High concentrations of ascorbic acid occur in scurvy-prone, teleost fish plasma. We quantified seasonal relationships between 1) dietary level of vitamin C and level of seminal plasma ascorbic acid, and 2) seminal plasma ascorbic acid concentration and sperm quality, in rainbow trout (Oncorhynchus mykiss). We maintained six groups of 2-yr-old rainbow trout on diets supplemented with 0, 30, 110, 220, 440, and 870 ppm ascorbyl monophosphate beginning in May 1992. Sperm were produced during the end of October 1992 through April 1993; we collected milt 13 times from 3-8 fish per treatment. We quan

While studying the spoilage microbiome of commercially manufactured marinated rainbow trout (Oncorhynchus mykiss), two strains, Mkasi1-1T and Mkahm2-6, that could not be identified as any known species of the family Lactobacillaceae by MALDI Biotyper MSP Identification Standard Method 1.1 analysis were isolated. In phylogenetic analyses, the two strains were positioned closest to Latilactobacillus (L.) fuchuensis. Genetically, Mkasi1-1T and Mkahm2-6 were highly similar to each other, having 100% average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) identities. The ANI and

Endocrine fibroblast growth factors (FGF19 subfamily) play a key role in regulating metabolic homeostasis in vertebrates. However, their functional diversification in salmonids remains poorly understood. In this study, we conducted an integrative characterisation of Fgf19 and Fgf21 signalling in rainbow trout (Oncorhynchus mykiss) by combining phylogenetic, structural and expression analyses. Phylogenetic analyses revealed the conservation of single fgf19 and fgf21 genes, despite the extensive expansion of receptors post-Ss4R (salmonid-specific fourth-round whole genome duplication). Structura

Intraspecific diversity enables populations to persist under stochastic and extreme environmental conditions. One system that is representative of notable intraspecific diversity is Chinook salmon (Oncorhynchus tshawytscha) in the Central Valley of California, USA. It is the only place in the species range where four distinct migration timings (Winter, Spring, Fall, Late-fall) co-occur. These populations are declining, with Winter run listed as Endangered and Spring run as Threatened under the Endangered Species Act (ESA). To quantify temporal changes in genetic diversity of the different runs

The potential of N-terminal mitochondrial-targeting sequences (MTSs) as potent antimicrobial peptides (AMPs) has been previously reported. Building on this, 3923 mitochondrial proteins were identified from various marine organisms, among which 470 MTSs were predicted using MitoFates. Of these, 25 MTSs were synthesized and assessed for antimicrobial activity. All MTSs exhibited antifungal activity against Candida albicans, while 22 and 20 MTSs demonstrated activity against Escherichia coli and Staphylococcus aureus, respectively. Notably, the MTS of methylcrotonyl-CoA carboxylase subunit 1 (MCC

Bony fish represent the most basal vertebrate branch with a dedicated mucosal immune system, which comprises immunologically heterogeneous microenvironments armed with innate and adaptive components. In rainbow trout (Oncorhynchus mykiss), a nasopharynx-associated lymphoid tissue (NALT) was recently described as a diffuse network of myeloid and lymphoid cells located in the olfactory organ of fish. Several studies have demonstrated high levels of protection conferred by nasal vaccines against viral and bacterial pathogens; however, the mechanisms underlying the observed protection are not well

This is the first study to isolate, identify and characterize Streptococcus iniae as the causative disease agent in two tilapia (Oreochromis aureus) populations. The populations were geographically isolated, of distinct origins, and did not share water sources. Affected fish showed various external (e.g., exophthalmia and cachexia, among others) and internal (e.g., granulomatous septicaemia and interstitial nephritis, among others) signs. All internal organ samples produced pure cultures, two of which (one from each farm, termed S-1 and S-2) were subjected to biochemical, PCR and 16S rRNA sequ

The present study aimed at investigating cytotoxicity and oxidative stress induced by silica-coated iron oxide nanoparticles functionalized with dithiocarbamate (Fe3O4 NPs) in Chinook salmon cells (CHSE-214) derived from Oncorhynchus tshawytscha embryos. A significant reduction in cell viability was evident in response to Fe3O4 NPs as revealed by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay after 24 h of exposure. Out of the tested concentrations (10, 20, and 30 μg/ml), the highest concentration has shown significant decrease in the viability of cells after 24

The freshwater trematode Nanophyetus salmincola has been demonstrated to impair salmonid immune function and resistance to the marine pathogen Vibrio anguillarum, potentially resulting in ocean mortality. We examined whether infection by the parasite N. salmincola similarly increases mortality of juvenile Chinook Salmon Oncorhynchus tshawytscha when they are exposed to the freshwater pathogens Flavobacterium columnare or Aeromonas salmonicida, two bacteria that juvenile salmonids might encounter during their migration to the marine environment. We used a two-part experimental design where juve

Three distinct microsporidia were identified from parasitic copepods in the northeast Pacific Ocean. Sequencing and phylogenetic analysis of a partial small subunit ribosomal RNA gene (SSU rDNA) sequence identified a genetically distinct variety of Desmozoon lepeophtherii from Lepeophtheirus salmonis on cultured Atlantic salmon Salmo salar, and this was confirmed by transmission electron microscopy. Phylogenetic analysis resolved the SSU rDNA sequence of the second organism in a unique lineage that was most similar to microsporidia from marine and brackish water crustaceans. The second occurre

The potential for xenobiotic compounds to bioconcentrate is typically expressed through the bioconcentration factor (BCF), which has gained increased regulatory significance over the past decade. Due to the expense of in vivo bioconcentration studies and the growing regulatory need to assess bioconcentration potential, BCF is often calculated via single-compartment models, using K(OW) as the primary input. Recent efforts to refine BCF models have focused on physiological factors, including the ability of the organism to eliminate the compound through metabolic transformation. This study looks

Several studies have demonstrated the toxicity of rubber leachate, mainly from rubber tires, to aquatic organisms. In the present study rainbow trout (Oncorhynchus mykiss) were exposed to water provided to aquaria through a rubber hose. Increased hepatic ethoxyresorufin-O-deethylase (EROD) activity, and glutathione reductase (GR) activity were observed in the exposed fish. Two common rubber additives, 2-mercaptobenzothiazole (MBT) and diphenylamine (DPA) and structurally related compounds, were identified by chemical analyses of water samples as were hydroxylated polycyclic aromatic hydrocarbo

Miniaturized bioassays offer many advantages in exploring the toxic potential of chemicals, including small sample volumes and compatibility with high-throughput screening. One problem common to miniaturized systems, however, is the loss of test chemicals because of sorption. The idea of the current study was to use the sorption phenomenon in a positive way. It was found that contaminants sorbed to the growth surface in wells of tissue-culture plates or to the surface of selected sorbent bead materials are available to vertebrate cells growing in direct contact with the contaminant-coated surf

A method has been developed suitable for the study of the binding of salmon (Oncorhynchus tsawytscha) gonadotropic hormone (GtH) to brown trout (Salmo trutta) ovarian particulate fractions. Several proteolytic enzyme inhibitors were tested to preserve the binding capacity of particulate fractions (1000, 20,000, and 110,000g) from yolky oocytes. The best preservation was obtained with trypsin inhibitor. The effects on binding of the pH, time, and temperature of the incubation were investigated. Binding increased linearly with amount of particulate fraction, and is saturable. In both the 20,000

This study examined the intestinal effects of polyethylene (PE; size 46.6 ± 11.3 μm) and polystyrene (PS; size 52.5 ± 11.5 μm) microparticles at three different concentrations (0.5%, 2%, and 5%) on juvenile rainbow trout (Oncorhynchus mykiss) using an integrative approach combining transcriptomics, proteomics, and microbiome analysis. After a six-week experiment, molecular analyses identified concentration-dependent transcriptional responses, including downregulation of genes involved in ion exchange (slc9a1b) and appetite regulation (ghrl), alongside upregulation of immune- and stress-related

Rainbow trout (Oncorhynchus mykiss) is one of the most widely farmed salmonid species globally. To investigate the mechanisms regulating muscle fat content and enhancing tolerance to high-lipid diets, transcriptomic changes in muscle tissues (myotomes and myosepta) were analyzed after treatment with an elevated dietary lipid level (30 %). The results revealed that myotomes and myosepta employed distinct strategies to maintain lipid homeostasis under elevated dietary lipids. Myotomes suppressed lipid deposition by upregulating lipoic acid synthesis to enhance lipolysis and reducing lipoprotein

We compared the sensitivity of closely related Pacific salmon and steelhead (Oncorhynchus spp.) to untreated urban stormwater runoff across three storm events. Juvenile coho, sockeye, steelhead, and Chinook were exposed for 24 h to untreated urban runoff and then transferred to clean water for 48 h. As anticipated from previous studies, coho were highly susceptible to runoff toxicity, with cumulative mortality rates ranging from 92%-100% across the three storms. By contrast, juvenile sockeye were unaffected (100% survival), and cumulative mortality rates were intermediate for steelhead (4%-42%

A wide variety of environmental contaminants has been shown to disrupt immune functions of fish and may compromise their defense capability against pathogens. Immunotoxic effects, however, are rarely considered in ecotoxicological testing strategies. The aim of this study was to systematically evaluate the suitability of an in vitro immuno-assay using selected fish immune parameters to screen for chemicals with known immunotoxic potential and to differentiate them from non-immunotoxicants. Non-stimulated and lipopolysaccharide-stimulated head kidney leukocytes of rainbow trout (Oncorhynchus my

The aim of the present study was to characterize two gram-negative bacterial strains that were isolated from diseased Atlantic Horse Mackerel Trachurus trachurus in 2017. Based on the results obtained from the biochemical and chemotaxonomic characterization, the isolates were identified as Lacinutrix spp. The highest similarity of the 16S rRNA gene sequences was obtained with the strain L. venerupis CECT 8573T (99.1%), while other species showed similarities of 98% (L. jangbogonensis) and 97% (L. algicola and L. mariniflava). Molecular characterization by repetitive element (REP)-PCR and enter

RATIONALE: Diltiazem, a calcium channel blocker drug, is widespread in the environment because of its incomplete elimination during water treatment. It can cause negative effects on aquatic organisms; thus, a rapid and sensitive liquid chromatography/mass spectrometry (LC/MS) method to detect its presence was developed. Our approach is based on accurate mass measurements using a hybrid quadrupole-orbital trap mass spectrometer that was used to measure diltiazem and its metabolites in fish tissue.
METHODS: Blood plasma, muscle, liver, and kidney tissues of rainbow trout (Oncorhynchus mykiss),
