscle disorder may vary from trivial myalgias or elevations of creatine kinase in asymptomatic individuals to rhabdomyolysis with myoglobinuria, renal failure, and death. Fortunately, significant myopathy occurs at a low rate. However, the large number of individuals taking these medications renders it a significant problem. Although the pathophysiology of the myopathy remains speculative, its occurrence appears to be dose related. Consequently, the total dosage of lipid-lowering drug consumed, the conco

Strain-dependent effects of phenobarbital on liver tumor promotion in inbred mice.

Inbred strains of mice were found to differ significantly in their susceptibility to liver tumor promotion by PB. The susceptibility to promotion of hepatocarcinogenesis by this drug was a dominant trait in crosses between the sensitive DBA and resistant C57 mice. The reciprocal F1 hybrids responded similarly to tumor promotion by PB. PB promoted the development of hepatoblastomas in D2B6F1 males but not in B6D2F1 mice. Female mice of the D2B6F1 cross, similarly exposed to PB, failed to develop hepatoblastomas. These results suggested a sex-linked differential response to the development of he

Mercury vapor uptake and hydrogen peroxide detoxification in human and mouse red blood cells.

The uptake of Hg vapor (Hg0) by suspensions of human and BALB-c mouse erythrocytes was studied in a closed exposure system. The formation of catalase-compound-I and thereby the oxidation of Hg0 was initiated by microinfusion of hydrogen peroxide. The degradation of H2O2 by the glutathione (GSH)/GSH peroxidase system was reduced by t-butyl-hydroperoxide (t-BOOH) or by 1-chloro-2,4-dinitrobenzene (CDNB). In human red blood cells, CDNB and t-BOOH increased the rate of Hg vapor oxidation at the low and intermediate H2O2 supplementation rates. In mouse erythrocytes, Hg uptake was increased by CDNB
