BACKGROUND: Male infertility has emerged as a global concern, with dysfunctional spermatogenesis being a major cause. A significant proportion of agricultural pests belong to lepidopteran species, making it valuable to explore the conserved genes involved in spermatogenesis within the lepidopteran model, Bombyx mori. Identifying these genes could reveal potential targets for pest control and enhance insect sterility technologies. Although the ovarian tumor (otu) gene is essential for oogenesis in Drosophila melanogaster, its role in Lepidoptera remains largely unexplored.

In their non-native ranges, invasive plant species may escape specialist herbivores and thus experience exclusive pressure from generalists. This is predicted to shape the defense strategies of successful non-native species, as proposed by the Shifting Defense Hypothesis. However, most research to date has quantified only limited sets of defense traits of non-native invasive plant species, and thus our understanding of their defense syndromes, which encompass suites of both physical and chemical traits, remains poorly understood. We evaluated a comprehensive array of leaf physicochemical trait

BACKGROUND: Gustatory receptors (GRs) are known to mediate responses to various chemical stimuli including sugars and bitter compounds and sex pheromones. Although numerous types of GRs have been identified across insect species, the physiological roles of most GRs remain largely unknown. The silkworm, Bombyx mori, is representative of the order Lepidoptera, which includes over 70% of agriculturally important pest species. A notable feature of GRs in B. mori is that most GRs are organized in clusters, making them highly suitable for large-scale genetic studies.

BACKGROUND: Spodoptera frugiperda (Lepidoptera: Noctuidae) is one of the world's major agriculture pests, causing significant crop damage and serious economic losses. As S. frugiperda has developed widespread resistance, environmentally friendly insecticides are urgently needed. Therefore, searching for natural antifeedant or insecticidal lead compounds has become imperative.
RESULTS: Three new indole diterpenoids, paspamine A (1), paspamine B (2), and paspalitrem D (3), along with nine known ones (compounds 4-12), were isolated from an endophytic Penicillium brefeldianum F4a. Their structure

Eggplant fruit and shoot borer, Leucinodes orbonalis (Lepidoptera: Crambidae), is a destructive borer pest that infests eggplant crop from nursery stage to fruiting. This study aimed to evaluate the current insecticide resistance status of L. orbonalis collected from ten different locations of Tamil Nadu, India, using a fruit dip bioassay. Late second instar larvae were exposed to spinosad, spinetoram, thiacloprid and emamectin benzoate. Median lethal concentration (LC50) and resistance ratios (RR) were estimated for each population using a susceptible NBAIR population as a reference. The resu

BACKGROUND: Nodular ophthalmia, a rare and specific inflammatory ocular disease, is typically triggered by contact with setae from Lepidoptera larvae. Type V according to the Cadera classification represents the rarest and prognostically poorest variant. CASE REPORT: A 55-year-old male developed immediate ocular pain, foreign body sensation, and blurred vision in his left eye following contact with a Thaumetopoea pityocampa (pine processionary caterpillar). Despite prompt removal of visible setae and initiation of topical non-steroidal anti-inflammatory drug (NSAID) therapy at a local facility

Oogenesis in insects is a carefully orchestrated process, facilitating the formation of female gametes, which is regulated by multiple extrinsic and intrinsic factors, including ovary serine protease (Osp). As a member of the serine protease family, Osp is a homolog of Nudel, a maternally required protease defining embryonic dorsoventral polarity in Drosophila. In this study, we used CRISPR/Cas9-mediated mutagenesis to functionally characterize Osp in the Asian corn borer, Ostrinia furnacalis, a devastating maize pest throughout Asia and Australia. Building on previous knowledge, we hypothesiz

Detection of pest infestations in fresh produce traded internationally could offer improved prospects for reducing the movement of unwanted pests. Because immature stages of some pests can be difficult to find visually, other cues such as herbivore-induced volatiles that can potentially be detected at the early stages of infestation are worth investigating. In this study, we artificially infested postharvested apples (Malus × domestica 'Royal Gala') with two economic apple pests, the specialist codling moth (CM, Cydia pomonella Linnaeus, Lepidoptera: Tortricidae) and the generalist Queensland

Persicaria maculosa (Polygonaceae) (known as lady's thumb) is an annual morphologically variable weed that is widely distributed in Chile. The purpose of this study was to investigate the antifeedant potential of methanolic (MeOH), ethanolic (EtOH), and dichloromethane (DCM) extracts from the aerial parts of this plant collected in the Valparaíso and Curicó provinces (Chile) and relate this activity to the antioxidant capacity and the presence of phenolic compounds in the extracts. A phenolic profile based on HPLC-ESI-MS/MS allowed the identification of 26 phenolic compounds, most of them glyc

The dynamic variability of transcription factors (TFs) and their binding sites makes it challenging to conduct genome-wide transcription regulation research. The silkworm Bombyx mori, which produces silk, is one of the most valuable model insects in the order Lepidoptera. The "opening" and "closing" of chromatin in different silk yield strains is associated with changes in silk production, making this insect a good model for studying the transcriptional regulation of genes. However, few studies have examined the open chromatin regions (OCRs) of silkworms, and studying OCR synergism and their f

Fresh market sweet corn (Zea mays L., convar. saccharata var. rugosa, Poales: Poaceae) ears produced in Florida are damaged by the larvae of Euxesta stigmatias Loew, Euxesta eluta Loew, and Chaetopsis massyla Walker (Diptera: Ulidiidae) that renders ears unmarketable. No standard lure exists for monitoring these pests. Oviposition substrate and attractant bioassays were designed to identify attractive substrates for further semiochemical investigation. Frass from the fall armyworm, Spodoptera frugiperda J.E. Smith (Lepidoptera: Noctuidae), was more attractive than other ovipositional substrate

Yersinia entomophaga MH96, which was originally isolated from the New Zealand grass grub, Costelytra zealandica, produces an orally active proteinaceous toxin complex (Yen-Tc), and this toxin is responsible for mortality in a range of insect species, mainly within the Coleoptera and Lepidoptera. The genes encoding Yen-Tc are members of the toxin complex (Tc) family, with orthologs identified in several other bacterial species. As the mechanism of Yen-Tc activity remains unknown, a histopathological examination of C. zealandica larvae was undertaken in conjunction with cultured cells to identif

Cordyceps sinensis (Chinese caterpillar fungus) is a fungus parasitic on the larvae of Lepidoptera and has been considered to be a precious tonic food and herbal medicine since ancient times in China. Recently, some fungal strains have been isolated from the fruiting bodies of wild C. sinensis, and some of them have been reported to show the same properties as the natural product. In the present study, an EPSF (exopolysaccharide fraction) was prepared from cultivated C. sinensis and its effects on B16 melanoma-bearing mice were investigated. Three doses of EPSF were intraperitoneally administe

Simulium blackfly larvae (Diptera: Simuliidae) were collected from rivers and streams at 500-1500 m a.s.l. in Chiapas State of southern Mexico. Among 45 sites surveyed over an area of 2300 km2 (around 15 degrees 15'N 92 degrees 20'W), some Simulium larvae from three sites were opalescent violet-blue, interpreted as patent infection with invertebrate iridescent virus (IIV). Dissection confirmed the presence of putative Iridovirus particles, 130nm diameter, but no IIV isolates were obtained from homogenates injected into Galleria mellonella (L) larvae (Lepidoptera: Pyralidae). A clear link between mating type and virulence has been demonstrated for some fungal pathogens, but not for Aspergillus fumigatus as of yet. An association between mating type and invasiveness has recently been established. The mating type proportion (MAT1-1:MAT1-2) of 213 A. fumigatus strains was determined (48.5%:51.5%) and results were in agreement with previous studies. However, these percentages changed when the strain collection was divided into azole-susceptible and -resistant strains. The 163 susceptible strains kept these proportions, but among the 50 azole-resistant strains 60.0% MAT

Cryptococcus neoformans is an environmental fungus that can cause lethal meningoencephalitis in immunocompromised individuals. The mechanisms by which environmental microbes become pathogenic to mammals are still obscure, but different studies suggest that fungal virulence evolved from selection imposed by environmental predators. The soil-living Acanthamoeba castellanii is a well-known predator of C. neoformans. In this work, we evaluated the participation of C. neoformans virulence-associated structures in the interaction of fungal cells with A. castellanii. Fungal extracellular vesicles (EV

Bacterial trans-translation is the main quality control mechanism employed to relieve stalled ribosomes. Trans-translation is mediated by the small protein B (SmpB) and transfer-mRNA (tmRNA) ribonucleoprotein complex, which interacts with translational complexes stalled at the 3' end of non-stop mRNAs to release the stalled ribosomes thereby targeting the nascent polypeptides and truncated mRNAs for degradation. The trans-translation system exists with a few exceptions in all bacteria. In the present study, we assessed the contribution of SmpB to the growth and virulence of Listeria monocytoge

Galleria mellonella is an excellent invertebrate model for the study of diseases that involve interactions with cells from the innate immune system, since they have an innate immune system capable of recognizing the pathogens. Here we present for the first time, an alternative model for an in vitro phagocytic assay using hemocytes of G. mellonella larvae to study infection by Leishmania (Viannia) braziliensis. We showed that the insect phagocytic cells were able to engulf promastigotes. Furthermore, this infective form differentiated into the amastigote form inside those cells. However, the ce

The introduction of an agricultural pest species into a new environment is a potential threat to agroecosystems of the invaded area. The phytosanitary concern is even greater if the introduced pest's phenotype expresses traits that will impair the management of that species. The invasive tomato borer, Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae), is one such species and the characterization of the insecticide resistance prevailing in the area of origin is important to guide management efforts in new areas of introduction. The spinosad is one the main insecticides currently used in Brazil

Although the lepidopteran larva Pseudaletia separata is attacked by the gregarious ectoparasitoid Euplectrus separatae, it continues to feed and grow. Lipid concentration in the hemolymph of the parasitized host was higher than that of the nonparasitized host from 3 to 8 days after parasitization. Artificial injection of parasitoid venom also elevated lipid concentration in the host hemolymph. One day after venom injection the host's fat body contained many lipid particles, but most of the lipid particles disappeared 7 days later. Light microscopy and transmission electron microscopy showed th
