Lipidomic analysis reveals disturbances in glycerophospholipid and sphingolipid metabolic pathways in benzene-exposed mice.

Benzene, a known occupational and environmental contaminant, has been recognized as the hematotoxin and human carcinogen. Lipids have a variety of important physiological functions and the abnormal lipid metabolism has been reported to be closely related to the occurrence and development of many diseases. In the present study, we aim to utilize LC-MS/MS lipidomic platform to identify novel biomarkers and provide scientific clues for mechanism study of benzene hematotoxicity. Results showed that a total of 294 differential metabolites were obtained from the comparison of benzene-treated group a

Maternal lipidomic signatures in relation to spontaneous preterm birth and large-for-gestational age neonates.

Lipidome-wide metabolites may be useful biomarkers of pregnancy outcomes. We sought to characterize maternal lipidomic signatures associated with preterm birth and neonatal anthropometric parameters. Plasma samples were collected 24-28 weeks gestation, and lipidomic profiling was quantified using high-performance liquid chromatography tandem mass spectrometry. Lipid metabolites were analyzed individually and as whole lipid classes and subgroups based on degree of hydrocarbon chain saturation. Associations were estimated using linear and logistic regression. After false discovery adjustment (q

Cerebrospinal fluid lipidomic biomarker signatures of demyelination for multiple sclerosis and Guillain-Barré syndrome.

Multiple sclerosis (MS) and Guillain-Barré syndrome (GBS) are demyelinating disorders affecting the central nervous system and peripheral nervous system (PNS), respectively. Cerebrospinal fluid (CSF) is one of the most valuable sources of diagnostic biomarkers in neurological diseases. In the present study high sensitivity shotgun mass spectrometry was used to characterise the CSF lipidome of patients with MS, GBS and controls with non-demyelinating diseases. The quantification of 222 CSF lipid molecular species revealed characteristic changes in the absolute and relative lipid concentrations

Comparison of Single Phase and Biphasic Extraction Protocols for Lipidomic Studies Using Human Plasma.

Lipidomic profiling of plasma is an emerging field, given the importance of lipids in major cellular pathways, and is dependent on efficient lipid extraction protocols. Recent attention has turned to plasma lipidomics as a means to identify potential diagnostic and prognostic biomarkers related to dementia, neuropsychiatric health and disease. Although several solvent-based lipid extraction protocols have been developed and are currently in use, novel and more efficient methods could greatly simplify lipid analysis in plasma and warrant investigation. Human plasma from normolipidemic adult vol…

Plasma lipidome reveals critical illness and recovery from human Ebola virus disease.

Ebola virus disease (EVD) often leads to severe and fatal outcomes in humans with early supportive care increasing the chances of survival. Profiling the human plasma lipidome provides insight into critical illness as well as diseased states, as lipids have essential roles as membrane structural components, signaling molecules, and energy sources. Here we show that the plasma lipidomes of EVD survivors and fatalities from Sierra Leone, infected during the 2014-2016 Ebola virus outbreak, were profoundly altered. Focusing on how lipids are associated in human plasma, while factoring in the state

Plasma lipidomic profiling of treated HIV-positive individuals and the implications for cardiovascular risk prediction.

BACKGROUND: The increased risk of coronary artery disease in human immunodeficiency virus (HIV) positive patients is collectively contributed to by the human immunodeficiency virus and antiretroviral-associated dyslipidaemia. In this study, we investigate the characterisation of the plasma lipid profiles of treated HIV patients and the relationship of 316 plasma lipid species across multiple lipid classes with the risk of future cardiovascular events in HIV-positive patients. METHODS: In a retrospective case-control study, we analysed plasma lipid profiles of 113 subjects. Cases (n = 23) were

Lipidome analysis in multiple sclerosis reveals protein lipoxidative damage as a potential pathogenic mechanism.

Metabolomic and lipidomic analyses have been used for the profiling of neurodegenerative processes, both in targeted and untargeted approaches. In this work we have applied these techniques to the study of CSF samples of multiple sclerosis (MS) patients (n = 9), compared with samples of non-MS individuals (n = 9) using mass-spectrometry. We have used western-blot and analyzed cell culture to confirm pathogenic pathways suggested by mass-spectrometric measurements. The results of the untargeted approach of metabolomics and lipidomics suggest the existence of several metabolites and lipids discr…

Lipidomics Reveals Potential Mechanisms of Intestinal-Targeted Astaxanthin in Ameliorating Metabolic Dysfunction-Associated Steatotic Liver Disease in ApoE-/- Mice.

Metabolic dysfunction-associated steatotic liver disease (MASLD) represents a major global health burden with an urgent need for effective interventions. Although astaxanthin (AST) exhibits hepatoprotective potential, its efficacy is limited by poor oral bioavailability. This study evaluated an intestine-targeted astaxanthin delivery system (AST-LPs) in HFHC-diet-induced MASLD mice. AST-LPs administration exhibited superior efficacy over free AST in attenuating hepatic steatosis, improving lipid profiles and glucose tolerance, and suppressing inflammation. Lipidomics analysis revealed that AST…

Triphenyl phosphate exposure at physiologically/environmentally relevant concentrations promotes malignant progression of breast cancer: Insights from metabolomics and lipidomics.

Triphenyl phosphate (TPHP) is one of the main alternatives to brominated flame retardants and a typical environmental endocrine disruptor. Although the in vitro tumor-promoting effect of TPHP has been partially emphasized in previous studies, its impact on breast cancer, particularly at physiological/environmental relevant concentrations, had received inadequate attention. To investigate the pro-breast cancer effects of TPHP, both three-dimensional cancer cell spheroids models and chronic murine exposure models were established. Spheroid growth analysis, bioluminescent imaging, and multi-omics

Method-Dependent Variability in Hempseed Lipidomics: Combined Influence of Extraction, Derivatisation, and Genotype on Fatty Acid Composition and Stability.

Hempseed (Cannabis sativa L.) is recognised as a valuable source of nutritionally important lipids, particularly polyunsaturated fatty acids (PUFA). Despite growing interest in hempseed oil, the combined influence of extraction and derivatisation on fatty acid profiling remains poorly understood, limiting comparability and standardisation of lipidomic analyses, particularly for fibre-type hemp cultivars. This study evaluated the effects of microwave-assisted and Soxhlet extraction, combined with acidic and basic derivatisation, on fatty acid composition, lipid quality indices, and oxidative st

Mechanism-guided untargeted-to-targeted lipidomics identifies phosphatidylcholine 38:4 in rat bile as an Abcb4/Mdr2 inhibition marker.

Drug-induced liver injury can occur when the canalicular phospholipid floppase multidrug resistance protein 3 (Mdr2 in rodents) is inhibited, but there is still a lack of early biomarkers to detect this risk. In this study, bile duct-cannulated rats were dosed with multidrug resistance protein 3/Mdr2 inhibitor itraconazole (ITZ; 100 mg/kg/d for 3 days) to assess phospholipid changes via an untargeted-to-targeted lipidomics workflow. Untargeted profiling of bile and liver samples identified 1347 and 2475 tentative lipids, of which 221 and 404 were phosphatidylcholines (PCs) in bile and the live…

ExchangeXplorer: an R/Shiny application for processing and visualization of LC-HDX-MS data in metabolomics, lipidomics, and exposomics.

BACKGROUND: Reliable annotation remains a major challenge in untargeted LC-MS-based metabolomics, lipidomics, and exposomics. Liquid chromatography-hydrogen/deuterium exchange-mass spectrometry (LC-HDX-MS) provides orthogonal structural information by revealing the number of exchangeable hydrogens within a molecule, thereby supporting functional-group assignment, distinguishing isomeric structures, and reducing false-positive annotations. However, broader adoption of LC-HDX-MS for small-molecule analysis has been limited by the lack of dedicated software for systematic data processing and inte…

Targeted Lipidomics Reveals Citrus aurantifolia Peel Extract's Potential to Increase Lipid Catabolism With PC34:1 as a Discriminative Metabolite.

Regulating lipid metabolism is the main mechanism of exercise to control blood lipids, body weight, and prevent metabolic diseases. However, the effects of complex mixtures of phytochemicals, particularly from plant-derived extracts, on lipid metabolism remain poorly understood. Recently, we developed a green extraction method for lime (Citrus aurantifolia) peel, yielding an extract rich in three major compounds (hesperidin, limonin, and bergaptol). In this study, the effect of the extract on lipid metabolism was investigated in normal human hepatocytes (THLE-2) using a targeted lipidomics app…
