Autophagy activation is required for myofibroblast differentiation during healing of oral mucosa.

AIM: It is known that periodontal tissues heal faster that skin, and gingiva in particular heal without scar formation. The mechanisms regulating this behaviour are still unclear. The aim of our work was to compare wound healing in oral mucosa and gingiva, investigating the role of α-smooth muscle actin (αSMA)-expressing myofibroblasts and autophagy. MATERIALS AND METHODS: Biopsies were obtained from seven patients immediately before and 24 hr after vertical releasing incision in oral mucosa and attached gingiva. Both whole biopsies and primary cultures of fibroblasts derived from the same ti…

Low-grade myofibroblastic sarcoma of the orbit: A case report and literature review.

RATIONALE: Low-grade myofibroblastic sarcoma (LGMS) is a malignant lesion composed of myofibroblasts. It is an uncommon tumor of unknown etiology that mainly develops in the bone or soft tissue and is most often reported in the head and neck, particularly in the tongue and oral cavity. PATIENT CONCERNS: A 2-year-old girl, previously well and with no significant medical history or family history of other diseases, presented with a 2-week painless swelling of the right orbit. DIAGNOSES: Preoperative computed tomography (CT) revealed a large homogeneous enhanced mass, 21 × 13 mm in size, locate…

Integrin alpha 11 in the regulation of the myofibroblast phenotype: implications for fibrotic diseases.

Tissue fibrosis, characterized by excessive accumulation of aberrant extracellular matrix (ECM) produced by myofibroblasts, is a growing cause of mortality worldwide. Understanding the factors that induce myofibroblastic differentiation is paramount to prevent or reverse the fibrogenic process. Integrin-mediated interaction between the ECM and cytoskeleton promotes myofibroblast differentiation. In the present study, we explored the significance of integrin alpha 11 (ITGA11), the integrin alpha subunit that selectively binds to type I collagen during tissue fibrosis in the liver, lungs and kid…

Pediatric Endobronchial Inflammatory Myofibroblastic Tumor: A Case Report and Review of the Literature.

Inflammatory myofibroblastic tumor (IMT) belongs to the group of soft tissue tumor and could occur at any anatomical site from the central nervous system to gastrointestinal tract. The lung and abdomen are commonly affected sites, however, pulmonary IMT is predominantly located within the parenchyma rather than presenting as endobronchial lesion. IMTs may occur in any age group, but they are observed most commonly in children and adolescents. Here, we present a case of IMT arising from the left main stem bronchus in a 10-year-old girl.

[Intestinal intussusception secondary to myofibroblastic tumor in an elderly patient. Case report].

BACKGROUND: Intussusception is defined as a segment of the gastrointestinal tract and mesentery within the lumen of an adjacent segment. It is a rare condition in adults that can occur anywhere in the gastrointestinal tract from the stomach to the rectum. Only 5% of all intussusceptions are presented in adults, and in 1-5% of all cases of intestinal obstruction. Inflammatory myofibroblastic tumour is rare, and is usually found in the lung, and rarely detected in some intestinal portions. It causes a variety of non-specific symptoms, with those that present as an intussusception being uncommon.

CT Manifestations of Inflammatory Myofibroblastic Tumors (Inflammatory Pseudotumors) of the Urinary System.

OBJECTIVE: The purpose of this study is to characterize the CT manifestations of inflammatory myofibroblastic tumors (IMTs) of the urinary system in eight patients. MATERIALS AND METHODS: The CT images of eight pathologically confirmed IMTs were retrospectively reviewed. Two of the eight IMTs occurred in the kidney, and six occurred in the bladder. Seven patients underwent both unenhanced CT and contrast-enhanced CT, and one of the patients who had a bladder tumor underwent unenhanced CT only. The site, shape, size, boundary, internal structure, and enhancement pattern of the lesions were ass

Periostin regulates fibrocyte function to promote myofibroblast differentiation and lung fibrosis.

Fibrocytes are circulating mesenchymal precursors (CD45+, col 1+) recruited to fibrotic areas. Fibrocytes secrete profibrotic mediators including periostin; a matricellular protein that regulates cellular interactions with extracellular matrix (ECM) components. In bleomycin-induced fibrosis, periostin deficiency in structural or hematopoietic cells limits development of pulmonary fibrosis. To determine if hematopoietic-derived fibrocytes might secrete soluble factors to activate structural myofibroblast differentiation, wild-type (WT) fibroblasts were treated with conditioned medium from fibro

Genetic lineage tracing defines myofibroblast origin and function in the injured heart.

Cardiac fibroblasts convert to myofibroblasts with injury to mediate healing after acute myocardial infarction (MI) and to mediate long-standing fibrosis with chronic disease. Myofibroblasts remain a poorly defined cell type in terms of their origins and functional effects in vivo. Here we generate Postn (periostin) gene-targeted mice containing a tamoxifen-inducible Cre for cellular lineage-tracing analysis. This Postn allele identifies essentially all myofibroblasts within the heart and multiple other tissues. Lineage tracing with four additional Cre-expressing mouse lines shows that periost…

A rare case of inflammatory myofibroblastic tumor of the diaphragmatic parietal pleura with dissemination.

Inflammatory myofibroblastic tumor (IMT) is a rare neoplasm that occurs at different sites in the body. Pleural IMT in particular is especially rare. IMTs infrequently tend to have malignancy. We report a rare case of advanced diaphragmatic parietal pleural IMT with dissemination. A 30-year-old woman complained of right upper abdominal pain. Computed tomography showed a large lobulated mass over the right diaphragm, but no disseminated nodules were noted. Intraoperatively, we found the primary tumor arising from the diaphragmatic parietal pleura and a dozen disseminated nodules, and we removed

miR-145 Contributes to Hypertrophic Scarring of the Skin by Inducing Myofibroblast Activity.

Hyperthrophic scarring of the skin is caused by excessive activity of skin myofibroblasts after wound healing and often leads to functional and/or aesthetic disturbance with significant impairment of patient quality of life. MicroRNA (miRNA) gene therapies have recently been proposed for complex processes such as fibrosis and scarring. In this study, we focused on the role of miR-145 in skin scarring and its influence in myofibroblast function. Our data showed not only a threefold increase of miR-145 levels in skin hypertrophic scar tissue but also in transforming growth factor β1 (TGF-β1)-ind…

Misdiagnosis of Behçet's disease presented with intracardiac mass as inflammatory myofibroblastic tumor.

Behçet's disease is a chronic multisystem inflammatory disorder. There are limited data about cardiac involvement, but it is seen rarely. Herein, we present a 33-year-old male patient with heart failure secondary to a right ventricular mass. It was first diagnosed as inflammatory myofibroblastic tumor (IMT) histopathologically. During the postoperative follow-up, a thrombus was detected at the interatrial septum, and the patient was reevaluated. The diagnosis was possible Behçet's disease, and the mass, previously reported as IMT, was determined to be an organizing thrombus with a mixture of g

Distribution of myofibroblast and tenascin-C in cystic adventitial disease: comparison with ganglion.

Cystic adventitial disease (CAD) is a rare peripheral artery disorder which shows the development of gelatinous cysts in the adventitia. Alt … ndle-shaped cells demonstrated to be positive for α-smooth muscle actin, indicating the presence of myofibroblasts in the cyst wall. A focal tenascin-C-positive area was observed in the cyst wall of our CAD cases. Presence of two different cell types, proliferation of myofibroblasts and expression of tenascin-C were consistent with those of cyst walls of 20 surg …

Carboplatin plus paclitaxel in the successful treatment of advanced inflammatory myofibroblastic tumor.

A 26-year-old man with unresectable inflammatory myofibroblastic tumor (IMT) presented with multiple metastases in the thoracic vertebra and lymph nodes as detected by positron emission tomography (PET) received chemotherapy with carboplatin plus paclitaxel. After three cycles of chemotherapy, fluorine-18-fluorodeoxyglucose (FDG)-PET/CT revealed tumor regression and significant reduction of FDG uptake in all lesions. The patient received six cycles of chemotherapy without any severe adverse event, and there was no sign of disease progression for seven months. This regimen is well tolerated and

Multiple inflammatory myofibroblastic tumor of the duodenum: case report and literature review.

INTRODUCTION: Inflammatory myofibroblastic tumor (IMT) is a rare low-grade malignant mesenchymal tumor, which can occur at any location, although the lung is the most commonly affected organ. It is extremely rare in the duodenum and only two cases have been reported previously. We report, to our knowledge, the first case of multiple neoplastic lesions. CASE REPORT: A 20-year-old male presented with the chief complaints of intermittent right epigastric pain, nausea and vomiting. Imaging examination, electronic gastroscopy and preoperative biopsy revealed undefined lesions in the duodenum. Panc
