CARM1-mediated hypoxanthine-enriched exosomes rewire inosine metabolism and impair CD8+ T cell antitumor function.

Cancer cells utilize tumor-derived exosomes to suppress antitumor immunity. Herein, we identify co-activator-associated arginine methyltransferase 1 (CARM1) as a key regulator of exosome biogenesis and metabolite sorting that inhibiting CD8+ T cell-mediated antitumor responses. Genetic ablation of CARM1 in breast cancer cells impairs immunosuppressive exosome secretion, enhancing CD8+ T cell infiltration, proliferation, and effector function. Mechanistically, CARM1 dimethylates apoptosis-linked gene-2 interacting protein X (ALIX) at arginine 757, facilitating its interaction with endosomal sor

Host analysis-guided selection and targeted engineering (HASTE) of Lipomyces tetrasporus for the conversion of CO2-derived feedstocks.

Cost-competitive bioproduction calls for utilizing cellulosic sugars or acetate from electro-reduction of CO2. Here, we identified a novel Lipomyces tetrasporus strain (i.e., NRRL Y-11562 from 24 Lipomyces species) capable of effectively assimilating diverse substrates (acetate, glucose, and xylose). We characterized this new chassis by integrative 13C-metabolic flux analysis, metabolomic analysis, and RNA sequencing with large language model-based data processing. During acetate metabolism, L. tetrasporus demonstrated a highly active TCA cycle, rapid citrate and malate accumulation, strong NA

Hydrogen sulfide orchestrates compartmentalized carbon flux reprogramming to sustain NADPH-dependent redox homeostasis under salt stress in rice.

Hydrogen sulfide (H2S) is recognized as an important redox signaling molecule in plant stress responses, yet how it coordinates metabolic regulation across subcellular compartments remains poorly understood. Here, using rice as a model system, we investigated whether H2S functions as a spatial metabolic signal that integrates carbon flux reprogramming with NADPH-dependent redox homeostasis under salt stress. By combining subcellular H2S quantification, persulfidation profiling, enzyme activity assays, and targeted metabolite analyses, we identified fructose-1,6-bisphosphate aldolase (FBA) as a

Nickel from olivine dissolution induces diel photodamage and metabolic adjustment in Gracilariopsis lemaneiformis.

Nickel (Ni) released from olivine weathering during ocean alkalinity enhancement (OAE) may threaten the carbon sequestration capacity of nearshore primary producers. Here, the holobiont of Gracilariopsis lemaneiformis was exposed to a Ni gradient (20-200 μg/L) for 15 days to simulate near-field exposure, combined with 16S rRNA sequencing and targeted metabolomics, to investigate the effects of Ni stress on algal-bacterial carbon metabolism and its light-dark dependent responses.Ni exposure disrupted carbon fixation through altered energy acquisition, metabolic allocation, and growth accumulati

Comprehensive Lipidomic Profiling Reveals Distinct Metabolic Remodeling during Differentiation of Human Monocyte-Derived Macrophages.

Macrophages are central players of innate immunity with diverse functions and involvement in numerous diseases, making it essential to understand their metabolic regulation. Here, we provide a comprehensive analysis of lipid metabolic dynamics during human monocyte-to-macrophage differentiation. Primary blood monocytes were differentiated for 5 days with either M-CSF or GM-CSF. Lipidomic profiling using the differential mobility spectrometry-shotgun lipidomics assistant (DMS-SLA) platform reliably quantified ∼400 lipids across 16 classes, while spectral flow cytometry was used to assess metabo

Glucose metabolic reprogramming in systemic lupus erythematosus and lupus nephritis: theoretical foundations and therapeutic implications.

Lupus nephritis (LN) represents the most severe and frequent complication of systemic lupus erythematosus (SLE), yet its treatment remains a significant unmet clinical need. Recent advances in immunometabolism have revealed that glucose metabolic reprogramming-including shifts in glycolysis, the pentose phosphate pathway (PPP), and the tricarboxylic acid (TCA) cycle-plays a central role in driving pathogenic immune cell activation in SLE. However, a critical gap persists in understanding how these metabolic alterations specifically operate within the renal microenvironment to promote immune ce

Glucose selectively drives a rapid oxidative burst and immunometabolic reprogramming in human neutrophils during Mycobacterium tuberculosis infection.

Neutrophil functions have been linked to tuberculosis (TB)-associated tissue damage; however, the mechanisms driving immunopathology in the human TB lung remain poorly understood, due partly to the scarcity of human tissue for study. Here, we examine the metabolic and bioenergetic reprogramming of human neutrophils in response to Mycobacterium tuberculosis ( Mtb ) infection. In human necrotic TB granulomas, levels of NETosis-associated proteins are increased and co-localize with GLUT3, linking nutrient uptake to tissue damage. In vitro , Mtb elicits an immediate, contact-dependent oxidative bu

Beyond biochemical cascades: the biophysical execution of disulfidptosis via actin network collapse.

The discovery of disulfidptosis has identified a distinct form of regulated cell death in which metabolic redox failure is translated into biophysical disruption of the actin cytoskeleton. This review synthesizes current evidence on the molecular machinery of actin remodeling under disulfide stress, with particular emphasis on the Rac1-WRC-Arp2/3 signaling axis and other actin-regulatory nodes. Mechanistically, disulfidptosis can be conceptualized as a redox-to-mechanics transition. In SLC7A11-high cells, glucose deprivation limits pentose phosphate pathway-derived NADPH production and weakens

Hydrogen sulfide-driven Janus nanoplatforms for comprehensive disruption of intracellular glucose metabolism and activation of disulfidptosis.

Glucose metabolism (GM) supplies energy and maintains redox homeostasis to support tumor proliferation, while the compensatory crosstalk among multiple GM pathways enables tumor cells to adapt to metabolic perturbations and sustain growth. Therefore, comprehensively blocking GM may dismantle this compensatory network and improve antitumour efficacy. Here, Janus-structured dendritic silica containing disulfide bonds and ZIF-90 loaded with quercetin (Qu) were synthesized to form DZQPP nanoparticles for systematic GM disruption. After reaching the tumor site, cleavage of the tetrasulfide bridges

Sheep-derived probiotics alleviate weaning-induced oxidative stress in lambs via the gut-liver axis.

Weaning is a critical transition in lamb production that can disrupt intestinal homeostasis, promote oxidative stress, and contribute to liver injury. This study investigated whether sheep-derived probiotics could mitigate weaning-associated hepatic injury through the gut-liver axis. Microbiota profiling of six intestinal segments from healthy Hu sheep led to the isolation of nine lactic acid bacterial strains. After in vitro screening for antimicrobial and antioxidant activities, Lactiplantibacillus plantarum M1 and Limosilactobacillus reuteri K4 were selected and administered daily to early-

PFAS penetrate the bone marrow microenvironment and alter immunometabolic homeostasis.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants associated with cancer risk, but their relevance to multiple myeloma remains poorly understood. In an unmatched case-control study of 173 participants, we integrated PFAS biomonitoring, immunoglobulin profiling, and untargeted metabolomics to characterize PFAS-associated immune and metabolic alterations. Higher circulating concentrations of perfluorodecanoic acid and perfluoroheptane sulfonate were associated with increased odds of multiple myeloma and lower polyclonal immunoglobulin G (IgG). Elucidation of Sulforaphane-Mediated Effects on the Cellular Human Metabolome Using Metabolic Profiling.

Sulforaphane (SFN) is an isothiocyanate derived from glucoraphanin, which occurs in broccoli. Several human health-promoting effects are attributed to the consumption of cruciferous vegetables or food supplements containing SFN. Its described cancer-preventive, chemoprotective, and antioxidant properties made SFN an increasingly important research topic. The antioxidant properties have previously been connected to stimulation of the Nuclear factor erythroid-2-related factor 2 (Nrf2)/Kelch-like ECH-associated protein 1 (Keap1) signaling pathway. However, the global effects of SFN on the primary

Safety, Tolerability, Pharmacokinetics, Food Effect of Ribitol, and Its Effect on QTcF in Healthy Adults: First-in-Human, Randomized, Double-Blind (Sponsor Unblinded), Placebo-Controlled Studies.

Limb-girdle muscle dystrophy Type R9 (LGMDR9), also known as LGMD Type 2I, is a rare genetic disease caused by partial loss of function of fukutin-related protein (FKRP) enzyme which glycosylates alpha-dystroglycan, thereby stabilizing myocytes during contraction. Hypoglycosylation leads to progressive muscle injury and impaired function including loss of ambulation. Ribitol is an endogenous pentose alcohol and precursor to CDP-ribitol, the substrate of FKRP. This first-in-human study demonstrated that ribitol was well tolerated when administered as single or multiple oral doses over 6 days to

Disulfidptosis in heart failure: an emerging mechanism awaiting exploration.

Heart failure (HF) is a major cardiovascular syndrome with increasing global incidence and mortality rates. Cardiomyocyte injury and ventricular remodeling are central pathological processes driving HF progression. Although several forms of regulated cell death have been implicated in HF, the mechanisms underlying cardiomyocyte injury remain poorly understood. Disulfidptosis, proposed in 2023, is a metabolism-related form of regulated cell death triggered by the depletion of the cellular reducing capacity. Evidence from tumor cell models indicates that under glucose-restricted conditions, cell

Biallelic Variants in KMO Cause a Novel Form of Congenital NAD Deficiency.

Congenital NAD deficiency disorder (CNDD) is a gene × environment disorder caused by disruptions of the kynurenine pathway. To date, CNDD has been associated with biallelic variants in three kynurenine pathway genes: KYNU, HAAO, and NADSYN1 . We identified two sisters with congenital anomalies overlapping with CNDD who have biallelic variants in a gene encoding a different kynurenine pathway enzyme, KMO . The surviving child also has elevated levels of metabolites upstream of KMO with low NAD + levels in plasma, suggesting that KMO deficiency is a novel CNDD. To explore the pathogenicity of KM

Biochemical deciphering of an Atypical methemoglobinemia.

BACKGROUND: Methemoglobin is formed when the iron in hemoglobin is oxidized from the ferrous to the ferric state, preventing oxygen transport. This potentially life-threatening condition requires prompt diagnosis and treatment. METHOD: We report the case of a 30-year-old Senegalese man with newly diagnosed acute leukemia, admitted to the emergency department for suspected leukostasis syndrome. Induction chemotherapy and rasburicase were initiated. Unexpectedly, 24-h later, he developed acute hemolysis and a methemoglobinemia of 17%. RESULTS: Investigation suggested rasburicase-induced methem

Metabolic adaptation of Yersinia pestis during flea colonization.

Yersinia pestis, the causative agent of plague, has evolved a dual lifestyle enabling survival in both mammalian hosts and hematophagous flea vectors. While mammalian virulence has been extensively studied, the flea stage represents a highly selective and metabolically constraining environment that plays a decisive role in transmission. Following ingestion during a blood meal, Y. pestis establishes within the flea digestive tract, where it must withstand nutrient limitation, oxidative stress, osmotic pressure, and antimicrobial factors while forming a proventricular biofilm essential for gut b

Transcriptomic signatures predict severe linezolid-induced peripheral neuropathy and reveal pre-treatment immune and antioxidant dysregulation.

Linezolid is an essential drug for treating multidrug-resistant and extensively drug-resistant tuberculosis, yet its long-term use is frequently limited by peripheral neuropathy, which in severe cases may be irreversible. To investigate biological factors underlying susceptibility, we analyzed transcriptomic profiles of peripheral blood mononuclear cells from 51 MDR/XDR-TB patients before and after six months of linezolid treatment, stratifying individuals into asymptomatic, mild, and severe neuropathy groups. Patients who later developed severe neuropathy already exhibited distinct transcript

6-aminonicotinamide dismantles proliferative and migratory capacities of oral cancer cells via PPP blockade.

OBJECTIVE: Cancer cells frequently exploit the Pentose Phosphate Pathway (PPP) to fuel antioxidant defenses and the synthesis of essential biomolecular precursors. It is hypothesized that sustained upregulation of glucose flux through the PPP is crucial for meeting the anabolic demands of cancer cells and mitigating oxidative stress. We investigated whether pharmacological inhibition of the rate-limiting PPP enzyme, glucose 6-phosphate dehydrogenase (G6PD), could decrease proliferation, disrupt cell-cell interactions, and impede migration in oral squamous cell carcinoma (OSCC). DESIGN: Non-tu
