Pandemic-associated disruption of seasonal mortality patterns in Lusaka, Zambia.

INTRODUCTION: Climate-sensitive mortality in rapidly urbanising tropical Africa is poorly characterised, and how pandemics disrupt established seasonal patterns remains underexplored. We analysed long-term all-cause mortality in Lusaka, Zambia, to quantify pre-COVID-19 seasonality and examine pandemic-associated deviations. METHODS: We conducted an observational, exploratory longitudinal time-series analysis of routinely collected monthly all-cause mortality data from Lusaka, Zambia (January 2013 to December 2023; n=180 276). Seasonal dynamics were assessed using classical decomposition and K

Improving patient and clinician safety during COVID-19 through rapidly adaptive simulation and a randomised controlled trial: a study protocol.

INTRODUCTION: COVID-19 required healthcare systems to iteratively adapt for safe and up-to-date care as knowledge of the disease rapidly evolved. Rates of COVID-19 infections continue to fluctuate and patients without COVID-19 increasingly return to the emergency department (ED) for care. This leads to new challenges and threats to patient and clinician safety as suspected patients with COVID-19 need to be quickly detected and isolated among other patients with non-COVID-19-related illnesses. At the front lines, emergency physicians also face continued personal safety concerns and increased wo

Molecular Mechanisms of Zika Virus Entry into Host Cells.

The World Health Organization classifies mosquito-borne viruses as a major global public health threat with pandemic risk and urges all nations to strengthen pandemic preparedness. As a representative mosquito-borne flavivirus, ZIKV bears prominent pandemic capacity. Its distinctive neurotropic property and capability of vertical transmission create unique pathogenic hazards and transmission risks. Owing to the absence of approved vaccines and targeted antiviral therapeutics, the prevention and management of ZIKV outbreaks remain extremely challenging. Viral entry into host cells marks the ver…

Highly specific SARS-CoV-2 main protease (Mpro) mutations against the clinical antiviral ensitrelvir selected in a safe, VSV-based system.

In the SARS-CoV-2 pandemic, the so far two most effective approved antivirals are the protease inhibitors nirmatrelvir, in combination with ritonavir (Paxlovid) and ensitrelvir (Xocova). However, antivirals and indeed all antimicrobial drugs are sooner or later challenged by resistance mutations. Studying such mutations is essential for treatment decisions and pandemic preparedness. At the same time, generating resistant viruses to assess mutants is controversial, especially with pathogens of pandemic potential like SARS-CoV-2. To circumvent gain-of-function research with non-attenuated SARS-C…

Informed decisions about public health and social measures.

Evidence, communication, critical thinking and participation are the cornerstones of informed decisions. In this article we discuss each of these in relation to decisions about public health and social measures (PHSM) during the coronavirus disease 2019 (COVID-19) pandemic and implications for future research. Reliable research evidence of the effects of interventions is particularly important for decisions about what to do because it provides the best basis for estimating the wanted and unwanted effects of doing something. There was little reliable research of the effects of PHSM during the p

Perspectives on Assembling Coronavirus Spikes on Fiber Optics to Reveal Broadly Recognizing Antibodies and Generate a Universal Coronavirus Detector.

In time of COVID-19 biological detection technologies are of crucial relevance. We propose here the use of state of the art optical fiber biosensors to address two aspects of the fight against SARS-CoV-2 and other pandemic human coronaviruses (HCoVs). Fiber optic biosensors functionalized with HCoV spikes could be used to discover broadly neutralizing antibodies (bnAbs) effective against known HCoVs (SARS-CoV, MERS-CoV and SARS-CoV-2) and likely future ones. In turn, identified bnAbs, once immobilized onto fiber optic biosensors, should be capable to detect HCoVs as diagnostic and environmenta
