Three dimensional structure of the anthrax toxin translocon-lethal factor complex by cryo-electron microscopy.

We have visualized by cryo-electron microscopy (cryo-EM) the complex of the anthrax protective antigen (PA) translocon and the N-terminal domain of anthrax lethal factor (LF(N) inserted into a nanodisc model lipid bilayer. We have determined the structure of this complex at a nominal resolution of 16 Å by single-particle analysis and three-dimensional reconstruction. Consistent with our previous analysis of negatively stained unliganded PA, the translocon comprises a globular structure (cap) separated from the nanodisc bilayer by a narrow stalk that terminates in a transmembrane channel (incom

Effect of coating electrolytes on two-tailed surfactant bilayer coatings in capillary electrophoresis.

Surfactants such as dioctadecyldimethylammonium bromide (DODAB) form semi-permanent coatings that effectively prevent adsorption of cationic proteins onto the fused silica capillary in capillary electrophoresis (CE). The bilayer coating is generated by flushing the capillary with a 0.1 mM surfactant solution. However, formation of the bilayer is strongly dependent on the coating electrolyte. The effect of counter-ions, electrolyte concentrations and buffer co-ions were monitored based on: the separation of basic model proteins; the adsorption kinetics of DODA(+) onto fused silica; and dynamic

Molecular modelling of specific and non-specific anaesthetic interactions.

There has been rapid progress in molecular modelling in recent years. The convergence of improved software for molecular mechanics and dynamics, techniques for chimeric substitution and site-directed mutations, and the first x-ray structures of transmembrane ion channels have made it possible to build and test models of anaesthetic binding sites. These models have served as guides for site-directed mutagenesis and as starting points for understanding the molecular dynamics of anaesthetic-site interactions. Ligand-gated ion channels are targets for inhaled anaesthetics and alcohols in the centr

Histological structure of human nail as studied by synchrotron X-ray microdiffraction.

Three layers (characterized by different orientations of the keratin molecules) from the outer to the inner side of human nail were observed … high mechanical rigidity and hardness, both in the curvature direction and in the growth direction. Lipid bilayers (49 A thick) parallel to the nail surface fill certain ampullar dilations of the dorsal plate and intercellular spaces in the ventral plate. Using X-ray micro-diffraction, we show that onychomycosis disrupts the keratin structure, probably during th …

Exchange and interactions between lipid layers at the surface of a liposome solution.

Lipid organization and lipid transport processes occurring at the air-water interface of a liposome (lipid vesicle) solution are studied by conventional surface pressure-area measurements and interpreted by an adequate theory. At the interface of a dioleoyl phosphatidylcholine vesicle solution, used for demonstration, a well defined two layer structure selfassembles: vesicles disintegrate at the interface forming a surface-adsorbed lipid monolayer, which prevents further disintegration beyond about 1 dyne/cm surface pressure. A layer of vesicles now assembles in close association with the mono

Membrane properties of the sarcolemma and sarcoplasmic reticulum of pigs susceptible to malignant hyperthermia. Action of halothane.

The role of sarcolemma and sarcoplasmic reticulum in malignant hyperthermia was studied by the esr technique using the trapezius muscle membrane of both normal and genetically susceptible pigs. Normal and malignant hyperthermia membranes from sarcolemma as well as from sarcoplasmic reticulum did not show significant differences near the polar heads of the phospholipidic bilayer. In contrast, the fluidity and activation energy of normal membranes differed from those in malignant hyperthermia; in both sarcolemma and sarcoplasmic reticulum the mobility of the label was greater than the controls.

Intravesical liposome drug delivery and IC/BPS.

Intravesical therapy has previously shown to be effective in delaying or preventing recurrence of superficial bladder cancer. This local route of drug administration is now demonstrating promise in the treatment of interstitial cystitis/bladder pain syndrome (IC/BPS) with the benefit of minimal systemic side effects. Liposomes (LPs) are lipid vesicles composed of phospholipid bilayers surrounding an aqueous core. They can incorporate drug molecules, both hydrophobic and hydrophilic, and vastly improve cellular uptake of these drug molecules via endocytosis. Intravesical LPs have therapeutic ef…

Microstructure controlled electrostatic and channel mediated interactions between bacterial membrane and copper and carbon surfaces.

The outer membrane of Gram-negative bacteria acts as a key physical barrier against antibacterial agents. While some antibiotics traverse this barrier through porins, contact-killing metallic surfaces bypass molecular transport entirely by directly compromising membrane integrity, a process still poorly understood at the atomic scale. Here, Reactive Force-Field (ReaxFF) molecular dynamics simulations were employed to elucidate how metal and carbon surfaces modulate bacterial membrane destabilisation. Using an Escherichia coli lipid bilayer as a model, we investigated its interactions with nano

Surfactant-associated proteins perturb lipid membrane architecture: insights from nanoplasmonic sensing and differential scanning calorimetry.

This study investigates the biophysical interactions between pulmonary surfactant proteins (SP) and model lipid membranes. Using a combination of state-of-the-art nanoplasmonic sensing (NPS) and micro differential scanning calorimetry (MicroCal DSC), the effects of surfactant protein B (SP-B), surfactant protein C (SP-C), and their mixture on the structural and thermodynamic properties of liposomes composed of neutral phosphatidylcholines and anionic phosphatidylglycerol, (POPC : POPG and DPPC : DMPG, 80 : 20 molar ratio) were assessed. Dual-Interface Hierarchical Assembly of Asymmetric ICG-Lipid Conjugates Enables Finite J-type Aggregation and Two-Stage NIR-II Fluorescence Enhancement.

Aggregation-induced quenching severely limits the imaging properties of indocyanine green (ICG), despite its FDA approval and its emission in the near-infrared II (NIR-II) region. Herein, we introduce a dual-interface assembly strategy that orchestrates the sequential assembly of asymmetric ICG-lipid conjugates at lipid and liposome-protein interfaces to achieve two-stage NIR-II fluorescence enhancement. The ICG-sphingosine conjugates without sulfonate groups (ICG-C-Sp) form finite, weakly coupled J-type aggregates within lipid bilayers, producing markedly enhanced NIR-II fluorescence. Upon se…

[Changes in spatial organization in sarcoplasmic reticulum membranes in rabbits with experimental thyrotoxicosis].

The structure of sarcoplasmic reticulum membranes (SR) of skeletal muscles from normal and thyrotoxic rabbits was studied with the use of the fluorescent probe pyrene. It was found that protein globules in SR preparations of thyrotoxic animals were submerged into the lipid bilayer to a greater extent than in the reticulum of normal animals. Electrophoresis in polyacrylamide gel revealed no detectable differences between SR of normal and thyrotoxic rabbits. The ratio protein/lipid in SR membranes remained 2:1 in rabbits with thyrotoxicosis.

Novel biomimetic hybrid plasmonic nanocavity-based ECL strategy for the detection of extracellular vesicles.

Tumor-derived extracellular vesicles detection has emerged as an important clinical liquid biopsy approach for cancer diagnosis. In this work, we developed a novel hybrid plasmonic nanocavity consisting of hexagonal Au nanoplates nanoarray, SnS2/Au nanosheet layer and biomimetic lipid bilayer. Firstly, the hybrid plasmonic nanocavity combined the optical confinement for the ECL regulation and the biological recognition for the detection of extracellular vesicles. Secondly, MXene-derived Ti2N QDs have been prepared as ECL nanoprobe to label extracellular vesicles. Moreover, biomimetic lipid bil

In silico pharmacology: Drug membrane partitioning and crossing.

Over the past decade, molecular dynamics (MD) simulations have become particularly powerful to rationalize drug insertion and partitioning in lipid bilayers. MD simulations efficiently support experimental evidences, with a comprehensive understanding of molecular interactions driving insertion and crossing. Prediction of drug partitioning is discussed with respect to drug families (anesthetics; β-blockers; non-steroidal anti-inflammatory drugs; antioxidants; antiviral drugs; antimicrobial peptides). To accurately evaluate passive permeation coefficients turned out to be a complex theoretical …

Importance of lipids for upper motor neuron health and disease.

Building evidence reveals the importance of maintaining lipid homeostasis for the health and function of neurons, and upper motor neurons (UMNs) are no exception. UMNs are critically important for the initiation and modulation of voluntary movement as they are responsible for conveying cerebral cortex' input to spinal cord targets. To maintain their unique cytoarchitecture with a prominent apical dendrite and a very long axon, UMNs require a stable cell membrane, a lipid bilayer. Lipids can act as building blocks for many biomolecules, and they also contribute to the production of energy. Ther

[Study of channel-forming properties of latrotoxin in liposomes].

Interaction of latrotoxin with phospholipid vesicles and bilayer lipid membranes is shown to proceed differently. Latrotoxin when interacting with liposomes is sorbed on the membrane surface forming no ionic channels in this case. Only latrotoxin fragments obtained due to the toxin hydrolysis by pronase or trypsin are able to form channels. These fragments being inserted into liposomes are coupled strongly with the membrane and are not subjected to the further splitting by proteinases. The electrophoretic spectrum of peptides bound with liposomes is presented by protein components with a molec

Membrane lipid homeostasis in bacteria.

The ability of bacteria to control the biophysical properties of their membrane phospholipids allows them to thrive in a wide range of physical environments. Bacteria precisely adjust their membrane lipid composition by modifying the types of fatty acids that are produced by the biosynthetic pathway and altering the structures of pre-existing phospholipids. The recycling of phospholipids that are used as intermediates in the biosynthesis of other major membrane components is also crucial to bilayer stability in dividing cells. Here, the principal genetic and biochemical processes that are resp

Negatively charged phospholipids accelerate the membrane fusion activity of the plant-specific insert domain of an aspartic protease.

Various plants use antimicrobial proteins/peptides to resist phytopathogens. In the potato, Solanum tuberosum, the plant-specific insert (PSI) domain of an aspartic protease performs this role by disrupting phytopathogen plasma membranes. However, the mechanism by which PSI selects target membranes has not been elucidated. Here, we studied PSI-induced membrane fusion, focusing on the effects of lipid composition on fusion efficiency. Membrane fusion by the PSI involves an intermediate state whereby adjacent liposomes share their bilayers. We found that increasing the concentration of negativel

Thyroid hormone interactions with DMPC bilayers. A molecular dynamics study.

The structure and dynamics of thyroxine (T4), distal and proximal conformers of 3',3,5-triiodo-l-thyronine (T3d and T3p), and 3,5-diiodo-l-thyronine (T2) upon interaction with DMPC membranes were analyzed by means of molecular dynamics simulations. The locations, the more stable orientations, and the structural changes adopted by the hormones in the lipid medium evidence that the progressive iodine substitution on the beta ring lowers both the possibility of penetration and the transversal mobility in the membrane. However, the results obtained for T3d show that the number of iodine atoms in t

Electrochemical and PM-IRRAS characterization of cholera toxin binding at a model biological membrane.

A mixed phospholipid-cholestrol bilayer, with cholera toxin B (CTB) units attached to the monosialotetrahexosylganglioside (GM1) binding sites in the distal leaflet, was deposited on a Au(111) electrode surface. Polarization modulation infrared reflection absorption spectroscopy (PM-IRRAS) measurements were used to characterize structural and orientational changes in this model biological membrane upon binding CTB and the application of the electrode potential. The data presented in this article show that binding cholera toxin to the membrane leads to an overall increase in the tilt angle of t
