Development and validation of a nomogram to predict recurrence after hepatectomy in overweight patients with hepatocellular carcinoma: a retrospective cohort study.

BACKGROUND: Even after curative hepatectomy, postoperative recurrence remains a major challenge for patients with hepatocellular carcinoma (HCC). Body mass index (BMI) has been associated with the risk of HCC development; however, the prognostic impact of high BMI on postoperative recurrence in patients with HCC has not been fully elucidated. OBJECTIVE: To evaluate the risk factors associated with postoperative recurrence in overweight HCC patients with negative surgical margins and to develop a prognostic nomogram for recurrence prediction. METHODS: This single-center retrospective study in

Drug targets in stress-related disorders.

Nervous and immune systems mutually cooperate via release of mediators of both neurological and immunological derivation. Adrenocorticotropin hormone (ACTH) is a product of the hypothalamus-pituitary adrenal axis (HPAA) which stimulates secretion of corticosteroids from adrenals. In turn, corticosteroids modulate the immune response in virtue of their anti-inflammatory activity. On the other hand, catecholamines, products of the sympathetic nervous system (SNS), regulate immune function by acting on specific beta-adrenergic receptors. Conversely, cytokines released by monocytes/macrophages and

A multimodal deep learning radiomics nomogram integrating clinicopathological features for predicting pathological complete response after neoadjuvant therapy in triple-negative breast cancer: a multicenter retrospective study.

BACKGROUND: Accurate noninvasive estimation of pathological complete response (pCR) after neoadjuvant therapy (NAT) in triple-negative breast cancer (TNBC) may support multidisciplinary discussions, but multimodal MRI-ultrasound evidence remains limited. We developed and externally validated a clinicopathological deep learning radiomics nomogram and explored its potential auxiliary value in a strictly hypothetical post-hoc surgical-planning scenario. METHODS: With institutional review-board approval and waived consent, this retrospective multicenter diagnostic accuracy study included 436 biop

Prognostic Value of Pretreatment Modified Glasgow Prognostic Score in Nivolumab-Treated Advanced Non-Small Cell Lung Cancer.

Background/Objectives: Immune checkpoint inhibitors have become a standard treatment option for advanced/metastatic non-small cell lung cancer (NSCLC); however, considerable heterogeneity exists among patients regarding treatment response and survival. This study aimed to determine whether the pretreatment modified Glasgow Prognostic Score (mGPS), a marker of systemic inflammation and nutritional status, is prognostically associated with progression-free survival (PFS) and overall survival (OS) in patients with advanced/metastatic NSCLC treated with nivolumab at a single tertiary oncology cent

Treatment of metastatic uveal melanoma with adoptive transfer of tumour-infiltrating lymphocytes: a single-centre, two-stage, single-arm, phase 2 study.

BACKGROUND: Uveal melanoma is a rare tumour with no established treatments once metastases develop. Although a variety of immune-based therapies have shown efficacy in metastatic cutaneous melanoma, their use in ocular variants has been disappointing. Recently, adoptive T-cell therapy has shown salvage responses in multiple refractory solid tumours. Thus, we sought to determine if adoptive transfer of autologous tumour-infiltrating lymphocytes (TILs) could mediate regression of metastatic uveal melanoma. METHODS: In this ongoing single-centre, two-stage, phase 2, single-arm trial, patients (a

Can Nutritional and Inflammatory Markers Be Combined to Develop an Accurate Survival Model for Surgical Decision-making in Patients With Lung Cancer Bone Metastases?

BACKGROUND: Targeted and immunotherapies have improved the survivorship of patients with lung cancer and bone metastases. However, most existing models were developed during the chemotherapy era and do not accurately reflect survival outcomes in the current therapeutic context, leading to limited clinical applicability. Additionally, a wide range of inflammatory and nutritional markers has been identified as useful for cancer survival assessment. The most effective selection or combination of these markers for evaluating survival in patients with lung cancer and bone metastases, especially wit

Thymic commitment of regulatory T cells is a pathway of TCR-dependent selection that isolates repertoires undergoing positive or negative selection.

The seminal work of Le Douarin and colleagues (Ohki et al. 1987; Ohki et al. 1988; Salaun et al. 1990; Coutinho et al. 1993) first demonstrated that peripheral tissue-specific tolerance is centrally established in the thymus, by epithelial stromal cells (TEC). Subsequent experiments have shown that TEC-tolerance is dominant and mediated by CD4 regulatory T cells (Treg) that are generated intrathymically by recognition of antigens expressed on TECs (Modigliani et al. 1995; Modigliani et al. 1996a). From these and other observations, in 1996 Modigliani and colleagues derived a general model for

Dynamics of minimal residual disease in patients with multiple myeloma on continuous lenalidomide maintenance: a single-arm, single-centre, phase 2 trial.

UNLABELLED: Background Lenalidomide maintenance improves progression-free survival for patients with multiple myeloma, although its optimal duration is unknown. Clearance of minimal residual disease (MRD) in the bone marrow results in superior outcomes, although its attainment or sustainment does not alter clinical decision-making. Studies that have evaluated MRD serially are limited in length. We therefore aimed to evaluate longitudinal changes in MRD-status (dynamics) and their association with progression-free survival in patients with multiple myeloma. METHODS: In this single-centre, sing

A Retrospective Study of Best Supportive Care as Initial Treatment for Oral Squamous Cell Carcinoma.

Objective Patients with oral squamous cell carcinoma (OSCC) may choose best supportive care (BSC) as initial treatment over active treatment for several reasons. However, no previous reports have compared the clinical characteristics of OSCC patients opting for BSC as initial treatment with those receiving standard treatment. This study retrospectively analyzed these differences to identify the factors influencing this decision. Materials and methods This retrospective study was conducted at Niigata University Medical and Dental Hospital, using medical records within January 2018-December 2022
