In silico evaluation of corneal patch eluting anti-VEGF agents concept.

This study introduces a novel approach utilizing a temporary drug-eluting hydrogel corneal patch to prevent neovascularization, alongside a numerical predictive tool for assessing the release and transport kinetics of bevacizumab (BVZ) after the keratoplasty. A key focus was investigating the impact of tear film clearance on the release kinetics and drug transport from the designed corneal patch. The proposed tear drug clearance model incorporates the physiological mechanism of lacrimal flow (tear turnover), distinguishing itself from previous models. Validation against experimental data confi

[Expression and function of microRNAs in the cornea].

MicroRNAs (miRNAs) are a class of small (21-2i nucleotictes), single-stranded, noncoding RNA molecules that regulate gene expression at the post-transcriptional or translational level by binding to the 3'-untranslated region of the target mRNAs. miRNAs ubiquitously exist in the genome of an organism. More than two hundred miRNA species are expressed in the eye, of which 25% are found in the cornea. miRNAs play important roles in corneal development, differentiation, glycogen metabolism, post-injury regeneration, and maintenance of homeostasis. On the other hand, miRNAs are involved in the regu

Controlling the angiogenic switch: a balance between two distinct TGF-b receptor signaling pathways.

Biochemical studies in endothelial cells (ECs) and genetic studies in mice and humans have yielded major insights into the role of transforming growth factor beta (TGF-beta) and its downstream Smad effectors in embryonic vascular morphogenesis and in the establishment and maintenance of vessel wall integrity. These studies showed that TGF-beta signaling is of critical importance for normal vascular development and physiology. They also indicated the involvement of two distinct TGF-beta signaling cascades within ECs, namely the activin receptor-like kinase 5 (ALK5)-Smad2/3 pathway and the ALK1-

Jugular vein wall repair after intravenous injection in equids.

The evolution of the healing process of venous puncture wounds was studied macroscopically and light-microscopically in order to determine the time of venipuncture. Seven small equids of various ages, all in normal healthy condition, were experimentally injected in the external jugular vein with a physiological Hartmann solution at different times before euthanasia. Two types of needle diameters i.e. 16.5 G and 19 G, were used. A fairly good uniformity in the healing process was seen in the 7 animals. In most cases the lesions induced within 5 weeks before death were macroscopically visible, w

microRNAs Distinctively Regulate Vascular Smooth Muscle and Endothelial Cells: Functional Implications in Angiogenesis, Atherosclerosis, and In-Stent Restenosis.

Endothelial cells (EC) and vascular smooth muscle cells (VSMC) are the main cell types within the vasculature. We describe here how microRNAs (miRs)--noncoding RNAs that can regulate gene expression via translational repression and/or post-transcriptional degradation--distinctively modulate EC and VSMC function in physiology and disease. In particular, the specific roles of miR-126 and miR-143/145, master regulators of EC and VSMC function, respectively, are deeply explored. We also describe the mechanistic role of miRs in the regulation of the pathophysiology of key cardiovascular processes i

Epidermal growth factor-like domain 7 is a marker of the endothelial lineage and active angiogenesis.

Epidermal growth factor-like domain 7 (Egfl7) expression in the developing embryo is largely restricted to sites of mesodermal progenitors of angioblasts/hemangioblasts and the vascular endothelium. We hypothesize that Egfl7 marks the endothelial lineage during embryonic development, and can be used to define the emergence of endothelial progenitor cells, as well as to visualize newly-forming vasculature in the embryo and during the processes of physiologic and pathologic angiogenesis in the adult. We have generated a transgenic mouse strain that expresses enhanced green fluorescent protein (e

Endothelial cell FGF signaling is required for injury response but not for vascular homeostasis.

Endothelial cells (ECs) express fibroblast growth factor receptors (FGFRs) and are exquisitely sensitive to FGF signals. However, whether the EC or another vascular cell type requires FGF signaling during development, homeostasis, and response to injury is not known. Here, we show that Flk1-Cre or Tie2-Cre mediated deletion of FGFR1 and FGFR2 (Fgfr1/2(Flk1-Cre) or Fgfr1/2(Tie2-Cre) mice), which results in deletion in endothelial and hematopoietic cells, is compatible with normal embryonic development. As adults, Fgfr1/2(Flk1-Cre) mice maintain normal blood pressure and vascular reactivity and

Vascular environment-responsive DNA nanoswitch controls the positive feedback system for spatiotemporal coupling of angiogenesis and osteogenesis.

Vascularization-ossification coupling is pivotal for bone defect repair. Neovascularization guides the migration of osteogenic precursor cells, and osteoblasts in the perivascular area support vascular regeneration through the secretion of cytokines. Delivery systems are being engineered to orchestrate the coupling of vascularization and ossification precisely. However, most delivery systems lack sensitivity to the inherent repair environment, hampering timely bone formation. This study developed a vascular environment-responsive biomaterial: a true bone ceramic/small intestine submucosa (TBC/

Unveiling the Angiogenic Potential and Functional Decline of Valve Interstitial Cells During Calcific Aortic Valve Stenosis Progression.

Valve interstitial cells (VICs) play a critical role in aortic valve calcification and angiogenic processes associated with calcific aortic valve stenosis (CAVS). Within the same valve, VICs from differently calcified regions can exhibit diverse phenotypic and functional properties. We hypothesised that VICs isolated from noncalcified (NC-VICs) and calcified (C-VICs) areas of human aortic valves possess distinct angiogenic characteristics. In this study, we isolated C-VICs and NC-VICs from 23 valves obtained after aortic valve replacement due to CAVS. Both VIC types exhibited similar phenotype

Sulfide oxidation promotes hypoxic angiogenesis and neovascularization.

Angiogenic programming in the vascular endothelium is a tightly regulated process for maintaining tissue homeostasis and is activated in tissue injury and the tumor microenvironment. The metabolic basis of how gas signaling molecules regulate angiogenesis is elusive. Here, we report that hypoxic upregulation of ·NO in endothelial cells reprograms the transsulfuration pathway to increase biogenesis of hydrogen sulfide (H2S), a proangiogenic metabolite. However, decreased H2S oxidation due to sulfide quinone oxidoreductase (SQOR) deficiency synergizes with hypoxia, inducing a reductive shift and

Microscopic technique for the detection of nitric oxide-dependent angiogenesis in an animal model.

Nitric oxide (NO) plays an important role in maintaining vascular homeostasis. The importance of NO in the vasculature is demonstrated by several experimental conditions, such as vascular endothelial growth factor (VEGF)-induced angiogenesis. Thus, the NO metabolic pathway in endothelial cells could be one of the main contributing factors for angiogenesis. Although several methods have been used for measuring in vitro angiogenesis, a proper technique has not been developed for identifying in vivo NO-dependent angiogenesis. This chapter provides a new intravital microscopic method for detecting

Vascular remodeling is airway generation-specific in a primate model of chronic asthma.

RATIONALE: Changes in the density of bronchial vessels have been proposed as a part of airway remodeling that occurs in chronic asthma. OBJECTIVES: Using an established nonhuman primate model of chronic allergic asthma, we evaluated changes in vascular density as well as the contribution of bronchial epithelium to produce vascular endothelial growth factor (VEGF). METHODS: Eight juvenile rhesus macaques were divided into two groups of four. One group was exposed to 11 cycles of aerosolized house dust mite allergen (HDMA), whereas the other was exposed to filtered air. Bronchial wall vasculat

Electrical-Stimulation Hydrogel Electronic Skin for Sustainable Hybrid Biomechanical-Electromagnetic Energy Harvesting and Accelerating Wound Healing.

External electrical stimulation (ES) plays a crucial role in promoting wound healing. However, conventional strategies depend on external power sources and exhibit insufficient responsiveness to the physiological microenvironment, which impedes precise and efficient healing. Herein, a hybrid self-powered ES electronic skin (HSESES) is designed by integrating a gelatin-based outer matrix with a zwitterionic conductive inner layer. This electronic skin effectively harvests both biomechanical energy (from body motion) and ambient electromagnetic energy (from surrounding appliances) to provide rea

Shhedding New Light on the Role of Hedgehog Signaling in Corneal Wound Healing.

The cornea, an anterior ocular tissue that notably serves to protect the eye from external insults and refract light, requires constant epithelium renewal and efficient healing following injury to maintain ocular homeostasis. Although several key cell populations and molecular pathways implicated in corneal wound healing have already been thoroughly investigated, insufficient/impaired or excessive corneal wound healing remains a major clinical issue in ophthalmology, and new avenues of research are still needed to further improve corneal wound healing. Because of its implication in numerous ce

Increased angiogenic response but deficient arteriolization and abnormal microvessel ultrastructure in critical leg ischaemia.

BACKGROUND: Ischaemia is known to induce angiogenesis, but the effects of critical leg ischaemia (CLI) on angiogenesis remain unclear. The aim of this study was to examine the physiological angiogenic response in CLI by investigating the extent of neovascularization, characterizing microvessel subtypes and determining the microvessel ultrastructure. METHODS: Gastrocnemius muscles were biopsied from 12 patients with CLI and 12 without leg ischaemia. Microvessels were evaluated immunohistochemically using three endothelial markers (anti-CD31, anti-CD34 and PAL-E) and anti-alpha smooth muscle ac

Enhanced Early Vascularization and Tissue Formation in a Biphasic Collagen Hydrogel Dermal Regeneration Template.

Reconstruction of full-thickness wounds remains a major clinical challenge. Full-thickness skin grafts (FTSG) are considered an ideal reconstructive option as they reconstitute both epidermal and dermal layers of skin; however, their use is limited by donor site availability and potential donor site morbidity. Dermal regeneration templates (DRTs) were introduced decades ago to reduce dermal harvesting needs and promote formation of a 'neodermal' layer, improving both functional and cosmetic outcomes. However, current DRTs are limited by slow vascularization, typically requiring 2-4 weeks for i

Proangiogenic Mechanisms and Modifications of Mesenchymal Stem Cells with a Focus on Neurological Disorders.

Mesenchymal stem cells (MSCs) are adult stem cells with extensive differentiation potential, sourced from bone marrow, adipose tissue, umbilical cord blood, and other tissues. MSCs from different origins exhibit distinct functional characteristics. These cells have demonstrated therapeutic efficacy in various neurological disorders, primarily by modulating immune responses, promoting neovascularization, and aiding neural circuit reconstruction. Notably, the strong proangiogenic properties of MSCs play a crucial role in disease treatment and regression. This review focuses on the application of

Microglia-vascular interactions after spinal cord injury: regulatory mechanisms and therapeutic advances.

The repair process following spinal cord injury (SCI) involves intricate crosstalk between neuroimmune and vascular systems, with microglia-vascular interactions increasingly recognized as an important regulatory interface that may shape both secondary injury progression and neural regeneration. This review delineates the dual role of angiogenesis in SCI: functionally mature neovessels can facilitate restoration of blood supply, provide neurotrophic support, and offer guidance cues for axonal regrowth; in contrast, structurally abnormal and hyperpermeable vessels can exacerbate blood-spinal co
