Polysialylation of Glioblastoma Cells Is Regulated by Autophagy Under Nutrient Deprivation.

Glioblastoma (GBM) is a highly aggressive brain tumor marked by invasive growth and therapy resistance. Tumor cells adapt to hostile conditions, such as hypoxia and nutrient deprivation, by activating survival mechanisms including autophagy and metabolic reprogramming. Among GBM-associated changes, hypersialylation, particularly, the aberrant expression of polysialic acid (PSA), has been linked to increased plasticity, motility, and immune evasion. PSA, a long α2,8-linked sialic acid polymer typically attached to the NCAM, is abundant in the embryonic brain and re-expressed in cancers, correla

Exploratory analysis of nutrient composition of adult and senior dog diets.

INTRODUCTION: Senior dog foods are often marketed as distinct from adult formulations, yet no specific nutrient profiles exist for this life … two pet food stores in Fort Collins, Colorado (one national retailer and one locally owned store). Nutrients were expressed per 1,000 kcal metabolizable energy, and descriptive statistics were generated. Adult vs. senior, dry vs. canned, and combined categories were compared using parametric or non-parametric tests depending on distribution. Within each co …

Predators can facilitate herbivory in nutrient-limited marine ecosystems.

Predators influence ecosystem functioning through consumptive and non-consumptive effects. Recent studies suggest that predators can also be an essential source of limiting nutrients in ecosystems such as coral reefs, potentially influencing prey ecology through nutrient input via their excreta. With rising commercial fishery, mesopredatory fishes are being selectively harvested from reefs. Yet, there is incomplete knowledge of the consequences of this extraction on essential ecosystem processes. Using field experiments and observations, we examined how mesopredatory fishes influence herbivory…

γ-Oryzanol: A nutrient-rich ingredient for promoting wound healing.

γ-Oryzanol (Orz) is a powerful antioxidant found in rice bran and oil, known for its numerous health benefits. Nevertheless, its precise impact on skin wound healing remains largely unexplored. This research focuses on comprehensively examining the ability of Orz to promote tissue repair through laboratory-based and animal model experiments. The research further examines its antimicrobial activity and its role in promoting wound healing through anti-inflammatory effects. To evaluate the effect of Orz on tissue repair, scratch assays performed in vitro were used to analyze its impact on cellula

Nutrient-derived signals regulate eosinophil adaptation to the small intestine.

Eosinophils are well recognized as effector cells of type 2 immunity, yet they also accumulate in many tissues under homeostatic conditions. However, the processes that govern homeostatic eosinophil accumulation and tissue-specific adaptation, and their functional significance, remain poorly defined. Here, we investigated how eosinophils adapt to the small intestine (SI) microenvironment and the local signals that regulate this process. We observed that eosinophils gradually migrate along the crypt-villus axis, giving rise to a villus-resident subpopulation with a distinct transcriptional sign

Microplastic pollution and nutrient enrichment shift the diet of freshwater macroinvertebrates.

Microplastic pollution poses a global threat to freshwater ecosystems, with laboratory experiments indicating potential toxic impacts through chemical toxicity, physical abrasion, and false satiation. Bioplastics have emerged as a potential greener alternative to traditional oil-based plastics. Yet, their environmental effects remain unclear, particularly at scales relevant to the natural environment. Additionally, the interactive impacts of microplastics with other environmental stressors, such as nutrient enrichment, are poorly understood and rarely studied. Under natural conditions organism

Dynamic ammonium retention for nutrient separation from manure digestate.

Extensive nitrogen emissions with negative impact on nature and the environment urge effective valorization of manure and fractionation of nutrients to enable precision fertilization. Typically, manure is fed to a digester to produce biogas. The remaining digestate is then mechanically separated into a solid phosphorous-rich fraction and a liquid fraction containing both NH4+ and K. These ions are difficult to separate due to their very identical size and charge. We show that with smart tuning of the pH to control the NH3/NH4+ equilibrium, membranes can produce dedicated N and K-rich streams. …

Effect of pregnancy on the expression of nutrient-sensors and satiety hormones in mice.

Small intestinal satiation pathways involve nutrient-induced stimulation of chemoreceptors leading to release of satiety hormones from intestinal enteroendocrine cells (ECCs). Whether adaptations in these pathways contribute to increased maternal food intake during pregnancy is unknown. To determine the expression of intestinal nutrient-sensors and satiety hormone transcripts and proteins across pregnancy in mice. Female C57BL/6J mice (10-12 weeks old) were randomized to mating and then tissue collection at early- (6.5 d), mid- (12.5 d) or late-pregnancy (17.5 d), or to an unmated age matched

Bacterial spore germination receptors are nutrient-gated ion channels.

Bacterial spores resist antibiotics and sterilization and can remain metabolically inactive for decades, but they can rapidly germinate and resume growth in response to nutrients. Broadly conserved receptors embedded in the spore membrane detect nutrients, but how spores transduce these signals remains unclear. Here, we found that these receptors form oligomeric membrane channels. Mutations predicted to widen the channel initiated germination in the absence of nutrients, whereas those that narrow it prevented ion release and germination in response to nutrients. Expressing receptors with widen…

Control of nutrient uptake by IRF4 orchestrates innate immune memory.

Natural killer (NK) cells are innate cytotoxic lymphocytes with adaptive immune features, including antigen specificity, clonal expansion an … K cells. This defect was partially attributed to the inability of IRF4-deficient NK cells to uptake nutrients required for survival and memory generation. Altogether, these data suggest that IRF4 is a signal integrator that acts as a secondary metabolic checkpoint to orchestrate the adaptive response of NK cells during viral infection.

Diet-Induced Metabolic Dysfunction of Hypothalamic Nutrient Sensing in Rodents.

A sedentary lifestyle and excessive nutrient intake resulting from the consumption of high-fat and calorie-rich diets are environmental factors contributing to the rapid growth of the current pandemic of type 2 diabetes mellitus (DM2). Fasting hyperglycemia, an established hallmark of DM2, is caused by excessive production of glucose by the liver, resulting in the inability of insulin to suppress endogenous glucose production. To prevent inappropriate elevations of circulating glucose resulting from changes in nutrient availability, mammals rely on complex mechanisms for continuously detecting

Cropland degradation and nutrient overload on Hainan Island: A review and synthesis.

As the only "tropical base of agricultural production" in China, Hainan lsland is vigorously developing high-value agriculture and is becoming the province with the highest proportion of cash crops. However, this intensive farming with large nutrient inputs has caused cropland degradation, nitrogen (N) and phosphorus (P) overloads and water pollution, which have been reversed to initiate the construction of free trade ports. Here, we systematically review the status, driving factors, and environmental impacts of cropland degradation and nutrient overload with quantified evaluations and compare

Gene Expression Responses to Sequential Nutrient Deficiency Stresses in Soybean.

Throughout the growing season, crops experience a multitude of short periods of various abiotic stresses. These stress events have long-term impacts on plant performance and yield. It is imperative to improve our understanding of the genes and biological processes underlying plant stress tolerance to mitigate end of season yield loss. The majority of studies examining transcriptional changes induced by stress focus on single stress events. Few studies have been performed in model or crop species to examine transcriptional responses of plants exposed to repeated or sequential stress exposure, w

Nutrient availability regulates proline/alanine transporters in Trypanosoma brucei.

Trypanosoma brucei is a species of unicellular parasite that can cause severe diseases in livestock and humans, including African trypanosomiasis and Chagas disease. Adaptation to diverse environments and changes in nutritional conditions is essential for T. brucei to establish an infection when changing hosts or during invasion of different host tissues. One such adaptation is the ability of T. brucei to rapidly switch its energy metabolism from glucose metabolism in the mammalian blood to proline catabolism in the insect stages and vice versa. However, the mechanisms that support the parasit

Association between polygenic propensity for psychiatric disorders and nutrient intake.

Despite the observed associations between psychiatric disorders and nutrient intake, genetic studies are limited. We examined whether polygenic scores for psychiatric disorders are associated with nutrient intake in UK Biobank (N = 163,619) using linear mixed models. We found polygenic scores for attention-deficit/hyperactivity disorder, bipolar disorder, and schizophrenia showed the highest number of associations, while a polygenic score for autism spectrum disorder showed no association. The relatively weaker obsessive-compulsive disorder polygenic score showed the greatest effect sizes sugg
