Alteration of transcriptional networks in the entorhinal cortex after maternal immune activation and adolescent cannabinoid exposure.

Maternal immune activation (MIA) and adolescent cannabinoid exposure (ACE) have both been identified as major environmental risk factors for schizophrenia. We examined the effects of these two risk factors alone, and in combination, on gene expression during late adolescence. Pregnant rats were exposed to the viral infection mimic polyriboinosinic-polyribocytidylic acid (poly I:C) on gestational day (GD) 15. Adolescent offspring received daily injections of the cannabinoid HU210 for 14days starting on postnatal day (PND) 35. Gene expression was examined in the left entorhinal cortex (EC) using

Systematic Review and Meta-Analyses of Perinatal Death and Maternal Exposure to Tobacco Smoke During Pregnancy.

We conducted a systematic review and 3-part meta-analysis to characterize the relationship between smoking and perinatal death, defined as the combination of stillbirth and neonatal death. The PubMed database was searched (1956-August 31, 2011) with keywords, and manual reference searches of included articles and Surgeon Generals' reports were conducted. The full texts of 1,713 articles were reviewed, and 142 articles that examined the associations between active or passive smoking and perinatal death were included in the meta-analyses. Data were abstracted by 2 reviewers. Any active maternal

Exposure to stimulatory CpG oligonucleotides during gestation induces maternal hypertension and excess vasoconstriction in pregnant rats.

Bacterial infections increase risk for pregnancy complications, such as preeclampsia and preterm birth. Unmethylated CpG DNA sequences are present in bacterial DNA and have immunostimulatory effects. Maternal exposure to CpG DNA induces fetal demise and craniofacial malformations; however, the effects of CpG DNA on maternal cardiovascular health have not been examined. We tested the hypothesis that exposure to synthetic CpG oligonucleotides (ODNs) during gestation would increase blood pressure and cause vascular dysfunction in pregnant rats. Pregnant and nonpregnant female rats were treated wi…

Maternal exposure to diluted diesel engine exhaust alters placental function and induces intergenerational effects in rabbits.

BACKGROUND: Airborne pollution is a rising concern in urban areas. Epidemiological studies in humans and animal experiments using rodent models indicate that gestational exposure to airborne pollution, in particular diesel engine exhaust (DE), reduces birth weight, but effects depend on exposure duration, gestational window and nanoparticle (NP) concentration. Our aim was to evaluate the effects of gestational exposure to diluted DE on feto-placental development in a rabbit model. Pregnant females were exposed to diluted (1 mg/m(3)), filtered DE (NP diameter ≈ 69 nm) or clean air (controls) fo

Maternal exposure to air pollutant PM2.5 and PM10 during pregnancy and risk of congenital heart defects.

Maternal exposure to ambient air pollution has increasingly been linked to congenital heart defects (CHDs). The objective of this study was to evaluate whether high levels of maternal exposure to PM2.5 and PM10 are related to increased risk of CHDs in Wuhan, China. We conducted a cohort study with a total of 105,988 live-born infants, stillbirths, and fetal deaths. The study included mothers living in the urban district of Wuhan during pregnancy over the 2-year period from 10 June 2011 to 9 June 2013. For each study participant, we assigned 1-month and 1-week averages of PM10 and PM2.5 exposur…

Maternal exposure to intimate partner abuse before birth is associated with autism spectrum disorder in offspring.

We sought to determine whether maternal (a) physical harm from intimate partner abuse during pregnancy or (b) sexual, emotional, or physical abuse before birth increased risk of autism spectrum disorder. We calculated risk ratios for autism spectrum disorder associated with abuse in a population-based cohort of women and their children (54,512 controls, 451 cases). Physical harm from abuse during pregnancy was not associated with autism spectrum disorder. However, autism spectrum disorder risk was increased in children of women who reported fear of partner or sexual, emotional, or physical abu

Maternal L-glutamine supplementation prevents prenatal alcohol exposure-induced fetal growth restriction in an ovine model.

Prenatal alcohol exposure is known to cause fetal growth restriction and disturbances in amino acid bioavailability. Alterations in these parameters can persist into adulthood and low birth weight can lead to altered fetal programming. Glutamine has been associated with the synthesis of other amino acids, an increase in protein synthesis and it is used clinically as a nutrient supplement for low birth weight infants. The aim of this study was to explore the effect of repeated maternal alcohol exposure and L-glutamine supplementation on fetal growth and amino acid bioavailability during the thi

Congenital microtia in a neonate due to maternal isotretinoin exposure 1 month before pregnancy: Case Report.

Isotretinoin is a drug used for treating severe cystic/nodular acne. Severe malformations have been documented in neonates whose mothers had taken isotretinoin during pregnancy. Women who became pregnant one cycle after completing therapy are believed to be at teratogenic risk not higher than baseline. We describe the case of a newborn whose mother had taken the drug for 4 weeks. The woman then had contraception for 4 weeks (after the drug treatment had finished), and became pregnant after that period. The newborn had isolated bilateral microtia due to suspected isotretinoin exposure. His moth

Prenatal methylmercury exposure through maternal rice ingestion: insights from a feasibility pilot in Guizhou Province, China.

Maternal hair and blood were investigated as biomarkers for prenatal methylmercury (MeHg) exposure among seventeen mothers recruited at parturition in Wanshan, Guizhou Province, China, where rice ingestion was the primary MeHg exposure pathway and atmospheric mercury (Hg) levels were elevated. For all three trimesters (n = 51), hair total Hg (THg) and MeHg concentrations ranged from 0.27 to 4.9 μg/g (median: 0.96 μg/g) and 0.077 to 2.3 μg/g (median: 0.43 μg/g), respectively, while blood THg levels ranged from 1.7 to 11 μg/L (median: 3.0 μg/L, n = 17). Despite adequate hair washing procedures,

Maternal diet, prenatal exposure to dioxins and other persistent organic pollutants and anogenital distance in children.

We investigated the potential endocrine disruptive effect of prenatal exposure to persistent organic pollutants (POPs) through maternal diet, by measuring anogenital distance in newborns and young children. We included 231 mothers and their newborns measured at birth from the Rhea study in Crete, Greece and the Hmar study in Barcelona, Spain and 476 mothers and their children measured between 1 and 2 years from the Rhea study. We used food frequency questionnaires to assess maternal diet and estimated plasma dioxin-like activity by the Dioxin-Responsive Chemically Activated LUciferase eXpressi

[Relationship of maternal malnutrition caused by Di(2-ethylhexyl) phthalate exposure with lifestyle disease in offspring].

The hypothesis that offspring growing up malnourished during their fetal period have a high risk of lifestyle diseases in later life has been attracting great attention. Although animal experiments and epidemiological studies have been reported, most of them focused on the deficiency of maternal malnutritional elements or starvation. We found that di(2-ethylhexyl) phthalate (DEHP) decreased maternal plasma triglyceride levels, which is a significant source of nutrients for fetuses, in mice. Therefore, we analyzed how offspring exposed to malnutritional status during their fetal period develop

Impairment of rat fetal beta-cell development by maternal exposure to dexamethasone during different time-windows.

AIM: Glucocorticoids (GCs) take part in the direct control of cell lineage during the late phase of pancreas development when endocrine and exocrine cell differentiation occurs. However, other tissues such as the vasculature exert a critical role before that phase. This study aims to investigate the consequences of overexposure to exogenous glucocorticoids during different time-windows of gestation for the development of the fetal endocrine pancreas. METHODS: Pregnant Wistar rats received dexamethasone acetate in their drinking water (1 µg/ml) during the last week or throughout gestation. Fet

Preadolescent behavior problems after prenatal cocaine exposure: Relationship between teacher and caretaker ratings (Maternal Lifestyle Study).

BACKGROUND: We previously reported an association between prenatal cocaine exposure (PCE) and childhood behavior problems as observed by the parent or caretaker. However, these behavior problems may not manifest in a structured environment, such as a school setting. OBJECTIVE: We determined whether there is an association between PCE and school behavior problems and whether ratings of behavior problems from the teacher differ from those noted by the parent or caretaker. METHODS: The Maternal Lifestyle Study, a multicenter study, enrolled 1388 children with and without PCE at one month of age

Maternal exposure to ambient levels of benzene and neural tube defects among offspring: Texas, 1999-2004.

BACKGROUND: Previous studies have reported positive associations between maternal exposure to air pollutants and several adverse birth outcomes. However, there have been no studies assessing the association between environmental levels of hazardous air pollutants, such as benzene, and neural tube defects (NTDs), a common and serious group of congenital malformations. OBJECTIVE: Our goal was to conduct a case-control study assessing the association between ambient air levels of benzene, toluene, ethylbenzene, and xylene (BTEX) and the prevalence of NTDs among offspring. METHODS: The Texas Bir…

Long-term effect of prenatal exposure to maternal microfilaraemia on immune responsiveness to filarial parasite antigens.

To identify long-term effects of prenatal exposure to maternal filarial-parasite infection, we assessed lymphocyte responses in 21 Polynesian children born 17-19 years previously to mothers diagnosed as being microfilaraemic or infection-free. All children lived on an island endemic for bancroftian filariasis but were free from infection at the time of study. While children (n = 10) of infection-free mothers responded vigorously to microfilarial antigen with lymphocyte proliferation, production of interleukin 2 (IL-2), IL-5, IL-10, granulocyte macrophage colony-stimulating factor (GM-CSF), and

Maternal exposure to volatile organic compounds and the risk of congenital anomalies: A systematic review and meta-analysis.

Volatile organic compounds (VOCs) are widespread environmental and occupational pollutants with potential teratogenic effects. Growing evidence indicates that maternal exposure to VOCs during pregnancy may increase the risk of congenital anomalies in offspring. This systematic review aims to evaluate and synthesize the association between maternal exposure to benzene, toluene, xylene, and formaldehyde during pregnancy and the occurrence of congenital anomalies, including neural tube defects (NTDs), congenital heart defects (CHDs), and cleft lip and/or palate (CL/P). A systematic search was con…

Associations of maternal exposure to nonylphenol and bisphenols with precocious puberty in girls: A nested case-control study.

Few studies have explored the association of prenatal exposure to typical endocrine disrupting chemicals such as nonylphenol (NP) and bisphenols with the odds of precocious puberty (PP) in girls. To evaluate the associations of maternal exposure to NP and bisphenols during pregnancy with the odds of PP [including central PP (CPP) and incomplete PP (IPP) subtypes] in girls, this study included 87 CPP cases, 109 IPP cases, and 588 matched (1: 3) controls based on a Wuhan birth cohort (2012-2014). Maternal urinary concentrations of three bisphenols and two NP metabolites [including oxo-NP and hyd…

The effects of maternal wildfire exposure: a 28-year longitudinal study of Chile wildfires and neonatal outcomes.

BACKGROUND: Wildfires are an increasingly important source of air pollution and environmental stress, yet their impacts on perinatal health in middle-income countries remain poorly understood. Chile has experienced a marked rise in wildfire activity over the last three decades, providing a unique setting to study these associations. METHODS: We conducted a nationwide panel data analysis using a municipality-month panel for 1990-2017 by aggregating individual birth records to municipality-month cells and linking them to municipality-month wildfire indicators in the mother's municipality of res
