Heme oxygenase isoform expression in cellular and antibody-mediated models of acute inflammation in the rat.

Heme oxygenase (HO) is the rate-limiting enzyme in the catabolism of heme to biliverdin, carbon monoxide (CO), and free iron. The enzyme exists as a constitutive isoform (HO-2) and an inducible isoform (HO-1), which is also a stress protein (HSP32). HO-1 has previously been shown to be associated with the resolution phase of a non-immune model of acute inflammation. In addition, elevation of the enzyme was markedly anti-inflammatory. In the present study, these observations have been extended to two pleural models of immune-driven inflammation in the rat, an immediate type III hypersensitivity

Colchicine-induced elevation of tissue metallothionein contents is mediated by inflammation-independent serum factor.

Subcutaneous injection of colchicine caused dose-dependent and time-dependent induction of hepatic MT in mice. Other than colchicine, similar MT induction was observed in vincristine- or vinblastine-injected mice, but not in beta-lumicolchicine-injected mice. MT contents were also elevated in the kidney, spleen, lung and heart by colchicine injection. Isoforms of colchicine-induced MT in the liver were identified to be MT-I and II by immunoblot analysis. Unlike turpentine-induced MT synthesis, dexamethasone, an anti-inflammatory agent, could not block the MT-inducing activity of colchicine. Th

Bidirectional, time-ordered association network linking psychiatric disorders and immune-mediated inflammatory diseases: Inflammation-sleep mediation and genetic modification.

BACKGROUND: Marked comorbidity between mental disorders and immune-mediated inflammatory diseases (IMIDs) suggests a bidirectional interplay, yet most prior work has tackled a single analytic direction, leaving the overall structure of time-ordered associations unclear. METHODS: Using the large, longitudinal UK Biobank cohort, we fitted Cox proportional-hazards models to quantify bidirectional, time-ordered associations between eight prototypical mental disorders and eight common IMIDs. Genetic susceptibility was quantified with polygenic risk scores (PRSs), and mediation models incorporated

UHPLC-DAD-ESI-MS/MS and HPTLC profiling of ash leaf samples from different commercial and natural sources and their in vitro effects on mediators of inflammation.

INTRODUCTION: In European traditional medicine, common ash leaf infusion is recommended by European Medicines Agency to treat minor articular pain and to increase the amount of urine for flushing minor urinary complaints. However, a comprehensive ultra-high-performance liquid chromatography diode array detector electrospray ionisation tandem mass spectrometry (UHPLC-DAD-ESI-MS/MS) analysis of this pharmacopeial plant material has never been performed. Moreover, the number of biological and pharmacological investigations proving the usefulness of this plant material in recommended traditional u

TYMP upregulation mediated by the hyperactivated IL-17/NF-κB1 axis promotes psoriasis through enhancing aberrant keratinization and neutrophil-mediated inflammation.

Psoriasis is a common, chronic, and recurrent immune-mediated disorder with global prevalence, underscoring the need for novel biomarkers to improve diagnosis and treatment. In this study, differentially expressed genes (DEGs) in psoriatic tissues were comprehensively identified through integrated single-cell and bulk RNA-seq analyses. Thymidine phosphorylase (TYMP) emerged as one of the most significantly upregulated biomarkers in psoriasis. Multiplex immunohistochemistry (mIHC) and IHC assays jointly confirmed marked overexpression of TYMP in psoriatic keratinocytes. Mechanistically, we demo

Gomisin M2 Inhibits Mast Cell-Mediated Allergic Inflammation via Attenuation of FcεRI-Mediated Lyn and Fyn Activation and Intracellular Calcium Levels.

Mast cells are effector cells that induce allergic inflammation by secreting inflammatory mediators. Gomisin M2 (G.M2) is a lignan isolated from Schisandra chinensis (Turcz). Baill. exhibiting anti-cancer activities. We aimed to investigate the anti-allergic effects and the underlying mechanism of G.M2 in mast cell-mediated allergic inflammation. For the in vitro study, we used mouse bone marrow-derived mast cells, RBL-2H3, and rat peritoneal mast cells. G.M2 inhibited mast cell degranulation upon immunoglobulin E (IgE) stimulation by suppressing the intracellular calcium. In addition, G.M2 in

Nanoparticle-Mediated Delivery of Pitavastatin to Monocytes/Macrophages Inhibits Left Ventricular Remodeling After Acute Myocardial Infarction by Inhibiting Monocyte-Mediated Inflammation.

Left ventricular (LV) remodeling after myocardial infarction (MI) causes heart failure. Although medical therapies including angiotensin converting enzyme inhibitors show inhibitory effects on post-infarct LV remodeling, the prognosis of patients with post-infarct heart failure is still poor. Accumulating evidence suggests that an inflammatory response is implicated in the process of post-infarct LV remodeling. Therefore, we hypothesized that anti-inflammatory therapy by nanoparticle-mediated monocyte/macrophage-targeting delivery of pitavastatin may protect the heart from post-infarct LV remo

In vitro anti-nephrotoxic potential of Ammi visnaga, Petroselinum crispum, Hordeum vulgare, and Cymbopogon schoenanthus seed or leaf extracts by suppressing the necrotic mediators, oxidative stress and inflammation.

BACKGROUND: The kidney is an essential organ required by the body to perform several important functions. Nephrotoxicity is one of the most prevailing kidney complications that result from exposure to an extrinsic or intrinsic toxicant, which increase the need for the acquisition of proper remedies. Recently, natural remedies are gaining great attention owed to the fact that they have fewer side effects than most conventional drugs. METHODS: The current study recorded a new therapeutic role of the well-known medicinal plants for kidney stones [Ammi visnaga (AVE), Petroselinum crispum (PCE), H

Association of pulmonary inflammation and increased microvascular permeability during the development of bronchopulmonary dysplasia: a sequential analysis of inflammatory mediators in respiratory fluids of high-risk preterm neonates.

OBJECTIVE: Bronchopulmonary dysplasia (BPD) of preterm neonates is associated with an increased recruitment of inflammatory cells into the airways. To evaluate further the role of inflammation in the pathogenesis of BPD, tracheobronchial aspirate fluid of neonates with birth weight < 1200 g (n = 59) was sequentially analyzed in a prospective study. METHODS: Tracheobronchial aspirate fluid was assessed for chemotactic activity, neutrophil cell count, concentrations of elastase-alpha 1-proteinase inhibitor and activity of free elastase, concentrations of chemoattractants (complement component C

Naringin attenuates the development of carrageenan-induced acute lung inflammation through inhibition of NF-κb, STAT3 and pro-inflammatory mediators and enhancement of IκBα and anti-inflammatory cytokines.

Naringin has been reported to possess diverse pharmacological properties, including anti-arthritic and anti-inflammatory activities. The aim of the present study was to determine the potential anti-inflammatory effect of naringin in a mouse model of carrageenan-induced pleurisy. A single dose of naringin (40 and 80 mg/kg) was administered per oral (p.o.) 1 h before carrageenan (Cg) administration. Pro- and anti-inflammatory cytokines were analysed in pleural fluid. We also assessed the effects of naringin on the expression levels of iNOS, inducible cyclooxygenase isoform (COX-2), ICAM-1, MIP-2

GALNT6-mediated O-glycosylation on PTGS2 regulates microglia inflammation and ferroptosis following intracerebral hemorrhage.

Intracerebral hemorrhage (ICH) is a common type of stroke characterized by high mortality and disability rates. Polypeptide N-acetylgalactosaminyl transferase-6 (GALNT6), which can catalyze the O-GalNAc glycosylation, is related to the development of various diseases. Nevertheless, the action of GALNT6 on ICH pathogenesis remains elusive. Hence, this study clarified the role of GALNT6 in ICH pathogenesis and investigated the possible underlying mechanism. The in vivo ICH model was created using collagenase in mice. BV2 microglial cells were subjected to oxygen and glucose deprivation combined

Myocardial injury mediated by acute inflammation after abdominal surgery in mice: a mechanistic translational study.

BACKGROUND: Perioperative myocardial injury has often been attributed to type 2 myocardial infarction, but acute systemic inflammation could also contribute. We examined the role of systemic inflammation in myocardial injury after abdominal surgery using a murine model. METHODS: Male C57BL/6 mice underwent standardised abdominal surgery or anaesthesia alone. Myocardial injury (serum cardiac troponin I [cTnI]) and systemic inflammation (serum interleukin-6 [IL-6]) were quantified (enzyme-linked immunoassay) at 1 h to 7 days after surgery with anaesthesia or anaesthesia alone. Transcriptomic ch

The relationship between maternal adiposity during pregnancy and toddler ADHD symptoms is mediated by gestational inflammation.

BACKGROUND: Increased adiposity during pregnancy is associated with increased risk for child attention-deficit/hyperactivity disorder (ADHD), though few studies have examined the mechanisms responsible for conveying this risk. The current study tested whether increased inflammation during pregnancy was a statistical mediator of the association between maternal adiposity and child ADHD symptoms during toddlerhood. It also examined whether this association was independent of maternal distress and dietary quality during pregnancy, important confounding factors that have previously been linked wit

Acupuncture Attenuates Allergic Airway Inflammation via CC10-Mediated Modulation of Lung Dendritic Cells.

BACKGROUND: Acupuncture has emerged as a promising complementary therapy for allergic asthma, but the molecular mechanisms underlying its therapeutic effects remain unclear. This study aimed to investigate whether Clara cell 10-kDa protein (CC10), an airway epithelial immunoregulatory protein, mediates the anti-inflammatory actions of acupuncture through the modulation of pulmonary dendritic cells (DCs). METHODS: We conducted a clinical study in patients with allergic asthma who underwent acupuncture at GV14 (Dazhui), BL12 (Fengmen), and BL13 (Feishu) acupoints to evaluate changes in asthma c

β-Carboline alkaloid harmaline alleviates hyperuricemia-mediated renal inflammation by suppressing oxidative stress.

Hyperuricemia is a metabolic disorder marked by elevated serum uric acid levels and is closely linked to the development of gout and progressive renal dysfunction. It is increasingly recognized as a major public health concern due to its association with chronic kidney disease and cardiovascular complications. Hyperuricemia induces kidney impairment through a complex interplay of oxidative and inflammatory stress. Harmaline, a β-carboline alkaloid primarily found in Peganum harmala, exhibits antioxidant and anti-inflammatory activities. Considering the roles of oxidative and inflammatory stres

Chaperone-Mediated Autophagic Degradation of USP9X in Macrophages Exacerbates Postmyocardial Infarction Inflammation and Cardiac Dysfunction.

Excessive macrophage-mediated inflammation following myocardial infarction (MI) exacerbates infarct expansion and impairs cardiac repair; however, the regulatory mechanisms remain poorly understood. Here, it is reported that ubiquitin-specific peptidase 9 X-linked (USP9X) was significantly downregulated in macrophages during early post-MI inflammation. Macrophage-specific deficiency of USP9X enhanced expression of pro-inflammatory genes, thereby impeding cardiac functional recovery. Mechanistically, USP9X deubiquitinated and stabilized tumor necrosis factor receptor-associated factor (TRAF)-ty

Associations of serum metal/metalloids with ovarian reserve and inflammation mediation: a repeated-measures longitudinal study.

BACKGROUND: Little is known about associations of metal/metalloids (the widespread environmental pollutants) with ovarian reserve, and the underlying mechanisms. METHODS: We conducted a longitudinal study of 897 women (20-49 years) with 1958 repeated observations from two infertility centers in Hubei, China, and measured 22 serum metal/metalloids, C-reactive protein, cell-based inflammatory biomarkers, and anti-müllerian hormone (AMH) at baseline and follow-up surveys. We examined associations of metal/metalloid exposures with AMH levels and diminished ovarian reserve (DOR, defined as AMH < 1

Uhrf1-Mediated PKM2 Degradation via Ubiquitination Alleviates Inflammation and Pyroptosis in Inflammatory Bowel Disease.

Inflammatory bowel disease (IBD) is characterized by chronic relapsing intestinal inflammation and closely associated with persistent inflammatory responses and impairment of the mucosal barrier integrity. In this work, we demonstrated that Th17/Treg immune imbalance drove inflammation and pyroptosis in IBD, as revealed by single-cell RNA sequencing, and pyruvate kinase M2 (PKM2) and ubiquitin-like with PHD and RING finger domains 1‌ (Uhrf1) displayed a negative correlation in IBD clinic samples. Target fishing analysis revealed that eupatolide (EPT) covalently bound to C165 of PKM2 with a dis
