Magnesium deficiency in patients with chronic pancreatitis identified by an intravenous loading test.

Magnesium deficiency is a common clinical condition that may exist despite a normal serum magnesium concentration. Patients with chronic pancreatitis could develop magnesium deficiency due to either malabsorption, diabetes mellitus, or chronic alcoholism. Since serum levels of magnesium are a poor indicator of magnesium deficiency, the retention of a low-dose intravenous magnesium load (0.1 mmol/kg body weight) was determined in 13 patients with chronic pancreatitis (10 due to alcoholism) and eight healthy controls. Percentage magnesium retention was greater in patients with chronic pancreatit…

Compartmental analysis of magnesium kinetics in Mg-sufficient and Mg-deficient rats.

In this study, we determined magnesium kinetic values in normal rats using stable-isotope techniques. Additionally, we calculated the mass of the exchangeable pools of Mg in Mg-deficient rats to determine whether it can be used as a marker of Mg status. Rats were fed either a control diet (1,000 mg Mg/kg) or a Mg-deficient diet (60 mg Mg/kg). After 2 weeks on the experimental diets, each rat received an intravenous injection of 26Mg. The plasma Mg disappearance curve over the next 7 days was used to measure the mass and fractional transport rate of 3 rapidly exchanging Mg metabolic pools. In c

Early morphological and immunological alterations in the spleen during magnesium deficiency in the rat.

Dietary magnesium deficiency in rodents, and especially in rats, causes inflammation and leads to alterations in the immune response. One of the characteristics of magnesium deficiency in the rat is a marked enlargement of the spleen. Considering the importance of the spleen for the immune response, in this study we have evaluated histological, cytological and immunological changes in this organ of rats in early stages of this deficiency. For this purpose, male weaning Wistar rats were pair-fed with either control or magnesium-deficient diet, for 2, 4 or 8 days. Results indicate that after 8 d…

[Effects of dietary magnesium deficiency in the rat: with special reference to ultrastructural examination].

Epidemiologically, it has been suggested that dietary magnesium/calcium imbalance is associated with the risk of heart diseases. In the present study, the effects of magnesium deficiency and/or calcium over intake were investigated in rats. Male Sprague-Dawley rats were divided into 4 groups, and respectively fed basal diet (AIN-76) alone (Group 1), calcium-doubled AIN-76 diet (Group 2), magnesium-deficient AIN -76 diet (Group 3) and magnesium-deficient/calcium-doubled AIN-76 diet (Group 4) for 19 days. A biochemical assay using inductively coupled plasma showed that the magnesium concentratio…

Iron accumulation in tissues of magnesium-deficient rats with dietary iron overload.

The mineral imbalances in magnesium-deficient rats with dietary iron overload were studied. Forty-four male Wistar rats were divided into six groups and fed six diets, two by three, fully crossed: magnesium adequate or deficient, and iron deficient, adequate, or excess. The concentrations of iron, magnesium, calcium, and phosphorus in tissues of the rats were measured. The results were as follows: (1) The excess iron intake reinforced the iron accumulation in liver and spleen of magnesium deficient rats; (2) The saturation of iron binding capacity was enormously elevated in the magnesium defic

Change in glucose homeostasis in rats by long-term magnesium-deficient diet.

It is widely known that hypomagnesemia is one of the symptoms observed in diabetic patients. This study was performed to assess the effect of chronic magnesium (Mg) deficiency on glucose metabolism in rats. Male Sprague-Dawley rats (at the age of four weeks) were given a Mg-deficient diet or a control diet for two to eight weeks. The rats were orally administered sucrose solution (2 g/kg BW) every two weeks, and blood was drawn from a tail vein before and 15 min after sucrose loading to determine the concentrations of blood glucose and plasma insulin. At the same time, other rats in a non-fast

Magnesium deficiency in alcoholism: possible contribution to osteoporosis and cardiovascular disease in alcoholics.

Magnesium (Mg) deficiency occurs frequently in chronic alcoholism and may contribute to the increased incidence of osteoporosis and cardiovascular disease seen in this population. Mg deficiency is primarily due to renal Mg-wasting and is exacerbated by dietary Mg deprivation, gastrointestinal losses with diarrhea or vomiting, as well as concomitant use of drugs such as diuretics and aminoglycosides. Osteoporosis is prevalent in the alcoholic population. Mg deficiency may contribute to increased bone loss by its effects on mineral homeostasis. In Mg depletion, there is often hypocalcemia due to

[Characteristics of the audiogenic convulsive crisis in mice made sensitive by magnesium deficiency].

Mice of the OF1, C57BL/6, AKR, C3H/He, DBA/2, BALB/C, B6D2F1 and CBA strains are susceptible to audiogenic seizures after 40 or 20 days of acute magnesium deficiency. The duration of the various phases of the audiogenic seizure response (at 100 dBA) (wild running latency period, convulsions latency period and the clonic and tonic convulsions) among these mice was measured. Using the Kruskall-Wallis test, no difference was recorded using these measurements and those obtained for genetically audio-susceptible animals. This shows that acute magnesium deficiency-induced audiogenic seizures develop…

[The relation of physical and mental stress to magnesium deficiency in patients with variant angina].

In this study we assessed the role of psychological factor in the etiology of coronary vasospasm using the Cornell Medical Index (CMI), focusing attention on the relationship between stress and serum magnesium (Mg). The study subjects consisted of 25 patients with variant angina (VA), 32 with old myocardial infarction without vasospasm (OMI), and 34 healthy men (controls). On a neurosis-discriminative diagram of CMI, areas I and II were considered as normal and areas III and IV were considered to be a neurotic disorder. The stress test included exercise and a quiz. Exercise test was performed

Dementias: the role of magnesium deficiency and an hypothesis concerning the pathogenesis of Alzheimer's disease.

Evidence is presented indicating that dementias are associated with a relative insufficiency of Magnesium (Mg) in the brain. Such insufficiency may be attributable to low intake or retention of Mg; high intake of a neurotoxic metal, such as aluminum (Al), which inhibits activity of Mg-requiring enzymes; or impaired transport of Mg and/or enhanced transport of the neurotoxic metal into brain tissue. It is proposed that Alzheimer's disease (AD) involves a defective transport process, characterized by both an abnormally high incorporation of Al and an abnormally low incorporation of Mg into brain

Magnesium deficiency inhibits biosynthesis of blood glutathione and tumor growth in the rat.

Previously we found that blood glutathione (GSH) levels increase in response to tumor growth in the rat and that this increase is not prevented with zinc deficiency. We also found that zinc deficiency which inhibited tumor growth did not prevent this increase in blood GSH. Therefore, the objectives of this study were to determine the effects of another nutritional modification, namely magnesium deficiency, on blood GSH status and on tumor growth. Magnesium was selected because it is an obligatory cofactor in GSH synthesis and in all biosynthetic reactions involving ATP. To this end, magnesium-…

Magnesium deficiency results in increased suberization in endodermis and hypodermis of corn roots.

The composition of the aliphatic components of suberin in the stele and cortex of young corn (Zea mays L.) roots was determined by combined gas-liquid chromatography/mass spectrometry of the LiAlD(4) depolymerization products. omega-Hydroxy acids were shown to be the major class of the aliphatic components of both the hypodermal (35%) and endodermal (28%) polymeric materials with the dominant chain length being C(24) in the former and C(16) in the latter. Nitrobenzene oxidation of the roots generated p-hydroxybenzaldehyde and vanillin with much less syringaldehyde. Electron microscopic examina

Effect of magnesium deficiency and excess on renal tubular potassium transport in the rat.

1. Dietary magnesium deficiency is commonly associated with significant potassium depletion although the mechanisms responsible are unknown. Because the kidney has an important role in both magnesium and potassium homeostasis, clearance and micropuncture experiments were performed on thyroparathyroidectomized magnesium-deficient, normal and hypermagnesaemic rats to study the effect of body magnesium status on renal potassium handling. 2. Dietary magnesium restriction that reduced total-body magnesium by 30% did not alter renal potassium excretion despite a 10% reduction in total-body potassium…

Light microscopic and ultrastructural changes of the exocrine pancreas in magnesium deficient rats.

In magnesium deficient rats with a clinical picture of protein malnutrition, pancreatic changes typical of protein malnutrition were expected. However, in rats fed for four weeks on a low magnesium diet (protein content 25%), light and electron microscopic studies revealed that the acinar cells of the pancreas were packed with zymogen granules, suggesting a disturbance in the discharge (rather than in the production) of the pancreatic enzymes. The mitochondria and the lumina of the RER were swollen. The nuclei had an irregular outline, the chromatin was aggregated into irregular granules and t

Effect of magnesium deficiency and food restriction on the immune response in young mice.

Similar batches of five-week-old C57 Bl/6 mice were given either a magnesium-deficient diet (4mg Mg/100 g), or a control diet (40 mg/100 g). … od limitation might therefore mainly affect immature IgM producing cells with a high dividing rate. Magnesium deficiency produced a drastic fall in the primary and secondary immune responses, as measured by the number of spleen AFC. The number RFC was also much lower in the spleen of deficient animals. Consequently, the spleen immune system is deeply affected by this …

The postmortem diagnosis of magnesium deficiency: studies in an animal model for the human infant.

Weanling rats were studied as a model for the human infant to determine the optimal tissue in which to assess the status of magnesium after death. Control rats were fed laboratory chow or purified diets that provided a surfeit of magnesium and accommodated a normal rate of growth. Other rats were fed diets that resulted in two degrees of magnesium deficiency: one that might result in spontaneous death within one week, and the other, within two weeks. These times may correlate with six months and one year in the human infant, the period during which the sudden infant death syndrome usually occu…

Plasma parathyroid hormone and calcitonin levels in hypocalcaemic magnesium-deficient calves.

Mg deficiency was experimentally induced in 3 calves given Mg-deficient semi-synthetic milk (10 mg of Mg/l of milk). Three control calves received the same milk supplemented with Mg (190 mg of Mg/l of milk). Calcium and vitamin D contents of this milk were similar to that of natural milk. Daily weight gain in control calves was similar to that observed in calves given natural milk. The daily weight gain of Mg-deficient calves was not statistically different from that of control calves during the first 6 weeks of Mg-deficiency. Then clinical symptoms of Mg-deficiency appeared (loss of appetite,

The influence of magnesium ions on pyruvate kinase-deficient red blood cells.

The effect of intravenous and oral administration of magnesium containing drug (Magnesium Diasporal, Protina, München) on the erythrocyte pyruvate kinase (PK) activity in two cases of congenital nonspherocytic hemolytic anemia associated with PK deficiency has been examined. Intravenous injection of the drug has been followed by transient increase in magnesium content. Positive correlation between erythrocyte magnesium ions concentration and PK activity has been found. The authors propose a hypothesis that the observed correlation may be due to the erythrocyte accumulation of some PK activator

Ultrastructural changes of the hypothalamo-hypophysial neurosecretory system in the magnesium deficient rats.

A magnesium deficient diet caused transient but marked degenerative changes in the rat hypothalamo-hypophysial neurosecretory system which strongly resembled in many ultrastructural respects those induced by a prolonged administration of aldosterone as previously reported by us. The possible mechanism for this selective alteration in the neurosecretory neurons has been briefly discussed with regard to aldosterone secretion.
