Differential effect of human interferon on normal and neoplastic T- and B-cell lymphocytes.

The effect of human interferon (HIF) on the growth and function of normal and neoplastic proliferating human T and B cells was studied. Cell cycle analysis by flow-cytometry showed that up to 2000 units of HIF had little effect on the proliferating fraction (that is, the present S + G2 + M phases of the cell cycle) of normal T cells grown continuously wit growth-promoting factor from PHA-stimulated lymphocytes, while 500 units of HIF suppressed the percent S + G2 + M of the acute lymphocytic leukemia (ALL) T-cell line, MOLT, by 36%. Up to 2000 units of HIF had a moderate enhancing effect on th

Silencing Br-C impairs larval development and chitin synthesis in Lymantria dispar larvae.

In insects, 20-hydroxyecdysone (20E) mediates developmental transitions and regulates molting processes through activation of a series of transcription factors. Broad-Complex (Br-C), a vital gene in the 20E signalling pathway, plays crucial roles during insect growth processes. However, whether Br-C affects chitin synthesis in insects remains unclear. In the present study, the Br-C gene from Lymantria dispar, a notorious defoliator of forestry, was identified based on transcriptome data, and subjected to bioinformatic analysis. The regulatory functions of LdBr-C in chitin synthesis and metabol…

Integrated computational and experimental identification of a novel ML-IAP-targeting peptide with antiproliferative activity in leukaemia cells.

Melanoma inhibitor of apoptosis protein (ML-IAP) is an IAP family member involved in tumour cell survival and a potential target in ML-IAP-expressing leukaemia. Here, 59 319 tetrapeptides were screened against ML-IAP by molecular docking, and four top-ranked peptides were selected. Microscale thermophoresis confirmed binding of peptides 1-4 to ML-IAP, with peptide-1 showing the lowest Kd value and a lower Kd value than the Smac peptide. Structure-activity relationship (SAR) analysis indicated that peptide-1 had more favourable ML-IAP recognition features. A molecular dynamics simulation, molec

Biological parameters of Triatoma longipennis recently introduced into Central Mexico.

There are many invasive alien insect species, several of these species can affect human health by causing disease. These include vectors of vector-borne diseases, such as the triatomines that transmit Chagas disease. Triatoma longipennis Usinger (Hemiptera: Reduviidae) is one of the most epidemiologically important vector species of Trypanosoma cruzi Chagas (Trypanosomatida: Trypanosomatidae) in Mexico. This study reports the third collection of T. longipennis from the state of Hidalgo, Mexico, where the prevalence of T. cruzi infection in humans has historically exceeded 3.38%. This study eva

Anticancer effect of piperine, a black pepper compound, regulating apoptosis mediated through extracellular vesicles and cathepsin D in acute leukemia.

Extracellular vesicles (EVs) contribute to tumorigenesis in acute leukemia, while natural compounds can counteract these EVs-mediated mechanisms. Piperine, a natural black pepper compound, exerts anticancer activity toward many cancer cells with minimal effect in normal PBMC cells via the apoptosis signaling pathway, a key programmed cell death pathway. The aim of this study was to investigate the involvement of EVs in piperine-induced apoptosis in acute leukemia. NB4 and MOLT-4 leukemic cell lines were treated with various concentrations of piperine. Cell viability, apoptosis, and cell cycle

Dual Hedgehog/GLI1 and PI3K/Akt/mTOR Targeting Possesses Higher Efficacy to Inhibit T-Cell Acute Lymphoblastic Leukemia Growth.

While the PI3K/Akt/mTOR pathway is a well-established drug target in T-cell acute lymphoblastic leukemia (T-ALL), the contribution of the Hedgehog (Hh) pathway in T-ALL malignancy remains poorly defined. We investigated the effects of pharmacological inhibition of key signaling nodes in these pathways using T-ALL cell lines (Jurkat, Molt-4, DND-41, and ALL-SIL). Cells were treated with the Gli1 inhibitor Gant-61, the Smoothened inhibitors GDC-0449 and Glasdegib, the Akt inhibitor MK-2206, and the mTOR inhibitor RAD001, both alone and in combination. Analyses of cell viability, cell cycle progr

Genome Expression Dynamics Reveal the Parasitism Regulatory Landscape of the Root-Knot Nematode Meloidogyne incognita and a Promoter Motif Associated with Effector Genes.

Root-knot nematodes (genus Meloidogyne) are the major contributor to crop losses caused by nematodes. These nematodes secrete effector proteins into the plant, derived from two sets of pharyngeal gland cells, to manipulate host physiology and immunity. Successful completion of the life cycle, involving successive molts from egg to adult, covers morphologically and functionally distinct stages and will require precise control of gene expression, including effector genes. The details of how root-knot nematodes regulate transcription remain sparse. Here, we report a life stage-specific transcript

Discovery of meta-sulfamoyl N-hydroxybenzamides as HDAC8 selective inhibitors.

In the past decade, although research and development of histone deacetylase (HDAC) inhibitors as therapeutic agents have achieved great accomplishments, especially in oncology field, there is still an urgent need for the discovery of isoform-selective HDAC inhibitors considering the side effects caused by nonselective HDAC inhibitors. HDAC8, a unique class I zinc-dependent HDAC, is becoming a potential target in cancer and other diseases. In the current study, a novel series of N-hydroxy-3-sulfamoylbenzamide-based HDAC8 selective inhibitors (12a-12p) were designed and synthesized, among which

T-type calcium channel antagonists, mibefradil and NNC-55-0396 inhibit cell proliferation and induce cell apoptosis in leukemia cell lines.

BACKGROUND: T-type Ca(2+) channels are often aberrantly expressed in different human cancers and participate in the regulation of cell cycle progression, proliferation and death. METHODS: RT-PCR, Q-PCR, western blotting and whole-cell patch-clamp recording were employed to assess the expression of T-type Ca(2+) channels in leukemia cell lines. The function of T-type Ca(2+) channels in leukemia cell growth and the possible mechanism of the effect of T-type Ca(2+) channel antagonists on cell proliferation and apoptosis were examined in T-lymphoma cell lines. RESULTS: We show that leukemia cell

Aberrant methylation and decreased expression of the RIZ1 gene are frequent in adult acute lymphoblastic leukemia of T-cell phenotype.

Retinoblastoma protein-interacting zinc finger, RIZ1, is a tumor suppressor gene that is inactivated in various solid tumors. However, the role of the RIZ1 gene has not been well examined in adult acute lymphoblastic leukemia (ALL). We analyzed the expression and promoter methylation status of the RIZ1 gene in patients with newly diagnosed ALL by quantitative real-time reverse transcription polymerase chain reaction (PCR) and methylation-specific PCR, respectively. RIZ1 expression in 67 cases of ALL (mean 1.043) was decreased compared with that in normal bone marrow (mean 1.471) (p = 0.030). M

A novel tick antigen shows high vaccine efficacy against the dog tick, Rhipicephalus sanguineus.

Ticks are acaridae ectoparasites that, while taking a blood meal, can transmit viruses, bacteria, protozoa and filarial nematodes, which cause a variety of human and animal illnesses. The use of chemical pesticides constitutes the primary measure for control of these ectoparasites. However, the intensive use of these chemicals has drawbacks such as the contamination of food, environmental pollution and development of resistance by ectoparasites. Vaccination is considered a promising alternative for controlling infestations by ectoparasites. Although emerging tick proteins have been identified

Gene expression profiling of the cephalothorax and eyestalk in Penaeus monodon during ovarian maturation.

In crustaceans, a range of physiological processes involved in ovarian maturation occurs in organs of the cephalothorax including the hepatopancrease, mandibular and Y-organ. Additionally, reproduction is regulated by neuropeptide hormones and other proteins released from secretory sites within the eyestalk. Reproductive dysfunction in captive-reared prawns, Penaeus monodon, is believed to be due to deficiencies in these factors. In this study, we investigated the expression of gene transcripts in the cephalothorax and eyestalk from wild-caught and captive-reared animals throughout ovarian mat

Domestic dogs (Canis familiaris) as reservoir hosts for Rickettsia conorii.

Rickettsia conorii is the causative agent of Mediterranean spotted fever (MSF) and Israeli spotted fever (ISF) transmitted by the brown dog tick Rhipicephalus sanguineus. In areas where MSF or ISF are prevalent, dogs have high prevalence of R. conorii -neutralizing antibodies. However, the true role of dogs in the persistence of the R. conorii transmission cycle is unknown, and their reservoir competence for this pathogen has remained untested. We assessed the ability of dogs infected with R. conorii to transmit the pathogen to previously uninfected Rh. sanguineus ticks. Dogs were infected eit

Mercury in the blood and eggs of American kestrels fed methylmercury chloride.

American kestrels (Falco sparverius) were fed diets containing methylmercury chloride (MeHg) at 0, 0.6, 1.7, 2.8, 3.9, or 5.0 µg/g (dry wt) starting approximately eight weeks before the onset of egg laying. Dietary treatment was terminated after 12 to 14 weeks, and unhatched eggs were collected for Hg analysis. Blood samples were collected after four weeks of treatment and the termination of the study (i.e., 12-14 weeks of treatment). Clutch size decreased at dietary concentrations above 2.8 µg/g. The average total mercury concentration in clutches of eggs and in the second egg laid (i.e., egg

West Nile virus detection in nonvascular feathers from avian carcasses.

West Nile virus (WNV) is a public health threat and has caused the death of thousands of North American birds. As such, surveillance for WNV has been ongoing, utilizing numerous biological specimens and testing methods. Nonvascular (i.e., fully grown) feathers would provide a simple method of collection from either dead or live birds of all ages and molt cycles, with presumably less biosafety risk compared with other specimen types, including feather pulp. The current study evaluates WNV detection in nonvascular feathers removed from naturally infected avian carcasses of several species groups

Ixodes scapularis ticks (Acari: Ixodidae) from Louisiana are competent to transmit Borrelia burgdorferi, the agent of Lyme borreliosis.

The principal vector of Borrelia burgdorferi, the Lyme borreliosis spirochete, in the Northeast and Midwestern regions of the United States is the blacklegged tick Ixodes scapularis. Because of a favorable environment, I. scapularis is also plentiful in the South; however, a correlation with Lyme borreliosis cases does not exist in this region of the United States. Concern existed that something intrinsic to ticks found in Louisiana could mitigate their ability to transmit B. burgdorferi. Therefore, we set out to assess the ability of I. scapularis ticks from Louisiana to become infected with
