Fear conditioning: Insights into learning, memory and extinction and its relevance to clinical disorders.

Fear, whether innate or learned, is an essential emotion required for survival. The learning, and subsequent memory, of fearful events enhances our ability to recognise and respond to threats, aiding adaptation to new, ever-changing environments. Considerable research has leveraged associative learning protocols such as contextual or auditory forms of fear conditioning in rodents, to understand fear learning, memory consolidation and extinction phases of memory. Such assays have led to detailed characterisation of the underlying neurocircuitry and neurobiology supporting fear learning processe…

Dynamic brain states during encoding and their post-encoding reinstatement predicts episodic memory in children.

Episodic memory, the ability to rapidly learn and explicitly remember past events and experiences, plays a critical role in children's academic learning and knowledge acquisition. The formation of lasting memories relies on the brain's ability to dynamically organize its activity. How these neural configurations unfold moment-by-moment across encoding and offline phases remains poorly understood. To probe these dynamics, we applied a novel Bayesian Switching Dynamic Systems approach, a hidden Markov model with automatic state detection, to fMRI data from children performing scene encoding foll

Rapamycin prevents drug seeking via disrupting reconsolidation of reward memory in rats.

The maladaptive drug memory developed between the drug-rewarding effect and environmental cues contributes to difficulty in preventing drug relapse. Established reward memories can be disrupted by pharmacologic interventions following their reactivation. Rapamycin, an inhibitor of mammalian target of rapamycin (mTOR) kinase, has been proved to be involved in various memory consolidation. However, it is less well characterized in drug memory reconsolidation. Using a conditioned place preference (CPP) procedure, we examined the effects of systemically administered rapamycin on reconsolidation of…

A prolonged stress rat model recapitulates some PTSD-like changes in sleep and neuronal connectivity.

Chronic post-traumatic stress disorder (PTSD) exhibits psychological abnormalities during fear memory processing in rodent models. To simulate long-term impaired fear extinction in PTSD patients, we constructed a seven-day model with multiple prolonged stress (MPS) by modifying manipulation repetitions, intensity, and unpredictability of stressors. Behavioral and neural changes following MPS conveyed longitudinal PTSD-like effects in rats for 6 weeks. Extended fear memory was estimated through fear retrieval induced-freezing behavior and increased long-term serum corticosterone concentrations

Direct comparison of reconsolidation of traumatic memories and prolonged exposure therapy: A randomized controlled trial.

New therapies are needed for posttraumatic stress disorder (PTSD), particularly in military service members (SMs). Reconsolidation of traumatic memories (RTM) is a novel treatment with promising results in small clinical trials. We randomized 94 active duty or retired SMs (Mage = 45.80, 31.0% women) with PTSD to receive up to ten 90-min sessions of RTM (n = 48) or prolonged exposure (PE; n = 46) to ascertain whether RTM achieved a greater and/or faster response. The Clinician-Administered PTSD Scale for DSM-5 (CAPS-5) was used to establish eligibility and assess response at 2 weeks and 2, 6, a

Disrupting fear memory reconsolidation in individuals with fear of spiders with cTBS: A Proof-of-Concept Study.

Anxiety disorders can be effectively treated with both cognitive behavioural therapy and psychopharmacological interventions. However, only 50 % of patients demonstrate significant benefits from these approaches. Therefore, investigating strategies to improve treatment effectiveness or develop novel therapeutic approaches remains an important research objective. Current therapeutic modalities may leave the original fear memory intact, potentially leading to symptom recurrence over time. In contrast, the disruption of reconsolidation processes can facilitate permanent modifications to fear memo

The role of frontal cortex in novel-word learning and consolidation: Evidence from focal transcranial direct current stimulation.

Previous studies have demonstrated that conventional transcranial direct current stimulation (tDCS) can enhance novel-word learning. However, because of the widespread current that is induced by these setups and lack of appropriate control conditions, little is known about the underlying neural mechanisms. In the present double-blinded and sham-tDCS controlled study, we investigated for the first time if regionally precise focal tDCS targeting two key nodes of the novel-word learning network at different time points would result in regionally and temporally distinct effects. 156 participants c

Movie-watching evokes ripple-like activity within events and at event boundaries.

Ripples are fast oscillatory events widely recognized as crucial markers for memory consolidation and neural plasticity. These transient bursts of activity are thought to coordinate information transfer between the hippocampus and neocortical areas, providing a temporal framework that supports the stabilization and integration of new memories. However, their role in human memory encoding during naturalistic scenarios remains unexplored. Here, we recorded intracranial electrophysiological data from ten epilepsy patients watching a movie. Ripples were analyzed in the hippocampus and neocortical …

A critical perspective on updating drug memories through the integration of memory editing and brain stimulation.

Addiction is a persistent, recurring condition characterized by repeated relapses despite the desire to control drug use or maintain sobriety. The attainment of abstinence is hindered by persistent maladaptive drug-associated memories, which drive drug-seeking and use behavior. This article examines the preliminary evidence supporting the combination of non-invasive brain stimulation (NIBS) techniques and memory editing (or reconsolidation) interventions as add-on forms of treatment for individuals with substance-related disorders (SUD). Studies have shown that NIBS can modestly reduce drug us

Sotalol Treatment may Interfere With Retrieval, Expression, and/or Reconsolidation Processes Thus Disrupting Traumatic Memories in a Post-Traumatic Stress Disorder Mice Model.

The processes by which fear memory is encoded, consolidated, and re-consolidated are extremely complex and appear to require the release of stress hormones, especially adrenaline (AD). AD improves contextual fear memory, acting specifically on peripheral β2-adrenoceptors. Propranolol (peripheral and central β-adrenoceptor antagonist) treatment was shown to prevent post-traumatic stress disorder (PTSD) development and reduce its symptoms. However, propranolol has several side effects. Thus, we aimed to evaluate if sotalol (a peripheral β-adrenoceptor antagonist) treatment interferes with retrie

The histone demethylase KDM6B in the medial prefrontal cortex epigenetically regulates cocaine reward memory.

Epigenetic remodeling contributes to synaptic plasticity via modification of gene expression, which underlies cocaine-induced long-term memory. A prevailing hypothesis in drug addiction is that drugs of abuse rejuvenate developmental machinery to render reward circuitry highly plastic and thus engender drug memories to be highly stable. Identification and reversal of these pathological pathways are therefore critical for cocaine abuse treatment. Previous studies revealed an interesting finding in which the mRNA of histone lysine demethylase, KDM6B, is upregulated in the medial prefrontal corte

Implicit and explicit memory in amnesia: some explanations and predictions by the TraceLink model.

After a brief overview of some of the characteristics and neuroanatomy of amnesia, a new model of amnesia is described: the TraceLink model. One novel aspect of the model is that it makes specific and testable predictions regarding semantic dementia, a recently described disorder that is viewed here as being related to amnesia. The TraceLink model consists of: a trace system (roughly the neocortix), a link system (hippocampus and adjacent areas), and a modulatory system (certain basal forebrain nuclei). Different forms of learning in the TraceLink model are explained, followed by a discussion

Astrocytes amplify cortical dopamine to encode winning memory in male mice.

Memories of winning experiences reinforce dominance hierarchies, yet mechanisms encoding these long-lasting "winning memories" remain unclear. Here, we introduce a combined Tube & Conditioned Place Preference (CPP) paradigm and applied fiber photometry, genetic manipulations, and optogenetics to characterize social competition between adult male mice. We found that initial victories trigger dopamine D2 receptor-mediated calcium activities of astrocytes in the medial prefrontal cortex (mPFC) and subsequent glutamate release. This mechanism enhances dopaminergic responses in mPFC neurons project

Cellular mechanisms of contextual fear memory reconsolidation: Role of hippocampal SFKs, TrkB receptors and GluN2B-containing NMDA receptors.

Memories are stored into long-term representations through a process that depends on protein synthesis. However, a consolidated memory is not static and inflexible and can be reactivated under certain circumstances, the retrieval is able to reactivate memories and destabilize them engaging a process of restabilization known as reconsolidation. Although the molecular mechanisms that mediate fear memory reconsolidation are not entirely known, so here we investigated the molecular mechanisms in the hippocampus involved in contextual fear conditioning memory (CFC) reconsolidation in male Wistar ra

Selective, retrieval-independent disruption of methamphetamine-associated memory by actin depolymerization.

BACKGROUND: Memories associated with drugs of abuse, such as methamphetamine (METH), increase relapse vulnerability to substance use disorder. There is a growing consensus that memory is supported by structural and functional plasticity driven by F-actin polymerization in postsynaptic dendritic spines at excitatory synapses. However, the mechanisms responsible for the long-term maintenance of memories, after consolidation has occurred, are largely unknown. METHODS: Conditioned place preference (n = 112) and context-induced reinstatement of self-administration (n = 19) were used to assess the

Circuit mechanism underlying fragmented sleep and memory deficits in 16p11.2 deletion mouse model of autism.

Sleep disturbances are prevalent in children with autism spectrum disorder (ASD). Strikingly, sleep problems are positively correlated with the severity of ASD symptoms, such as memory impairment. However, the neural mechanisms underlying sleep disturbances and cognitive deficits in ASD are largely unexplored. Here, we show that non-rapid eye movement sleep (NREMs) is fragmented in the 16p11.2 deletion mouse model of ASD. The degree of sleep fragmentation is reflected in an increased number of calcium transients in the activity of locus coeruleus noradrenergic (LC-NE) neurons during NREMs. In

A proposed attention-based model for spatial memory formation and retrieval.

Animals use sensory information and memory to build internal representations of space. It has been shown that such representations extend beyond the geometry of an environment and also encode rich sensory experiences usually referred to as context. In mammals, contextual inputs from sensory cortices appear to be converging on the hippocampus as a key area for spatial representations and memory. How metric and external sensory inputs (e.g., visual context) are combined into a coherent and stable place representation is not fully understood. Here, I review the evidence of attentional effects alo
