Effect of vitamin C upon gastric mucosal O6-alkyltransferase activity and on gastric vitamin C levels.

The repair enzyme O6-alkyltransferase will repair O6-methylguanine adducts in human DNA. In gastric mucosal DNA these adducts may be formed … ose displaying evidence of chronic atrophic gastritis. We speculate that inhibition of intragastric nitrosation by vitamin C results in decreased formation of O6-methylguanine-DNA. In consequence, less O6-alkyltransferase is consumed in repairing these adducts resulting in higher tissue levels of this enzyme.

Tumor induction in a rat model for ureterosigmoidostomy without evidence of nitrosamine formation.

Twenty rats were randomized into a vesicosigmoidostomy and an unoperated control group. In both groups the 24 hour excretion of secondary am … ine formation. This and the missing effect of nitrosamine antidotes suggest that other factors than nitrosation must be responsible for colon carcinogenesis following urinary diversion via intestine.

Diet and chronic atrophic gastritis: a case-control study.

A hospital-based case-control study of gastric cancer precursor lesions was conducted in a high-risk black population in southern Louisiana. … C is consistent with findings for gastric cancer and with the etiologic hypothesis of intragastric nitrosation. A twofold increased risk was associated with cigarette smoking. Gastric juice pH, NO3-, and NO2- were determined for subjects undergoing gastroscopy, and comparisons were made between this high-risk U.S. group and a Colombian population with a much …

Garcinia cowa bark extract induces oxidative stress mediated cellular apoptosis in Leishmania donovani parasite modulated by its active phytosterol constituent.

Visceral leishmaniasis still remains a leading cause of parasitic deaths, with modern pentavalent antimonials showing limited efficacy and health risks. The methanolic bark extract of the Northeastern Indian plant, Garcinia cowa, demonstrated potent leishmanicidal effects against the parasite Leishmania donovani, demonstrating IC50 values of 20-36 µg/ml, with selective toxicity for parasites over healthy cells. It induced parasite death through elevated oxidative and nitrosative stress elements, reduced arginase activity, nuclear fragmentation, cell cycle arrest, and apoptosis. A GC-MS study a

Increased serum nitric oxide and malondialdehyde levels in patients with acute intestinal amebiasis.

OBJECTIVE: To determine the level of oxygen-nitrogen stress parameters in the pathogenesis of amebiasis. METHODS: Twenty-four acute intestinal amebiasis patients and 20 healthy controls were enrolled in the present study. Serum malondialdehyde and nitric oxide levels were determined spectrophotometrically. RESULTS: Serum malondialdehyde and nitric oxide levels were significantly higher in acute intestinal amebiasis patients than healthy controls (P<0.001). CONCLUSIONS: These results suggest that oxidative and nitrosative stress may play a major role in tissue damage in acute intestinal ameb

Essential roles of S-nitrosothiols in vascular homeostasis and endotoxic shock.

The current perspective of NO biology is formulated predominantly from studies of NO synthesis. The role of S-nitrosothiol (SNO) formation and turnover in governing NO-related bioactivity remains uncertain. We generated mice with a targeted gene deletion of S-nitrosoglutathione reductase (GSNOR), and show that they exhibit substantial increases in whole-cell S-nitrosylation, tissue damage, and mortality following endotoxic or bacterial challenge. Further, GSNOR(-/-) mice have increased basal levels of SNOs in red blood cells and are hypotensive under anesthesia. Thus, SNOs regulate innate immu

Hepatic encephalopathy.

Hepatic encephalopathy (HE) is a prognostically relevant neuropsychiatric syndrome that occurs in the course of acute or chronic liver disease. Besides ascites and variceal bleeding, it is the most serious complication of decompensated liver cirrhosis. Ammonia and inflammation are major triggers for the appearance of HE, which in patients with liver cirrhosis involves pathophysiologically low-grade cerebral oedema with oxidative/nitrosative stress, inflammation and disturbances of oscillatory networks in the brain. Severity classification and diagnostic approaches regarding mild forms of HE ar

Photodissociation dynamics of dinitrite at 355 nm: initiation of a reactive pathway.

Dinitrites are effective nitrosating reagents in preparative chemistry and their decomposition products are involved in the atmospheric reaction of volatile organic compounds with nitrogen oxides (NOx). In this work, photodissociation dynamics of five alkyl dinitrites were investigated by detecting the LIF spectra of the dissociation products and theoretical calculations. The results showed that the C-C bond connecting the two nitrosooxy (-ONO) functional groups was the weakest bond in vicinal dinitrites and aldehydes were the dissociation products. For dinitrites with two ONO groups separated

In vivo damage of CNS myelin and axons induced by peroxynitrite.

In multiple sclerosis (MS) the mechanisms of injury caused by peroxynitrite remain uncertain. To study histological, ultra structural and molecular alterations caused by peroxynitrite in brain, the peroxynitrite donor 3-morpholinosydnonimine was injected in rat corpus callosum. Peroxynitrite induces strong primary axonal damage with characteristics of primary acute axonopathy, together with severe myelin alteration, myelin vacuolation and demyelination, and nitrotyrosine formation as confirmed by detection of nitrosated target proteins. Administration of the peroxynitrite scavenger uric acid i

Microplastics in motion: Genotoxic and redox imbalance impacts of systemic exposure in a murine model.

The increasing dissemination of microplastics (MPs) in the environment and their potential adverse effects on human and animal health have raised significant concerns in the scientific community. In this context, we aimed to investigate the toxicity induced by polyethylene (PE) MPs in the blood of Swiss mice (Mus musculus), focusing on biomarkers of oxidative and nitrosative stress and genotoxicity. Thirty mice were distributed across three experimental groups: two groups intravenously inoculated with polyethylene MPs at target systemic blood concentrations of 7.1 µg/mL and 355 µg/mL, respecti

De novo nitric oxide synthesis drives tactile hypersensitivity induced by ATP-sensitive potassium channel opening in mice: relevance to migraine and other headache disorders.

Adenosine triphosphate-sensitive potassium (K ATP ) channel opener levcromakalim is a potent inducer of vasodilation, headache, and migraine attacks in humans and tactile hypersensitivity in mice. Other migraine-inducing agents, such as nitric oxide (NO) donors, calcitonin gene-related peptide, and pituitary adenylate cyclase-activating polypeptide, are thought to activate second messengers leading to K ATP opening. However, the mechanism by which K ATP channel opening leads to migraine remains unclear. In this study, we investigated the contribution of nitric oxide synthase (NOS) isoforms and

Neuroprotective potential of eugenol against acrylamide-induced brain toxicity by regulating Nrf2/NQO1/HO-1 and NLRP3/NF-κB/IL-1β signaling cascades.

Acrylamide (ACR), a common environmental and dietary neurotoxicant, exerts profound deleterious effects on the central nervous system by triggering oxidative stress, neuroinflammation, apoptosis, and motor impairments. Eugenol (EU), a natural phenolic compound known for its antioxidant and anti-inflammatory properties, was evaluated for its neuroprotective efficacy in ACR-induced brain toxicity in rats. Male Wistar rats were orally administered ACR (20 mg/kg/day) for four weeks to induce neurotoxicity, with concurrent administration of EU at two doses (50 and 100 mg/kg/day). Behavioral assessm

Lipoprotein(a) and Vascular Redox State in Patients With Advanced Coronary Atherosclerosis.

BACKGROUND: Lp(a) (lipoprotein[a]) is associated with cardiovascular disease, but neither the causal nature nor the underlying mechanisms are fully documented. This study investigated whether Lp(a) triggers atherogenesis by dysregulating vascular redox-sensitive inflammatory state. METHODS: Plasma Lp(a) was measured in 1027 patients with advanced coronary artery disease undergoing cardiac surgery. These patients were genotyped, and a modified LPA genetic risk score (LPA GRS) determining Lp(a) levels was generated. RNA sequencing and vascular superoxide measurements were performed in internal

β-blockade prevents coronary macro- and microvascular dysfunction induced by a high salt diet and insulin resistance in the Goto-Kakizaki rat.

A high salt intake exacerbates insulin resistance, evoking hypertension due to systemic perivascular inflammation, oxidative-nitrosative stress and endothelial dysfunction. Angiotensin-converting enzyme inhibitor (ACEi) and angiotensin receptor blockers (ARBs) have been shown to abolish inflammation and redox stress but only partially restore endothelial function in mesenteric vessels. We investigated whether sympatho-adrenal overactivation evokes coronary vascular dysfunction when a high salt intake is combined with insulin resistance in male Goto-Kakizaki (GK) and Wistar rats treated with tw

MiADMSA ameliorate arsenic induced urinary bladder carcinogenesis in vivo and in vitro.

BACKGROUND AND PURPOSE: Arsenicosis is a major threat to public health and is a major cause of the development of urinary bladder cancer. Oxidative/ nitrosative stress is one of the key factors for these effects but the involvement of other associated factors is less known. There is a lack of data for the efficacy of chelator against urinary bladder carcinogenesis. The present study demonstrates the early signs of arsenic exposed urinary bladder carcinogenesis and its attenuation by Monoisoamyl dimercaptosuccinic acid (MiADMSA). METHODS: Male rats were exposed to 50 ppm of sodium arsenite and

Thymus algeriensis and Thymus fontanesii exert neuroprotective effect against chronic constriction injury-induced neuropathic pain in rats.

We have previously demonstrated that the Thymus algeriensis and Thymus fontanesii extracts have powerful anti-inflammatory, antipyretic, and analgesic effects against acute pain models. We profiled their chemical composition and found many phenolic acids, flavonoids, and phenolic diterpenes. In this work, we investigated their antioxidant properties on HaCaT cells exposed to UVA-induced oxidative stress and examined their effects against chronic neuropathic pain and the underlying mechanisms. Through a rat chronic constriction injury (CCI) model, we induced chronic neuropathic pain by placing

Attenuation of autoimmune responses to oxidative specific epitopes, but not nitroso-adducts, is associated with a better clinical outcome in Myalgic Encephalomyelitis/chronic fatigue syndrome.

OBJECTIVES: There is evidence that inflammatory, oxidative and nitrosative stress (IO&NS) pathways participate in the pathophysiology of a subgroup of patients with Myalgic Encephalomyelitis/chronic fatigue syndrome (ME/CFS). Increased IgM-related autoimmune responses to oxidative specific epitopes (OSEs), including malondialdehyde (MDA), oleic acid and phosphatidyl inositol (Pi), and nitroso-(NO)-adducts, including NO-tryptophan (NOW), NO-arginine and NO-cysteinyl, are frequently observed in ME/CFS. Autoimmune responses in ME/CFS may be driven by increased bacterial translocation as measured

UV Light-Driven Nitric Oxide Release from Porous Nitrogen Heterocyclic Polymers.

In this study, porous polymers with nitrogen heterocyclic core structures are synthesized through the condensation of enaminonitrile and terephthalaldehyde monomers. These polymers are used as a platform to store bioactive nitric oxide (NO) and control its release. NO loading is achieved by nitrosating the polymers with acidified nitrite, a process that also imparts photoresponsivity to the polymers. Polymer composition and porosity affect NO storage and release. It is observed that under UV light at 365 nm in a PBS solution, the polymers (NO@DHP-POP) can release NO in a manner fully controlle

Moderate exercise-induced dynamics on key sepsis-associated signaling pathways in the liver.

BACKGROUND: There is a clear relationship between quantitative measures of fitness (e.g., VO2 max) and outcomes after surgical procedures. Whether or not fitness is a modifiable risk factor and what underlying biological processes drive these changes are not known. The purpose of this study was to evaluate the moderate exercise training effect on sepsis outcomes (survival) as well as the hepatic biological response. We chose to study the liver because it plays a central role in the regulation of immune defense during systemic infection and receives blood flow directly from the origin of infect
