During development and evolution, the morphology of ectodermal organs can be modulated so that an organism can adapt to different environments. We have proposed that morphoregulation can be achieved by simply tilting the balance of molecular activity. We test the principles by analyzing the effects of partial downregulation of Bmp signaling in oral and dental epithelia of the keratin 14-Noggin transgenic mouse. We observed a wide spectrum of tooth phenotypes. The dental formula changed from 1.0.0.3/1.0.0.3 to 1.0.0.2(1)/1.0.0.0. All mandibular and M3 maxillary molars were selectively lost beca

The frequency, gender, and maxillary/mandibular occurrences of third molar agenesis have been studied in a Danish population of Down syndrome individuals (39 F and 31 M). The study was based on radiological examination and compared with 2 control groups of normal Caucasian populations. We found that the occurrence of third molar agenesis in Down syndrome individuals was around 4 times greater than in a normal population and higher than in corresponding previous studies. The agenesis occurred more often in the maxilla than in the mandible, and the frequency was not significantly higher in femal

The zebrafish, Danio rerio, is a small, freshwater teleost that only began to be used as a vertebrate genetic model by the late George Streisinger in the early 1980s. The strengths of the zebrafish complement genetic studies in mice and embryological studies in avians. Its advantages include high fecundity, externally fertilized eggs and transparent embryos that can be easily manipulated, inexpensive maintenance, and the fact that large-scale mutagenesis screens can be performed. Here we review studies that have used the zebrafish as a model for craniofacial development. Lineage studies in zeb

Cleidocranial dysplasia (CCD), a rare, inherited, generalized, skeletal and dental dysplasia, exhibiting an autosomal dominant mode of transmission, may be associated with delays during tooth maturation. To test whether permanent tooth formation is delayed in patients with CCD and if the presence of supernumerary teeth adversely influences maturation of the dentition, a group of CCD patients (eight females, three males) was compared to an equal number of control subjects matched for age and gender. Dental maturity was assessed using panoramic radiographs and the Dental Maturity Ratio, (DMR = m

Vitamin D is required for the normal development of teeth and bones. When there is excess vitamin D, systemic and dental changes may occur. This is a case report of a girl who experienced hypercalcemia secondary to excess vitamin D derived from the consumption of milk that was incorrectly fortified. The changes in the permanent dentition to date are enamel hypoplasia and focal pulp calcification. These changes correspond to the timing of the toxemia caused by hypervitaminosis D.

Dental casts from 2159 black and white Americans with detailed neurological data available from the Collaborative Perinatal Study were examined to investigate the relationship of maternal smoking during pregnancy and delayed motor development at 1 year of age to morphological traits in the dentition. Earlier results have indicated that maternal smoking during pregnancy may cause selected tooth size metric reductions in the deciduous dentition and at least in some of the permanent teeth with prenatal crown formation, these features being influenced by sex and race differences. The present resul

A prospective study about the development of the mandibular second right molar, has been performed with 17 chemotherapy treated children in the Paediatric Oncology department of the Centre Oscar-Lambret (Lille). Our method is based upon the measure of the crown or root length for different periods: periods with or without chemotherapy. These measurements have been performed every 6 months for 18 months with orthopantomograms. We notice for both the crown and the root: a significant difference in growing speed between periods with an without chemotherapy; a speeding up of the mineralization at

Osteopetrosis, a general increase in bone density, is well-known as an autosomal recessive disease in several mammalian species. Compared with normal littermates, the osteopetrotic rats suffered from a complete failure of tooth eruption. Histologic and microradiographic studies show that the molars and the incisors developed a progressive ankylosis, the bone invades the germs and finally dissociates them. As early as the first days after birth, the incisors and the first molars are affected by the osteopetrotic process while the second and the third molars remain untouched. The periodontal mem

Roentgenographic studies of the jaws and teeth in a group of forty-eight pituitary dwarfs showed the following abnormalities in the development of the teeth: 1. Delayed shedding of the deciduous teeth. 2. Absence of resorption of the roots of the deciduous teeth at the usual time. 3. Marked delay in eruption of the permanent teeth. 4. Retention of permanent teeth in the maxillary and mandibular shafts. 5. Development of the apical parts of roots of the retained permanent teeth and their growth toward the lower mandibular edge. 6. Displacement of the first molars from the mandibular shaft to ra

Tooth formation has been studied in the fry and adult trout (Salmo fario L.) in order to describe their structure and investigate the links between histogenetic and architectural features. Odontoblasts show typical polarized kinetics during their activity; they shrink back from the tooth wall they have elaborated, without leaving any cytoplasmic process (Tomes' fibres); so the dentine in a characteristic way lacks any cellular or canalicular inclusion, at any stage of its development. The tooth as a whole grows from an apical to basal direction; its organic matrix is formed of coarse fibres fr

Dental-derived mesenchymal stem cells (DMSC), owing to their self-renewal capacity and multi-lineage differentiation potential, have demonstrated significant promise in the field of oral tissue regeneration. Mitochondria, as essential organelles within cells, can not only be responsible for energy metabolism but also influence the maintenance of stem cell stemness and the direction of differentiation by regulating cellular metabolic processes, apoptosis, and signal transduction pathways. In recent years, the mechanisms by which mitochondria participating in the osteogenic and odontogenic diffe

OBJECTIVE: To investigate the effects of quercetin (QU), hesperetin (HT), and taxifolin (TX) on human dental pulp cells (hDPCs) chronically exposed to lipopolysaccharide (LPS).
METHODS: First, the cytotoxicity (alamarBlue) and bioactivity (biomineralization, Alizarin Red) of QU, HT, and TX concentrations were evaluated on healthy hDPCs. Then, the effects of non-cytotoxic and bioactive concentrations were investigated on hDPCs after previous stimulation with E. coli LPS (10 µg/mL) for 7 days. Cell culture media with and without LPS were used as positive and negative controls, respectively. Cel

Pulp exposure often leads to pulp necrosis, root fractures, and ultimate tooth loss. The repair of the exposure site with pulp capping treatment is of great significance to preserving pulp vitality, but its efficacy is impaired by the low bioactivity of capping materials and cell injuries from the local accumulation of oxidative stress. This study develops a Wnt3a-loaded hydroxyapatite nanowire@mesoporous silica (Wnt3a-HANW@MpSi) core-shell nanocomposite for pulp capping treatments. The ultralong and highly flexible hydroxyapatite nanowires provide the framework for the composites, and the mes

The tooth root transmits and balances occlusal forces through the periodontium to the alveolar bone. The periodontium, including the gingiva, the periodontal ligament, the cementum and the partial alveolar bone, derives from the dental follicle (DF), except for the gingiva. In the early developmental stages, the DF surrounds the tooth germ as a sphere and functions to promote tooth eruption. However, the morphological dynamics and factors regulating the differentiation of the DF during root elongation remain largely unknown. Miniature pigs are regarded as a useful experimental animal for model

Recently, the involvement of PIN1, a peptidyl-prolyl cis/trans isomerase, has been reported in age-related bone homeostasis and adipogenesis. However, the role of PIN1 during odontogenic and adipogenic differentiation remains to be fully understood, particularly regarding human dental pulp stem cells (HDPSCs). Thus, in the present study, we have investigated the role of PIN1 in odontogenic and adipogenic differentiation of HDPSCs and signaling pathways possibly involved. PIN1 mRNA and protein level were upregulated in a time-dependent manner during adipogenic differentiation, increasing until

OBJECTIVE: To test the null hypotheses that children with unilateral agenesis of the mandibular second premolar (MnP2) do not show (1) greater distal angulation of the unerupted antimere and (2) delayed tooth formation compared with children without agenesis.
MATERIALS AND METHODS: Panoramic radiographs of 38 patients with unilateral aplasia of MnP2 were retrospectively examined and compared to a non-agenesic control group of 82 patients. Ages ranged from 8 to 15 years. Contralateral mandibular deciduous second molars were present for all participants. Each unerupted MnP2 was traced, and its

Tooth loss adversely affects not only mouth functions but also the esthetics of one's face. To repair these defects, current treatment methods mainly depend on nonbiological materials or artificial implants that also can, sometimes, reduce the quality of life because of their limited physiological function, or elicit an immunological rejection. Theoretically, a biological tooth (bio-tooth) that is made from the patient's own cells and grows in its intended location should be the best choice for treating tooth loss, although such bioengineered teeth have been nothing more than a dream for many
