Transcriptome Analysis of Deltamethrin Resistance Mechanisms in Aedes aegypti Populations from Metro Manila, Philippines.

Background: Arboviral transmission by Aedes aegypti is exacerbated by insecticide resistance, which undermines effective vector control strategies. In the Philippines, deltamethrin resistance in several Ae. aegypti populations has been reported and prompts an investigation into the possible molecular mechanisms involved. Methods: Two Ae. aegypti populations were subjected to susceptibility assays to deltamethrin and were RNA-sequenced for transcriptome analysis. Results: The two (2) Ae. aegypti populations were resistant to deltamethrin and had LC50 values of 1.26 and 2.15 μg/mL at the zeroth …

Influence of testing modality on bioefficacy for the evaluation of Interceptor® G2 mosquito nets to combat malaria mosquitoes in Tanzania.

BACKGROUND: Insecticide-treated net (ITN) durability is evaluated using longitudinal bioefficacy and fabric integrity sampling post-distribution. Interceptor® G2 was developed for resistance management and contains two adulticides: alpha-cypermethrin and chlorfenapyr; it is a pro-insecticide that is metabolized into its active form by mosquito-detoxifying enzymes and may be enhanced when the mosquito is physiologically active. To elucidate the impact of bioassay modality, mosquito exposures of the alphacypermethrin ITN Interceptor® and dual adulticide Interceptor® G2 were investigated. METHOD

The great potential of entomopathogenic bacteria Xenorhabdus and Photorhabdus for mosquito control: a review.

The control of insects of medical importance, such as Aedes aegypti and Aedes albopictus are still the only effective way to prevent the transmission of diseases, such as dengue, chikungunya and Zika. Their control is performed mainly using chemical products; however, they often have low specificity to non-target organisms, including humans. Also, studies have reported resistance to the most commonly used insecticides, such as the organophosphate and pyrethroids. Biological control is an ecological and sustainable method since it has a slow rate of insect resistance development. Bacterial spec

Recombinant bacteria for mosquito control.

Bacterial insecticides have been used for the control of nuisance and vector mosquitoes for more than two decades. Nevertheless, due primarily to their high cost and often only moderate efficacy, these insecticides remain of limited use in tropical countries where mosquito-borne diseases are prevalent. Recently, however, recombinant DNA techniques have been used to improve bacterial insecticide efficacy by markedly increasing the synthesis of mosquitocidal proteins and by enabling new endotoxin combinations from different bacteria to be produced within single strains. These new strains combine

Livestock and avermectins in sub-Saharan Africa: a restricted systematic review of the impacts on productivity and documentation of resistance.

There is growing interest in using avermectins in livestock as a vector control tool for mosquitoes involved in the transmission of human malaria in sub-Saharan Africa (SSA). If implemented, the potential health and productivity impacts across the livestock sector would need to be considered, as avermectins are already commonly used in veterinary medicine to treat gastrointestinal helminths and parasitic insects. Here we present the results of a restricted systematic review that summarizes what is known about the effects of avermectins on cattle and swine productivity in SSA and the presence o

Optimizing Spinosyn Insecticide Applications for Allium Leafminer (Diptera: Agromyzidae) Management in Allium Crops.

Allium leafminer, Phytomyza gymnostoma Loew (Diptera: Agromyzidae), is an invasive pest of allium crops in North America. Spinosyn insecticides, spinetoram and spinosad, have been effective choices for managing P. gymnostoma infestations in allium crops, but their use should be optimized for economical and resistance management purposes. In New York from 2018 through 2020, performance of each spinosyn insecticide was evaluated by making two applications spaced either 1 or 2 wk apart beginning at various intervals after P. gymnostoma was first detected in the field; a weekly spray program also

Spinosad-associated modulation of select cytochrome P450s and glutathione S-transferases in the Colorado potato beetle, Leptinotarsa decemlineata.

The Colorado potato beetle (Leptinotarsa decemlineata (Say)) is an insect pest that threatens potato crops. Multiple options exist to limit the impact of this pest even though insecticides remain a primary option for its control. Insecticide resistance has been reported in Colorado potato beetles and a better understanding of the molecular players underlying such process is of utmost importance to optimize the tools used to mitigate the impact of this insect. Resistance against the insecticide spinosad has been reported in this insect and this work thus aims at exploring the expression of targ…

Inhibiting the proteasome reduces molecular and biological impacts of the natural product insecticide, spinosad.

BACKGROUND: Insecticide targets are often identified by mutations that confer resistance, but the intricacies of insecticide binding and downstream processes leading to insect death often remain obscure. Mutations in α6-like nicotinic acetylcholine receptor subunit genes have been associated with high levels of resistance to spinosad in many insect species, including Drosophila melanogaster. Here, we aimed to expand our understanding of the effects of the natural product insecticide spinosad on its protein target, the α6 subunit, using genetic tools available in D. melanogaster. RESULTS: Func

Potential Risks Associated with Japanese Encephalitis Prevalence in Shandong Province, China.

Japanese encephalitis (JE), which is caused by the Japanese encephalitis virus (JEV), is a zoonotic, vector-borne neurotropic disease that remains a major cause of viral encephalitis in Asia. JEV is spread to humans through mosquitoes, and its primary transmission vector is Culex tritaeniorhynchus. Mosquitoes were sampled from three collection sites: Hanzhuang town in Weishan County, Taibai Lake in Jining city, and Dongping Lake in Shandong Province, China. Pyrethroid insecticide resistance bioassays were conducted using adult mosquitoes. Cx. tritaeniorhynchus and C. pipiens pallens population…

Significant variations in tolerance to clothianidin and pirimiphos-methyl in Anopheles gambiae and Anopheles funestus populations during a dramatic malaria resurgence despite sustained indoor residual spraying in Uganda.

BACKGROUND: A dramatic malaria resurgence occurred in areas of Uganda between 2020 and 2022, coinciding with the switch to clothianidin-based formulations for indoor residual spraying. During the resurgence, Anopheles funestus sensu lato (s.l.) numbers increased more than those of Anopheles gambiae s.l., but when an alternative insecticide, pirimiphos-methyl, was reintroduced in 2023, both malaria cases and An. funestus mosquito density fell. METHODS: In this study, we investigated possible causes of the resurgence by assessing (1) whether sufficient quantities of insecticide were sprayed, (2

Genetic control strategies for population suppression in the Anopheles gambiae complex: a review of current technologies.

Malaria continues to pose a critical public health threat, with mosquitoes from the Anopheles gambiae complex acting as the main vectors of the disease in sub-Saharan Africa, where approximately 95% of malaria-related deaths occur. Despite significant advancements in vector control, such as insecticide-treated bed nets and indoor spraying, the effectiveness of these interventions is increasingly compromised by various challenges, including rising levels of insecticide and pathogen resistance, mosquito behavioural adaptations, and persistent funding gaps. In this context, genetic vector control

The WHO Onchocerciasis Control Programme: retrospect and prospects.

The history of onchocerciasis control in Africa and the genesis of the WHO Onchocerciasis Control Programme in West Africa (OCP) are briefly reviewed. The importance of experience gained in anti-locust campaigns in helping to plan the OCP is stressed. Members of the Simulium damnosum species complex are the vectors of onchocerciasis, which OCP is controlling with insecticide treatments on the stretches of rivers where the Simulium breed. Migrations of flies have been responsible for reinfestations of controlled areas and the spread of insecticide resistance. The management of these problems an…

Screening of broad spectrum natural pesticides against conserved target arginine kinase in cotton pests by molecular modeling.

Cotton is an economically important crop and its production is challenged by the diversity of pests and related insecticide resistance. Identification of the conserved target across the cotton pest will help to design broad spectrum insecticide. In this study, we have identified conserved sequences by Expressed Sequence Tag profiling from three cotton pests namely Aphis gossypii, Helicoverpa armigera, and Spodoptera exigua. One target protein arginine kinase having a key role in insect physiology and energy metabolism was studied further using homology modeling, virtual screening, molecular do…

Sodium Channel Mutations and Pyrethroid Resistance in Aedes aegypti.

Pyrethroid insecticides are widely used to control insect pests and human disease vectors. Voltage-gated sodium channels are the primary targets of pyrethroid insecticides. Mutations in the sodium channel have been shown to be responsible for pyrethroid resistance, known as knockdown resistance (kdr), in various insects including mosquitoes. In Aedes aegypti mosquitoes, the principal urban vectors of dengue, zika, and yellow fever viruses, multiple single nucleotide polymorphisms in the sodium channel gene have been found in pyrethroid-resistant populations and some of them have been functiona

Consolidating strategic planning and operational frameworks for integrated vector management in Eritrea.

BACKGROUND: Contemporary malaria vector control relies on the use of insecticide-based, indoor residual spraying (IRS) and long-lasting insecticidal nets (LLINs). However, malaria-endemic countries, including Eritrea, have struggled to effectively deploy these tools due technical and operational challenges, including the selection of insecticide resistance in malaria vectors. This manuscript outlines the processes undertaken in consolidating strategic planning and operational frameworks for vector control to expedite malaria elimination in Eritrea. CASE DESCRIPTION: The effort to strengthen s…

Depleting SfCarE: Innovative strategies for enhancing chlorantraniliprole toxicity through mixing with carbaryl in Spodoptera frugiperda (Lepidoptera: Noctuidae).

The sustained application of insecticides has led to the development of resistance. Carboxylesterases (CarEs) play important roles in the resistance and synergism of chlorantraniliprole and carbaryl. In this study, the key Spodoptera frugiperda CarEs gene, SfCarE, was identified. SfCarE was highly expressed at the 4th instar and in the midgut of the 6th instar larvae. Knockdown of SfCarE resulted in significant decrease in CarE activity and an increase in susceptibility to chlorantraniliprole, as well as to the mixture of chlorantraniliprole and carbaryl. Metabolic assays demonstrated that the
