Antimicrobial peptide exposure selects for Staphylococcus aureus resistance to human defence peptides.

BACKGROUND: The clinical development of antimicrobial peptides (AMPs) is currently under evaluation to combat the rapid increase in MDR bacterial pathogens. However, many AMPs closely resemble components of the human innate immune system and the ramifications of prolonged bacterial exposure to AMPs are not fully understood. OBJECTIVES: We show that in vitro serial passage of a clinical USA300 MRSA strain in a host-mimicking environment containing host-derived AMPs results in the selection of stable AMP resistance. METHODS: Serial passage experiments were conducted using steadily increasing c…

(R)-α-trifluoromethylalanine containing short peptide in the inhibition of amyloid peptide fibrillation.

The extracellular deposition of insoluble amyloid fibrils resulting from the aggregation of the amyloid-β (Aβ) is a pathological feature of neuronal loss in Alzheimer's disease (AD). Numerous small molecules have been reported to interfere with the process of Aβ aggregation. Compounds containing aromatic structures, hydrophobic amino acids and/or the α-aminoisobutyric acid (Aib) as β-sheet breaker elements have been reported to be effective inhibitors of Aβ aggregation. We synthesized two peptides, one containing the Aib amino acid and the other including its trifluoromethylated analog (R)-α-T…

Macrolide-peptide conjugates as probes of the path of travel of the nascent peptides through the ribosome.

Despite decades of research on the bacterial ribosome, the ribosomal exit tunnel is still poorly understood. Although it has been suggested … tunnel has a relaxed preference for the directionality (N → C or C → N orientation) of the nascent peptides. Moreover, the X-ray crystal structure of one peptolide derived from a positively charged, reverse Nuclear Localization Sequence peptide, bound to the 70S bacterial ribosome, reveals that the macrolide ring of the peptolide binds in the same positio …

Short peptide amyloid organization: stabilities and conformations of the islet amyloid peptide NFGAIL.

Experimentally, short peptides have been shown to form amyloids similar to those of their parent proteins. Consequently, they present useful systems for studies of amyloid conformation. Here we simulate extensively the NFGAIL peptide, derived from the human islet amyloid polypeptide (residues 22-27). We simulate different possible strand/sheet organizations, from dimers to nonamers. Our simulations indicate that the most stable conformation is an antiparallel strand orientation within the sheets and parallel between sheets. Consistent with the alanine mutagenesis, we find that the driving forc…

Calreticulin synthetic peptide analogues: anti-peptide antibodies in autoimmune rheumatic diseases.

Autoantibodies in sera from patients with systemic lupus erythematosus (SLE) and onchocerciasis recognize calreticulin (CaR), a calcium-binding protein, as antigen. In this study we present the immunological properties of two synthetic peptides prepared to correspond to the 1-24 and 7-24 amino acid sequence of CaR. In contrast to information previously reported for the recombinant protein, the CaR-peptide analogues appeared immunoreactive to anti-Ro/SSA autoimmune sera. Human sera from patients with SLE, Sjögren's syndrome (SS), rheumatoid arthritis (RA), as well as mixed connective tissue dis…

Dual-Peptide Nanoplatform: Mesoporous Silica Nanoparticles Functionalized With a Cell-Penetrating Peptide and Loaded With Rationally Designed Antimicrobial Peptides for Tuberculosis Therapy.

Tuberculosis remains a major infectious disease worldwide and demands new therapeutic approaches capable of targeting intracellular Mycobacterium tuberculosis. Using a machine learning (ML)-guided design strategy based on the plectasin scaffold, a peptide library was generated and DC05 was identified as the most active analog. As a free peptide, DC05 inhibited the H37Rv strain with an MIC90 of 12.46 ± 0.06 µm, displayed low cytotoxicity in MRC-5 fibroblasts (IC50 >250 µm) and J774A.1 macrophages (115.18 ± 8.49 µm), and increased ethidium bromide accumulation to levels comparable to verapamil,

AI-Driven Design and Clinical Translation of Nucleotide-Peptide and Peptide-Drug Conjugates.

While nucleotide-peptide conjugates and peptide-drug conjugates demonstrate a considerable evolution as next-generation therapeutics, the successful translation of these complex agents relies on advances in bioconjugation strategies. By anchoring nucleotides or pharmaceutical payloads to multifaceted peptide scaffolds, researchers have engineered molecular platforms with dual mechanisms of action. However, to fully realize the potential of these hybrid systems, further refinements are required to ensure both in vivo stability and retention of the desired biological activities over time. Optimizing delivery of an anti-cytomegalovirus inhibitory peptide using a cell-penetrating peptide.

Cytomegalovirus (CMV) is a beta herpesvirus that persists quiescently in healthy individuals but causes severe disease in immunocompromised patients and congenitally infected newborns. Side effects posed by the currently available antivirals necessitate the development of new antivirals with improved safety profiles. This study aims to optimize the potency of an anti-CMV peptide (P10) mimicking conserved region 2 of tegument pp150, fused with an elastin-like polypeptide (ELP). Our previous study has demonstrated that ELP-P10 inhibits murine cytomegalovirus (MCMV) growth in vitro and in vivo wi

Regulation of response to antigen peptides is independent of peptide distribution in lymph node therapeutics.

Autoimmune disease occurs when immune cells mistakenly identify specific molecules, termed antigens, on healthy cells. There are no cures fo … ese depots to encapsulate peptide antigens in distinct loading configuration - either with multiple peptides in a single particle or a single peptide in distinct particles. We show that both formulations are taken up by cells, and drive equivalent T cell responses both in vitro and in vivo. Notably, when loaded with an immunomodulatory cue, both formulatio …

Peptide Toxin Diversity and a Novel Antimicrobial Peptide from the Spider Oxyopes forcipiformis.

Spider venoms are emerging as a rich source of bioactive peptide toxins with therapeutic potential. Lynx spiders of the genus Oxyopes are small, cursorial hunters that employ complex venom to subdue arthropod prey. However, extracting crude venom from these diminutive arachnids poses significant challenges. This study presents a transcriptome analysis of venom glands from an undescribed Oxyopes forcipiformis species, revealing 339 putative protein and peptide toxin sequences categorized into seven functional groups. The venom composition was dominated by membrane-active peptides (40.71%), veno…

Development of a Therapeutic Peptide for Cachexia Suggests a Platform Approach for Drug-like Peptides.

During the development of a melanocortin (MC) peptide drug to treat the condition of cachexia (a hypermetabolic state producing lean body mass wasting), we were confronted with the need for peptide transport across the blood-brain barrier (BBB): the MC-4 receptors (MC4Rs) for metabolic rate control are located in the hypothalamus, i.e., behind the BBB. Using the term "peptides with BBB transport", we screened the medical literature like a peptide library. This revealed numerous "hits"-peptides with BBB transport and/or oral activity. We noted several features common to most peptides in this cl…

A review on anti-tuberculosis peptides: Impact of peptide structure on anti-tuberculosis activity.

Antibiotic resistance is a major public health problem globally. Particularly concerning amongst drug-resistant human pathogens is Mycobacterium tuberculosis that causes the deadly infectious tuberculosis (TB) disease. Significant issues associated with current treatment options for drug-resistant TB and the high rate of mortality from the disease makes the development of novel treatment options against this pathogen an urgent need. Antimicrobial peptides are part of innate immunity in all forms of life and could provide a potential solution against drug-resistant TB. This review is a critical…

Divergent Peptide Presentations of HLA-A*30 Alleles Revealed by Structures With Pathogen Peptides.

Human leukocyte antigen (HLA) alleles have a high degree of polymorphism, which determines their peptide-binding motifs and subsequent T-cell receptor recognition. The simplest way to understand the cross-presentation of peptides by different alleles is to classify these alleles into supertypes. A1 and A3 HLA supertypes are widely distributed in humans. However, direct structural and functional evidence for peptide presentation features of key alleles (e.g., HLA-A*30:01 and -A*30:03) are lacking. Herein, the molecular basis of peptide presentation of HLA-A*30:01 and -A*30:03 was demonstrated b…

PEG-PGA enveloped octaarginine-peptide nanocomplexes: An oral peptide delivery strategy.

The objective of this work was the development of a new drug nanocarrier intended to overcome the barriers associated to the oral modality of administration and to assess its value for the systemic or local delivery of peptides. The nanocarrier was rationally designed taking into account the nature of the intestinal barriers and was loaded with insulin, which was selected as a model peptide. The nanocarrier consisted of a complex between insulin and a hydrophobically-modified cell penetrating peptide (CPP), enveloped by a protecting polymer. The selected CPP was octaarginine (r8), chemically c…

From a pro-apoptotic peptide to a lytic peptide: One single residue mutation.

Further discovery and design of new anticancer peptides are important for the development of anticancer therapeutics, and study on the detailed acting mechanism and structure-function relationship of peptides is critical for anticancer peptide design and application. In this study, a novel anticancer peptide ZXR-1 (FKIGGFIKKLWRSKLA) derived from a known anticancer peptide mauriporin was developed, and a mutant ZXR-2 (FKIGGFIKKLWRSLLA) with only one residue difference at the 14th position (Lys→Leu) was also engineered. Replacement of the lysine with leucine made ZXR-2 more potent than ZXR-1 in …
