Comparison of 99mTc-HYNIC-PSMA-11, bone scan and post radioligand therapy images in mCRPC patients: A single center experience.

BACKGROUND: Prostate-Specific Membrane Antigen is overexpressed in primary and metastatic prostate cancer tissues. While PSMA PET agents have gained their role in the prostate cancer guidelines, Technetium-based PSMA agents have left behind. Given the novelty of 99mTc-HYNIC-PSMA-11, we conducted this study to compare its detection rate in tracing skeletal metastases in comparison to bone scans and also compared the number of metastatic lesions which showed PSMA uptake in the diagnostic scan with those detected in post-treatment scans after radioligand therapy (RLT) with 177Lu-PSMA-617-617. Peptide Receptor Radionuclide Therapy Improves Survival in Patients Who Progress After Resection of Gastroenteropancreatic Neuroendocrine Tumors.

BACKGROUND: Peptide receptor radionuclide therapy (PRRT) is an effective treatment for advanced gastroenteropancreatic (GEP) neuroendocrine tumors (NETs). We investigated a 2-decade experience with PRRT to determine whether PRRT confers a survival advantage to patients who progress after surgery versus other therapies. METHODS: We identified patients from our clinic who had resection/cytoreduction of GEP-NETs, then disease progression by Response Evaluation Criteria in Solid Tumors (RECIST) 1.1. The Kaplan-Meier method assessed progression-free survival (PFS) and overall survival (OS), calcul

PSMA and Sigma-1 receptor dual-targeted peptide mediates superior radionuclide imaging and therapy of prostate cancer.

Radionuclide therapy, in particular peptide receptor radionuclide therapy (PRRT), has emerged as a valuable means to combat malignant tumors. The specific affinity of ACUPA peptide toward prostate-specific membrane antigen (PSMA) renders the successful development of PRRT for prostate cancer. The clinical outcome of PRRT is, however, generally challenged by moderate tumor uptake and off-target toxicity. Here, we report on a novel design of Sigma-1 receptor and PSMA dual-receptor targeted peptide (S1R/PSMA-P) for superior radionuclide imaging and therapy of prostate cancer. S1R/PSMA-P was acqui

Development and Preclinical Evaluation of Dual-Target 68Ga/177Lu-Labeled FAP-Bisphosphonate Radioligands with Enhanced Osteoma Retention.

Bone metastases and associated skeletal-related events (SREs) severely compromise the survival and quality of life of patients with advanced cancer. Although bisphosphonates enable efficient bone targeting, their radiolabeled analogues exhibit limited uptake in osteolytic lesions due to the low hydroxyapatite content. Fibroblast activation protein, highly expressed in cancer-associated fibroblasts within the tumor microenvironment, provides tumor specificity and may enhance tracer accumulation in osteolytic bone metastases. To overcome this limitation, we developed two heterodimeric [68Ga]Ga/[

Development of a CD9-Targeted Radiopharmaceutical for Imaging and Radionuclide Therapy in CD9-Positive Glioma.

High expression of CD9 (cluster of differentiation 9) is closely associated with the poor prognosis of glioma, and it is necessary to develop targeted radiopharmaceuticals for diagnosis and treatment. The therapeutic efficacy of peptide-based drugs is often limited by their rapid metabolism. This study aims to develop an integrated theranostic radiopharmaceutical capable of in vivo CD9 targeting by modifying peptides with maleimide. The CD9-binding molecule DOTA-M-P was synthesized and labeled with 68Ga and 177Lu by using an indirect labeling method. Construction of a CD9-overexpressing cell l
